Abstract
The present invention relates to a cosmetic composition comprising a mixed extract of unripe fruits as an effective ingredient, and provides a cosmetic composition comprising a mixed extract of unripe oranges, unripe apples, unripe tomatoes, and one kind of unripe fruits selected from the group consisting of unripe strawberries, unripe bananas, unripe lemons, unripe persimmons, unripe figs, unripe jubes, and unripe plums as an effective ingredient. The cosmetic composition comprising the above-mentioned unripe fruit mixed extract as an effective ingredient has excellent effects of improving skin wrinkles, improving skin elasticity, regenerating skin and relieving irritation.
Description
Technical Field
The present invention relates to an anti-aging cosmetic composition comprising a mixed extract of unripe fruits, and more particularly, to a cosmetic composition having excellent anti-aging activity comprising unripe oranges, unripe apples, unripe tomatoes, and a mixed extract selected from the group consisting of unripe strawberries, unripe bananas, unripe lemons, unripe persimmons, unripe figs, unripe jubes, and unripe plums as an effective ingredient.
Background
All organisms undergo the aging process from birth, which is an indispensable part of life. Skin also ages with age, but the first site to appear is the face. Aging of skin is classified into intrinsic aging and light aging caused by exposure to environmental factors. Environmental factors include ultraviolet rays, fine dust, smoking, and the like, and particularly, sunlight is known as a representative factor causing skin aging. Intrinsic aging is mainly caused by active oxygen produced by the body's metabolic processes, and the accumulation of active oxygen promotes aging by causing skin damage.
Wrinkles of the skin are common symptoms of skin aging, and as the skin ages, the amount of collagen produced in the body decreases, so that facial wrinkles increase and the feeling of elasticity and firmness gradually disappears. Therefore, if skin aging is to be slowed, it is important to maintain the amount of collagen. Collagen is a protein that occupies most of the dermal layer under the epidermis of the skin, and has the effect of helping cells in the skin to support each other. Elastin, which is an elastic fiber component that participates in skin elasticity together with collagen, is localized in skin tissue together with collagen, thereby acting to soften skin tissue and improve stretchability. Therefore, when the skin is aged, the production amount of collagen and elastin is reduced, thereby forming wrinkles, resulting in deterioration of skin elasticity, and finally, in the face being more prominent.
As society develops, the aging population increases, and the need to maintain skin health and youth increases, with an increasing focus on healthy skin care and skin aging. Management of skin aging under such concerns is rapidly advancing.
In connection with this, studies are being conducted to apply all of the discarded unripe fruits, which are not commercially available and cannot be eaten, to the field of foods or cosmetics. In korean laid-open patent No. 10-2006) -0104054 "whitening cosmetic composition comprising an immature citrus extract", it is disclosed that 'the immature citrus extract is excellent in the effect of inhibiting pigmentation by inhibiting melanin production and thus has excellent dark spot or freckle improving effect and skin whitening effect', in korean laid-open patent No. 10-2011-0066042 "cosmetic composition comprising an immature apple extract", it is disclosed that 'the tyrosinase inhibitory