Abstract
The invention relates to an anti-aging skin care product composition with wrinkle removing effect, which comprises carpet bugle, astragalus root and citron fruit peel; the preparation method adopts a new fermentation method; and applying it to lotions, milky lotions, creams, essences, foundations and face washes. According to the invention, astragalus root, citron fruit peel and carpet bugle are adopted for compatibility, and aiming at the wrinkle-removing and anti-aging effects, all the substances have the mutual promotion effect, so that the generation of wrinkles is effectively delayed, and the generated fine wrinkles are reduced; by adopting a new fermentation mode, more active substances can be provided, the activity of the plant can be utilized more greatly, partial toxin in the plant can be reduced, and the permeation and the utilization of the activity of the plant are promoted; the application of the anti-aging skin care product composition with the wrinkle removing effect has good effects of delaying the generation of wrinkles and fading the generated fine wrinkles.
Description
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to an anti-aging skin care product composition, a preparation method and application thereof.
Background

The wrinkles are the chief reason for showing people's age, and are always a difficult problem for beauty lovers, women are over 25 years old, the wrinkles can violently grow out on the face carelessly, so that the beauty lovers are very worried, the youthful vigor is not enjoyed, the wrinkles climb over the brow tips, the beauty lovers are concerned, and therefore, the method for delaying the generation of the wrinkles and fading the generated fine wrinkles is the direction of active research of various cosmetic manufacturers.
Disclosure of Invention
Based on the above, there is a need to provide an anti-aging skin care composition with wrinkle removing effect, a preparation method thereof and applications thereof, aiming at the problems of delaying the generation of wrinkles and lightening the generated fine wrinkles.
The invention relates to an anti-aging skin care product composition with wrinkle removing effect, which comprises carpet bugle, astragalus root and citron fruit peel.
Further, the anti-aging skin care product composition with the wrinkle removing effect comprises, by weight, 1-10 parts of carpet bugle, 1-10 parts of astragalus root and 1-5 parts of citron fruit peel.
The invention relates to a preparation method of an anti-aging skin care product composition with wrinkle removing effect, which comprises the following steps:
s1, respectively pulverizing dried herba Ajugae and radix astragali, and sieving with a 20-mesh sieve to obtain herba Ajugae powder and radix astragali powder; cutting fresh pericarp of fructus Citri;
s2, fermenting with ajuga reptans, radix astragali and citron fruit peel as substrates, with bifidobacterium as a strain and with a bifidobacterium bacterial liquid of 105-108 CFU/mL;
s3, centrifuging the fermentation liquor to obtain supernatant and bacterial sludge;
s4, adding protective agent mannitol into the bacterial sludge according to the ratio of 1: 1-10, adding phosphate buffer to adjust the pH value to 6.5-7.5, uniformly stirring, treating for 3-5 minutes by a homogenizer, and then sterilizing at high temperature to obtain a bifidobacterium lysate;
s5, mixing the supernatant with the bifidobacterium lysate, and freeze-filtering to obtain a clear and transparent liquid.
Further, in the step S2, 1-10 parts by weight of carpet bugle powder, 1-10 parts by weight of astragalus root powder and 1-5 parts by weight of citron fruit peel fragments are used as substrates, 1-10 parts by weight of bifidobacterium liquid and 40-350 parts by weight of water are added, and after the substrates are completely mixed uniformly, the substrates are subjected to oscillation fermentation at 40-45 ℃ and 140-160 rpm for 20-28 hours.
Further, the step of freezing and filtering in the step S5 is that the temperature is reduced to 1-5 ℃, standing is carried out for 12-24 hours, and then filtering is carried out by using 1000-mesh filter cloth.

