Abstract
本发明公开了一种增强免疫力的组合物及其制备方法和应用,属于保健品、医药技术领域,包括A组份和B组份,其中A组份包括以下重量百分比的物质:担子菌1‑20%、裸藻1‑10%、酵母β‑葡聚糖1‑20%、刺梨1‑10%、金银花1‑10%和西兰花种子提取物1‑5%;B组份包括以下重量百分比的物质:大豆肽10‑80%和胶原肽10‑50%;并通过放入搅拌机中混合搅拌,放料备存,混合罐装,得到成品。本发明的组合物能够有效提高免疫能力,且成分温和安全;而本发明的组合物的制备方法能够保持各物质的活性,并使各成分充分反应,协同提升整体的功效和作用,降低成分的刺激性,达到温和无刺激的效果。
Description
技术领域
本发明涉及保健品、医药技术领域,具体属于一种增强免疫力的组合物及其制备方法和应用。
背景技术
免疫力是人体自身的防御机制,是人体识别和消灭外来侵入的任何异物(病毒、细菌等);处理衰老、损伤、死亡、变性的自身细胞以及识别和处理体内突变细胞和病毒感染细胞的能力,现代医学研究证明,免疫力低下是造成疾病的根本原因。
在免疫能力低下时,系统不能正常发挥保护作用,这种情况下,极易招致细菌、病毒、真菌等感染,因此容易导致人体很是容易生病。为了避免人体免疫力低下容易生病,或者提高人体对疾病的抵抗能力,则需要额外补充相应的药物或者保健品等以增强人体的免疫能力。随着人们对人体免疫能力的重视,各大医药和保健品厂家也推出了各类增强免疫力的产品,但是其免疫效果不佳,无法有效提高人体的免疫力,或者有些产品满足了增强免疫力的作用,却由于所用的配方成分刺激性较大,导致人体更加敏感甚至还有一定的副作用。
此外,目前市场上增强免疫力的药物或者保健品的免疫效果不够温和、全面、稳定,不适合长期服用,无法达到良好的养生保健和预防疾病的效果。因此,如何获得具有全面、显著增强免疫效果,且温和无刺激的组合物,成为本领域一直以来焦点关注的技术难题。
发明内容
针对背景技术中存在的缺陷和不足,本发明提供了一种增强免疫力的组合物,能够有效提高免疫能力,且成分温和安全。
本发明的目的之二是提供一种增强免疫力的组合物的制备方法,通过该制备方法能够保持各物质的活性,并使各成分充分反应,协同提升整体的功效和作用,降低成分的刺激性,达到温和无刺激的效果。
本发明的目的之三是提供一种保健品及一种药物,使得上述增强免疫力的组合物得到更广泛的推广和使用。
为实现上述目的,本发明采用以下技术方案予以实现:
一种增强免疫力的组合物,包括A组份和B组份,
其中A组份包括以下重量百分比的物质:担子菌1-20%、裸藻1-10%、酵母β-葡聚糖1-20%、刺梨1-10%、金银花1-10%和西兰花种子提取物1-5%;
B组份包括以下重量百分比的物质:大豆肽10-80%和胶原肽10-50%。
进一步所采取的措施是:所述A组份包括以下重量百分比的物质:担子菌3-5%、裸藻3-4%、酵母β-葡聚糖2-20%、刺梨5-10%、金银花3-10%和西兰花种子提取物2-5%;
所述B组份包括以下重量百分比的物质:大豆肽30-60%和胶原肽12-40%。
进一步所采取的措施是:所述西兰花种子提取物采用干燥的西兰花,经过超微粉碎机粉碎为超微颗粒,再经过水提取超临界萃取处理,最后经过浓缩制得,且纯度为99.8%以上。
进一步所采取的措施是:所述A组份还包括紫芝1-3%和巴茅果1-5%。
进一步所采取的措施是:所述紫芝和巴茅果的重量比例为1:(1-1.5)。
进一步所采取的措施是:所述A组份包括以下重量百分比的物质:担子菌3%、裸藻3%、酵母β-葡聚糖2%、刺梨10%、金银花3%、西兰花种子提取物2%、紫芝2%和巴茅果3%;
所述B组份包括以下重量百分比的物质:大豆肽60%和胶原肽12%。
一种增强免疫力的组合物的制备方法,包括以下步骤:
(1)将A组份中相应重量百分比的各物质放入搅拌机中,设定温度为18-26℃,搅拌速度为75转/分钟,混合45分钟,得到A相,放料备存;
(2)将B组份中相应重量百分比的各物质放入搅拌机中,温度为25-30度,搅拌速度为45转/分钟,混合30分钟,得到B相,放料备存;
(3)将备存的A相和B相放入搅拌机中,温度为30℃,搅拌速度为45转/分钟,混合30分钟,放料备存,进行灌装,得到成品。
