Technical Field

The western medicine theory considers that: one of the important causes of acne is the massive proliferation of propionibacterium acnes and other microorganisms, the production of lysolipases, the breakdown of sebum, the production of free acids, the resulting damaged rupture of the hair follicle wall, the entry of accumulated sebum into the surrounding dermal tissue, and the consequent clinical manifestations of lesions ranging from inflammatory papules to cysts.
Acne is a disease difficult to be cured radically, and needs to be controlled by long-term use of medicines, some products can be added with harmful components such as metronidazole, chloramphenicol, clindamycin and the like, and the antibiotics have remarkable treatment effect on the acne, but have great influence on skin after long-term use. The traditional Chinese medicine treatment mainly adopts traditional Chinese medicines such as scutellaria baicalensis, coptis chinensis, honeysuckle, witch hazel and the like, and is matched with certain active ingredients for oral administration and external application; the western medicine treatment mainly uses chemical substances such as hormones, antibiotics and the like, and the skin of a patient is greatly damaged by long-term use, so that the skin is stimulated and damaged. The living standard of people is continuously improved, the demand on cosmetics is increased, the demands on safety, efficacy and quality of products are gradually improved, and the traditional Chinese medicine cosmetics have small toxic and side effects, mild effects, stability and safety and are favored by more consumers. The comparative patent cn201410298553.x discloses a cosmetic containing aloe vera extract, natto extract and asiaticoside components, but the cosmetic has a weak inhibitory effect on propionibacterium acnes.

