技术领域
本发明涉及饲料添加剂技术领域,具体涉及一种畜用益生菌发酵中药饲料添加剂及其制备方法与应用。
背景技术
我国是世界上最大的禽养殖国,畜禽养殖业的健康可持续发展关乎国计民生。随着技术发展,高密度、大面积的养殖导致家禽病害频发,抗生素滥用和乱用的问题,并因此造成耐药,导致更长的治疗过程、更强的致病原和更差的治疗效果,形成了恶性循环。
中药是我国传统医学的中药组成部分,近年来,畜禽养殖领域也得到了广泛应用,中药成分在饲料中的具有抗菌、提高动物免疫能力、促进生长等作用,与此同时,与其他化学合成类药物相比,中药具有环境友好、生物安全、无依赖性的特点,可以有效的减少化学合成类药物和添加剂的使用。
采用益生菌发酵中药制备畜用饲料可以有效解决上述问题。一方面,有益微生物对中药进行发酵,使中药的有效活性成分充分发挥作用;另一方面,益生菌产生多种酶和抗菌肽等次级代谢产物可以有助于禽对饲料中营养成分的消化、吸收和代谢,有效促进禽的生长、增重,提高饲料转换率,维持禽肠道微生物的稳态,进而提高家禽家畜的免疫能力和抗病能力。
发明内容
本发明的目的是提供一种畜用益生菌发酵中药饲料,以替代化学合成类药物在畜养殖中的使用,同时提供所述益生菌发酵中药饲料的制备和饲喂方法。
本发明为解决其技术问题,采用得技术方案是:一种畜用益生菌中药发酵饲料,所述中药发酵饲料有效成分包括中药、混合益生菌剂和辅料;
所述的中药包括:黄芪、党参、陈皮、板蓝根、金银花、茯苓、葛根、蒲公英、白芷;
所述混合益生菌包括:动物双歧杆菌、粪肠球菌、解淀粉芽孢杆菌、植物乳杆菌、酿酒酵母、产朊假丝酵母、纳豆芽孢杆菌;
所述辅料包括:玉米粉、豆粕、鱼粉。
一种畜用益生菌发酵中药饲料的制备和饲喂方法,包括步骤如下:
(1)配制所述中药:黄芪、党参、陈皮、板蓝根、金银花、茯苓、葛根、蒲公英、白芷进行干燥、粉碎,过40目筛,按重量比称取各组分,搅拌均匀,备用;
所述各中药占中药组分总量的重量比为:
黄芪4-5份、党参3.5-5.5份、陈皮1-1.5份、板蓝根2-3份、金银花2-3份、茯苓4-5份、葛根2.5-3.5份、蒲公英2.5-4.5份、白芷1-2份;
(2)将所述益生菌:动物双歧杆菌、粪肠球菌、解淀粉芽孢杆菌、植物乳杆菌、酿酒酵母、产朊假丝酵母、纳豆芽孢杆菌接种到相应培养基中,进行单菌种菌株富集培养,使得每种益生菌的菌浓度为1×107CFU/ml。从而获得各益生菌的单菌种一级种子液;
(3)将上述各种益生菌的单菌种一级种子液按重量比混合,获得多菌种混合二级种子液;
所述各益生菌单菌种一级种子液混合重量比为:
动物双歧杆菌1.5-2.5份、粪肠球菌2-3份解淀粉芽孢杆菌0.5-1份、植物乳杆菌2-3份、酿酒酵母1-3份、产朊假丝酵母1-3份、纳豆芽孢杆菌0.5-1份;
(4)将上述多菌种混合二级种子液按1:50接种到含有(1)中组分的中药发酵基质中进行中药发酵,在发酵罐中,设置搅拌转速为160-180rpm,温度为30-37℃,发酵24h,从而获得多菌种混合中药发酵液;
所述中药发酵基质中包括10%的中药组分,5%的红糖,0.5%氯化钠,0.2%的磷酸氢二钾,1%的蛋白胨和余量的水。
