Abstract
The invention relates to the technical field of skin external preparations, in particular to a composition, a preparation method thereof and an anti-aging cosmetic. The composition comprises the following components in percentage by mass; 25 to 35 percent of component A and 65 to 75 percent of component B; the component A comprises herba Bidentis Bipinnatae plant extract, and the component B comprises Concha Margaritifera fermented product. The composition has excellent fibroblast proliferation promoting activity and excellent anti-primary aging effect through the coordination and coordination of the components. The anti-aging cosmetic prepared from the composition has good anti-aging effect, and can remarkably reduce skin wrinkles, increase the moisture content of skin horny layer, reduce skin roughness, smoothen skin, increase skin elasticity and reduce transepidermal water loss after being used.

Background
The market scale of the anti-aging products shows a continuous acceleration trend from 2016, and according to statistics, the population proportion of the most active target user group of the anti-aging products between 25 and 49 years is gradually increased. In young consumers of 20-25 years old, some common skin aging manifestations such as dull and dull skin, insufficient skin moisture and the like begin to be concerned. Research shows that the skin gradually enters a natural aging state from the age of 20-25, and the aging speed is different according to different genetic genes and living habits of each person. Human skin is composed of epidermis, basement membrane and dermis, and fibroblasts in the dermis layer have the effects of producing collagen, elastin, glycosaminoglycan and the like, and the components have the effects of maintaining skin elasticity, moisturizing the dermis layer and the like. The activity of fibroblasts plays a crucial role in maintaining the healthy appearance of skin. It has been observed that with age, the proliferation of fibroblasts and the functional activity of synthesizing skin benefit agents gradually decrease. In the past, the fibroblast activity of old skin is improved by increasing the dosage of cell activity stimulator; there are different needs for improvement of the initial aging symptoms of young skin, and the method of increasing the amount of the irritant is not completely suitable for the initial aging skin.
In view of the above, the present invention is particularly proposed.
Disclosure of Invention
The invention aims to provide a composition which has excellent activity of promoting fibroblast proliferation and good anti-primary-aging effect through the coordinated cooperation of the components.
The invention also aims to provide a preparation method of the composition, which is simple to operate and environment-friendly.
It is another object of the present invention to provide an anti-aging cosmetic composition having excellent anti-aging effects, which can significantly reduce skin wrinkles, increase the moisture content of the stratum corneum of the skin, reduce skin roughness, smooth the skin, increase skin elasticity, and reduce transepidermal water loss after use.
In order to achieve the above purpose of the present invention, the following technical solutions are adopted:

Preferably, the component A also comprises 0.1 to 10 percent of water by mass percentage.
Preferably, the bidens plant is selected from at least one of bidens pilosa, bidens salicifolia, bidens tripartita and bidens alba, and is preferably bidens pilosa.
Preferably, the mother-of-pearl fermentation product is mainly prepared by fermenting strains and mother-of-pearl, wherein the strains are selected from at least one of yeast, aureobasidium pullulans and lactobacillus;
preferably, in the mother-of-pearl fermentation product, the content of polysaccharide is 30-300 mg/L, and the content of calcium ions is 10-200 mg/L;
preferably, the mother-of-pearl is selected from at least one of the group consisting of a queen phoenix, a hyriopsis cumingii, a crista plicata and a pearl oyster of mad.

Preferably, the preparation method of the component A comprises the following steps:
adding the lecithin into the mixture of the Bidens plant extract, the caprylic/capric triglyceride, the grass cotton seed oil, the linseed oil and the palm oil, heating, adding the glycerol and the water, homogenizing and cooling.
Preferably, the preparation method of the mother-of-pearl fermentation product comprises the following steps:
inoculating the strain into the mixture of the crushed nacre and the fermentation liquid culture medium, and performing fermentation culture;
preferably, the addition amount of the crushed nacre is 0.01-10% of the mass of the fermentation liquid culture medium;
preferably, the culture temperature is 27.8-28.2 ℃, and the culture time is 4-6 days;
preferably, the culture is shaking culture, and the rotation speed of the culture is 115-125 rpm.