activity of the immature apple extract is higher than that of a mature apple', and in korean laid-open patent No. 10-1194767 "cosmetic composition for shrinking pores and inhibiting sebum secretion comprising an immature apple extract", it is disclosed that 'the immature apple extract, particularly ethyl acetate fraction of immature apple peel, is excellent in the pore shrinking effect and sebum secretion inhibiting effect', in korean laid-open patent No. 10-2014-0141188 "skin external preparation composition comprising an unripe persimmon extract", it is disclosed that 'the unripe persimmon extract has effects of shrinking skin pores and reducing sebum secretion'.
The present inventors have studied about the activity of various unripe fruits by preparing a mixed extract of unripe fruits in the process of searching for natural substances having anti-aging activity, and at this time, confirmed that the mixed extract of unripe oranges and unripe apples and unripe tomatoes has excellent anti-aging effect as compared with each extract, and in the case of preparing here a mixed extract with specific other unripe fruits, the activity thereof is further increased to have excellent skin improvement effect, and thus completed the present invention by finding that it can be effectively used as an anti-aging cosmetic.
Disclosure of Invention
The present invention is directed to providing a cosmetic composition having excellent anti-aging activity by containing a mixed extract of unripe orange, unripe apple, unripe tomato, and one kind of unripe fruit selected from the group consisting of unripe strawberry, unripe banana, unripe lemon, unripe persimmon, unripe fig, unripe date, and unripe plum as an active ingredient.
In order to achieve the above object, in the present invention, there is provided a cosmetic composition comprising a mixed extract of unripe orange, unripe apple, unripe tomato, and one kind of unripe fruit selected from the group consisting of unripe strawberry, unripe banana, unripe lemon, unripe persimmon, unripe fig, unripe date, and unripe plum as an effective ingredient. In this case, the mixed extract is contained as an active ingredient in an amount of 0.01 to 90 wt% based on the total weight of the cosmetic composition.
Preferably, the mixed extract is prepared by mixing the following components in a ratio of 1-2: 1-2: 1-2: 1-2 by weight, mixing unripe orange, unripe apple, unripe tomato, and one kind of unripe fruit selected from the group consisting of unripe strawberry, unripe banana, unripe lemon, unripe persimmon, unripe fig, unripe date, and unripe plum, and most preferably, mixing the mixture of 2: 2: 2: 1 weight ratio of the raw materials and extracting.
The mixed extract is a hot water extract or a solvent extract obtained by using at least one solvent selected from the group consisting of water, ethanol, methanol, butanol, propanol, butylene glycol, glycerol, chloroform, ethyl acetate, dichloromethane, hexane, acetone, acetonitrile, petroleum ether and diethyl ether.
The cosmetic composition is characterized by being used for improving skin wrinkles and elasticity, for regenerating skin cells or for relieving skin irritation.
The mixed extract of unripe orange, unripe apple, unripe tomato, and one unripe fruit selected from the group consisting of unripe strawberry, unripe banana, unripe lemon, unripe persimmon, unripe fig, unripe date, and unripe plum of the present invention has excellent effects of improving skin wrinkle, improving skin elasticity, regenerating skin cells, and relieving skin irritation, and thus can be effectively used in the anti-aging cosmetic.

