长久以来,随着现代化饲料工业的发展,我们在畜禽饲养中广泛使用抗生素、化学合成药物、激素、防腐剂等人工合成的饲料添加剂。这些促生长保健剂带来了畜牧养殖业的革命,推动了规模化养殖的极大发展,但是随着研究和应用的深入,这些添加剂带来的负面效应渐渐显露,特别是抗生素的大量使用,甚至滥用,导致畜禽和水产产品中药物残留严重,病原菌耐药性增强,影响了产品质量,直接或间接威胁人类健康,甚至破坏生态环境。因此,亟待寻求可以替代抗生素并且没有药物残留等毒副作用的绿色添加剂。
中草药是中华医学瑰宝,它与食物同源,无毒副作用,无耐药性,不易残留。研究表明,中草药含有多糖、生物碱、萜类、酚类等多种活性抗菌物质,也富含氨基酸、维生素、微量元素等营养物质,可以同时用作药物和营养物质,提高动物免疫功能,促进生长发育,是极好的饲用抗生素替代物。但是中草药的药效需要经过一定的加工手段才能释放出来,在畜牧业中不便使用。为了便于动物吸收利用,提高中草药活性成分的生物利用率,需要对其进行有效处理。
目前,对中药材进行微生物发酵改性研究是天然药物化学研究的新领域。微生物具有十分丰富的酶系统,中药微生物发酵过程中,由于微生物的代谢作用,常常伴随着大量的生物转化反应,会引起中药有效活性成分的变化或药效、药性的改变。同时某些中药也有一定的毒性,使用微生物发酵可以进行减毒炮制,实现中药的减毒增效。此外,很多中草药由于自身结构特点,不能被机体直接吸收利用,而是作为前体物质,经肠道微生物处理后,才能发挥药效。在畜牧业中使用微生物发酵中草药是对传统养殖技术的革新。利用微生物发酵中草药,可以提高中草药的药性药效,同时益生菌发酵过程中也会转化生成酶、抗菌肽、有机酸等多种功能性物质,可以改善动物肠道健康,提高代谢功能。微生物与中草药的结合相得益彰,获得一种天然的饲料添加剂,使中药使用效果更显著,产品科技含量大幅提升,同时减少了抗生素等化学物质的使用,解决了药残和耐药性的问题,具有深远的意义和广阔的应用前景。但是,如何选用合适的微生物对中药草进行发酵一直是难以解决的技术问题。
发明内容
本发明的目的在于提供一种适用于猪的复方中草药益生菌发酵的饲料添加剂,再将中草药合理配伍后,经过复合益生菌的深度发酵,有效提高中草药中有效成分的生物利用率,获得一种高效代替抗生素的新型饲料添加剂,可以有效增强猪的机体免疫力,解决了现在饲料添加剂领域中抗生素残留、细菌耐药性增强的问题以及中草药有效成分提取处理的技术问题。
本发明所提供的饲料添加剂,是将黄芪、白术、茯苓和甘草使用枯草芽孢杆菌、产朊假丝酵母、植物乳杆菌和屎肠球菌进行发酵制备的;
所述的黄芪、白术、茯苓和甘草的质量份数比为4~8:2~4:茯苓4~8:甘草1~4;
本发明所提供的饲料添加剂,其制备方法如下:
1)将黄芪、白术、茯苓和甘草粉碎获得中草药粉;
2)将中草药粉中添加营养物质和水,搅拌混合均匀,获得含水量为35~45%的发酵培养基;
3)在步骤2)中制备的发酵培养基中添加活化的枯草芽孢杆菌、产朊假丝酵母、植物乳杆菌和屎肠球菌进行发酵
所述的发酵,是在30~40℃进行发酵;
所述的2)中的营养物质,以添加营养物的质量占发酵培养基总质量的百分比计,分别为:麸皮5.0~15.0%、玉米粉15.0~20.0%、糖蜜2.0~5.0%、硫酸铵0.02%、氯化钙0.01%、磷酸氢二钾0.02%。