Compared with the prior art, the invention has the beneficial effects that:
(1) the composition has excellent fibroblast proliferation promoting activity and good anti-aging effect through the coordination and coordination of the components.

According to one aspect of the invention, the invention relates to a composition, comprising the following components in percentage by mass;
component A25% -35% and component B65% -75%;
the component A comprises herba Bidentis Bipinnatae plant extract, and the component B comprises Concha Margaritifera fermented product.
The herba Bidentis Bipinnatae plant extract contains flavone and volatile oil, and has effects of clearing away heat and toxic materials, removing blood stasis, promoting blood circulation, whitening skin, and repairing tissue.
The nacre is the nacre layer of shells of shellfish, and common sources include hyriopsis cumingii, crista plicata, and Pinctada martensii, and the more rare Queen Phoenix shell producing a pearl. The mother-of-pearl comprises the following main components: 1) calcium: the main factors regulating nerves and muscles participate in various metabolisms in the body and are also the components of bones; 2) trace elements: contains higher lithium and magnesium elements; 3) amino acids: contains more than twenty kinds of amino acids, mainly including aspartic acid, threonine, serine, glycine and alanine; 4) a polysaccharide.
The composition has the advantages of promoting the proliferation activity of fibroblast, having good safety and no cytotoxicity by the coordination of the extract of the bidens and the fermentation product of the nacre.
In one embodiment, the component a is 25% to 35%, and may alternatively be 25%, 26%, 27%, 28%, 29%, 30%, 31%, 32%, 33%, 34%, or 35%.
In one embodiment, the component B is 65% to 75%, and may alternatively be 65%, 66%, 67%, 68%, 69%, 70%, 71%, 72%, 73%, 74%, or 75%.
Preferably, the component A comprises the following components in percentage by mass:
10-20% of bidens extract, 10-15% of lecithin, 40-60% of glycerol, 10-15% of caprylic/capric triglyceride, 1-10% of palm oil, 1-10% of grass cotton seed oil and 1-5% of linseed oil.
The components are coordinated and matched, so that the composition has a better effect of promoting fibroblast proliferation and has an excellent anti-aging effect.
In one embodiment, in the component a, the content of the bidens extract is 10% to 20% by mass, and may be 10%, 11%, 12%, 13%, 14%, 15%, 16%, 17%, 18%, 19% or 20% by mass.
In one embodiment, the content of lecithin in the component a is 10% to 15% by mass, and may be 10%, 11%, 12%, 13%, 14% or 15% by mass.
In one embodiment, the content of glycerin in the component a is 40% to 60% by mass, and may also be selected from 40%, 41%, 42%, 43%, 44%, 45%, 46%, 47%, 48%, 49%, 50%, 51%, 52%, 53%, 54%, 55%, 56%, 57%, 58%, 59% or 60%.
In one embodiment, the content of the caprylic/capric triglyceride in the component a is 10% to 15% by mass, and may be 10%, 11%, 12%, 13%, 14% or 15% by mass.

In one embodiment, in the component a, the content of the gossypium herbaceum seed oil is 1% to 10% by mass, and may be 1%, 2%, 3%, 4%, 5%, 6%, 7%, 8%, 9% or 10% by mass.
In one embodiment, in the component a, the linseed oil accounts for 1% to 5% by mass, and may also be selected from 1%, 2%, 3%, 4% or 5%.
Preferably, the component A also comprises 0.1 to 10 percent of water by mass percentage.

