百日咳是一种主要侵袭儿童的高度传染性疾病。除了引起呼吸并发症,还可导致神经损伤和高的死亡发生率,特别是在来自社会经济低阶层的孩子中以及不具有母亲抗-百日咳抗体的新生儿中。百日咳的病原体是革兰氏阴性球杆菌属,百日咳杆菌。人们确信这种细菌侵入呼吸道并引起一种毒性状态,这种毒性状态甚至能持续至细菌消失数天之后。
对这种疾病的控制通常是通过“全细胞”疫苗免疫。这种“全细胞”疫苗是通过在发酵罐中培养百日咳杆菌并经热处理和/或加入化学试剂对所得细胞进行灭活处理而得。虽然目前世界卫生组织(WHO)推荐对婴儿进行免疫以防止百日咳的发生和漫延,但所报道的由各种不同的疫苗配方所造成的一些不良事件已引起了人们的关注。随之而减少的对常规百日咳疫苗的使用已导致百日咳感染发病率的增加。因此,人们意识到需要一种百日咳疫苗,它能够避免所报道的由全细胞疫苗引起的不良事件。因而,已投入了相当大的研究力量,以期研制出一种有效的包含少数几个高度纯化了的抗原性蛋白组分的无细胞疫苗。
已有几种抗原被提仪作为无细胞疫苗成分,例如淋巴细胞增多激刺因子(lymphocytosis promoting factor,LPF)(亦称为组胺敏感因子(histamine sensitising factor)、岛激活蛋白(isletactivating protein)、一般称为百日咳毒素(pertussis toxin,PT)),丝状血凝素(filamentous hemagglutinin,FHA)以及菌毛凝集素(fimbrial agglutinogens)。
一种更有潜力的抗原是一种细菌的外膜蛋白,分子量约为69000道尔顿(称为pertactin),发现于百日咳杆菌的所有有毒株中。这种百日咳杆菌69kD蛋白与电泳迁移率稍有差异的人类病原体副百日咳博代氏杆菌及动物病原体支气管败血性博代氏杆菌所产生的类似蛋白质有免疫学相关性。虽然在百日咳杆菌培养过程中,只有相当少量的69kD蛋白分泌到发酵液中,但发现大部分的69kD蛋白结合在细胞膜上,很容易从中提取出来。然而,已发表的69kD蛋白的提取和纯化程序均不能用于大规模的工业性生产高度纯化的和稳定的抗原。
EP-0162639描述的程序是,将细胞进行酸-甘氨酸提取后,进行几步纯化步骤,最后利用一种特异的单克隆抗体进行亲和层析分离。不过,业已报道,所获得的蛋白质不仅稳定性差而且有腺苷酸环化酶活性,而且这种下游的纯化程序不适合于大规模生产。
Brennan等人(Infection and Immunity 56,3189-3195,1988)描述了一种更进一步的方法,通过热处理使蛋白质从细胞上释放下来,得到一种蛋白质提取液,然后通过胎球蛋白-Sepharose(琼脂糖)层析和一种单克隆抗体亲和层析进行纯化。
美国专利号(US Setial No.7/308864)描述的提取和纯化程序包括热处理和离心,随后是进行DEAE(二乙氨乙基)-Sepharose离子交换层析和蛋白质-特异的,染料-配体凝胶层析。这一方法避免了昂贵的单克隆抗体的使用,但仍被认为不适用于大规模生产。已经查出的一个特殊问题是,如果按占热处理后释放的总蛋白的百分率计算,释放到溶液中的69kD蛋白量很少。EP-A-0437687通过一个涉及许多步提取步骤的重复提取过程,使从培养液中释放69kD蛋白的效率有了改善。
本发明提供一个处理过程,经过单一提取步骤后,可增强从微生物细胞释放的蛋白质产量,从而克服了蛋白回收效率低的问题。本发明也免除了随后的离心去除细胞“碎片”的需要。此外,本发明方法的另一个优点是,它有效地去掉了大部分的大分子内毒素(脂多糖)。这些内毒素存在于发酵后的液体培养基中,当利用热处理提取膜蛋白时,又进一步释放到溶液中。
发酵后对微生物悬液的热处理,是对那些在发酵过程中很少被微生物分泌到液体培养基中的蛋白质(如外膜蛋白质)的分离过程中的一个重要步骤。百日咳杆菌69kD蛋白即是这种典型的外膜蛋白质,它的分泌量不足以保证能以生产规模从液体培养基的上清液中直接分离出来。在这一点上,它不同于无细胞百日咳疫苗其它的候选抗原:百日咳毒素(PT)和丝状血凝素(FHA)。