进一步所采取的措施是:所述步骤(3)中的灌装过程包括:开启10列粉剂灌装机,进行预热30分钟,真空上料到10列粉剂灌装机料斗,调整装量为5克/袋,控制生产速度300条/分钟,开始灌装,进行检查气密性和装量均一性,检验不合格的产品重新灌装,检验合格的产品则收集转入外包车间,按每盒15-30条,装条成盒,烟包机过膜完成灌装,得到成品。
一种保健品,所述保健品包括权利要求1-8所述增强免疫力的组合物。
一种药物,所述药物包括权利要求1-8所述增强免疫力的组合物。
西兰花种子提取物含特殊成分萝卜硫苷和萝卜硫素,能使产品的功能有所提升,但原料稳定性和有效率都比较差,无法真正全面、有效增强免疫力的效果。本发明通过对西兰花种子提取物的生产工艺做了改进,利用前期的超微粉碎和水提取超临界萃取技术的结合,并进一步浓缩,从而充分提取出西兰花种子中包括萝卜硫苷和萝卜硫素等有效成分,得到提取完全且纯度高的西兰花种子提取物,大大提高了西兰花种子提取物的原料稳定性和有效率,从而在配方中发挥出显著提高免疫力,以及治疗和预防肿瘤的效果。
裸藻具有抗氧化和抗病毒的作用,但使用不当会产生副作用,而酵母β葡聚糖具有激活免疫系统的作用,但作用发挥不太稳定,本发明通过裸藻和酵母β葡聚糖与西兰花种子提取物采用特定配比进行搭配,使得三者的功能相辅相成,取得了显著增强免疫力,预防人体疾病发生的效果,同时通过本发明特殊的制备方法,使得裸藻、酵母β葡聚糖和西兰花种子提取物进行前期的预混,促使三者充分反应后再与大豆肽等成分混合,很好的解决了原料的稳定性问题,并且提升整体的协调功效,促进和增强人体免疫力的稳定发挥。
紫芝,一般用于安神健胃,滋补强壮,主治神经衰弱和食欲不振;巴茅果一般用于补中强智、宁心益胃,以及治疗消化不良和解菌毒,但通过紫芝与巴茅果特定比例的搭配,二者协调作用,加速人体代谢,并起到显著提高人体抵抗力,并具有突出的抗癌作用,并通过与本发明的其他成分配伍,特别是与裸藻和经过本发明特殊提取方式的西兰花种子提取物,使得大大提高整体抵抗病毒的能力,并维持整体的稳定性,使得整体作用的发挥更加持续有效,并且各成分温和,搭配合理,不会产生副作用。
还有大豆肽和胶原蛋白肽,具有抗氧化、调节免疫力的作用,并且容易被人体吸收,给人体提供营养的同时增强人体的抵抗能力;而刺梨和金银花起到舒缓的作用,降低整体的刺激性,随着对组方的研究越来越深入,也越来越发现大豆肽与本发明其他成分以特定比例进行搭配,再配合本发明独特的制备方法,具有人们无法预料的特殊生理功能,使得整体配方协同作用,功效得到最大化的发挥,起到优异的增强免疫力的作用,产生极好的养生保健预防效果,并且长期使用也温和无刺激。
本发明经过长期深入的研究,效果突出显著,适合推广应用,可用于保健品,医药甚至添加到食用产品中。
通过上述技术方案,本发明与现有技术相比,所具有的有益效果如下:
1、本发明成分温和安全,整体搭配新颖合理,使得各成分之间相互协调作用,从而稳定、有效的提高免疫能力,且效果优异、安全无刺激,达到良好的养生保健和预防疾病的效果。
2、本发明制备方法根据本发明的配方进行设计,制备步骤精简,且能够使得各功能成分都能够发挥出最佳的功效,并抑制成分之间作用产生的刺激性,从而达到显著增强免疫力和预防肿瘤等疾病发生的作用,且整体温和无刺激。
3、通过将本发明的组合物应用于保健品及药物中,使得本发明增强免疫力的组合物能够得到更广泛的推广和使用。
具体实施方式
为了更清楚的了解本发明所采用的技术方案,下面对本发明的优选实施例进行说明,应当理解,此处所描述的实施例仅用于说明和解释本发明,并不用于限定本发明。
实施例1-5:一种增强免疫力的组合物,所包含的具体成分及相应重量百分比如下表1所示。

上述实施例1-5均通过以下制备方法制得,包括以下步骤:
(1)将A组份中相应重量百分比的各物质放入搅拌机中,设定温度为18-26℃,搅拌速度为75转/分钟,混合45分钟,得到A相,放料备存;
(2)将B组份中相应重量百分比的各物质放入搅拌机中,温度为25-30度,搅拌速度为45转/分钟,混合30分钟,得到B相,放料备存;
