The invention provides an anti-dandruff and anti-itching composition, which comprises the following components in parts by weight: 13g of mixed extract of rheum palmatum and curcuma longa, 3g of mint extract, 2g of dandelion extract, 9g of schizonepeta extract, 25g of propylene glycol, 15g of 1, 2-hexanediol and 33g of water.
The mixed extract of rheum palmatum and curcuma longa described in example 5 was prepared by the following steps in sequence:
(1) pretreatment of raw materials: respectively cleaning the rheum palmatum and the turmeric root, drying, crushing, sieving with a 80-mesh sieve, and uniformly mixing according to the mass ratio of 3:2 to obtain mixed powder;
(2) enzymolysis: adding the mixed powder in the step (1) into an aqueous solution according to the material-liquid ratio of 1g to 40mL, then adding alpha-amylase (the enzyme activity is 7000U/g) and papain (4 ten thousand U/g) which are 0.2% by weight of the mixed powder and have the mass ratio of 1:4, carrying out enzymolysis for 4 hours under the conditions that the pH value is 4.5 and the temperature is 40 ℃, and inactivating the enzyme to obtain an enzymolysis solution;
(3) high-pressure homogeneous extraction: adding an absolute ethanol solution with the volume 5 times that of the enzymolysis solution into the enzymolysis solution in the step (2), uniformly stirring, and then carrying out normal-temperature extraction by using a high-pressure homogeneous extractor, wherein the high-pressure homogeneous extraction time is 20min, and the high-pressure homogeneous extraction pressure is 60 Mpa.
(4) And (3) purification: filtering the extracting solution obtained in the step (3), concentrating under reduced pressure until no alcohol smell exists, then adsorbing the concentrated solution by using an H-20 type macroporous adsorption resin column, eluting by using water until the concentrated solution is colorless, then eluting by using an ethanol solution with the volume 3 times that of the column and 70 percent, collecting the eluent, filtering, and freeze-drying to obtain the mixed extract of the rheum palmatum and the turmeric root.
In the above examples 1-5, the mint extract was prepared by washing, drying and pulverizing mint, and mixing the following raw materials in a ratio of 1g: adding 60% ethanol into 40mL, heating, refluxing, extracting for 2h, cooling the extractive solution, filtering, and concentrating to dry.
The dandelion extract is prepared by cleaning, drying and crushing dandelion, and mixing the following raw materials in a material-liquid ratio of 1g: adding 70% ethanol into 35mL, heating, refluxing, extracting for 3h, cooling the extractive solution, filtering, and concentrating to dry.
The schizonepeta extract is prepared by cleaning, drying and crushing schizonepeta, and mixing the raw materials according to a feed-liquid ratio of 1g: adding 50% ethanol into 25mL of the mixture, heating and refluxing for extraction for 2.5h, cooling the extracting solution, filtering, and concentrating to dryness to obtain the product.
The preparation method of the anti-dandruff and anti-itching composition of any one of the above examples 1 to 5 is as follows:
(1) uniformly mixing phenoxyethanol and ethylhexyl glycerol with a small amount of polyhydric alcohol in advance for later use (when the phenoxyethanol and the ethylhexyl glycerol are not used, the step is omitted);
(2) heating water to 40-45 ℃, adding the rest of polyhydric alcohol, uniformly stirring, adding the mixture obtained in the step (1) and uniformly stirring, adding other materials, and uniformly stirring to obtain the product.
The composition can be used as active ingredient for preparing daily product for washing and caring hair. When the composition is used specifically, the composition prepared by the invention is directly added into a daily necessity matrix, and the addition amount is 1-8 wt%.
To demonstrate the effect of the present invention, comparative experimental examples of the present application are given below:
comparative example 1
A composition comprising the following components in amounts by weight: 5g of mixed extract of rheum palmatum and curcuma longa, 22g of mint extract, 25g of glycerol and 48g of water.
The mixed extract of rheum palmatum and curcuma longa in comparative example 1 was prepared in the same manner as in example 1, and the mint extract was prepared in the same manner as in example 1.
Only the dandelion extract was replaced with the same amount of mint extract as in example 1, and the other ingredients were the same.
Comparative example 2
A composition comprising the following components in amounts by weight: 6g of mint extract, 5g of dandelion extract, 16g of schizonepeta extract, 18g of glycerol, 15g of 1, 2-hexanediol, 0.45g of phenoxyethanol, 0.05g of ethylhexyl glycerol and 39.5g of water.