热处理过程包括,在例如60℃下加热缓冲液中的细胞悬液约1小时,在此期间,外膜蛋白质从细胞膜脱落进入溶液中。然而这种溶液并不是一种能自由流动的液体,而主要是一种粘性的胶状物质,由死细胞及其内含物组成,这种胶状物质虽然能小规模(5-10ml等份试样)地从自由流动的液体中分离出去(如通过过滤或离心),但很困难。这种高度水合的胶状物常含有高于50%体积的原液,如果弃掉,将明显地降低69kD蛋白的产率。而且粗滤也不尽人意,因为胶状物会很快堵塞滤器,有效地阻止液体的通过。
本发明提供了一种可在热处理过程中免除胶状团块形成的有效方法。业已发现,在热处理之前向细胞悬液中加入一种絮凝剂可使细胞发生强烈的絮凝作用。可很容易地把这种絮凝了的细胞块从悬浮液体培养基中分离出来,并可在热处理之前进行洗涤以除去非必需的液体培养基成分。热处理后,将死细胞进行再絮凝,从而留下一种实际上是无细胞的上清液,内含极大部分的可溶性蛋白。通过这一程序可获得高回收率的目的蛋白溶液,用于随后的纯化。
本发明另外的优点是,由于在热处理后,大多数高分子量的内毒素已随同死细胞絮凝块从含蛋白溶液中清除出去,所以它所提供的回收蛋白可允许其只需进行较不严格的随后的纯化。因此,有可能减少为达到预防和治疗目的所要求的蛋白纯度标准所需的随后纯化步骤。
本发明因此提供了一种细菌来源的细胞结合蛋白的提取方法,包括把细胞结合蛋白悬液与一种絮凝剂接触。
本领域中熟知的多种絮凝剂均可用于本发明的方法中,以改善热处理后细胞悬液的处理质量。用于本发明的优选絮凝剂是那些具有二价阳离子的物质。用于本发明的合适的二价阳离子可来自钡盐、钙盐和锶盐,例如卤化盐:氯化钡、氯化钙、或氯化锶,优选氯化钡。
这种絮凝剂适合于在pH控制的条件下加入到细胞悬液中,加入适当的缓冲液以调整液体体积。混匀后,将絮凝的细胞静置。将絮凝细胞用适当的沉降方法(或离心)来收集,并在进行热处理之前可用盐水或缓冲液洗涤。
本发明的提取方法,如果结合随后的进一步处理,可从百日咳杆菌获得纯度很高的69kD蛋白。例如,它不含有可检出水平的PT或热不稳定毒素,并且内毒素水平降至每毫克蛋白几毫微克的水平。此外,此蛋白没有酶活性,尤其是没有以前的生产方法带来的腺苷酸环化酶活性。
在本发明的一个优选实施方案中,69kD蛋白是从百日咳杆菌的液体发酵培养基中制备而来。并对用于本发明的合适菌株作了描述,这些菌株可从一些商业性的保藏单位获得,如美国马里兰州洛克费勒市的美国典型培养物保藏中心(the American Type Culture Col-lection,Rockville Maryland,USA)。优选的菌株是那些能在液体培养基中生长,并具有高产量的69kD蛋白的菌株。
可使用的菌株包括(但不限于):百日咳杆菌工期(B.pertus-sis phase I)、百日咳杆菌II期、百日咳杆菌I期CS(B.pertussisphase I CS)、百日咳杆菌Tohama株、百日咳杆菌185-30株、百日咳杆菌18323株、百日咳杆菌134株、百日咳杆菌509株、百日咳杆菌Wellcome 28株、以及生物制剂办公室(Office of Biologics)百日咳杆菌165株。本发明中使用的优选菌株是百日咳杆菌I期,Tohama,可从日本大阪的发酵研究所获得,登记号为IFO-14073。
为了用于本发明,可用本领域技术人员所知的各种不同方式培养所选择的百日咳杆菌菌株。已知有各种不同的培养方法,根据种子培养物的数量、来源或保存方法,可采用不同的培养步骤以及液体或固体培养基。不过,只要能提供用于大规模生产常规所能接受的种菌量,已知的任何一种方法都可用于本发明。
本领域的任何-个技术人员都能选择出一种用于百日咳杆菌种菌培养的合适培养基。合适的培养基包括(并不限于):Gengou培养基(EP-A-0077646);N.Andorn等人所描述的培养基(Appi.Micro-biol.Biotechnol.,28,356-360,1988)以及其所引文献中的培养基;Stainer-Scholte培养基(J.Gen.Microbiol.,63,211-220,1971);A.Imaizumi等人所描述的改良的Stainer-Scholte培养基(Infect.Immun.,41,3,1138-1143,1983及J.Microbiol.Methods,2,339-347,1984);Verway培养基(US4784589);合成培养基B2(P.Van Hemert;Prog.Indust.Microbiol.;(Bull,M.J.ed.),Vol 13,p.151,Elsevier Sci.,Amsterdam(1977))或其所描述的改良培养基。