本发明所提供的饲料添加剂用于添加到猪饲料中。
本发明的有益效果如下:
(1)本发明以动物营养学及中兽医学理论为基础,精心挑选黄芪、白术、茯苓、甘草等多种可饲用天然植物中草药,经过合理配伍,具有利湿补气、清热解毒、健脾保肝、消食健胃、刺激肠道蠕动增强胃肠动力的作用,可以有效增强猪的脾胃消化吸收功能,提高食欲,促进生长发育,提高免疫力。本发明的饲料添加剂的应用可以降低仔猪腹泻率,克服因断奶、饲料改变、生长环境变化而产生的应激现象,改善母猪亚健康问题,去除泪斑死皮,解决母猪便秘问题,改善母猪的繁殖性能。
(2)本发明选用包括枯草芽孢杆菌、产朊假丝酵母、植物乳杆菌、屎肠球菌在内的复合益生菌对中草药进行发酵精制,提高了天然中草药的药效,提高了活性物质的生物效价,可有效提高猪对中草药中活性成分的吸收利用率,从而迅速发挥疗效。发酵后的中草药中大大提高黄芪多糖、黄酮、皂苷的含量。
(3)本发明使用的复合益生菌在发酵过程中可产生大量有机酸、酶、维生素、抗菌肽等代谢产物,可以补充营养物质,调节肠道微生态平衡,改善肠道环境,促进动物消化吸收,增强动物免疫力。此外,微生物发酵可以产生一些风味物质,降低中草药的苦味,提高中草药饲料添加剂的适口性。

具体实施方式
以下通过具体实施例来进一步描述本发明,但不应以此限定本发明的保护范围。
实施例1:猪用复方中草药益生菌发酵
猪用复方中草药益生菌发酵饲料添加剂,具体制备步骤如下:
(1)按重量配比准确称量中草药,黄芪1600g、白术800g、茯苓800g、甘草400g;
(2)中草药的预处理:将按步骤(1)称取的中草药分别粉碎,过30目筛,混合均匀,获得复方中草药粉,备用;
(3)配置15%的糖蜜水,按10%比例加入益生菌组合A(含枯草芽孢杆菌、产朊假丝酵母),按0.2%的比例加入益生菌组合B(含植物乳杆菌、屎肠球菌),搅拌均匀,维持水温37℃,活化1h,获得复合益生菌种子液。
(4)往步骤(2)制得的粉碎中草药中添加麸皮1000g、玉米粉2000g、糖蜜495g、硫酸铵2g、氯化钙1g、磷酸氢二钾2g,培养基自然pH值,添加水分2.97kg使物料含水量达到41%,混合机中搅拌混合均匀,获得发酵培养基;
(5)往步骤(4)制备得到的发酵培养基中按1%的比例缓慢倒入按步骤(3)活化好的复合益生菌种子液,并于混合机中混合均匀。
(6)将步骤(5)所得的中草药与益生菌发酵物料分装入呼吸发酵袋中,密封发酵,控制环境温度在37℃,发酵时间72h,即获得复合中草药发酵饲料添加剂10.07kg。
实施例2:
一种猪用复方中草药益生菌发酵饲料添加剂,具体制备步骤如下:
(1)按重量配比准确称量中草药,黄芪1200g、白术800g、茯苓1200g、甘草400g;
(2)中草药的预处理:将按步骤(1)称取的中草药分别粉碎,过30目筛,混合均匀,获得复方中草药粉,备用;
(3)配置15%的糖蜜水,按10%比例加入益生菌组合A(含枯草芽孢杆菌、产朊假丝酵母),按0.2%的比例加入益生菌组合B(含植物乳杆菌、屎肠球菌),搅拌均匀,维持水温37℃,活化1h,获得复合益生菌种子液。