Preferably, the bidens plant is selected from at least one of bidens pilosa, bidens salicifolia, bidens tripartita and bidens alba, and is preferably bidens pilosa.
The invention adds proper amount of bidens pilosa extract, and has at least the following excellent effects: clearing away heat and toxic material, removing blood stasis, promoting blood circulation, whitening skin, and repairing tissue.
The bidens tripartita contains components such as flavonoid, tannin, polysaccharide, amino acid, ascorbic acid, organic acid and the like, and has remarkable anti-inflammatory, analgesic and antipyretic activities.
Preferably, the mother-of-pearl fermentation product is mainly prepared by fermenting a strain and mother-of-pearl, wherein the strain is at least one of yeast, aureobasidium pullulans and lactobacillus.
The invention selects at least one of yeast, aureobasidium pullulans and lactobacillus as a fermentation strain, and the fermentation strain is mixed with the mother-of-pearl for fermentation, so that the obtained mother-of-pearl fermentation product can further improve the activity of the effective components, and can further promote the skin fibroblast proliferation and the anti-primary aging activity by matching with the bidens plant extract.
Preferably, in the mother-of-pearl fermentation product, the content of polysaccharide is 30-300 mg/L, and the content of calcium ions is 10-200 mg/L;

The component A prepared by the method is uniformly mixed, and can be better coordinated with the mother-of-pearl fermentation product to play the role of primary aging resistance.
Preferably, the preparation method of the mother-of-pearl fermentation product comprises the following steps:
inoculating the strain into the mixture of the crushed nacre and the fermentation liquid culture medium, and performing fermentation culture.
Preferably, the addition amount of the crushed nacre is 0.01-10% of the mass of the fermentation liquid culture medium;
preferably, the temperature of the culture is 27.8-28.2 ℃, and the time of the culture is 4-6 days.
Preferably, the culture is shaking culture, and the rotation speed of the culture is 115-125 rpm.
In one embodiment, the temperature of the culture is 27.8-28.2 ℃, and can be 27.8 ℃, 27.9 ℃, 28 ℃, 28.1 ℃ or 28.2 ℃.
In one embodiment, the culturing time is 4-6 days, and further 4 days, 5 days or 6 days can be selected.

According to another aspect of the invention, the invention also relates to an anti-aging cosmetic comprising a composition as described above.

Example 1
The composition consists of the following components in percentage by mass;
30% of component A and 70% of component B;
the component A comprises the following components in percentage by mass: bidens pilosa extract 15%, lecithin 12%, glycerin 50%, caprylic/capric triglyceride 13%, palm oil 5%, gossypium herbaceum seed oil 3%, linseed oil 2 and water 5%;
the component B is mother-of-pearl fermentation product;
the preparation method of the composition comprises the following steps:
(a) preparation of component a (bidens pilosa liposomes): stirring herba Bidentis Tripartitae extract, caprylic/capric triglyceride, palm oil, semen gossypii seed oil and oleum Lini at room temperature to disperse uniformly, adding lecithin dissolved by heating and stirring, mixing uniformly, heating, adding glycerol and water, homogenizing, and cooling to room temperature to obtain yellowish viscous liquid herba Bidentis Tripartitae liposome;