The present invention will be described in further detail below.
The present invention relates to a cosmetic composition comprising a mixed extract of unripe orange, unripe apple, unripe tomato, and one kind of unripe fruit selected from the group consisting of unripe strawberry, unripe banana, unripe lemon, unripe persimmon, unripe fig, unripe date, and unripe plum as an effective ingredient.
The unripe orange (Citrus unshiu) used in the present invention is an orange whose outer skin is green due to unripe orange. It is known that immature oranges have a total polyphenol and total flavonoid content higher than that of mature oranges by a factor of 2 or more, and contain 1.5 to 2% of citric acid for decomposing lactic acid, which is a causative substance of fatigue, and thus contribute to skin protection.
Unlike ripe apples, unripe apples (apples) are known to have a polyphenol content 10 times higher, and since they are rich in antioxidant components, they can prevent skin aging by preventing cell oxidation and cell aging. Tannin and rutin, which are components that impart astringency to unripe apples, are effective in removing active oxygen and shrinking pores, and thus contribute to preventing skin aging as a whole.
Tomato (tomato Lycopersicum) is a main supply source of lycopene having excellent antioxidant and anti-inflammatory effects, and is also known as a longevity food. Unripe tomatoes are hard and astringent and rich in lycopersicin, tomatidine and tomatidine, which promote muscle growth and prevent muscle wasting due to aging.
Strawberry (Fragaria Ananassa) is a perennial plant of the family Rosaceae of the order Rosales of the order Heterocotyledonous plant. The strawberry (strawberry) has good combination of sour taste and sweet taste, and is rich in vitamin C, anthocyanin, flavonoid, polyphenol and other antioxidant substances. These antioxidant substances are known to have the functions of inhibiting carcinogenic cells produced by active oxygen and reducing cholesterol including resistance to mutation. On the other hand, the kind or content of these functional plant compounds (Phytochemicals) contained in plants varies depending on the degree of maturity of fruits. In the present invention, the unripe strawberry means a fruit before ripening into a red fruit.
Banana (Musa Paradisica) is a generic term for plants belonging to the genus Musa of the family Musaceae of the order Zingiberales, monocotyledonous. Unripe banana refers to an unripe banana fruit in green color, which is known to benefit our body by being rich in resistant starch (resiststarch), and it is known that starch, which is the most important element of water-soluble dietary fiber in the body, is gradually absorbed by the human body and provides energy for a long time. Bananas contain vitamin C and vitamin B6, thereby preventing aging by preventing premature aging of skin induced by active oxygen.
Lemon (Citrus Limon) is a representative fruit rich in vitamin C, which is a representative antioxidant vitamin for preventing aging of the human body, and thus functions to slow down aging by preventing cellular oxidation, and immature lemon is rich in this vitamin and mineral, thus preventing skin aging.
Tannin is a component for giving astringent taste to persimmon (Diospyros Kaki), and has excellent astringent effect, elasticity to skin and protection of skin by preventing active oxygen through strong antioxidant ability. Persimmon is rich in vitamin A, and thus enhances immunity and makes skin elastic. The unripe persimmon is green persimmon.
Fig (Ficus carica L.) tree, a subtropical deciduous tree belonging to the family Moraceae, is a plant in which over 600 varieties are distributed in common worldwide, and is called a fruit that is the earliest utilized by humans in long historical years. The fig contains abundant carbohydrates, proteins, vitamins and inorganic matters, and the fig skin contains polyphenol, thereby playing a role in antioxidation. Fig helps to prevent aging and improve elasticity, and vitamin C and vitamin E contained in fig also help to whiten. The extract of unripe fig has excellent protease activity, and is thus being developed as a meat softener for food processing.
The fructus Jujubae is fruit of Ziziphus jujuba Mill (Zizyphus jujuba Mill) of Rhamnaceae. Beta-carotene, which is known to be rich in various vitamins and minerals and known as an excellent antioxidant ingredient, has excellent effects of promoting metabolism, and preventing and curing aging. Initially green and reddish brown or reddish brown by 9-10 months of maturity. It is known that jujube has excellent detoxifying and anti-inflammatory effects, thus helping to remove inflammation and relieve pain in the body. The unripe date suitable for use in the present invention refers to unripe green fruit.
Plum (Prunus Salicina) is rich in vitamin C and water-soluble dietary fiber, and is beneficial for skin beauty, and vitamin E of plum prevents harmful active oxygen, thereby having excellent anti-aging effect. Immature plums suitable for use in the present invention refer to immature green plums.
In one embodiment of the present invention, the above-mentioned unripe fruit mixed extract is prepared by the following method.
Respectively mixing 1-2: 1-2: 1-2: 1-2 weight ratio of the extract, mixing the washed and dried unripe orange, unripe apple, unripe tomato, and one kind of unripe fruit selected from the group consisting of unripe strawberry, unripe banana, unripe lemon, unripe persimmon, unripe fig, unripe date, and unripe plum. The excellent skin improving effect is exhibited from the above mixing ratio. In this case, at least one solvent selected from the group consisting of water, ethanol, methanol, butanol, propanol, butylene glycol, glycerol, chloroform, ethyl acetate, dichloromethane, hexane, acetone, acetonitrile, petroleum ether, and diethyl ether may be used as the solvent. After extraction, the mixed extract can be prepared by sterilization, filtration, and concentration.
The mixed extract of immature fruits thus prepared had excellent wrinkle-related factor expression increasing effects (test examples 1 and 3), wrinkle-formation factor expression inhibiting effects (test examples 2 and 4), elasticity-related factor expression increasing effects (test example 5), elasticity reduction-related factor inhibiting effects (test example 6), skin cell regeneration effects (test example 7), and skin irritation relieving effects (test example 8).