(5)将所述发酵辅料:玉米粉、豆粕和鱼粉按重量比称取,在121℃条件下灭菌15-20分钟后,将(4)中的混合益生菌中药发酵液按重量比(30-40):(70-80)与辅料混合进行二次发酵,发酵温度为室温,发酵48h,然后得到畜用益生菌发酵中药饲料;
所述各发酵辅料重量比为:
玉米粉10-15份、豆粕10-15份、鱼粉1-2份;
(6)饲喂,与家畜或家禽的常用预混料混合均匀饲喂,按饲料总重量的6%-8%混拌添加发酵饲料。
本发明还提供了上述饲料在家畜养殖中的应用
本发明提供的技术方案具有以下优点:
1.本发明提供的的益生菌发酵中药饲料,采用益生菌发酵中药和家畜饲料常用辅料作为原料,充分保障了饲料原料的安全性,所选用的中药具有调节免疫平衡、抗菌消炎、调节消化能力等作用;
2.本发明提供的益生菌发酵中药饲料,将中药干燥粉碎后,与富集后的混合益生菌进行发酵,缩短了中药的发酵周期的同时使中药的活性成分充分发挥作用,
3.本发明提供的益生菌中药发酵饲料可以提高饲料的利用率,提高采食量,促进动物的营养吸收。益生菌产生的淀粉酶、蛋白酶、纤维素酶、脂肪酶等消化酶有助于将原料中的淀粉、蛋白制等大分子分解,降低养殖动物的消化负担,提高饲料的吸收利用率;
4.本发明提供的益生菌发酵中药饲料,可以调节动物肠道菌群平衡,提高动物机体免疫能力和抗病能力。粪肠球菌等是健康畜禽动物肠道内的优势菌,可以有效抑制致病菌如大肠杆菌、伤寒沙门氏菌和痢疾杆菌的繁殖,同时可以促进有益菌双歧杆菌、乳酸菌和拟杆菌的增殖。部分益生菌可以产生抗菌、抑菌的功能肽等活性物质,这些物质被动物吸收利用可以有效地提高动物的抗病能力和免疫能力。
具体实施方式
以下将以具体优选的实施例对本发明进行进一步的描述。
实施例一
(1)配制中药粉末,将黄芪4份、党参3.5份、陈皮1份、板蓝根2份、金银花2份、茯苓4份、葛根2.5份、蒲公英2.5份、白芷1份,进行干燥、粉碎,过40目筛,得到中药粉;
(2)制备益生菌单菌种一级子液,将动物双歧杆菌、粪肠球菌、解淀粉芽孢杆菌、植物乳杆菌、酿酒酵母、产朊假丝酵母和纳豆芽孢杆菌分别接种到各自的培养基中,培养获得单菌的一级种子液。
具体的,各菌种培养基如下:
解淀粉芽孢杆菌和纳豆芽孢杆菌,分别分离纯化后,接种至牛肉膏蛋白胨液体培养基中,培养基包括:0.05%牛肉膏、1.5%蛋白胨、0.5%氯化钠、0.2%葡萄糖和余量的水,培养基pH值为7.0-7.2,接种量按照体积比1:100接种,37℃培养12-18h,得到菌浓度为1×107CFU/ml解淀粉芽孢杆菌和纳豆芽孢杆菌的一级种子液;
动物双歧杆菌、植物乳杆菌和粪肠球菌,分别纯化后,接种至MRS液体培养基中,培养基包括:1%蛋白胨、1%牛肉膏、0.5%酵母膏、0.2%K2HPO4·3H2O、0.5%乙酸钠、2%葡萄糖、0.1%吐温80、0.2%柠檬酸二铵、0.058%MgSO4·7H2O、0.025%MnSO4·4H2O和余量的水,培养基pH为6.2-6.4,接种量按照体积比1:100接种,37℃培养12-18h,得到菌浓度为1×107CFU/ml动物双歧杆菌、植物乳杆菌和粪肠球菌的一级种子液;