the component A comprises the following components in percentage by mass: 15% of bidens tripartita extract, 12% of lecithin, 50% of glycerol, 13% of caprylic/capric triglyceride, 5% of palm oil, 3% of grass cotton seed oil, 2% of linseed oil and 5% of water;
the component B is mother-of-pearl fermentation product;
the preparation method of the composition comprises the following steps:
(a) preparation of component a (tripartite liposome): stirring the bidens tripartita extract, caprylic/capric triglyceride, palm oil, grass cotton seed oil and linseed oil at room temperature until the mixture is uniformly dispersed, adding lecithin which is heated and stirred to be dissolved, uniformly mixing, heating, adding glycerol and water, homogenizing, and cooling to room temperature to obtain a yellow viscous liquid bidens tripartita liposome;
(b) preparing mother-of-pearl fermentation: removing horny layer of Cristaria plicata and Pinctada martensii (mass ratio of 1:1), retaining pearl layer, crushing to obtain micron-sized powder, adding into fermentation broth culture medium according to 0.2%, sterilizing, inoculating lactobacillus, placing on a constant temperature shaking table, fermenting and culturing at 28 deg.C and 125rpm for 4 days, after fermentation, dissolving bacteria, decolorizing and filtering to obtain mother-of-pearl fermentation product, wherein polysaccharide content is 50mg/L, and calcium ion content is 20 mg/L;
(c) sequentially adding the component A obtained in the step (a) and the mother-of-pearl fermentation product obtained in the step (b) into a stirring kettle, stirring at a low speed until the components are uniformly mixed, and cooling to room temperature.
Example 5
The anti-aging essence is mainly prepared from the following components in percentage by mass:
0.6% of sodium polyacryloyldimethyl taurate, 2% of diglycerol, 4% of glycerol, 2% of betaine, 0.8% of hydroxyethyl piperazine ethanesulfonic acid, 0.1% of acetylated sodium hyaluronate, 0.01% of sodium hyaluronate, 0.1% of inulin lauryl carbamate, 0.2% of octyldodecanol, 1% of polydimethylsiloxane, 0.5% of the composition, 0.25% of p-hydroxyacetophenone, 0.15% of phenoxyethanol, 0.02% of essence, and the balance of water.
Examples of the experiments
Fibroblast activity promoting effect
3T3 cells in logarithmic growth phase were seeded in 96-well plates and placed in CO2After 3 days of incubator culture, cells were treated with a series of different samples of different concentrations, solvent blank and positive control were set, CO was added2After the incubator is incubated for 24 hours, 0.5mg/mL MTT is added for continuous culture for 4 hours, 100 mu L DMSO is added, and an enzyme-labeling instrument is used for detecting OD550 and carrying out statistical analysis; the results are shown in Table 1.