为了培养本发明的起始材料百日咳杆菌,把种菌加到一合适的液体培养基中,并用本领域所知的常规发酵方法和发酵装置进行发酵。本领域内的技术人员懂得,基于所选择的常规发酵装置、发酵培养基、方法及参数的不同组合,会获得不同的结果。用于本发明的优选组合是那些适用于大规模生产的组合。关于方法、装置及培养基的这种组合的例子示于EP-A-0077646,更好的例子是在EP-A-0121249及EP-A-0239504中。
发酵完成后,对这种百日咳杆菌发酵液进行适当的处理以除去直接分泌到液体培养基中的抗原性因子PT和FHA,例如,用EP-A-0427462中所描述的方法,使抗原性因子吸附到羟基磷灰石上,其方法引入本文作为参考。
余下的含有69kD细胞结合蛋白的微生物悬液,在进行热处理以实现蛋白质释放之前,先按照本发明用一种絮凝剂处理。
业已发现,通过控制上清液的pH值,可使残留的内毒素含量降到最小值。通过常规试验,即可选择出任一所选絮凝剂的最适pH。因此,在加入例如钡、钙或锶的卤化盐(优选氯化钡)水溶液之前或之后,用碱把上清液的pH调整至pH4-10之间,最好是在pH8.5-9.5之间。液体体积可通过加入缓冲液进行调整,如用TRIS缓冲液。适当混匀约5分钟后,让所得的悬浮液絮凝约1小时。然后分离并弃掉上清液,此上清液中含有未用完的生长培养基成分(盐、氨基酸、矿物质等)、以及在培养过程中释放到液体培养基中的其它蛋白物质和内毒素,所获絮凝物可进一步用缓冲液或盐水洗涤。或在洗涤之前离心把絮凝细胞分离出来。
由于存在于发酵液中的高浓度的盐、氨基酸和矿物质会干扰随后的纯化步骤,所以这种絮凝/洗涤处理过程可能需要重复多次,直至所获得的絮凝细胞悬浮液中卤化盐含量降低至一个可接受的水平(如约0.02%w/v)。
然后把絮凝细胞悬浮液于约60℃下热处理15-60分钟,在此期间,69kD蛋白被释放到液体悬液中,细菌被杀死。冷却(如在约4℃下)适当的时间(最好过夜)后,把死细胞去掉(如用倾析或离心),上清液过滤后,于无菌状态下低温(如约4℃下)保存。
然后用在此领域已知的适当技术,把69kD蛋白进行下游纯化。优选采用离子交换、疏水作用和分子阻隔层析三者的组合,对69KD蛋白进行纯化。合适的层析载体包括:阴离子交换,如DEAE-Se-pharose、Q-Sepharose、SP-Sepharose、CM-Sepharose;疏水作用,如Butyl-(丁基-)、Phenyl-(苯基-)、Octyl-(辛基-)Sepharose、TSK;以及凝胶过滤,如Sephacryl、Sepharose、Sephadex(交联葡聚糖)、Superose、Superdex等等。
这种高度纯化了的69kD蛋白可采用过滤除菌,如有必要,可进行双过滤(diafiltration)。
对以上所描述的方法分离而得的纯化的69kD外膜蛋白抗原,可用此领域内熟知的技术来鉴定,例如用SDS-PAGE(十二烷基磺酸钠-聚丙烯酰胺凝胶电泳)并用单克隆抗体进行Western印迹(Western blot)分析而确定。其纯度可用SDS-PAGE进行定性分析,定量分析则可用ELISA(酶联免疫吸附试验)或HPLC(高压液相层析)方法。
按照本发明所生产的纯化的69kD蛋白,ELISA检测表明其PT和FHA含量低于检出灵敏度极限值。没有可检出的PT活性(CHO细胞)。用LAL-CS(鲎属变形细胞溶解产物-生色底物,Limulus AmoebocyteLysate-Chromogenic Substrate)检测表明内毒素含量低于0.1单位/mcg(μg)蛋白。该69kD蛋白没有可检出的腺苷酸环化酶活性。用Western印迹方法分析其腺苷酸环化酶为阴性。