酿酒酵母和产朊假丝酵母,分别纯化后,接种至PDA液体培养基中,培养基包括:20%土豆、2%葡萄糖、0.3%KH2PO4、0.15%MgSO4和余量的水,接种量按照体积比1:100接种,37℃培养12-18h,得到菌浓度为1×107CFU/ml的酿酒酵母和产朊假丝酵母的一级种子液。
(3)配制多菌种混合二级种子液,将上述单菌种一级种子液按照比例混合,得到多菌种混合二级种子液。
具体的,各单菌种子液占混菌种子液总量按重量比为:
动物双歧杆菌1.5份、粪肠球菌2份、解淀粉芽孢杆菌0.5份、植物乳杆菌2份、酿酒酵母1份、产朊假丝酵母1份、纳豆芽孢杆菌0.5份;
(4)将多菌种混合二级种子液接种至中药发酵基质中进行中药发酵,获得多菌种混合中药发酵液;
具体的,多菌种混合二级种子液接种量为1:50,所述中药发酵基质包括10%的中药组分,5%的红糖,0.5%氯化钠,0.2%的磷酸氢二钾,1%的蛋白胨和余量的水;
进一步的,所述中药组分为(1)中配制的混合中药粉末;
更进一步的,将中药发酵基质中各组分按配比加入发酵罐中,121℃对发酵罐灭菌30min,以有效杀灭发酵基质中的有害菌,再冷却到30℃,设置搅拌转速为160-180rpm,发酵24h,从而获得多菌种混合中药发酵液;
(5)与发酵辅料混合进行二次发酵,获得畜用益生菌发酵中药饲料;
具体的,将所述发酵辅料:玉米粉、豆粕、鱼粉按重量比称取,加入发酵罐中,在121℃条件下灭菌30min,冷却至室温;
进一步的,所述各发酵辅料重量比为:玉米粉10份、豆粕15份、鱼粉1份;
更进一步的,将(4)中的混合益生菌中药发酵液按重量比30:80与发酵辅料混合进行二次发酵,设置搅拌转速为120rpm,发酵温度为室温,发酵48h,然后得到畜用益生菌发酵中药饲料;
(6)饲喂,畜用益生菌中药发酵饲料与家禽的常用预混料混合均匀饲喂,按饲料总重量的4%混拌添加发酵饲料。
实施例二
(1)配制中药粉末,将黄芪5份、党参5.5份、陈皮1.5份、板蓝根3份、金银花3份、茯苓5份、葛根3.5份、蒲公英4.5份、白芷2份进行干燥、粉碎,过40目筛,得到中药粉;
(2)制备益生菌单菌种一级子液,将动物双歧杆菌、粪肠球菌、解淀粉芽孢杆菌、植物乳杆菌、酿酒酵母、产朊假丝酵母和纳豆芽孢杆菌分别接种到各自的培养基中,培养获得单菌的一级种子液。
具体的,各菌种培养基如下:
解淀粉芽孢杆菌和纳豆芽孢杆菌,分别分离纯化后,接种至牛肉膏蛋白胨液体培养基中,培养基包括:0.05%牛肉膏、1.5%蛋白胨、0.5%氯化钠、0.2%葡萄糖和余量的水,培养基pH值为7.0-7.2,接种量按照体积比1:100接种,37℃培养12-18h,得到菌浓度为1×107CFU/ml解淀粉芽孢杆菌和纳豆芽孢杆菌的一级种子液;