As shown in Table 1, in comparison with NT group, Bidens pilosa liposome has no cytotoxicity in the concentration range of 0.05% -0.001%, and has slight but not obvious effect of promoting fibroblast proliferation, and the maximum effect of promoting cell proliferation is 103.7%. Compared with NT group, the fermentation product of nacre has no cytotoxicity in the concentration range of 0.05-0.001%, and has certain effect of promoting the proliferation of fiber cells, the maximum effect of promoting the proliferation of cells is 116.6%. The active composition of the external anti-aging skin preparation prepared in example 3 has no cytotoxicity in the concentration range of 0.05% -0.001%, and has strong fibroblast proliferation promoting effect, and the effect values of promoting cell proliferation under the same concentration are all higher than those of the individual bidens pilosa liposome and the mother-of-pearl fermentation product, which shows that the compound of the bidens pilosa liposome and the mother-of-pearl fermentation product has obvious cell proliferation promoting effect, and the cell proliferation promoting effect is superior to that of the two when acting on cells alone.
Second, evaluation of anti-aging ability
The essence prepared in example 5 was subjected to a test of the aging resistance before and after use.
Volunteers: female aged 27-50 years; the moisture value of the cheek is less than or equal to 60c.u., the canthus wrinkles are more than or equal to grade 1, and the facial skin is rough; actually recruiting 33 people, valid data 33 people;
test area: a face;
the using method comprises the following steps: the volunteers use the liquid once in the morning and at night every day, and 0.8-1 g of 2 dropper essence liquid is uniformly smeared on the whole face until the liquid is absorbed;
the service cycle is as follows: 28 days;
frequency of use: the medicine is used once in the morning and at night;
and (3) testing period: before use, 14 days after use, 28 days after use;
testing parameters: visia CR is used for acquiring facial images; primos is used for facial image acquisition and skin wrinkle analysis; 3. skin moisture testerCM 825 is used for detecting the moisture content of stratum corneum; the Visioscan VC98 is used for carrying out facial image acquisition and skin texture analysis; 5. skin elasticity testerMPA580 is used to detect skin elasticity; 6. skin water loss testerTM300 is used for detecting the transepidermal water loss rate of the skin; 7. volunteers assessed themselves.
Before use: the volunteers clean the face, wait for 30min in a laboratory at the temperature of 21 +/-1 ℃ and the relative humidity of 50 +/-5 percent, collect facial images and test the moisture content of the stratum corneum, the elasticity of the skin and the water loss rate of the skin through the epidermis.
After 14 days of use: cleaning face of volunteer, waiting for 30min in laboratory at 21 + -1 deg.C and 50 + -5% relative humidity, collecting facial image, and testing stratum corneum water content, skin elasticity, and skin transepidermal water loss rate.
After 28 days of use: cleaning the face of a volunteer, waiting for 30min in a laboratory at the temperature of 21 +/-1 ℃ and the relative humidity of 50 +/-5%, carrying out facial image acquisition, testing the moisture content of the stratum corneum, the elasticity of the skin and the water loss rate of the skin through epidermis, and carrying out self-evaluation.
And (3) data statistics: data were counted using SPSS19.0 and compared using paired T-test with significance difference levels all P < 0.05.
Compared with the prior use, the experimental results are as follows;
1) skin wrinkles: after 14 days of use, the Ra value of skin wrinkles in the tested area was significantly reduced by 5.72%, the number of skin wrinkles was significantly reduced by 7.50%, the mean volume value of skin wrinkles was not significantly reduced by 8.28%, the mean area value of skin wrinkles was significantly reduced by 8.33%, and the mean length value of skin wrinkles was significantly reduced by 16.14%; after 28 days of use, the Ra value of skin wrinkles in the tested area was significantly reduced by 8.49%, the number of skin wrinkles was significantly reduced by 10.00%, the mean volume value of skin wrinkles was not significantly reduced by 5.03%, the mean area value of skin wrinkles was significantly reduced by 11.75%, and the mean length value of skin wrinkles was significantly reduced by 22.44%.
2) Moisture content of skin stratum corneum: after 14 days of use, the moisture content value of the stratum corneum of the tested area is remarkably increased by 28.64 percent; after 28 days of use, the moisture content of the stratum corneum in the test area was significantly increased by 34.33%.
3) Skin texture: after 14 days of use, the skin roughness value of the tested area is significantly reduced by 12.42%, and the skin smoothness value is not significantly increased by 0.62%; after 28 days of use, there was a significant reduction in skin roughness by 13.15% in the test area and no significant increase in skin smoothness value by 0.01%.
4) Skin elasticity: after 14 days of use, the R2 value for skin elasticity in the test area increased significantly by 7.98%; after 28 days of use, the R2 value for skin elasticity in the test area increased significantly by 8.19%.