(4)往步骤(2)制得的粉碎中草药中添加麸皮1200g、玉米粉2000g、糖蜜495g、硫酸铵2g、氯化钙1g、磷酸氢二钾2g,培养基自然pH值,添加水分2.91kg使物料含水量达到40%,混合机中搅拌混合均匀,获得发酵培养基;
(5)往步骤(4)制备得到的发酵培养基中按2%的比例缓慢倒入按步骤(3)活化好的复合益生菌种子液,并于混合机中混合均匀。
(6)将步骤(5)所得的中草药与益生菌发酵物料分装入呼吸发酵袋中,密封发酵,控制环境温度在37℃,发酵时间72h,即获得复合中草药发酵饲料添加剂10.2kg。
实施例3:
猪用复方中草药益生菌发酵饲料添加剂,具体制备步骤如下:
(1)按重量配比准确称量中草药,黄芪1600g、白术400g、茯苓1200g、甘草600g;
(2)中草药的预处理:将按步骤(1)称取的中草药分别粉碎,过30目筛,混合均匀,获得复方中草药粉,备用;
(3)配置12%的糖蜜水,按10%比例加入益生菌组合A(含枯草芽孢杆菌、产朊假丝酵母),按0.2%的比例加入益生菌组合B(含植物乳杆菌、屎肠球菌),搅拌均匀,维持水温37℃,活化1h,获得复合益生菌种子液。
(4)往步骤(2)制得的粉碎中草药中添加麸皮800g、玉米粉2000g、糖蜜395g、硫酸铵2g、氯化钙1g、磷酸氢二钾2g,培养基自然pH值,添加水分3.07kg使物料含水量达到41%,混合机中搅拌混合均匀,获得发酵培养基;
(5)往步骤(4)制备得到的发酵培养基中按1%的比例缓慢倒入按步骤(3)活化好的复合益生菌种子液,并于混合机中混合均匀。
(6)将步骤(5)所得的中草药与益生菌发酵物料分装入呼吸发酵袋中,密封发酵,控制环境温度在37℃,发酵时间72h,即获得复合中草药发酵饲料添加剂10.07kg。
实施例4:复合中草药发酵饲料添加剂的检测:
(一)成分测定
将实施例1~3所制备的中草药发酵饲料样品进行营养成分测定,结果如下表1所示:

本发明的中草药发酵饲料添加剂,以中草药合理配伍,通过益生菌发酵,营养均衡并且含有丰富的有机酸、活性小肽和益生菌活菌。
(二)活性成分改变
中草药的活性药用成分非常复杂,主要为多糖类、皂苷类、黄酮类、生物碱、有机酸、挥发油以及矿物元素等,这些活性成分能有效抗菌抗病毒,增强机体免疫功能。研究发现黄芪的主要成分为黄芪多糖、黄酮和皂苷,甘草的有效成分有甘草皂苷、黄酮等,白术的有效成分有白术多糖、黄酮、氨基酸、挥发油等,茯苓的有效成分为茯苓多糖、茯苓糖、茯苓酸等。
粗多糖的检测方法采用苯酚硫酸法,黄酮的检测方法参考行业标准SN/T 4592-2016“出口食品中总黄酮的测定”,皂苷的检测方法采用香草醛高氯酸分光光度法,参考行业标准T/AHFIA 004-2018“食品中总皂苷含量的测定分光光度法”。以未经发酵的中草药组方为对照,分别测定发酵前后中药组方中粗多糖、总黄酮、总皂苷的含量,分析发酵前后中药中主要活性成分的变化,结果如下表3。

实验结果表明,实施例1发酵后中草药中的粗多糖、总黄酮、总皂苷含量分别比发酵前高34.9%、39.3%、20.2%;实施例2发酵后中草药中的粗多糖、总黄酮、总皂苷含量分别比发酵前高38.8%、35.7%、25.4%;实施例3发酵后中草药中的粗多糖、总黄酮、总皂苷含量分别比发酵前高44.4%、38.2%、34.6%。由此可知,中草药经复合益生菌发酵后,其中可溶性有效活性成分的含量显著提高。