动物双歧杆菌、植物乳杆菌和粪肠球菌,分别纯化后,接种至MRS液体培养基中,培养基包括:1%蛋白胨、1%牛肉膏、0.5%酵母膏、0.2%K2HPO4·3H2O、0.5%乙酸钠、2%葡萄糖、0.1%吐温80、0.2%柠檬酸二铵、0.058%MgSO4·7H2O、0.025%MnSO4·4H2O和余量的水,培养基pH为6.2-6.4,接种量按照体积比1:100接种,37℃培养12-18h,得到菌浓度为1×107CFU/ml动物双歧杆菌、植物乳杆菌和粪肠球菌的一级种子液;
酿酒酵母和产朊假丝酵母,分别纯化后,接种至PDA液体培养基中,培养基包括:20%土豆、2%葡萄糖、0.3%KH2PO4、0.15%MgSO4和余量的水,接种量按照体积比1:100接种,37℃培养12-18h,得到菌浓度为1×107CFU/ml的酿酒酵母和产朊假丝酵母的一级种子液。
(3)配制多菌种混合二级种子液,将上述单菌种一级种子液按照比例混合,得到多菌种混合二级种子液。
具体的,各单菌种子液占混菌种子液总量按重量比为:
动物双歧杆菌2.5份、粪肠球菌3份、解淀粉芽孢杆菌1份、植物乳杆菌3份、酿酒酵母3份、产朊假丝酵母3份、纳豆芽孢杆菌1份;
(4)将多菌种混合二级种子液接种至中药发酵基质中进行中药发酵,获得多菌种混合中药发酵液;
具体的,多菌种混合二级种子液接种量为1:50,所述中药发酵基质包括10%的中药组分,5%的红糖,0.5%氯化钠,0.2%的磷酸氢二钾,1%的蛋白胨和余量的水;
进一步的,所述中药组分为(1)中配制的混合中药粉末;
更进一步的,将中药发酵基质中各组分按配比加入发酵罐中,121℃对发酵罐灭菌30min,以有效杀灭发酵基质中的有害菌,再冷却到30℃,设置搅拌转速为160-180rpm,发酵24h,从而获得多菌种混合中药发酵液;
(5)与发酵辅料混合进行二次发酵,获得畜用益生菌发酵中药饲料;
具体的,将所述发酵辅料:玉米粉、豆粕、鱼粉按重量比称取,加入发酵罐中,在121℃条件下灭菌30min,冷却至室温;
进一步的,所述各发酵辅料重量比为:玉米粉15份、豆粕10份、鱼粉2份;
更进一步的,将(4)中的混合益生菌中药发酵液按重量比40:70与发酵辅料混合进行二次发酵,设置搅拌转速为120rpm,发酵温度为室温,发酵48h,然后得到畜用益生菌发酵中药饲料;
(6)饲喂,畜用益生菌中药发酵饲料与家畜的常用预混料混合均匀饲喂,按饲料总重量的8%混拌添加发酵饲料。
上述技术方案中,本发明所述的中药组分的各成分的功效为:
黄芪:味甘性微温,含有黄芪多糖、胆碱和多种维生素,具有调节机体免疫促进抗体形成,提高诱生干扰素,增强体液免疫的作用;党参:味甘性平,有补中益气、止渴、健脾益肺,含有活性成分党参皂苷和党参多糖,对细胞免疫、体液免疫和非特异性免疫的调节功能有增强作用,同时党参多糖能增加胃黏膜、胃壁厚度,促进肠绒毛生长,提高机体消化能力;陈皮:味苦辛性温,理气健脾,含有活性成分单萜烯挥发油具有抗氧化、抗菌等作用,类柠檬苦素具有促进消化吸收的作用;板蓝根:味苦性寒,可清热解毒,具有抗菌抗病毒作用,对畜禽常见致病菌金黄色葡萄球菌、大肠杆菌、肠炎杆菌等都有抑制作用;金银花:性甘寒气芳香,有清热解毒、抗菌消炎、保肝利胆等功效,含有挥发油、黄酮类、有机酸和三萜类等有效成分,具有抗病原微生物(金黄色葡萄球菌、绿脓杆菌、溶血性链球菌等)的作用,并可有效增强白细胞、炎症细胞的吞噬能力从而起到增强免疫力的作用;茯苓:味甘淡性平,所含茯苓酸具有增强免疫力、抗肿瘤以及镇静、降血糖等的作用。