Abstract

本发明提供一种检测人体液中胎盘生长因子含量的试剂盒。本发明将化学发光技术与免疫磁微粒相结合,提供了一种接近均相的反应体系,并且采用了一步法反应模式,使得检测灵敏度、精密性大大提高、检测范围扩大,反应时间大大缩短,从开始加样到检测结果,时间少于20min;并且可以在全自动化学发光仪上同时测定多个样本,实现胎盘生长因子(PIGF)的高通量快速化测定,准确度高,特异性强,精确度和检测效率有了较大的提高。

一种检测人体液中胎盘生长因子含量的试剂盒
技术领域
本发明属于检测试剂盒领域,特别是胎盘生长因子检测试剂盒。
背景技术
胎盘生长因子(PIGF)主要由胎盘合体滋养细胞分泌,促进滋养细胞增殖、活化,PIGF也可在绒毛间质内皮细胞中丰富表达,促进新生血管形成分枝、非分枝血管形成及建立有效的胎儿胎盘血管网,在胎盘形成及胎儿发育过程中发挥重要作用。正常妊娠时,PIGF的合成随孕周增加逐渐增加,这与妊娠过程中胎盘供氧的变化是一致的。妊娠15周之前PIGF水平较低,妊娠15~26周PIGF表达迅速增加,并在妊娠28~30周达高峰,妊娠后期至分娩期随胎盘成熟老化、功能减退,PIGF水平又逐渐下降。
发明内容
本发明目的在于提供一种采用磁微粒化学发光法测定人体液样本(包含但不限于血清/血浆/脑脊液/腹水/羊水等)中胎盘生长因子(PIGF)含量的试剂盒。
根据本发明的一方面,上述的一种检测人体液中胎盘生长因子含量的试剂盒包括校准品、质控品、抗试剂、磁微粒试剂、发光底物。
根据本发明的一方面,上述的一种检测人体液中胎盘生长因子含量的试剂盒,上述抗试剂为碱性磷酸酶标记的抗胎盘生长因子标记抗体。
根据本发明的一方面,上述的一种检测人体液中胎盘生长因子含量的试剂盒,上述磁微粒试剂为磁微粒-抗胎盘生长因子包被抗体连接物。
根据本发明的一方面,上述的一种检测人体液中胎盘生长因子含量的试剂盒,上述化学发光底物为AMPPD。
根据本发明的一方面,上述的一种检测人体液中胎盘生长因子含量的试剂盒,上述人体液的样本来源为血清、血浆、脑脊液、腹水或羊水其中一种或多种。
技术原理为:包被抗胎盘生长因子(PIGF)抗体的磁珠与碱性磷酸酶(AP)标记的胎盘生长因子(PIGF)配对抗体和样本、校准品或质控品中的胎盘生长因子结合形成“三明治”复合物。在外加磁场的作用下,将免疫反应形成的复合物与未结合的其他物质分离,清洗复合物后,加入酶促化学发光底物。底物在酶作用下被催化裂解,形成不稳定的激发态中间体,当激发态中间体回到基态时发出光子,形成发光反应,即可使用化学发光仪检测反应的发光强度。在检测范围内,发光强度与样本中的胎盘生长因子的含量成正比,使用改良的四参数Logistic方程拟合可计算出样本中胎盘生长因子浓度。
本发明将化学发光技术与免疫磁微粒相结合,提供了一种接近均相的反应体系,并且采用了一步法反应模式,使得检测灵敏度、精密性大大提高、检测范围扩大,反应时间大大缩短,从开始加样到检测结果,时间少于20min;并且可以在全自动化学发光仪上同时测定多个样本,实现胎盘生长因子(PIGF)的高通量快速化测定,准确度高,特异性强,精确度和检测效率有了较大的提高。
本发明使用基于磁微粒的化学发光(碱性磷酸酶催化)免疫分析检测胎盘生长因子的方法。

下面结合附图和实施例,对本发明的具体实施方式作进一步详细描述。以下实施例用于说明本发明,但不用来限制本发明的范围。
实施例中未注明具体条件的实验方法,通常按照常规条件,例如《精编分子生物学实验指南》(奥斯伯著)中所述的条件,或按照制造厂商所建议的条件。
实施例1、检测人体液中胎盘生长因子含量的试剂盒
本实施例提供检测人体液中胎盘生长因子含量的试剂盒,试剂孔中的试剂为预先封装进试剂条,采用铝塑复合材料进行封膜,这些试剂含有校准品、质控品、抗试剂、磁微粒试剂、发光底物(AMPPD)等试剂。
实施例2、胎盘生长因子检测
(一)样本-磁珠孵育
将100μL血浆样本转移至反应孔中,随后将试剂孔中的100μL含磁微粒-抗胎盘生长因子包被抗体连接物转移至样本所在反应孔,维持反应孔恒温状态,为37度。
(二)磁珠转移清洗
上一步温育结束,仪器的磁分离模块将反应孔中的磁微粒吸附富集,随后将不含磁微粒的废液吸去。然后,将试剂孔中的清洗液加入反应孔中,同时,磁分离模块关闭,对反应孔进行吹打混合,静止30秒,磁分离模块启动,将磁微粒吸附,移液模块将清洗液吸去,完成一次清洗。

当上一步清洗完成后,信号测定模块对信号进行信号采集分析,获得对应的浓度值,用时总时间小于20分钟,标准曲线图如图1。

1.一种检测人体液中胎盘生长因子含量的试剂盒,其特征在于,所述试剂盒包括校准品、质控品、抗试剂、磁微粒试剂、发光底物。
2.根据权利要求1所述的一种检测人体液中胎盘生长因子含量的试剂盒,其特征在于,所述抗试剂为碱性磷酸酶标记的抗胎盘生长因子标记抗体。
3.根据权利要求2所述的一种检测人体液中胎盘生长因子含量的试剂盒,其特征在于,所述磁微粒试剂为磁微粒-抗胎盘生长因子包被抗体连接物。
4.根据权利要求3所述的一种检测人体液中胎盘生长因子含量的试剂盒,其特征在于,所述化学发光底物为AMPPD。
5.根据权利要求4所述的一种检测人体液中胎盘生长因子含量的试剂盒,其特征在于,所述人体液的样本来源为血清、血浆、脑脊液、腹水或羊水其中一种或多种。