(三)抑菌实验
将实施例1~3所制备的中草药发酵饲料样品进行抑菌效果测定,以未经益生菌发酵的中草药组方为对照,各称取5g样品,加入50mL无菌水,室温提取1h,然后以牛津杯法进行体外抑菌试验(图1),结果如下表4。

注:抑菌圈直径>20mm为极敏,抑菌圈直径15~20mm为高敏,抑菌圈直径10~15mm为中敏,抑菌圈直径<10mm为低敏,无抑菌圈的为不敏感。
由以上结果可知,与未经发酵的中草药组方相比,复合益生菌发酵处理过的中草药对大肠杆菌、沙门氏菌、金黄色葡萄球菌的抑制效果有了显著的提高。
(四)保育猪养殖实验
以实施例1制得的产品进行保育猪养殖实验,实验选择体重15kg左右的三元商品猪180头,按3×3分组,每个重复20头猪,实验期共35天。对照组猪采用基础日粮饲喂,实验1组在基础日粮中添加2%发酵中草药饲料添加剂,实验2组在基础日粮中添加5%发酵中草药饲料添加剂,生猪自由采食。实验期内每天观察并记录猪的采食和生长情况,实验结束时称体重,计算每个实验组猪的日增重、日采食和料肉比。

与对照组相比,实验1组保育猪的日增重提高6.12%,料肉比降低3.89%(图2)。实验结果表明,本发明制得的饲料添加剂能有效增加保育猪的日增重和降低料肉比,能促进猪的生长,提高饲料利用率。
(五)断奶仔猪养殖实验
以实施例1制得的产品进行断奶仔猪养殖实验,实验选择体重10kg左右的三元商品猪断奶仔猪54头,按3×3分组,每个重复6头猪,公母各半。对照组猪采用基础日粮饲喂,实验1组在基础日粮中添加2%发酵中草药饲料添加剂,实验2组在基础日粮中添加5%发酵中草药饲料添加剂,乳猪自由饮水和采食。实验期内每天观察并记录乳猪的采食和健康状况,实验结束时称体重,对数据进行统计分析。

由统计结果可知,两个实验组的日增重和日采食量均高于对照组,两个实验组的仔猪腹泻率也明显低于对照组(图3)。结果表明,本发明制得的饲料添加剂能有效提高仔猪生长性能,降低腹泻率,提高养殖效益。
Claims (7)
1.一种饲料添加剂,其特征在于,所述的饲料添加剂是将黄芪、白术、茯苓和甘草使用枯草芽孢杆菌、产朊假丝酵母、植物乳杆菌和屎肠球菌进行发酵制备的。
2.如权利要求1所述的饲料添加剂,其特征在于,所述的黄芪、白术、茯苓和甘草的质量份数比为4~8:2~4:4~8:1~4。
3.权利要求1或2所述的饲料添加剂的制备方法,其特征在于,所述的方法的步骤如下:
1)将黄芪、白术、茯苓和甘草粉碎获得中草药粉;
2)将中草药粉中添加营养物质和水,搅拌混合均匀,获得含水量为35~45%的发酵培养基;
3)在步骤2)中制备的发酵培养基中添加活化的枯草芽孢杆菌、产朊假丝酵母、植物乳杆菌和屎肠球菌进行发酵。
4.如权利要求3所述的制备方法,其特征在于,所述的2)中的营养物质,以添加营养物的质量占发酵培养基总质量的百分比计,分别为:麸皮5.0~15.0%、玉米粉15.0~20.0%、糖蜜2.0~5.0%、硫酸铵0.02%、氯化钙0.01%、磷酸氢二钾0.02%。
5.如权利要求3所述的制备方法,其特征在于,所述的3)中的发酵,是在30~40℃进行发酵。
6.权利要求1或2所述的饲料添加剂在制备猪饲料中的应用。
7.一种猪饲料,其特征在于,所述的猪饲料中添加有权利要求1或2所述的饲料添加剂。