可松弛消化道平滑肌,抑制胃酸分泌,抗菌等功效;葛根:味甘辛性凉,可生津止渴,富含异黄酮类、葛根皂苷类以及多种生物碱类活性成分,具有较强的抗菌活性,尤以大肠杆菌和金黄色葡萄球菌,具有雌激素样作用,对哺乳期家畜具有一定的保健作用,同时具有显著改善糖和脂质代谢的作用;蒲公英:味甘性平,可清热解毒、消肿散结,活性成分蒲公英甾醇、萜类、胆碱、香豆素等,具有良好的广谱抗菌、抗自由基、抗病毒、抗感染和增强免疫力等功效;白芷:味辛性温,有效成分主要有香豆素和挥发油,并含有多种微量元素,具有有效的抗菌消炎及清除氧化自由基的能力。由此可见,本发明所提供的中药组分不仅有益于家畜的生长,而且能够增强家畜的免疫能力和抗病能力,提高家畜养殖过程中的存活率。
中试养殖试验
在家猪养殖厂,选择健康仔猪180头,60头为一组,分别为试验组一、试验组二和对照组,从仔猪断奶后开始间断饲喂,包括保育期前期饲喂34天,及出栏前育肥期饲喂60天,试验组饲喂添加实施例一和实施例二中的中药发酵饲料,对照组饲喂传统养殖无添加饲料,分别记录家猪的死亡量、始末体重和饲料投喂量。
经过保育期的34天饲喂,饲喂添加畜用益生菌发酵中药饲料可以显著提高仔猪的存活率,日增重较对照组相比也有显著提升;在育肥期饲喂60天后发现,益生菌发酵中药饲料的添加可以显著提高家猪的增重量;180天的养殖试验后发现,与对照组相比,饲喂了添加益生菌发酵中药饲料的家猪的死亡率最高降低6.6%,同时料肉比降低约10%。证明本发明提供的一种畜用益生菌发酵中药饲料可以显著提高家猪的存活率和出栏率,且对育肥猪的生长增重有显著的促进作用。

Claims (7)
1.一种畜用益生菌发酵中药饲料,其特征在于,以质量分数计,组份包括包括中药成分、混合益生菌剂和发酵辅料;
所述的中药成分包括:黄芪4-5份、党参3.5-5.5份、陈皮1-1.5份、板蓝根2-3份、金银花2-3份、茯苓4-5份、葛根2.5-3.5份、蒲公英2.5-4.5份、白芷1-2份。
2.如权利要求1所述的畜用益生菌发酵中药饲料,其特征在于,所述混合益生菌包括:动物双歧杆菌1.5-2.5份、粪肠球菌2-3份解淀粉芽孢杆菌0.5-1份、植物乳杆菌2-3份、酿酒酵母1-3份、产朊假丝酵母1-3份、纳豆芽孢杆菌0.5-1份。
3.如权利要求1所述的畜用益生菌发酵中药饲料,其特征在于,所述辅料包括:玉米粉10-15份、豆粕10-15份、鱼粉1-2份。
4.一种权利要求1-3所述的畜用益生菌发酵中药饲料的制备方法,其特征在于,包括步骤如下:
(1)配制所述中药成分:黄芪、党参、陈皮、板蓝根、金银花、茯苓、葛根、蒲公英、白芷,分别进行干燥、粉碎,分别过40目筛,按重量比称取各组分,搅拌均匀,备用;所述的中药成分重量份数:黄芪4-5份、党参3.5-5.5份、陈皮1-1.5份、板蓝根2-3份、金银花2-3份、茯苓4-5份、葛根2.5-3.5份、蒲公英2.5-4.5份、白芷1-2份;