The acne-removing and allergy-relieving composition is a multi-component mixture consisting of a centella extract, a bletilla extract, a honeysuckle extract, a algal polysaccharide extract, ascorbic acid, nicotinamide, a witch hazel extract, D-panthenol, potassium glycyrrhetate, bisabolol, tea tree oil and sodium hyaluronate.
An acne-removing allergy-relieving gel is prepared from the following raw materials in percentage by weight:
0.05-0.1% of centella asiatica extract, 0.05-0.1% of bletilla striata extract, 0.05-0.1% of honeysuckle extract, 0.05-0.1% of algal polysaccharide extract, 0.1-1% of ascorbic acid, 1-3% of nicotinamide, 0.1-1% of hamamelis virginiana extract, 0.1-0.5% of D-panthenol, 0.1-0.2% of potassium glycyrrhetinate, 0.1-0.2% of bisabolol, 0.1-0.2% of tea tree oil, 0.1-0.24% of sodium hyaluronate, 0.5-1% of carbomer, 0.5-1% of triethanolamine, 10-20% of glycerol, 0.1-0.2% of methylparaben and 71.06-87% of deionized water.
The preparation method of the acne-removing and allergy-relieving gel provided by the invention comprises the following steps:
(1) putting the centella asiatica extract, the bletilla striata extract, the honeysuckle extract and the algal polysaccharide extract into a 100mL beaker according to the weight percentage, adding water, stirring until the mixture is dissolved, and filtering to obtain a traditional Chinese medicine extract solution;
(2) adding appropriate amount of water into ascorbic acid, nicotinamide, Hamamelis virginiana extractive solution and D-panthenol, and heating for dissolving; sequentially adding potassium glycyrrhizate, bisabolol and tea tree oil, and continuously stirring for dissolving; adding the traditional Chinese medicine extract solution prepared in the step (1), stopping heating and stirring after the traditional Chinese medicine extract solution is completely dissolved, slowly adding sodium hyaluronate, stirring for dissolving, standing and cooling, and adding methyl paraben to prepare a water phase;
(3) mixing carbomer with water according to the weight percentage, and swelling for 24 hours; after complete swelling, slowly adding triethanolamine dropwise, and stirring continuously to obtain transparent colloid; slowly adding glycerol, and stirring to obtain gel matrix;
(4) and (3) mixing the water phase prepared in the step (2) with the gel matrix prepared in the step (3), uniformly stirring, dropwise adding triethanolamine to adjust the pH value to 6-7, and adding water to a constant volume to obtain the gel.
The glycerin used in the invention has the effects of moistening skin, protecting skin and reducing water loss.
The triethanolamine used in the invention is neutralized with carboxyl of carbomer to form a stable macromolecular structure, so as to achieve the effects of thickening and moisturizing and have moisturizing and oil controlling effects on skin.
The ascorbic acid used in the invention can resist oxidation, improve the immunity of human bodies and improve the emergency protection capability of organisms.
The niacinamide used in the invention can prevent and treat the pellagra, has strong whitening and moisturizing effects, improves the whelk symptom, reduces skin inflammation and acne, and is a skin care component with high safety.
The witch hazel extract specially added in the invention can alleviate and stimulate skin, does not cause skin dryness, is deep and clean, enables skin to be more young and bright, effectively regulates pore relaxation, prevents and treats acne, has unique effects of aging resistance and the like.
The dipotassium glycyrrhizinate used in the invention can prevent allergy, resist bacteria, diminish inflammation, repair skin wound, inhibit bacterial growth and reduce free radicals, and has the characteristics of mild action, safety, stability and the like.
D-panthenol used in the present invention can promote epithelial tissue growth and wound healing as well as anti-inflammation.
The bisabolol used in the invention can resist bacteria, diminish inflammation, relieve itching and resist allergy, and is suitable for being added into cosmetics.
The sodium hyaluronate used in the invention can care skin, keep skin moist and glossy, and has the effects of moisturizing, nourishing, repairing and preventing skin injury, lubricating skin and forming film.
The tea tree oil used in the invention can diminish inflammation, inhibit bacteria, relieve itching, remove acnes, relieve allergy, keep the moisture and oil content balance of skin, promote the growth of tissue cells, help repair damaged cells and keep the skin moist and glossy.
The centella asiatica extract used in the invention has the effects of removing pockmarks, inhibiting bacteria, resisting oxidation, whitening and the like;
the bletilla striata extract used in the invention has the effects of stopping bleeding, resisting oxidation and aging, repairing damaged skin, removing pockmarks and pox marks, inhibiting bacterial growth and the like;
the honeysuckle extract used in the invention has the effects of resisting bacteria and diminishing inflammation, repairing damaged skin, fading scars and the like.
The algal polysaccharide used in the invention can be combined with skin protein to form a moisture-retaining gel, and can also form a protective film on the surface of the skin through the cooperation of water to prevent water evaporation.
Has the advantages that: the invention screens out the acne-removing and allergy-relieving component consisting of multiple components through experiments, and the experimental results show that the acne-removing and allergy-relieving component provided by the invention adopts 12 active components in the optimal weight ratio for compatibility and combination, has obvious acne-removing and allergy-relieving effects, and particularly has a good acne-inhibiting effect. The compatibility of all the components plays a good role in synergy.
In particular, the screening of the components shows that when the effective components are deficient in centella asiatica extract, hamamelis virginiana extract and tea tree oil components, the anti-propionibacterium acnes and the anti-allergic effect are obviously weakened.
According to the acne-removing and allergy-relieving composition, the acne-removing and allergy-relieving composition is prepared into a gel, and components dissolved in a water phase are dissolved in a gel matrix and can be slowly released, so that the effects of removing acne, relieving allergy and the like can be maintained for a long time.

