本发明公开了一种回乳抑增颗粒回流提取方法,属于中药提取技术领域。所述方法包括以下步骤:S1.称取夏枯草20份、生麦芽120份、浙贝母10份、玄参5份、山慈菇3份、昆布5份、牡蛎10份;S2.采用6倍量40%乙醇于70℃下加热,回流提取3次,每次0.5h;S3.过滤,滤液于70℃下浓缩回收溶剂;S4.将浓缩后的药液在‑0.1MPa条件下真空干燥成干浸膏,即得。本申请制备得到的回乳抑增颗粒中迷迭香酸、哈巴俄苷含有量分别为40.35μg/g、8.03μg/g。本申请使用方便,所得成品便于携带和服用,便于保存,同时能够对药物的质量和药效进行准确控制,利于患者恢复,具有实用性。

Description

一种回乳抑增颗粒回流提取方法
技术领域
本发明涉及中药提取技术领域,特别涉及一种回乳抑增颗粒回流提取方法。
背景技术
高泌乳素血症为女性内分泌系统常见疾病,由下丘脑-垂体-性腺轴系统病变引起内分泌紊乱、激素失调所致。近年来研究显示,泌乳素含有量与肝郁有关,肝气郁结、情志不遂可使气血逆乱、脏腑功能失调,从而导致泌乳素异常升高,中医主要采用疏肝理气、调摄冲任、逐瘀化痰方法进行治疗。
复方回乳抑增一号是武汉市第三医院用于治疗高泌乳素血症的临床经验方,由于原方回乳抑增方药量过大,前期已对其进行优化,优化后的处方组成为生麦芽、夏枯草、郁金、丹参、玄参、浙贝母、白芥子、牡蛎、淡昆布、山慈菇7 味药材,但作为传统汤剂在临床使用时仍有很多不便之处,如不宜携带、服用麻烦、难以保存、质量不可控制等,故需要对该剂型进行改进。
发明内容
针对现有技术存在的不足,本发明的目的是提供一种回乳抑增颗粒回流提取方法。
本发明的上述技术目的是通过以下技术方案得以实现的:
一种回乳抑增颗粒回流提取方法,包括以下步骤:
S1.称取规定量的夏枯草、生麦芽、浙贝母、玄参、山慈菇、昆布、牡蛎;
S2.采用6倍量40%乙醇于70℃下加热,回流提取3次,每次0.5h;
S3.过滤,滤液于70℃下浓缩回收溶剂;
S4.将浓缩后的药液在-0.1MPa条件下真空干燥成干浸膏,即得。
进一步的,步骤S1中,7味中药的用量为:夏枯草20份、生麦芽120份、浙贝母10份、玄参5份、山慈菇3份、昆布5份、牡蛎10份。
进一步的,步骤S4中,制备得到的回乳抑增颗粒中迷迭香酸、哈巴俄苷含有量分别为40.35μg /g、8.03μg /g。
本发明具有以下有益效果:
本申请的回乳抑增颗粒回流提取工艺结合药效学实验加以验证,为后期相关研究奠定基础。本申请使用方便,所得成品便于携带和服用,便于保存,同时能够对药物的质量和药效进行准确控制,利于患者恢复,具有实用性。
具体实施方式
以下结合实施例对本发明作进一步详细说明。
实施例1
一种回乳抑增颗粒回流提取方法,包括以下步骤:
S1.称取夏枯草20g、生麦芽120 g、浙贝母10 g、玄参5 g、山慈菇3 g、昆布5 g、牡蛎10g;
S2.采用6倍量40%乙醇于70℃下加热,回流提取3次,每次0.5h;
S3.过滤,滤液于70℃下浓缩回收溶剂;
S4.将浓缩后的药液在-0.1MPa条件下真空干燥成干浸膏,即得。
性能检测
1. 迷迭香酸、哈巴俄苷含有量测定
色谱条件Wondasil C18色谱柱( 4.6 mm×250 mm,5μm) ;流动相乙腈(A)-0. 1%磷酸(B) ,梯度洗脱( 0~10 min,95%B;10~30 min,95%~70%B;30~55 min,70%~80%B) ;体积流量1.0 mL /min;柱温25℃;检测波长270 nm;进样量20μL。
对照品溶液制备:精密称取对照品迷迭香酸13.2 mg、哈巴俄苷8.86 mg,置于同一50 mL量瓶中,50%甲醇溶解并稀释至刻度,摇匀,0.2μm微孔滤膜过滤,即得。
经测定,实施例1的产品中迷迭香酸、哈巴俄苷含有量分别为40.35μg /g、8.03μg/g。
2. 浸膏得率测定
精密移取提取液50 mL,置于已干燥至恒重的蒸发皿中,水浴蒸干,105℃常压干燥至恒重,取出,置于干燥器中冷却30 min,迅速精密称定质量,计算浸膏得率。
经测定,实施例1的产品浸膏得率为12.03%。
3.动物实验
高泌乳素血症模型的建立方法为:为雌性未孕Wistar 大鼠(体质量180~200 g)注射盐酸甲氧氯普胺注射液,以皮下、肌肉注射为主,给药10 d,每天1 次,或者给药5 d,每天2次。本实验采用皮下注射给药以减少肌肉损伤,并为了节省时间采取给药5 d,每天2 次(早晚各1次) 的方案,除正常组外每组大鼠给药量均为75 mg /kg。
分组与给药:将大鼠随机分为正常组、模型组、实验组,每组8只,模型组、实验组进行高泌乳素血症模型构建,然后实验组灌胃给予药物,正常组、模型组大鼠灌胃给予生理盐水。给药30 d 后,大鼠眼角静脉取血,测定PRL、P、E2水平。
统计学分析:通过SPSS 16. 0软件进行处理,数据以( x±s) 表示,组间比较采用t 检验。P<0. 05 表示有显著性差异。
结果分析:如表1所示,模型组泌乳素水平较正常组有显著变化( P<0. 01) ,表明造模成功,而且在给予生理盐水30 d 后未恢复;实验组泌乳素水平较模型组有显著变化( P<0. 05,P<0. 01)。

1.一种回乳抑增颗粒回流提取方法,其特征在于:包括以下步骤:
S1.称取规定量的夏枯草、生麦芽、浙贝母、玄参、山慈菇、昆布、牡蛎;
S2.采用6倍量40%乙醇于70℃下加热,回流提取3次,每次0.5h;
S3.过滤,滤液于70℃下浓缩回收溶剂;
S4.将浓缩后的药液在-0.1MPa条件下真空干燥成干浸膏,即得。
2.根据权利要求1所述的一种回乳抑增颗粒回流提取方法,其特征在于:步骤S1中,7味中药的用量为:夏枯草20份、生麦芽120份、浙贝母10份、玄参5份、山慈菇3份、昆布5份、牡蛎10份。
3.根据权利要求1所述的一种回乳抑增颗粒回流提取方法,其特征在于:步骤S4中,制备得到的回乳抑增颗粒中迷迭香酸、哈巴俄苷含有量分别为40.35μg /g、8.03μg /g。