所得的69KD蛋白可用作一种无细胞疫苗的抗原成分,注射这种疫苗可激发对百日咳杆菌感染的一种保护性抗体反应。这样的疫苗可用常规方法制备,如,可制备一种疫苗,其水溶液中含有一些抗原因子和一种合适的常规载体,并把其pH缓冲至生理pH值,以便可直接使用。另一种方法是,也可把抗原因子吸附到一个常规的佐剂上,如氢氧化铝或磷酸铝。此外,也可以把一种或多种百日咳杆菌抗原(包括69kD蛋白)与其它免疫原组合起来制备成多价疫苗,可诱导对多于一种病原体的保护作用。
本发明还有其特殊的用途,它可作为从单一的百日咳杆菌发酵液中分离和纯化几种无细胞百日咳疫苗候选蛋白抗原的整个处理过程的一部分。因此,可以从同一份发酵液中分别分离获得纯化形式的PT、FHA和69kD蛋白,用于共同组成一个单一的百日咳疫苗。可按照EP-A-0427466中所描述的方法对百日咳杆菌发酵液进行适当处理以分离出PT和FHA,余下的上清液则用本发明所描述的方法进行处理以获得69kD蛋白。百日咳毒素可按照WO91/12020中所述的方法进行类毒素化,其方法引入本文作为参考。已分别纯化的抗原成分可被分别佐剂化,然后合并起来。
通过加入一种絮凝剂(优选的是一种二价阳离子),从溶液中提取细胞结合蛋白并去除内毒素的方法并不仅限于从致病的百日咳微生物(如百日咳杆菌)中分离外膜蛋白(如Pertactin)。本发明的方法在任何涉及预防的或治疗的外膜蛋白的生物学生产过程中都有其实用性。可以从本发明受益的其它生物学处理过程的典型例子有:
a)从脑膜炎属(Meningitidis)菌种如脑膜炎双球菌(Neisseriameningitidis)中生产外膜蛋白;
b)从大肠杆菌(E.Coli)中生产菌毛(Pili)和凝集素;
c)从嗜血杆菌属菌种如流感嗜血杆菌(Haemophilus influen-zae)中生产外膜蛋白;
d)从包柔氏螺旋体属菌种如Borrelia burgdorferi中生产外膜蛋白;
e)从链球菌属菌种如甲型和乙型链球菌(Streptococcus)菌株中生产外膜蛋白。
可根据本发明的方法获得的生物学产品并不仅局限于用于人类的蛋白质。可受益的另一些方法的典型例子是,分离外膜蛋白用于:
f)鼻炎疫苗的生产,此病可侵袭猪,是由支气管败血性博代氏杆菌(Bordetella bronchiseptica)和Haemophilus Pleuropneumo-niae引起的;
q)大肠杆菌病(Colibacillosis)疫苗的生产,此病可侵袭猪,是由大肠杆菌(E.Coli)引起的;
h)一种用于钩端螺旋体病(Leptospirosis)ELISA检测的抗原的生产(此病的侵袭对象是家畜、牛犊和人类)。
通过下面关于从百日咳杆菌中提取69kD外膜蛋白的实施例,举例说明了(但不是限定了)本发明的方法。

实施例1
絮凝试验
在控制的发酵条件下培养百日咳杆菌Tohama株,使获得的细胞悬液量足以用羟基磷灰石凝胶吸附方法从上清液中提取PT和FHA。剩余的细胞悬液用下列步骤处理:
1)使悬液冷却至4℃,并分成50ml的等份样品。
2)每份样品中分别加入下列絮凝剂之一使其达到所标明的终浓度:葡聚糖T500(10g/L),氯化钙(10g/L),甲醇(20%),及聚乙二醇50000(10g/L)。
3)在充分混匀下,将每份样品的pH调整至事先决定的pH值(葡聚糖T500,pH5;氯化钙,pH4和9;甲醇,pH6;聚乙二醇50000,pH5)。然后将样品于4℃下静置使其絮凝。
4)倾析掉上清液,并代之以等体积的125mM TRIS缓冲液。重新悬浮后,将样品置水浴中加热至60℃并在60℃保持1小时。为保证均匀的温度分布,定期(每隔5分钟)振摇每份样品。
5)在不摇动的情况下冷却至4℃后,死细胞重新絮凝。离心后,对每份样品的上清液进行以下测试:不仅用SDS-PAGE、Westernblot和ELISA检测69kD蛋白的存在,也用LAL-CS试验检测所污染的内毒素的存在。