0.5% of PEG/PPG-17/6 copolymer;

0.2% of PEG-60 hydrogenated castor oil;
0.1 percent of arginine;
0.1-10% of anti-aging skin care composition with wrinkle removing effect;
the balance of water.
The invention has the beneficial effects that:
1. according to the anti-aging skin care product composition with the wrinkle removing effect, the astragalus root, the citron fruit peel and the carpet bugle are matched, and aiming at the wrinkle removing and anti-aging effect, all the substances have the mutual promotion effect, so that the cell proliferation can be better promoted, the cell tissue function can be maintained and remolded, the over-expression of matrix metalloproteinase can be better inhibited, the reduction of the content of subcutaneous collagen and hyaluronic acid can be avoided, the generation of wrinkles can be effectively delayed, and the generated fine wrinkles can be reduced;
2. according to the anti-aging skin care product composition with the wrinkle removing effect, the action effect generated by the compatibility of the astragalus root, the citron fruit peel and the carpet bugle has a complex influence mechanism, and a long-term test determines that the specific proportion of the optimal mutual promotion effect is achieved, so that the effects of delaying the generation of wrinkles and fading the generated fine wrinkles are better;

The physicochemical properties of each substance in the anti-aging skin care product composition with wrinkle-removing effect are explained in detail as follows:
the carpet bugle used in the invention is a carpet bugle, belongs to the Xinjiang variety of the carpet bugle, is used as a medicine by whole herbs, is mainly used for treating symptoms such as lung heat hemoptysis, traumatic injury, tonsillitis, pharyngolaryngitis and the like in the traditional medicine, is an effective component capable of improving the activity of skin cell aquaporin, and can be used as a skin conditioner in a cosmetic formula.
It should be noted that the growth of the herbs is critical to the natural conditions, the quality of the herbs is different, the dosage may be different according to the action mechanism, and the effect may also be changed.
Astragalus membranaceus (Astragalus membranaceus) is dried root of Astragalus membranaceus (Fisch.) bge. Also named Huang Qi (Ben Jing) and mian Huang Qi (Ben Cao Tu Jing). Sweet in flavor and slightly warm in nature. Enter lung and spleen meridians. Unprocessed: benefiting defense, consolidating superficial resistance, inducing diuresis to alleviate edema, expelling toxin, and promoting granulation. Different dosages of some Chinese herbs have different actions, and clinical dosage depends on the requirement of efficacy and action, and different herbs with the same efficacy have different dosages, different dosages in compatibility and different effects. The following is a description of a method for preparing an anti-aging skin care composition having wrinkle-removing effect, comprising the steps of:

The anti-aging skin care product composition with the wrinkle removing effect is prepared by adopting a fermentation process, so that the irritation of the anti-aging skin care product composition with the wrinkle removing effect on skin can be reduced, more active substances can be provided, the activity of plants can be utilized more greatly, excessive waste is not caused, partial toxin in the plants can be reduced, partial macromolecular nutrient substances are decomposed into micromolecular nutrient substances, and the permeation and the utilization of the activity of the plants are promoted.
In S2 of the preparation method, 1-10 parts by weight of decumbent bugle herb powder, 1-10 parts by weight of astragalus root powder and 1-5 parts by weight of fragments of citron fruit peel are taken as substrates, 1-10 parts by weight of bifidobacterium liquid and 40-350 parts by weight of water are added, after all the materials are uniformly mixed, the mixture is subjected to oscillation fermentation at 40-45 ℃ and 140-160 rpm for 20-28 hours, and the bifidobacterium can be a strain with ATCC number 27917; and in the S5 of the preparation method, the freezing filtration is to cool the temperature to 1-5 ℃, stand for 12-24 h, and filter with 1000-mesh filter cloth.
The pH value influences the charge state of substances in the reaction liquid, the ionization state of amino acid in bifidobacterium cells and the activity of enzyme in the cells, and when the pH value is 6.5-7.5, the bifidobacterium strain with the ATCC number of 27917 has the best fermentation activity on the mixture of carpet bugle, astragalus root and citron fruit peel.
The fermentation temperature, time and inoculation amount are two important factors in the fermentation process, the influence of the temperature and time on the growth and fermentation of the thalli is the result of comprehensive expression of various factors, the influence of the proper temperature and time on the increment of the thalli and the yield of products is large, the highest biomass of the thalli can be achieved by oscillating and fermenting at 40-45 ℃ and 140-160 rpm for 20-28 h, and the formed product quality is better; the inoculation amount is related to the influence on the total number of live bacteria in the same time, and the highest live bacteria amount of the bacteria can be achieved by fermenting with 105-108 CFU/mL of bifidobacterium liquid.
The following description will be made on the application of the anti-aging skin care composition having wrinkle-removing effect, mainly to lotions, milky lotions, creams, essences, foundations, and facial cleansers.
In each application, the addition amount of the anti-aging skin care composition with the wrinkle removing effect is 0.1-10%.
The application of the anti-aging skin care product composition with the wrinkle removing effect has the good effects of delaying the generation of wrinkles and fading the generated fine wrinkles.
Some specific components of the application of the anti-aging skin care composition with wrinkle removing effect are exemplified below, but it should be noted that the following does not exhaust all possible cases of the application.
Application 1: emulsion and method of making
The emulsion comprises the following components in percentage by mass:
5% of glycerol;
caprylic/capric triglyceride 3%;
isononyl isononanoate 3%;
1% of glycerol stearate;
cetostearyl alcohol 1%;
0.5 percent of sodium stearoyl lactylate;
5% of butanediol;
3% of polydimethylsiloxane;
1% of cetyl polydimethylsiloxane;
1% of 1, 2-hexanediol;