5) Skin transepidermal water loss rate: after 14 days of use, the value of the transepidermal water loss of the skin in the tested area is significantly reduced by 11.27 percent; after 28 days of use, the transepidermal water loss rate of the skin in the test area was significantly reduced by 20.78%.
6) Self-assessment of volunteers: after 28 days of use, 94% of the volunteers were satisfied with the effect of the test sample, 94% of the volunteers were willing to continue using the test sample after the test was finished, 97% of the volunteers were willing to purchase the test sample without considering the price, and 100% of the volunteers considered to recommend the test sample to others.
7) Adverse reactions: during the use of the samples, 0 volunteers had local or systemic adverse skin reactions.
From the above results, it can be seen that the active composition for external anti-aging skin preparations of the present invention has a good anti-aging effect, and can significantly reduce skin wrinkles, increase the moisture content of the stratum corneum of the skin, reduce pachylosis, smooth the skin, increase the elasticity of the skin, reduce transepidermal water loss, and provide volunteers with satisfactory use effects without adverse reactions to the skin.
Finally, it should be noted that: the above embodiments are only used to illustrate the technical solution of the present invention, and not to limit the same; while the invention has been described in detail and with reference to the foregoing embodiments, it will be understood by those skilled in the art that: the technical solutions described in the foregoing embodiments may still be modified, or some or all of the technical features may be equivalently replaced; and the modifications or the substitutions do not make the essence of the corresponding technical solutions depart from the scope of the technical solutions of the embodiments of the present invention.
Claims (13)
1. A cosmetic composition is characterized by comprising the following components in percentage by mass;
25% -35% of the component A and 65% -75% of the component B;
the component A comprises the following components in percentage by mass:
10-20% of bidens plant extract, 10-15% of lecithin, 40-60% of glycerol, 10-15% of caprylic/capric triglyceride, 1-10% of palm oil, 1-10% of grass cotton seed oil and 1-5% of linseed oil;
the component B comprises mother-of-pearl leavening; the mother-of-pearl fermentation product is mainly prepared by fermenting strains and mother-of-pearl;
the preparation method of the mother-of-pearl fermentation product comprises the following steps:
inoculating the strain into the mixture of the crushed nacre and the fermentation liquid culture medium, and performing fermentation culture;
the addition amount of the crushed nacre is 0.01-10% of the mass of the fermentation liquid culture medium;
the culture temperature is 27.8-28.2 ℃, and the culture time is 4-6 days;
the culture is shaking culture, and the rotating speed of the culture is 115-125 rpm.
2. The cosmetic composition according to claim 1, wherein the component A further comprises 0.1 to 10 mass% of water.
3. The cosmetic composition of claim 1, wherein said plant of the genus Bidens is selected from at least one of Bidens pilosa, Bidens salicifolia, Bidens tripartita, and Bidens alba.
4. The cosmetic composition according to claim 1, wherein the bacterial species is at least one selected from the group consisting of yeast, aureobasidium pullulans and lactobacillus.
5. The cosmetic composition according to claim 1, wherein the mother-of-pearl fermentation product contains 30 to 300mg/L of polysaccharide and 10 to 200mg/L of calcium ion.
6. The cosmetic composition of claim 1, wherein said nacre is selected from at least one of the group consisting of queen phoenix, hyriopsis cumingii, crista plicata, and pearl oyster mazel.
7. A process for the preparation of a cosmetic composition according to any one of claims 1 to 6, comprising the steps of:
and uniformly mixing the component A and the component B.
8. The method for preparing a cosmetic composition according to claim 7, wherein the component A is prepared by the method comprising the steps of:
adding lecithin into mixture of Bidens plant extract, caprylic/capric triglyceride, semen gossypii oil, oleum Lini and palm oil, heating, adding glycerol and water, homogenizing, and cooling.
9. An anti-aging cosmetic comprising the cosmetic composition according to any one of claims 1 to 6.
10. The anti-aging cosmetic as claimed in claim 9, wherein the amount of the cosmetic composition is 0.001-10% by mass of the total mass of the anti-aging cosmetic.
11. The anti-aging cosmetic as claimed in claim 10, wherein the amount of the cosmetic composition is 0.01 to 5% by mass of the total mass of the anti-aging cosmetic.
12. The anti-aging cosmetic of claim 9, wherein the anti-aging cosmetic comprises serum.
13. The anti-aging cosmetic according to claim 12, wherein the essence is mainly prepared from the following components in percentage by mass:
0.1-2% of sodium polyacryloyl dimethyl taurate, 0.5-5% of diglycerol, 1-10% of glycerol, 0.5-5% of betaine, 0.1-4% of hydroxyethyl piperazine ethane sulfonic acid, 0.01-1% of acetylated sodium hyaluronate, 0.01-0.5% of inulin lauryl carbamate, 0.1-2% of octyldodecanol, 0.2-10% of polydimethylsiloxane, 0.1-10% of the cosmetic composition, 0.1-0.5% of p-hydroxyacetophenone, 0.1-0.5% of phenoxyethanol, 0.01-0.02% of essence and the balance of water.