The present invention will be described in detail below with reference to examples and test examples, but the present invention is not limited thereto.
Example 1: preparing mixed extracts of unripe tangerine, unripe apple, unripe tomato and unripe strawberry
Dried samples of green tangerine 3g, green apple 3g, green strawberry 3g and green tomato 3g (dry weight ratio 1: 1: 1: 1) were mixed with 70% aqueous ethanol 1kg, extracted by boiling in an extractor equipped with a condenser at 100 ℃ for 8 hours, and then filtered with Filter paper (qualitative Filter paper No.2) to obtain a filtrate.
Examples 2 to 11: preparing mixed extracts of unripe tangerine, unripe apple, unripe tomato and unripe strawberry
As shown in table 1 below, a mixed extract was prepared by extraction and filtration in the same manner as in example 1 by changing the mixing weight ratio of unripe oranges, unripe apples, unripe tomatoes, and unripe strawberries.

Dried 12g of unripe apples were added to 1Kg of 70% aqueous ethanol and boiled at a temperature of 100 ℃ for 8 hours in an extractor equipped with a condenser to extract, and then filtered by using filter paper (qualitative filter paper No.2) to obtain the extract in the state of filtrate.
Comparative example 3: preparation of unripe tomato extract
Dried 12g of unripe tomatoes are added to 1Kg of 70% aqueous ethanol and boiled at a temperature of 100 ℃ for 8 hours in an extractor equipped with a condenser to extract, and then filtered with filter paper (qualitative filter paper No.2) to obtain in the state of filtrate.
Comparative example 4: preparing unripe strawberry extract
Dried 12g of unripe strawberry was added to 1Kg of 70% aqueous ethanol and boiled at a temperature of 100 ℃ for 8 hours in an extractor equipped with a condenser to extract, and then filtered with filter paper (qualitative filter paper No.2) to obtain the extract in the state of filtrate.
Test example 1: confirming the effect of increasing the expression of wrinkle-associated factor
Fibroblast cells (HDFa) were conditioned to 5 × 10 using Medium106 Medium supplemented with Low Serum Growth Supplement (LSGS)6Cells/well (cell/well) were then seeded in 6-well plates (well plates) and cultured for 24 hours. After the incubation, the samples of the above examples and comparative examples were treated and incubated for 48 hours. Ribonucleic acid (RNA) was extracted from the cultured cells and the increased rate of collagen (collagen) expression was confirmed by Reverse transcription polymerase chain reaction (RT-PCR). As a control group, vitamin a palmitate (retinol palmitate) was used.

As confirmed in table 2 above, the treatment at the same concentration showed excellent collagen expression rates in examples 1 to 11, which were related to the mixed extracts obtained by mixing unripe strawberries with unripe oranges, unripe apples and unripe tomatoes, compared to comparative examples 1 to 4 related to the control group and the respective unripe fruit extracts. Thereby, a synergistic effect can be confirmed by mixing them. In particular, in the case of a reaction between 2: 2: 2: the mixed extract of example 11, which was extracted by mixing unripe oranges, unripe apples, unripe tomatoes, and unripe strawberries at a weight ratio of 1, showed the most excellent collagen expression rate.
Test example 2: inhibitory Effect on the expression of wrinkle-Forming factor
Fibroblast cells were adjusted to 5 × 10 using Medium106 Medium supplemented with low serum growth supplements6Cells/well were seeded in 6-well plates and cultured for 24 hours. After the culture, the cells were irradiated with ultraviolet b (uvb), and then the samples of the above examples and comparative examples were treated and cultured for 48 hours. Ribonucleic acid was extracted from the cultured cells and the rate of inhibition of matrix metalloproteinase 1(MMP-1) expression was confirmed by reverse transcription polymerase chain reaction. The inhibition ratio of matrix metalloproteinase 1 expression was calculated by the following formula, and the results are shown in table 3 below. As a control group, vitamin a palmitate was used.

As shown in table 3, it was confirmed that the treatment at the same concentration showed excellent matrix metalloproteinase 1 expression inhibiting effect in examples 1 to 11 of the mixed extracts obtained by mixing unripe oranges, unripe apples, unripe tomatoes, and unripe strawberries, as compared to comparative examples 1 to 4 of the control group and the respective unripe fruit extracts. In particular, the lowest expression rate of matrix metalloproteinase 1 is shown in example 11.
Examples 12 to 17: preparing mixed extract of unripe fruits comprising unripe tangerine, unripe apple, and unripe tomato
In the preparation of the mixed extracts of the above examples, unripe bananas, unripe lemons, unripe persimmons, unripe figs, unripe jubes, and unripe plums were mixed with the compositions shown in the following table 4, respectively, instead of the unripe strawberries of example 11, and the mixed extracts were prepared by extraction and filtration in the same manner as in example 1.