The preparation methods of the mint extract, dandelion extract and schizonepeta extract in comparative example 2 were the same as those in example 3.
Compared with example 3, the mixed extract of the rhubarb palmate and the turmeric root is only replaced by the same amount of schizonepeta extract, and other components are the same.
Comparative example 3
A composition comprising the following components in amounts by weight: 15g of mixed extract of rheum palmatum and curcuma longa, 6g of mint extract, 5g of dandelion extract, 1g of schizonepeta extract, 18g of glycerol, 15g of 1, 2-hexanediol, 0.45g of phenoxyethanol, 0.05g of ethylhexyl glycerol and 39.5g of water.
The mixed extract of rheum palmatum and curcuma longa in comparative example 3 is prepared by the following steps in sequence:
(1) pretreatment of raw materials: respectively cleaning the rheum palmatum and the turmeric root, drying, crushing, sieving with a 100-mesh sieve, and uniformly mixing according to a mass ratio of 1:2 to obtain mixed powder;
(2) enzymolysis: adding the mixed powder in the step (1) into an aqueous solution according to the material-liquid ratio of 1g to 20mL, then adding papain (4 ten thousand U/g) with the weight of 0.4% of the mixed powder, carrying out enzymolysis for 2h under the constant temperature condition that the pH value is 4.0 and the temperature is 45 ℃, and inactivating the enzyme to obtain an enzymolysis solution;
(3) high-pressure homogeneous extraction: adding an absolute ethyl alcohol solution with the volume 2 times that of the enzymolysis solution into the enzymolysis solution obtained in the step (2), uniformly stirring, and then carrying out normal-temperature extraction by using a high-pressure homogeneous extraction machine, wherein the high-pressure homogeneous extraction time is 30min, and the high-pressure homogeneous extraction pressure is 40 Mpa.
(4) And (3) purification: filtering the extracting solution obtained in the step (3), concentrating under reduced pressure until no alcohol smell exists, then adsorbing the concentrated solution by using an H-20 type macroporous adsorption resin column, eluting by using water until the concentrated solution is colorless, then eluting by using 4 times of column volume of 40% ethanol solution, collecting eluent, filtering, and freeze-drying to obtain the mixed extract of the rheum palmatum and the turmeric root.
In comparison with example 3, the composition had the same raw material composition. But the mixed extract of the rhubarb palmate and the turmeric root is only added with papain for enzymolysis in the step (2) during the preparation.
The preparation methods of the mint extract, dandelion extract and schizonepeta extract in comparative example 3 were the same as those in example 3.
Comparative example 4
A composition comprising the following components in amounts by weight: 5g of rheum palmatum extract, 10g of curcuma longa extract, 6g of mint extract, 5g of dandelion extract, 1g of schizonepeta extract, 18g of glycerol, 15g of 1, 2-hexanediol, 0.45g of phenoxyethanol, 0.05g of ethylhexyl glycerol and 39.5g of water.
The Rheum palmatum extract and Curcuma rhizome extract are commercially available. Wherein the Rheum palmatum extract is purchased from Beijing institute of biochemical and confirmatory technology, and the Curcuma rhizome extract is purchased from Guangzhou Norran Biotech, Inc.
The preparation methods of the mint extract, dandelion extract and schizonepeta extract in comparative example 4 were the same as those in example 3.
Comparative examples 1-4 were prepared in the same manner as examples 1-5 and were used in the same manner as examples 1-5.
In order to better illustrate the advantages of the present invention, the experimental results of the technical solution provided by the present invention are given below:
test 1: skin irritation test
The test method comprises the following steps: the chicken embryo chorioallantoic membrane blood vessel test verifies that the irritation is quantified according to the reaction of blood vessels, and the reaction of human skin to the irritation is objectively reflected. The test utilizes the characteristics of complete, obvious and transparent blood vessels of CAM (i.e. middle chorioallantoic membrane of fertilized chick embryo of 10-14 days old after hatching), the compositions of examples 1-5 diluted by 50 times are respectively contacted with 5 CAMs, after a period of action, the vascular injury of the CAMs is observed, the injury degree is scored, and the more serious the vascular injury is, the larger the score is. The judgment criteria are shown in Table 1, and the test results are shown in Table 2:

As can be seen from the results of the skin irritation test, the anti-dandruff anti-itch compositions prepared in examples 1 to 5 are safe to use, better mild, non-irritating, and superior to chemical anti-dandruff antipruritic agents.