0.05% of sodium hyaluronate;
0.3% of carbomer;

Application 2: cream
The cream comprises the following components in percentage by mass:
5% of glycerol;
4% of butanediol;
caprylic/capric triglyceride 3%;
5% of squalane;
2% of polydimethylsiloxane;
0.5% of hydrogenated lecithin;

Application 3: essence of plant
The essence comprises the following components in percentage by mass:

0.5% of PEG/PPG-17/6 copolymer;

0.1% of xanthan gum;
5% of 1, 2-pentanediol;
0.05% of laurocapram;
0.2% of PEG-60 hydrogenated castor oil;
0.1 percent of arginine;
0.1-10% of anti-aging skin care composition with wrinkle removing effect;
the balance of water.
Some specific examples are listed below, but it should be noted that the following examples are not exhaustive of all possible cases. And the materials used in the following examples are commercially available unless otherwise specified.
Example 1
S10, respectively pulverizing dried herba Ajugae and radix astragali, and sieving with a 20-mesh sieve to obtain herba Ajugae powder and radix astragali powder; cutting fresh pericarp of fructus Citri;
s20, taking 1 part of carpet bugle powder, 10 parts of astragalus root powder and 5 parts of citron fruit peel fragments (the total weight is X) as substrates by weight parts, adding 5 parts of 105CFU/mL bifidobacterium bacterial liquid and 300 parts of water, uniformly mixing, and then carrying out oscillatory fermentation for 25h at 45 ℃ and 150 rpm.
S30, centrifuging the fermentation liquor to obtain supernatant and bacterial sludge;
s40, adding mannitol serving as a protective agent into the bacterial sludge according to the ratio of 1:5, adding a phosphate buffer solution to adjust the pH value to 7.0, uniformly stirring, treating for 4 minutes by using a homogenizer, and then sterilizing at high temperature to obtain a bifidobacterium lysate;
s50, mixing the supernatant with the bifidobacterium lysate, cooling to 3 ℃, standing for 18h, and filtering with a 1000-mesh filter cloth to obtain clear transparent liquid.
Example 2

s20, taking 10 parts of carpet bugle powder, 1 part of astragalus root powder and 2.5 parts of citron fruit peel fragments (the total weight is X) as substrates by weight parts, adding 5 parts of 105CFU/mL bifidobacterium bacterial liquid and 300 parts of water, mixing uniformly, and then carrying out oscillatory fermentation for 25h at 45 ℃ and 150 rpm.
S30, centrifuging the fermentation liquor to obtain supernatant and bacterial sludge;
s40, adding mannitol serving as a protective agent into the bacterial sludge according to the ratio of 1:5, adding a phosphate buffer solution to adjust the pH value to 7.0, uniformly stirring, treating for 4 minutes by using a homogenizer, and then sterilizing at high temperature to obtain a bifidobacterium lysate;
s50, mixing the supernatant with the bifidobacterium lysate, cooling to 3 ℃, standing for 18h, and filtering with a 1000-mesh filter cloth to obtain clear transparent liquid.
Example 3
S10, respectively pulverizing dried herba Ajugae and radix astragali, and sieving with a 20-mesh sieve to obtain herba Ajugae powder and radix astragali powder; cutting fresh pericarp of fructus Citri;
s20, taking 5 parts of carpet bugle powder, 5 parts of astragalus root powder and 1 part of citron fruit peel fragments (the total weight is X) as substrates by weight parts, adding 5 parts of 105CFU/mL bifidobacterium bacterial liquid and 300 parts of water, mixing uniformly, and then carrying out oscillatory fermentation for 25h at 45 ℃ and 150 rpm.