(3)将备存的A相和B相放入搅拌机中,温度为30℃,搅拌速度为45转/分钟,混合30分钟,放料备存,进行灌装;
(4)灌装过程包括:开启10列粉剂灌装机,进行预热30分钟,真空上料到10列粉剂灌装机料斗,调整装量为5克/袋,控制生产速度300条/分钟,开始灌装,进行检查气密性和装量均一性,检验不合格的产品重新灌装,检验合格的产品则收集转入外包车间,按每盒15-30条,装条成盒,烟包机过膜完成灌装,得到成品。
一种保健品,该保健品包括上述实施例得到的增强免疫力的组合物。
一种药物,该药物包括上述实施例得到的增强免疫力的组合物。
针对以上实施例1-5得到的成品进行免疫能力试验。
首先选取SPF级健康ICR雌性小鼠,18-22g,288只,采购自北京阜康生物科技股份有限公司。将小鼠随机分成6个平行组,每平行组48只。第1-5平行组分别用1-5实例得到的成品进行喂养按照每天5g/60kg的推荐使用量折算,第6平行组不添加任何产品正常进行喂养,6个平行组的小鼠均在同一环境下喂养28天一个生理周期,然后进行以下免疫能力相关检测试验。
(一)体液免疫实验
采用抗体生成细胞检测(Jerne改良玻片法):取二氧化碳培养箱温育1.5h,然后用SA液稀释的补体(1:8)加入到玻片架凹槽内,继续温育1.5h后,计数溶血空斑数。
血清溶血素水平测定:随机在每组小鼠中抽取12只小鼠,进行腹腔注射SR-BC,6天后,摘眼球取血,离心取血清稀释100倍,进行HC50测定,记录下测量数据,结果如下表所示:

由上表可以看出,5个组别小鼠的半数溶血值(HC50)均处于较高水平,明显比对照组正常喂养饲料的小鼠的半数溶血值要高得多,说明喂养本发明实施例获得的组合物的各个小鼠的体液免疫能力都得到了显著、有效的提高,其中实施例1-4对应组别小鼠的半数溶血值均在70以上,体液免疫效果优异。
(二)巨噬细胞吞噬能力测定
碳廓清实验:随机在每组小鼠中抽取12只小鼠,进行尾静脉注射1:3稀释的印度墨汁,待墨汁注入立即计时,分别在注入墨汁后2、10min时(分别用t1、t2表示),从内眦静脉丛取血20μl,并将其加到2ml0.1%碳酸钠溶液中,在600nm波长处测光密度值,分别为OD1、OD2。
将小鼠处死,取肝和脾脏称重,计算吞噬指数。
其中,吞噬指数=结束体重×1/3K/(肝重+脾重);
K=(logOD1-logOD2)/(t2-t1)。

由上表可以看出,5个组别小鼠的溶血空斑数(×103/全脾)均处于较高水平,明显比对照组正常喂养饲料的小鼠的吞噬指数要高得多,说明各个喂养本实施例获得的组合物的小鼠的巨噬细胞都得到了显著、有效的提高,即免疫能力得到了明显提高,其中实施例1和实施例2的溶血空斑数高达29×103以上,具有优异的增强免疫能力的效果。
(三)NK细胞活性测定:将每组剩余的12只小鼠进行颈椎脱臼处死,无菌取脾,制备脾细胞悬液,用Hanks液洗2次,每次离心10min(1000r/min),将细胞悬浮于含10%小牛血清RPMI1640完全培养液中,调整细胞浓度为2×107个/ml。将YAC-1细胞(4×105个/ml)加入96孔培养板,每孔100μl,试验孔加入100μl脾细胞(效靶比50:1),自然释放孔加入100μl培养液,最大释放孔。
加入100μl1%NP40,37℃培养4h离心,取上清100μl置96孔酶标板中,加入LDH基质液100μl,反应3min,以1mol/L的HCL终止反应,酶标仪490nm处测定OD值。
NK细胞活性(%)=反应孔OD值-自然释放孔OD值/(最大释放孔OD值-自然释放孔OD值)×100
将表层培养基加热溶解后与等量双倍Hanks液混合,分装小试管,每管0.5ml,再向管内加50μl 10%SRBC(V/V,用SA液配制)、25μl脾细胞悬液,迅速混匀后,倾倒于已刷琼脂糖薄层的玻片上,待琼脂凝固后,将玻片水平扣放在玻片架上,放入,记录结果,如下表4所示。

由上表可以看出,5个组别小鼠的NK细胞活性均处于较高水平,明显比对照组正常喂养饲料的小鼠的NK细胞活性要高得多,说明各个喂养本实施例获得的组合物的小鼠的NK细胞活性都得到了显著、有效的提高,即免疫能力得到了明显提高,其中实施例1的NK细胞活性高达33.5%,效果最优。