And (3) testing 2: malassezia inhibition efficacy test
The experimental principle of the bacteriostatic zone is that the bacteriostatic agent is dissolved in a culture medium and continuously diffuses to form different concentration gradients so as to display the bacteriostatic action of the bacteriostatic agent, and whether the bacteriostatic ability of the bacteriostatic agent exists or not is judged according to the size of a bacteriostatic ring. If the diameter of the antibacterial ring is larger than 7mm, the antibacterial effect is achieved, and if the diameter of the antibacterial ring is smaller than or equal to 7mm, the antibacterial effect is not achieved.
(1) Test samples: the compositions of examples 1-5, comparative examples 1-4 were prepared as 2% sample solutions in sterile distilled water.
(2) The test method comprises the following steps: preparing activated Malassezia furfur into 106cfu/mL-107cfu/mL bacterial suspension, 0.3mL bacterial suspension is smeared on a Leeming with the length of 12cm&And (3) placing the bacterium-containing plate on a Notman culture medium plate for 10min, opening a plate cover in a sterile ultra-clean bench, and drying the plate. Dipping a small amount of sample to be detected on a sterile filter paper sheet with the diameter of 5mm by using sterile forceps, and smearing the sample uniformly as much as possible. 4 test filter paper samples and 1 negative control (containing no sample to be detected) sample are attached to each flat plate. After the sample is attached and placed, the sample wafer is lightly pressed by sterile forceps and is attached to the surface of the flat plate. And (4) covering a dish cover, placing the dish cover in a constant-temperature incubator, performing inverted culture for 7 days at 32 ℃, and measuring and recording the diameter of the inhibition zone by using a vernier caliper. The test results are shown in Table 3.

As can be seen from the experimental results in Table 3, the compositions of the examples of the invention have strong bacteriostatic effect on malassezia furfur, and the comparative example compositions have a smaller bacteriostatic circle on the malassezia furfur than the examples, which shows that the compositions of the invention generate synergistic effect and can effectively inhibit the malassezia furfur which causes dandruff. As shown by comparison between the example 5 and the example 4, when rheum palmatum L and turmeric root are extracted at a specific ratio and are subjected to enzymolysis by alpha-amylase and papain at a certain ratio, the bacteriostatic effect of the composition is better. As can be seen from the comparison between example 3 and comparative examples 3-4, the content of the active ingredients in the mixed extract of rhubarb palmate and turmeric root prepared by the method of the invention is higher, the ingredients are synergistic after the mixed extract is added into the composition, and the action of inhibiting the malassezia is also enhanced.
And (3) testing: human body trial evaluation
(1) Test samples: in order to evaluate the improvement effect of the composition on oil control and dandruff removal, the compositions of examples 1, 3, 4 and 5 and comparative examples 1 and 4 are respectively added into the shampoo base according to the mass fraction of 2% to prepare corresponding shampoos. The shampoo comprises the following components in percentage by mass:
phase A: adding water to 100%, guar hydroxypropyl trimethyl ammonium chloride 0.2%, and hydroxypropyl methyl cellulose 0.2%;
phase B: 10% of ammonium laureth sulfate and 10% of ammonium laureth sulfate;
and C phase: 3% of water// sodium lauroamphoacetate// sodium chloride, 2% of sodium lauroyl sarcosinate, 1% of ethylene glycol distearate, 0.2% of methylparaben, 0.1% of propylparaben and 0.05% of disodium EDTA;
phase D: dimethiconol// sodium dodecylbenzenesulfonate// water 2%, water// amodimethicone// stearyltrimethylammonium chloride 1.2%, fructose 1%, sodium PCA 0.8%, DMDM hydantoin 0.2%, PPG-15 stearyl ether 0.2%, pyridoxine hcl 0.1%, sodium chloride 0.1%, citric acid 0.1%, (daily) perfume 0.5%, examples 1, 3, 4, 5, composition 2% of comparative examples 1, 4;
phase E: 2% of acrylic acid (ester) copolymer.
The preparation method of the shampoo comprises the following steps:
putting water into a vacuum emulsifying pot, adding the phase A raw material, and stirring to fully dissolve the phase A raw material;
secondly, adding the phase B raw material into a vacuum emulsifying pot, heating to 60 ℃ while stirring, and stirring to fully dissolve the phase B raw material;
adding the E-phase raw material, and homogenizing for 20 minutes;
fourthly, when the temperature is raised to 80 ℃, adding the phase C raw material, fully stirring and dissolving the mixture evenly, and keeping the temperature for 30 minutes;
and fifthly, cooling to 40-45 ℃, adding the D-phase raw material, stirring for 30 minutes, discharging, standing, and checking to be qualified to obtain the product.