The anti-aging skin care product composition with the wrinkle removing effect is tested, and the specific test is as follows:
test 1: proliferation-promoting assay for human epidermal fibroblasts
Purpose of the experiment
The anti-aging skin care composition with wrinkle-removing efficacy was tested for its proliferation-promoting effect on human epidermal fibroblasts.
Test protocol
Test article and grouping: 5g of the anti-aging skin care product composition with the wrinkle removing effect prepared in the embodiments 1-3 of the invention are respectively used as test groups 1-3, 5g of the test group 4 (carpet bugle powder (total weight is X) is used as a substrate), 5g of the test group 5 (astragalus root powder (total weight is X) is used as a substrate), 6 g of citron fruit peel fragments (total weight is X) is used as a substrate, and 5g of the blank group (clear water) is obtained by the method of the embodiments 1-3.
Test procedure
1. Respectively adding substances of the test groups 1-6 and the blank group into 50mL volumetric flasks, adding phosphate buffer solution (PBS, pH7.4) into each volumetric flask for dilution and volume fixing, stirring at room temperature for 1h, centrifuging at 12000rpm for 30min, collecting supernatant, filtering with a 0.22 mu m microporous filter membrane to obtain solutions to be tested 1-7 respectively, and storing in a refrigerator at 5 ℃ for later use;
2. preparing 21 culture media, inoculating human epidermal fibroblasts in logarithmic growth phase into a 96-well plate at a density of 1 × 103/mL, and adding 200 μ L of a pre-prepared culture medium (MEM cell culture medium (from Nissan Shuichi, 05900 Eagle' sMEM));
3. respectively adding 1-7 solutions to be detected into 21 culture media (the types of the solutions to be detected in every 3 culture media are the same);
4. the relative number of viable cells in each well was measured at 490nm by MTT method at a fixed time every day for 4 days of culture.
Test results
The results of the experiment are shown in FIG. 1.
Conclusion of the experiment
As can be seen from FIG. 1, the absorbance (cell OD value) of the test groups 1-3 gradually exceeded that of the test groups 4-6 and the blank group with the prolonged action time of the antiaging composition, which indicates that the antiaging composition has a proliferation promoting effect on human epidermal fibroblasts, and the proliferation promoting effect of the test groups 1-3 is significantly higher than that of the test groups 4-6 under the condition of the same test dose. Further, the combination of the three components has better effect on promoting the proliferation of human epidermal fibroblasts than the combination of the three components used alone.
Test 2: matrix metalloproteinase inhibition assay
Purpose of the experiment
The anti-aging skin care composition having wrinkle-removing efficacy was tested for the inhibition rate of matrix metalloproteinase.
Principle of experiment
Inhibition rate is (a-B)/ax100%;
a is MMP-1 expression amount without adding a solution to be tested;
b is MMP-1 expression amount added with the solution to be tested.
Test protocol
Test article and grouping: 5g of the anti-aging skin care product composition with the wrinkle removing effect prepared in the embodiments 1-3 of the invention are respectively used as test groups 1-3, and 5g of the test group 4 (carpet bugle powder (total weight is X) is used as a substrate), the test group 5 (astragalus root powder (total weight is X) is used as a substrate) and the test group 6 citron fruit peel fragments (total weight is X) are respectively used as substrates obtained by the methods of the embodiments 1-3.
Test procedure
1. Respectively adding the substances of the test groups 1-6 into 50mL volumetric flasks, adding phosphate buffer solution (PBS, pH7.4) into each volumetric flask for dilution and volume fixing, stirring for 1h at room temperature, centrifuging for 30min at 12000rpm, collecting supernatant, passing through a 0.22 mu m microporous filter membrane to obtain solutions to be tested 1-6 respectively, and storing in a refrigerator at 5 ℃ for later use;
2. preparing 18 culture media, taking human epidermal fibroblasts in a logarithmic growth phase, inoculating the human epidermal fibroblasts into a 96-well plate at the density of 1 × 105 cells/mL, and carrying out starvation culture in a serum-free culture medium for 24 hours;
3. the starvation cultured cells were washed with PBS and then treated with 40mJ uv light;