SDS-PAGE和Western blot的结果分别见图1和图2。每份样品(对应于一种絮凝剂)被点到凝胶和滤膜的两个相邻的样品槽上,点样量分别为15μl和45μl。其余的3个样品槽被点上点样量连续增加(6.8μl、13.6μl和27.2μl)的纯化的69kD蛋白溶液,其浓度为55mg/L。关于在凝胶中和Western blot中的样品槽说明可见下表:

图1和图2表明,69kD蛋白存在于极大多数样品中,并显示其所在位置。列于上表2的ELISA检测结果证实了69kD蛋白的存在。
尽管在pH4和pH9下,CaCl2均可引起絮凝作用,但凝胶上的槽5和6(图1)显示了一个特别显著的例子,即用CaCl2在pH9下处理后,上清液中所存在的内毒素含量显著降低。这已被列于上面表2中的LAL-CS检测结果所证实。
实施例2
二价阳离子的选择
采用实施例1中所述的方法进行絮凝试验,使用浓度为10g/L、pH值为9的以下卤化盐:氯化钙、氯化钡及氯化锶。
SDS-PAGE和Western blot结果分别示于图3和图4。前3个样品槽加的是浓度为55mg/L的纯化69kD蛋白溶液,点样量连续增加(6.8μl、13.6μl和27.2μl),最后1个样品槽加的是细胞悬液的非絮凝化对照样品。关于凝胶中和Western blot中的样品槽说明如下表所示:

图3和图4显示了69kD蛋白存在于所有样品中及其所在位置。在对照样品中(条带7)观察到的69kD蛋白带的密度明显低于被二价阳离子絮凝过的样品中的69kD蛋白带(条带4、5和6),表明在非絮凝化样品的上清液中,69kD蛋白的浓度明显偏低。
表4中ELISA检测得出的低浓度69kD蛋白结果给人的印象是:锶不如钙或钡有效。但这一观察结果并未反映在凝胶中(图3)或印迹中(图4)。
然而,用钙或钡絮凝的样品,在其上清液中观察到的69kD蛋白的浓度,在ELISA检测中两者大致相等;在凝胶和印迹中得出的69kD蛋白带两者密度相似。
肉眼观察絮凝过程,根据絮凝作用的速度和浮块的形态,发现用钡比用钙和锶稍有效些。
实施例3
絮凝和洗涤程序
采用下列程序进行絮凝和洗涤:
1)把提取PT和FHA后剩下的细胞悬液冷却至4℃,并分成4等份样品,每份50ml。留出一份作为对照。
2)其余3份样品中均加入氯化钡,使其终浓度为10g/L。在充分混匀下,用一合适的碱把细胞悬液的pH值调整至pH9。
3)悬液于4℃下静置使其絮凝。
4)把沉淀物上的上清液倾析除去后,分别向沉淀物中加入3种不同浓度的TRIS缓冲液(125、50或20mM)(每份沉淀物加入一种浓度),使其最终体积为50ml。
5)重新悬浮后,把3份样品于4℃下静置,进行第二次絮凝。
6)重复上述4)和5)的程序多次,直至把氯化钡的浓度稀释至约0.2g/L。
7)将对照样品于10,000g离心1小时,弃去上清液,并代之以等体积的125mM TRIS缓冲液。
8)最后一次把样品重悬于所要求浓度的TRIS缓冲液中后,把所有样品(包括对照)置于水浴中加热至60℃,并维持1小时。定期(每隔5分钟)振摇每份样品,以保证有一个均匀的温度分布。
9)在不摇动下把所有样品冷却至4℃后,含氯化钡的样品中的死细胞发生重新絮凝并将对照样品中的死细胞于10,000g下重新离心1小时。
10)用SDS-PAGE、Western blot和ELISA方法检测来自每份样品(包括对照)的上清液中的69kD蛋白。
SDS-PAGE及Western blot的结果分别示于图5和图6。每份样品被点到凝胶和滤膜的两个相邻的样品槽上,点样量分别为15μl和45μl。关于凝胶中和Western blot中的样品槽说明可见下表:

图5和图6显示出所有样品中均存在69kD蛋白及其所在位置。上面表6显示的ELISA检测结果证实了69kD蛋白的存在。
经过如上所述的反复絮凝和洗涤步骤后,细胞悬液的上清液中仍存在69kD蛋白而且其浓度相似,表明:应用这一技术,可大大降低悬浮液体培养基的离子强度,而不会对从百日咳杆菌中69kD蛋白的提取及其产量产生不良的影响。
此外,氯化钡作为一种絮凝剂,即使它在上清液中的浓度低至0.2g/L,其有效性亦得到证实。
Claims (10)
1、一种可提高细菌来源的细胞外膜结合蛋白分离效果的方法,包括用热处理使这种蛋白质从宿主细胞中释放出来,其特征是,在热处理前,把这种细胞结合蛋白的悬浮液与一种絮凝剂相接触。