(2)将所述益生菌:动物双歧杆菌、粪肠球菌、解淀粉芽孢杆菌、植物乳杆菌、酿酒酵母、产朊假丝酵母、纳豆芽孢杆菌接种到相应培养基中,进行单菌种菌株富集培养,使得每种益生菌的菌浓度为1×107CFU/ml;从而获得各益生菌的单菌种一级种子液;所述混合益生菌包括:动物双歧杆菌1.5-2.5份、粪肠球菌2-3份解淀粉芽孢杆菌0.5-1份、植物乳杆菌2-3份、酿酒酵母1-3份、产朊假丝酵母1-3份、纳豆芽孢杆菌0.5-1份;
(3)将上述各种益生菌的单菌种一级种子液按上述份数混合,获得多菌种混合二级种子液;
(4)将上述多菌种混合二级种子液按1:50接种到含有(1)中组分的中药发酵基质中进行中药发酵,在发酵罐中,设置搅拌转速为160-180rpm,温度为30-37℃,发酵24h,从而获得多菌种混合中药发酵液;
所述中药发酵基质中包括10%的中药组分,5%的红糖,0.5%氯化钠,0.2%的磷酸氢二钾,1%的蛋白胨和余量的水;
(5)将所述发酵辅料:玉米粉、豆粕、鱼粉按重量比称取,在121℃条件下灭菌15-20分钟后,将(4)中的混合益生菌中药发酵液按重量比(30-40):(70-80)与辅料混合进行二次发酵,发酵温度为室温,发酵48h,然后得到畜用益生菌发酵中药饲料;所述辅料包括:玉米粉10-15份、豆粕10-15份、鱼粉1-2份。
5.按照权利要求4的制备方法,其特征在于:
所述动物双歧杆菌、粪肠球菌、解淀粉芽孢杆菌、植物乳杆菌、酿酒酵母、产朊假丝酵母、纳豆芽孢杆菌的相应培养基为:
解淀粉芽孢杆菌和纳豆芽孢杆菌,分别分离纯化后,接种至牛肉膏蛋白胨液体培养基中,培养基包括:0.05%牛肉膏、1.5%蛋白胨、0.5%氯化钠、0.2%葡萄糖和余量的水,培养基pH值为7.0-7.2,接种量按照体积比1:100接种,37℃培养12-18h,得到菌浓度为1×107CFU/ml解淀粉芽孢杆菌和纳豆芽孢杆菌的一级种子液;
动物双歧杆菌、植物乳杆菌和粪肠球菌,分别纯化后,接种至MRS液体培养基中,培养基包括:1%蛋白胨、1%牛肉膏、0.5%酵母膏、0.2%K2HPO4·3H2O、0.5%乙酸钠、2%葡萄糖、0.1%吐温80、0.2%柠檬酸二铵、0.058%MgSO4·7H2O、0.025%MnSO4·4H2O和余量的水,培养基pH为6.2-6.4,接种量按照体积比1:100接种,37℃培养12-18h,得到菌浓度为1×107CFU/ml动物双歧杆菌、植物乳杆菌和粪肠球菌的一级种子液;
酿酒酵母和产朊假丝酵母,分别纯化后,接种至PDA液体培养基中,培养基包括:20%土豆、2%葡萄糖、0.3%KH2PO4、0.15%MgSO4和余量的水,接种量按照体积比1:100接种,37℃培养12-18h,得到菌浓度为1×107CFU/ml的酿酒酵母和产朊假丝酵母的一级种子液。
6.一种权利要求1所述的禽用益生菌发酵中药饲料的应用,其特征在于,所述的饲料应用在家畜或家禽的饲料中。
7.按照权利要求6所述的禽用益生菌发酵中药饲料的应用,其特征在于,按饲料总重量的4%-6%混拌添加禽用益生菌发酵中药饲料。