4. the solutions to be tested were added to 18 media (each 3 media contained the same type of solution) 1-6 at intervals of 24h, 10. mu.L of the solution to be tested was added to the cells of each media, MMP-1 separated from the media was measured with a kit (MMP-1, Biotrak Amersham pharmacia Biotech, United Kingdom; RPN 2610) after 48h, and MMP-1 expression inhibition was calculated.
Test results
The results of the experiment are shown in FIG. 2.
Conclusion of the experiment
As can be seen from FIG. 2, the matrix metalloproteinase inhibitory rates of the test groups 1 to 3 were higher than those of the test groups 4 to 6 under the same test dose. Further summarize and show that the inhibition ratio of the three mutually matched used on the matrix metalloproteinase is better than the additive effect of the three used independently.
Test 3: volunteer efficacy evaluation test
Purpose of the experiment
The anti-aging skin care composition having wrinkle removing effect was tested for improvement of facial wrinkles.
Test protocol
Test article and grouping: the anti-aging skin care composition with the wrinkle removing effect prepared in the embodiments 1 to 3 of the present invention is added to an emulsion matrix in an amount of 5 wt% to prepare anti-aging emulsions as emulsions i to iii, respectively, and the emulsions iv (ajuga reptans powder (total weight X) as a substrate), the emulsions v (astragalus root powder (total weight X) as a substrate) and the emulsions vi (citron fruit peel fragments (total weight X) as a substrate) obtained by the methods for obtaining the emulsions i to iii are used.
Test procedure
1. 180 healthy women with age of 35-55 years and obvious fine wrinkles on the skin around the eyes are summoned, evenly distributed into test groups I-IV, and respectively and correspondingly used with anti-aging emulsions I-IV;
2. cleaning the face of a subject with clean water, wiping the face water, then statically sitting for 30min at the temperature of 20-22 ℃ and the relative humidity of 40-60%, and taking a picture of the skin around the eyes of the volunteer by a measurer by adopting VISIA-CR;
3. uniformly smearing 1mL of emulsion on the face, and lightly tapping for absorption;
4. smearing the paste once in the morning and at night (repeating for 2-3);
5. the wrinkle visibility coefficient and the skin wrinkle occupancy coefficient were calculated, and the facial wrinkle improvement before and after the use of the product was evaluated.
It is further noted that the inclusion criteria and exclusion criteria for 180 women aged 35-55 years are:
inclusion criteria were: the test can be well matched, and the life rule can be kept during the research period; all the contents of the informed consent can be read and understood and signed voluntarily; during the test period, the use of skin care products is stopped, the skin care products do not participate in any other clinical test of a research center, and cosmetics, medicines and health care products which have influence on the result are not used;
exclusion criteria: the skin disease at the tested part may affect the judgment of the test result; those with high allergic constitution; a female who is pregnant, lactating, or intended to be pregnant during the test; severe center of gravity, liver and kidney function damage and severe immunologic hypofunction; those with mental disorders, severe endocrine disorders, and oral contraceptives; adding drugs into clinical testers or other testers of the drugs within 30 days, or patients who have systemic effect on the test result within nearly 1 week; those who had oral and topical cosmetic products within 2 weeks that may have an effect on the outcome of the test; the test person can not be matched; researchers considered unsuitable participants.
Test results
The test results are shown in fig. 3 and 4.
Conclusion of the experiment
As can be seen from the above figures 3 and 4, the rate of reduction of the visibility coefficient of wrinkles and the rate of reduction of the wrinkle occupancy coefficient of the canthus skin were significantly increased and better than those of the test groups IV-VI after 2 weeks, 4 weeks, 6 weeks, and 8 weeks using the antiaging emulsions prepared by this test groups I-III. Further summarize and show that the improvement of skin wrinkles by the mutual cooperation of the three is better than the additive effect of the three when used alone.
The technical features of the above embodiments can be arbitrarily combined, and for the sake of brevity, all possible combinations of the technical features in the above embodiments are not described, but should be considered as the scope of the present specification as long as there is no contradiction between the combinations of the technical features.