并进行汇总结果,进行实验数据统计:数据均用Stata软件进行方差分析,P<0.05被认为差异有统计学意义,结果显示6个平行组小鼠的试验数据的方差分析结果均能达到P<0.05,说明本发明6个实施例均有统计学意义。
针对以上实施例1-5得到的成品进行家兔眼刺激性试验:选取20只成年、健康家兔,体重为1.8-2.1kg,雌雄各半,采购自北京阜康生物科技股份有限公司。将家兔随机分成5个平行组,每平行组4只。分别用1-5实例得到的成品对应滴入5个平行组家兔的左侧及右侧的眼结膜囊内,然后使被滴入实施例成品的家兔眼睛的眼睑被动闭合10秒,记录下受试家兔用药后的12h、24h、36h、72h至7天时,眼部的反应及变化情况。
实验结果:5组家兔用药后的12h、24h、36h、72h至7天时,5组家兔的眼角膜、虹膜、结膜均没有浑浊、充血、水肿等异常异常情况出现,全部家兔活动正常。因此,本实施例1-5得到的成品对家兔的眼刺激程度为无刺激。
综合上述实验结果显示,本发明实施例得到的组合物能够稳定、有效提高小鼠的体液免疫及巨噬细胞吞噬能力,也能提高小鼠脾淋巴细胞增殖能力,提高大鼠NK细胞活性,而且安全无刺激性,达到全面提高整体免疫能力,使得整体免疫系统更稳定,抵抗力更强,达到良好的养生保健和预防疾病的效果,造福人类。
以上所述仅用以说明本发明的技术方案而非限制,本领域普通人员对本发明的技术方案所做的均等修饰与变化,只要不脱离本发明创造整体构思的情况下,均仍属于本发明创造涵盖的范围之中。
Claims (10)
1.一种增强免疫力的组合物,其特征在于:包括A组份和B组份,
其中A组份包括以下重量百分比的物质:担子菌1-20%、裸藻1-10%、酵母β-葡聚糖1-20%、刺梨1-10%、金银花1-10%和西兰花种子提取物1-5%;
B组份包括以下重量百分比的物质:大豆肽10-80%和胶原肽10-50%。
2.根据权利要求1所述的一种增强免疫力的组合物,其特征在于:
所述A组份包括以下重量百分比的物质:担子菌3-5%、裸藻3-4%、酵母β-葡聚糖2-20%、刺梨5-10%、金银花3-10%和西兰花种子提取物2-5%;
所述B组份包括以下重量百分比的物质:大豆肽30-60%和胶原肽12-40%。
3.根据权利要求1所述的一种增强免疫力的组合物,其特征在于:所述西兰花种子提取物采用干燥的西兰花,经过超微粉碎机粉碎为超微颗粒,再经过水提取超临界萃取处理,最后经过浓缩制得,且纯度为99.8%以上。
4.根据权利要求1所述的一种增强免疫力的组合物,其特征在于:所述A组份还包括紫芝1-3%和巴茅果1-5%。
5.根据权利要求4所述的一种增强免疫力的组合物,其特征在于:所述紫芝和巴茅果的重量比例为1:(1-1.5)。
6.根据权利要求5所述的一种增强免疫力的组合物,其特征在于: 所述A组份包括以下重量百分比的物质:担子菌3%、裸藻3%、酵母β-葡聚糖2%、刺梨10%、金银花3%、西兰花种子提取物2%、紫芝2%和巴茅果3%;
所述B组份包括以下重量百分比的物质:大豆肽60%和胶原肽12%。
7.权利要求1-6中任一项所述的一种增强免疫力的组合物的制备方法,其特征在于:包括以下步骤:
(1)将A组份中相应重量百分比的各物质放入搅拌机中,设定温度为18-26℃,搅拌速度为75转/分钟,混合45分钟,得到A相,放料备存;
(2)将B组份中相应重量百分比的各物质放入搅拌机中,温度为25-30度,搅拌速度为45转/分钟,混合30分钟,得到B相,放料备存;
(3)将备存的A相和B相放入搅拌机中,温度为30℃,搅拌速度为45转/分钟,混合30分钟,放料备存,进行灌装,得到成品。
8.根据权利要求7所述的一种增强免疫力的组合物的制备方法,其特征在于:所述步骤(3)中的灌装过程包括:开启10列粉剂灌装机,进行预热30分钟,真空上料到10列粉剂灌装机料斗,调整装量为5克/袋,控制生产速度300条/分钟,开始灌装,进行检查气密性和装量均一性,检验不合格的产品重新灌装,检验合格的产品则收集转入外包车间,按每盒15-30条,装条成盒,烟包机过膜完成灌装,得到成品。
9.一种保健品,其特征在于:所述保健品包括权利要求1-8所述增强免疫力的组合物。