The preparation method of the acne-removing and allergy-relieving gel provided by the invention comprises the following steps:
(5) putting the centella asiatica extract, the bletilla striata extract, the honeysuckle extract and the algal polysaccharide extract into a 100mL beaker according to the weight percentage, adding water, stirring until the mixture is dissolved, and filtering to obtain a traditional Chinese medicine extract solution;
(6) adding appropriate amount of water into ascorbic acid, nicotinamide, Hamamelis virginiana extractive solution and D-panthenol, and heating for dissolving; sequentially adding potassium glycyrrhizate, bisabolol and tea tree oil, and continuously stirring for dissolving; adding the traditional Chinese medicine extract solution prepared in the step (1), stopping heating and stirring after the traditional Chinese medicine extract solution is completely dissolved, slowly adding sodium hyaluronate, stirring for dissolving, standing and cooling, and adding methyl paraben to prepare a water phase;
(7) mixing carbomer with water according to the weight percentage, and swelling for 24 hours; after complete swelling, slowly adding triethanolamine dropwise, and stirring continuously to obtain transparent colloid; slowly adding glycerol, and stirring to obtain gel matrix;
(8) and (3) mixing the water phase prepared in the step (2) with the gel matrix prepared in the step (3), uniformly stirring, dropwise adding triethanolamine to adjust the pH value to 6-7, and adding water to a constant volume to obtain the gel.
The extraction methods of the centella asiatica extract, the bletilla striata extract, the honeysuckle extract and the algal polysaccharide extract used in the following examples are respectively as follows:
centella asiatica extract: cutting herba Centellae into segments, extracting with 75% ethanol under reflux for 2 times, adding 12 times of water for 1 time, and extracting for 1 hr; adding 10 times of water for the 2 nd time, and extracting for 1 h; filtering with three layers of gauze each time, mixing filtrates, concentrating the filtrate under reduced pressure until no alcohol smell is produced, and adding appropriate amount of water to disperse. Extracting with n-butanol for 2 times, mixing n-butanol layers, and concentrating under reduced pressure to dry to obtain herba Centellae extract.
And (3) common bletilla pseudobulb extract: slicing rhizoma Bletillae, adding water, reflux-extracting for 2 times, adding 12 times of water for 1 time, and extracting for 1.5 hr; adding 8 times of water for 2 times, and extracting for 1.5 hr; filtering with three layers of gauze each time, mixing filtrates, and concentrating the filtrate under reduced pressure to dry to obtain rhizoma Bletillae extract.
And (3) honeysuckle extract: reflux-extracting flos Lonicerae decoction pieces with water for 2 times, adding 15 times of water for 1 time, and extracting for 1.5 hr; adding 8 times of water for 2 times, and extracting for 1.5 hr; filtering with three layers of gauze each time, mixing filtrates, and concentrating the filtrate under reduced pressure to dry to obtain flos Lonicerae extract.
Algal polysaccharide extract: extracting Sargassum decoction pieces with water under reflux for 2 times, and extracting with 15 times of water for 1.5 hr for 1 time; adding 10 times of water for 2 times, and extracting for 1.5 hr; filtering with three layers of gauze each time, mixing filtrates, concentrating the filtrate under reduced pressure to material-liquid ratio of 1/10, adding ethanol until the alcohol concentration is 75%, standing overnight, and filtering to obtain polysaccharide crude extract. Separating and purifying the crude polysaccharide extract with Sephadex-G100 gel chromatography to obtain algal polysaccharide extract.
Example 1
The preparation method of the acne-removing and allergy-relieving gel comprises the following steps:
step (1): preparing a traditional Chinese medicine extract solution, namely putting 0.05 percent of centella extract, 0.05 percent of bletilla striata extract, 0.05 percent of honeysuckle extract and 0.05 percent of algal polysaccharide extract in a 100mL beaker by mass, adding water, stirring until the mixture is dissolved, and filtering to obtain a filtrate for later use;
step (2): taking 0.5% ascorbic acid, 1% nicotinamide, 0.5% hamamelis virginiana extract and 0.1% D-panthenol in percentage by mass, adding a proper amount of water, and heating for dissolving; then sequentially adding 0.1% of potassium glycyrrhizate, 0.1% of bisabolol and 0.1% of tea tree oil, and continuously stirring for dissolving; adding the traditional Chinese medicine extract solution obtained in the step (1), stopping heating and stirring after the traditional Chinese medicine extract solution is completely dissolved, slowly adding 0.1% sodium hyaluronate, stirring for dissolving, standing and cooling, and adding 0.1% methyl paraben to obtain a water phase;
and (3): mixing 0.5% carbomer with water, and swelling for 24 hr; after complete swelling, slowly dripping 0.5 percent triethanolamine, and continuously stirring to form transparent colloid; slowly adding 10% glycerol, and stirring to obtain gel matrix;
and (3) mixing the water phase prepared in the step (2) with the gel matrix prepared in the step (3), uniformly stirring, dropwise adding triethanolamine to adjust the pH value to 6, and adding water to a constant volume to obtain the gel.
Example 2
The preparation method of the acne-removing and allergy-relieving gel comprises the following steps:
step (1): dissolving the traditional Chinese medicine extract, namely putting 0.08 percent of centella extract, 0.08 percent of bletilla extract, 0.08 percent of honeysuckle extract and 0.08 percent of algal polysaccharide extract in a 100ml beaker according to mass percentage, adding water, stirring until the mixture is dissolved, and filtering to obtain filtrate for later use;
step (2): taking 0.8% ascorbic acid, 2% nicotinamide, 0.8% hamamelis virginiana extract and 0.3% D-panthenol in percentage by mass, adding a proper amount of water, and heating for dissolving; then sequentially adding 0.2% of potassium glycyrrhizate, 0.2% of bisabolol and 0.2% of tea tree oil, and continuously stirring for dissolving; adding the Chinese medicinal extract solution, stopping heating and stirring after the Chinese medicinal extract solution is completely dissolved, slowly adding 0.2% sodium hyaluronate, stirring for dissolving, standing for cooling, and adding 0.2% methyl hydroxybenzoate to obtain water phase;
and (3): mixing 0.8% carbomer with water, and swelling for 24 hr; after complete swelling, slowly dripping 0.8% triethanolamine, and continuously stirring to obtain transparent colloid; slowly adding 15% glycerol, and stirring to obtain gel matrix;