Comparative example 5: preparation of unripe banana extract
12g of dried unripe banana was added to 1Kg of 70% aqueous ethanol and boiled at a temperature of 100 ℃ for 8 hours in an extractor equipped with a condenser to extract, and then filtered with filter paper (qualitative filter paper No.2) to obtain the banana-containing extract in the state of filtrate.
Comparative example 6: preparing unripe lemon extract
12g of dried unripe lemon was added to 1Kg of 70% aqueous ethanol and boiled at a temperature of 100 ℃ for 8 hours in an extractor equipped with a condenser to extract, and then filtered by using filter paper (qualitative filter paper No.2) to obtain a filtrate.
Comparative example 7: preparation of unripe persimmon extract
12g of dried unripe persimmon was added to 1Kg of 70% aqueous ethanol and boiled at a temperature of 100 ℃ for 8 hours in an extractor equipped with a condenser to extract, and then filtered with filter paper (qualitative filter paper No.2) to obtain a filtrate.
Comparative example 8: preparation of unripe fig extract
12g of dried unripe fig was added with 1Kg of 70% aqueous ethanol and boiled at a temperature of 100 ℃ for 8 hours in an extractor equipped with a condenser to extract, and then filtered with filter paper (qualitative filter paper No.2) to obtain the fig in the state of filtrate.
Comparative example 9: preparation of unripe date extract
12g of dried unripe date was added to 1Kg of 70% aqueous ethanol and boiled at a temperature of 100 ℃ for 8 hours in an extractor equipped with a condenser to extract, and then filtered with filter paper (qualitative filter paper No.2) to obtain the extract in the state of filtrate.
Comparative example 10: preparation of unripe plum extract
Adding 12g of dried unripe plum to 1Kg of 70% aqueous ethanolAnd boiled at a temperature of 100 ℃ for 8 hours in an extractor equipped with a condenser to extract, and then filtered with Filter paper (qualitative Filter paper No.2) to obtain a filtrate.

As confirmed in table 5 above, the mixed extracts of examples 12 to 17, which include unripe oranges, unripe apples and unripe tomatoes and are mixed with one kind of unripe fruits selected from the group consisting of unripe bananas, unripe lemons, unripe persimmons, unripe figs, unripe jubes and unripe plums, show more excellent collagen expression rate according to their synergistic effect, when treated at the same concentration as the respective unripe fruit extracts of the control and comparative examples (tables 1 and 5).

As confirmed in table 6 above, the mixed extracts of examples 12 to 17, which were extracted by mixing one kind of immature fruit selected from the group consisting of immature bananas, immature lemons, immature persimmons, immature figs, immature dates and immature plums with immature oranges, immature bananas, immature lemons, immature persimmons, immature figs, immature plums and immature plums, showed more excellent inhibitory effects on the expression of matrix metalloproteinase 1, based on their synergistic effects, when treated at the same concentrations as the respective immature fruit extracts of the control and comparative examples (tables 2 and 6).
Test example 5: increased effect of expression of elasticity-related factor
Fibroblast cells were adjusted to 5 × 10 using Medium106 Medium supplemented with low serum growth supplements6Cells/well were seeded in 6-well plates and cultured for 24 hours. After the incubation, the samples of the above examples and comparative examples were treated and incubated for 48 hours. Ribonucleic acid was extracted from the cultured cells and the rate of increase in the expression of elastin (Tropoelastin) was confirmed by reverse transcription polymerase chain reaction. The increase rate of elastin expression was calculated by the following formula, and the results are shown in table 7 below using vitamin a palmitate as a control group.