(2) The test method comprises the following steps: selecting 120 volunteers with more scurf and oily hair, wherein the age is 15-60 years, randomly dividing the volunteers into 6 groups of 20 persons each, respectively using the test samples to wash hair once in the morning every day, continuously washing hair for four weeks, observing the oily condition of the hair in the morning next day, and counting the number of the persons to table 5, wherein the evaluation standard of the effect is shown in table 4. Meanwhile, the volunteers were investigated for sensory evaluation of smell, vision and sensation of the shampoo, and the scores were scored at 1-10 points, with higher scores representing better sensory evaluation, and the scores were counted in table 4.

The experimental results in table 5 show that the shampoo prepared in the embodiment of the invention has good effects of controlling scalp oil secretion and dandruff, the total effective rate is more than 85%, the sensory score is more than 9 points, the effect is better than that of the comparative shampoo, and the composition has good effects of removing dandruff, relieving itching and controlling oil, and the feeling of experience of the public is good.
The above embodiments are preferred embodiments of the present invention, but the present invention is not limited to the above embodiments, and any other changes, modifications, substitutions, combinations, and simplifications which do not depart from the spirit and principle of the present invention should be construed as equivalents thereof, and all such changes, modifications, substitutions, combinations, and simplifications are intended to be included in the scope of the present invention.

Claims (8)

1. The anti-dandruff and anti-itching composition is characterized by comprising the following components in parts by weight: 5-15 parts of mixed extract of rheum palmatum and curcuma root, 3-10 parts of mint extract, 2-12 parts of dandelion extract, 25-40 parts of polyhydric alcohol and 33-48 parts of water.
2. The composition for removing dandruff and relieving itching according to claim 1, wherein the composition further comprises the following components in parts by weight: 1-9 parts of schizonepeta extract.
3. The composition as claimed in claim 1, wherein the mixed extract of Rumex palmate and Curcuma rhizome is prepared by the following steps:
(1) pretreatment of raw materials: respectively cleaning radix et rhizoma Rhei Palmati and Curcuma rhizome, oven drying, pulverizing, sieving with 60-100 mesh sieve, and mixing to obtain mixed powder;
(2) enzymolysis: adding aqueous solution into the mixed powder in the step (1) according to the ratio of material to liquid of 1g to (20-40) mL, adding complex enzyme for enzymolysis for 2-4h under the conditions that the pH value is 4.0-5.5 and the temperature is constant at 35-45 ℃, adding 0.2-0.4% of the weight of the complex enzyme to the mixed powder, and inactivating the enzyme to obtain enzymatic hydrolysate;
(3) high-pressure homogeneous extraction: adding an absolute ethanol solution with the volume 2-5 times of that of the enzymolysis solution into the enzymolysis solution in the step (2), uniformly stirring, and then carrying out normal-temperature extraction by using a high-pressure homogenizing extractor, wherein the high-pressure homogenizing extraction time is 10-30min, and the high-pressure homogenizing extraction pressure is 40-60 Mpa.
(4) And (3) purification: filtering the extracting solution obtained in the step (3), concentrating under reduced pressure until no alcohol smell exists, then adsorbing the concentrated solution by using an H-20 type macroporous adsorption resin column, eluting by using water until the concentrated solution is colorless, then eluting by using 40-70% ethanol solution with the volume of 3-5 times of the column volume, collecting the eluent, filtering, and freeze-drying to obtain the mixed extract of the rheum palmatum and the turmeric root.
4. The composition for removing dandruff and relieving itching as claimed in claim 3, wherein the mass ratio of the rhubarb palmate and the turmeric root in the step (1) is 3: 2.
5. The composition for removing dandruff and relieving itching according to claim 3, wherein the complex enzyme in the step (2) is composed of alpha-amylase and papain in a mass ratio of 1:4, wherein the enzyme activity of the alpha-amylase is as follows: 7000U/g, enzyme activity of papain is: 4 ten thousand U/g.
6. The composition as claimed in claim 1, wherein the polyhydric alcohol is one or more selected from propylene glycol, glycerin, butylene glycol, 1, 2-hexanediol, and ethoxydiglycol.
7. Use of the anti-dandruff and antipruritic composition according to any one of claims 1 to 6 for the preparation of daily care products.
8. The use of the anti-dandruff and anti-itching composition according to claim 7 for the preparation of daily necessities for washing and caring, wherein the daily necessities are one of shampoo, hair conditioner, hair essence, hair mask and shampoo.