Claims (10)
1. An anti-aging skin care product composition with wrinkle removing effect is characterized by comprising carpet bugle, astragalus root and citron fruit peel.
2. The anti-aging skin care composition with the wrinkle-removing effect according to claim 1, wherein the anti-aging skin care composition with the wrinkle-removing effect comprises, by weight, 1-10 parts of ajuga reptans, 1-10 parts of astragalus root and 1-5 parts of citron fruit peel.
3. The preparation method of the anti-aging skin care composition with wrinkle removing effect according to any one of claims 1-2, characterized by comprising the following steps:
s1, respectively pulverizing dried herba Ajugae and radix astragali, and sieving with a 20-mesh sieve to obtain herba Ajugae powder and radix astragali powder; cutting fresh pericarp of fructus Citri;
s2, fermenting with ajuga reptans, radix astragali and citron fruit peel as substrates, with bifidobacterium as a strain and with a bifidobacterium bacterial liquid of 105-108 CFU/mL;
s3, centrifuging the fermentation liquor to obtain supernatant and bacterial sludge;
s4, adding protective agent mannitol into the bacterial sludge according to the ratio of 1: 1-10, adding phosphate buffer to adjust the pH value to 6.5-7.5, uniformly stirring, treating for 3-5 minutes by a homogenizer, and then sterilizing at high temperature to obtain a bifidobacterium lysate;
s5, mixing the supernatant with the bifidobacterium lysate, and freeze-filtering to obtain a clear and transparent liquid.
4. The preparation method of the anti-aging skin care composition with the wrinkle-removing effect according to claim 3, characterized in that in S2, 1-10 parts by weight of Ajuga reptans powder, 1-10 parts by weight of radix astragali powder and 1-5 parts by weight of citron fruit peel fragments are used as substrates, 1-10 parts by weight of bifidobacterium bacterial liquid and 40-350 parts by weight of water are added, and after all the materials are uniformly mixed, the materials are subjected to oscillation fermentation at 40-45 ℃ and 140-160 rpm for 20-28 hours.
5. The preparation method of the anti-aging skin care product composition with the wrinkle removing effect according to claim 3, wherein the freezing filtration in S5 is cooling to 1-5 ℃, standing for 12-24 h, and then filtering with 1000-mesh filter cloth.
6. The anti-aging skin care composition with wrinkle-removing effect according to any one of claims 1 to 2 is applied to lotions, milky lotions, creams, essences, foundations and facial cleansers.
7. The use of the anti-aging skin care composition with wrinkle-removing effect according to claim 6, wherein in the use, the addition amount of the anti-aging skin care composition with wrinkle-removing effect is 0.1-10%.
8. The use of the anti-aging skin care composition with wrinkle removing effect according to claim 6, characterized in that the emulsion comprises, by mass percent:
5% of glycerol;
caprylic/capric triglyceride 3%;
isononyl isononanoate 3%;
1% of glycerol stearate;
cetostearyl alcohol 1%;
0.5 percent of sodium stearoyl lactylate;
5% of butanediol;
3% of polydimethylsiloxane;
1% of cetyl polydimethylsiloxane;
1% of 1, 2-hexanediol;
cetearyl alcohol wheat straw glycosides 0.5%;
0.05% of sodium hyaluronate;
0.3% of carbomer;
0.3 percent of triethanolamine;
0.1% of xanthan gum;
0.5 percent of p-hydroxyacetophenone;
0.1-10% of anti-aging skin care composition with wrinkle removing effect;
the balance of water.
9. The use of the anti-aging skin care composition with wrinkle removing effect according to claim 7, characterized in that the cream comprises, by mass percent:
5% of glycerol;
4% of butanediol;
caprylic/capric triglyceride 3%;
5% of squalane;
2% of polydimethylsiloxane;
0.5% of hydrogenated lecithin;
0.5% of polyglycerol-3 distearate;
1% of glycerol stearate citrate;
pentaerythritol tetra (ethyl hexanoate) 0.5%;
1% of mica;
1% of sorbitan sesquioleate;
ethylene diamine/stearyl alcohol dilinoleate copolymer 0.1%;
0.5 percent of C15-19 alkyl;
0.5 percent of p-hydroxyacetophenone;
3% of propylene glycol;
acrylic acid (ester)/C10-30 alkanol acrylate crosspolymer 0.5%;