As confirmed in Table 7 above, examples 1 to 17, which are the mixed extracts of immature fruits according to the present invention, showed more excellent elastin expression rate according to their synergistic effect, when treated at the same concentration, compared to the respective immature fruit extracts of the control group and comparative examples 1 to 10. In particular, the highest elastin expression rate is shown in example 11.
Test example 6: elastic reduction correlation factor inhibition effect
To confirm the inhibitory effect of Elastase (Elastase) activity, an Elastase congo red assay (elastin consistent red assay) was performed. 2.5% agar (agar) containing 0.1% elastin-congo red was prepared using 0.2M Tris (hydroxymethyl) aminomethane buffer (Tris HCl buffer). After wells (holes) of the same size were made in agar, 1800units/ml of an elastase solution (elastase enzyme) and a sample were mixed and dispensed into the wells, and then reacted for 24 hours. The elastase activity was calculated by measuring halo diameters (halosameters) using the following formula, and the results are shown in table 8 below. As a control group, vitamin a palmitate was used.

As confirmed in Table 8 above, the immature fruit mixed extracts of examples 1 to 17 of the present invention showed lower elastase activity when treated at the same concentration as compared to the control and comparative examples 1 to 10. In particular, the lowest elastase activity rate was observed in example 11.
Test example 7: skin cell regeneration effect
In order to confirm the skin cell regeneration effect, a wound healing test (wound healing assay) was performed. By adding hornsMedium154 Medium for Keratinocyte Growth Supplement (HKGK) adjusted keratinocytes (HEKa) to 1 × 106Cells/well were seeded in 6-well plates and cultured for 18 hours. After scraping off a part of the cells, the samples of the above examples and comparative examples were treated and cultured for 8 hours. The cell regeneration rate was calculated by measuring the wound gap (wound gap) using the following formula, and the results are shown in table 9 below. As a control group, madecassoside (madecassoside) was used.

As confirmed from the above Table 9, the examples 1 to 17, which are the mixed extracts of immature fruits according to the present invention, show more excellent cell regeneration effect by synergistic effect than the comparative examples 1 to 10, which are the control group and the respective extracts of immature fruits, when treated at the same concentration.
Examples 18 to 24: preparation of emulsion containing mixed unripe fruit extracts
Emulsions containing the mixed unripe fruit mixed extracts prepared in the above examples (examples 11 to 17) were prepared according to a conventional method with the compositions and contents shown in table 10 below.

Test example 8: confirmation of skin irritation alleviating Effect
The degree of sedation caused by the stimulation was relatively evaluated by applying the emulsions containing examples 18 to 24 and the emulsions containing the comparative examples described above after inducing skin irritation in the inner part of the lower wrist by a Tape Stripping Method (Tape striping Method) on 10 male and female subjects having no skin allergy symptoms at present, and by using the emulsions containing examples 18 to 24. First, skin erythema was induced by applying scotch tape 30 times to both sides of the inner part of the lower wrist. After the induction of erythema, the cosmetic containing the mixed unripe fruit extract of the present invention was applied, and the emulsion of the control example not containing the mixture of the present invention was applied. After 3 hours of application, the degree of skin irritation was evaluated.

As confirmed in the above 11, the emulsion containing the mixed extract of unripe fruits according to the present invention has an excellent skin irritation-relieving effect.
Claims (7)
1. A cosmetic composition comprising a mixed extract of unripe orange, unripe apple, unripe tomato, and one kind of unripe fruit selected from the group consisting of unripe strawberry, unripe banana, unripe lemon, unripe persimmon, unripe fig, unripe date, and unripe plum as an active ingredient.
2. The cosmetic composition according to claim 1, wherein the mixed extract is contained in an amount of 0.01 to 90 wt% based on the total weight of the cosmetic composition.
3. The cosmetic composition according to claim 1, wherein the mixed extract is prepared by mixing the following components in an amount of 1 to 2: 1-2: 1-2: 1-2 weight ratio of green orange, green apple, green tomato and one kind of green fruit selected from green strawberry, green banana, green lemon, green persimmon, green fig, green jujube and green plum.
4. The cosmetic composition of claim 1, wherein said cosmetic composition is used for improving skin wrinkles.
5. The cosmetic composition of claim 1, wherein said cosmetic composition is used for improving skin elasticity.
6. The cosmetic composition of claim 1, wherein said cosmetic composition is used for skin cell regeneration.
7. The cosmetic composition of claim 1, wherein said cosmetic composition is for alleviating skin irritation.