Example 3 preparation of Multi-component acne removing and allergy relieving gel

The preparation method of the acne-removing and allergy-relieving gel comprises the following steps:
step (1): dissolving the traditional Chinese medicine extract, namely putting 0.08 percent of bletilla striata extract, 0.08 percent of honeysuckle extract and 0.08 percent of algal polysaccharide extract in a 100ml beaker by mass, adding water, stirring until the mixture is dissolved, and filtering to obtain filtrate for later use;
step (2): taking 0.8 percent of ascorbic acid, 2 percent of nicotinamide and 0.3 percent of D-panthenol by mass percent, adding a proper amount of water, and heating for dissolving; then sequentially adding 0.2% of potassium glycyrrhizate and 0.2% of bisabolol, and continuously stirring for dissolving; adding the Chinese medicinal extract solution, stopping heating and stirring after the Chinese medicinal extract solution is completely dissolved, slowly adding 0.2% sodium hyaluronate, stirring for dissolving, standing for cooling, and adding 0.2% methyl hydroxybenzoate to obtain water phase;
and (3): mixing 0.8% carbomer with water, and swelling for 24 hr; after complete swelling, slowly dripping 0.8% triethanolamine, and continuously stirring to obtain transparent colloid; slowly adding 15% glycerol, and stirring to obtain gel matrix;
and (3) mixing the water phase prepared in the step (2) with the gel matrix prepared in the step (3), uniformly stirring, dropwise adding triethanolamine to adjust the pH value to 7, and adding water to a constant volume to obtain the gel.
Example 4 acne treatment screening test: inhibition of Propionibacterium acnes
Sample preparation: the samples prepared in example 1, example 2 and comparative example 1 are taken as 1g each, and the comparative patent 201410298553X cosmetic is 2g, and the salicylic acid single component is 20 mg. A certain amount of sample is taken and respectively added into 50mL of deionized water or 50mL of 1, 3-butanediol, and the mixture is uniformly mixed for later use.

Inoculation of LB plate medium: dipping bacteria liquid with sterilized cotton stick, squeezing off excess bacteria liquid on tube wall, uniformly coating on LB plate culture medium, sweeping a circle along the edge of the plate, covering the plate, and drying for 2-3 min. Measuring the diameter of the bacteriostatic zone: taking an average value of diameter values of 3 groups of inhibition zones, and grading the antimicrobial susceptibility: the diameter of the antibacterial ring is more than 20mm, the diameter is high sensitivity, the diameter is 10-20mm, the diameter is medium sensitivity, and the diameter is less than 10mm, the diameter is drug resistance.

Example 5 anti-allergy screening assay
Taking mouse mastocytoma cells in logarithmic growth phase, and counting 0.6 × 105Each well was suspended in 10% serum-containing RPMI-1640 medium, and the suspension was inoculated into a 24-well plate in a volume of 1 mL.
