肽对人的生老病死起着决定性的作用。肽是调节体内水、电解质平衡的重要物质,因而肽是维持细胞正常形态,保持维持细胞功能的基础;肽能为免疫系统制造对抗细菌、病毒的抗体,提高机体免疫力;肽在体内能参与酶代谢,促进机体代谢生长,促进伤口愈合,修复机体保持健康。小分子肽除了以上各种功能外还能透过血脑、胎盘屏障等人体保护性结构,营养修复人体大脑、胎儿等一般营养物质不能到达的组织器官或机体,对人体机能起到更好的修复和改善作用。
发明内容
本发明的目的在于提供一种富肽复方制剂及其制备方法,将富里酸与小分子肽协同,制备出一种富(源自富里酸)肽(源自蛋白肽粉)复方制剂及保健食品,从而对人体形态和机能调理康复起到安全、高效的作用。
为了实现上述目的,本发明采用以下技术方案:
本发明提供了一种富肽复方制剂,包括富里酸及蛋白肽粉。
优选地,所述富里酸与蛋白肽粉的重量比为4:1。
本发明还提供了一种富肽保健食品,包括上述富肽复方制剂。
本发明还提供了一种富肽复方制剂的制备方法,包括:
1)制备富里酸,又包括以下步骤:
1-4)将刺梨、复合微生物活菌剂及发酵配料混合均匀后,装入容器中,28-35℃,发酵8-15天;
1-5)固液分离,滤渣挤压出汁;
1-6)将分离液与压出的汁混合后,静置沉淀,取上清液,得富里酸;
2)制备蛋白肽粉,又包括以下步骤:
2-1)分别将大豆和核桃仁洗净、加水泡发后,磨成糊状备用;
2-2)将步骤2-1)准备好的两种糊状原料混合后加入玉米粉、糖和酶菌合剂,混合均匀,置于27-35℃、15天以上,完成水解;
2-3)生鸡蛋去壳后装入容器中搅拌均匀,再加入玉米粉和糖,最后加入酶菌合剂,置于27-35℃中,发酵20天以上;
2-4)将步骤2-2)获得的水解产物通过胶体磨研磨后与步骤2-3)获得的发酵产物混合,均质;
2-5)将步骤2-4)所得产物烘干,粉碎,过筛,得蛋白肽粉;
3)将步骤1)制备的富里酸和步骤2)制备的蛋白肽粉混合,得富肽复方制剂。
优选地,步骤1-1)中,所述发酵配料包括氮原、碳源、无机盐和水;每100公斤水和刺梨中加入1-3g复合微生物活菌剂。
更优选地,所述复合微生物菌剂包括菌种和玉米芯粉,所述菌种包括乳杆菌、酿酒酵母和枯草芽孢杆菌,所述菌种与玉米芯粉的重量比为30-40:60-70,乳杆菌、酿酒酵母和枯草芽孢杆菌的重量比为1:1:1。
优选地,步骤2-2)中,所述酶菌合剂中,酶包括纤维素酶、木聚糖酶和蛋白酶,菌种包括枯草芽孢杆菌、酿酒酵母和乳酸菌,每1g酶菌合剂中,枯草芽孢杆菌≥5.6×107cfu,乳酸菌≥4×106cfu,酿酒酵母≥3.0×107cfu,纤维素酶200U,木聚糖酶2000U,蛋白酶6000U;大豆糊状原料和核桃仁糊状原料按重量比9:1混合,两种糊状原料混合后的总重量与玉米粉、糖和酶菌合剂的重量比为10kg:2kg:200g:3g。
优选地,步骤2-3)中,生蛋液与玉米粉、糖和酶菌合剂的重量比为5000:1500:150:3。
优选地,步骤2-4)中,通过胶体磨研磨至细度为60-100μm;步骤2-2)获得的水解产物研磨后与步骤2-3)获得的发酵产物混合重量比为7:3。
优选地,步骤2-5)中,釆用低温烘干机50℃烘干,粉碎后过100目筛。
本发明所制备的富肽复方制剂集富里酸和小分子肽的理化优点,充分利用二者对人体的作用机理,趋利避害注重安全和有效性,着力开辟富里酸和小分子肽在医药保健应用领域的崭新广阔前景。富里酸与肽的复合制剂能够起到协同作用,从而对人体形态和机能调理康复起到安全、高效的作用。本发明在当前你生物科技应用中具有领先科技水平。
具体实施方式
为了更清楚的描述本发明,现结合具体实施例做进一步描述。可以理解的是,所述实施例为示例性说明,而非用作对本发明的限定。
实施例
(一)富里酸的制备
本制备流程中采用的原料如下:
I.刺梨(品种不限,以无籽刺梨为好)洗净、粉碎,备用。
II.复合微生物活菌剂:由乳杆菌、酿酒酵母和枯草芽孢杆菌混合而成,三个菌种均采用现有市售产品,菌落规格(CFU/g)均为2×107,重量配比为1:1:1;为了利于菌种生长,优选采用玉米芯粉作为载体。
III.配料:氮原(用豆粕或尿素)、碳源(蔗糖和/或玉米粉)、无机盐(食盐或食品级磷酸二氢钾)和水。按100公斤(水和剌梨的重量)为例:豆粕3公斤(或尿素0.5公斤),蔗糖2.5公斤、玉米粉5公斤、食品级磷酸二氢钾150克。以上配料亦均为现有市售产品。另,由于刺梨中维生素C含量较高,可省略通常作为生长因子的维生素C。
具体步骤如下:
1、将乳杆菌、酿酒酵母和枯草芽孢杆菌按照1:1:1的重量比混合后,取35g混合菌种与65g玉米芯细粉混合,得复合微生物活菌剂。
2、将40公斤刺梨洗净、粉碎后装入食品级塑料桶中,加水60公斤,加蔗糖2.5公斤,玉米粉5公斤,豆粕3公斤(要求细粉),食品级磷酸二氢钾150克,配合2g复合微生物活菌剂混合均匀后,盖紧盖子,桶上留三分之一空间以防发酵溢出,室温保持在28-35℃,经过8-15天发酵即可。
3、固液分离,发酵好后用纱布过滤,约得分离液58公斤,滤渣挤压出汁,约得22公斤(取决于压汁是否干)。
4、将58公斤分离液与22公斤压出的汁混合后,静置沉淀36小时,取上清液78公斤,既为可直接饮用含富里酸产品。
该工艺流程采用刺梨作发酵底物结合一种复合微生物活菌剂发酵产生富里酸。用刺梨生产的富里酸,除具有富里酸的保健价值外,还有刺梨的保健价值,所产生的刺梨富里酸的保健作用远大于单独使用富里酸和刺梨,具有很高的保健价值,产品可直接饮用,无需分离,是很好的保健产品和功能饮品的主要成份。且工艺简单、生产成本低,无有害物体排出,环保节能。
(二)蛋白肽粉的制备
本制备流程中,原料主要采用鸡蛋,优选采用鲜鸡蛋,配以少量核桃仁、大豆、玉米粉、糖和水。其中,“糖”可选用葡萄糖或蔗糖。
菌种选用枯草芽孢杆菌、酿酒酵母、乳酸菌,酶选用蛋白酶、纤维素酶、木聚糖酶,菌种和酶均为现有市售产品,可直接购买,只要混合后每1g酶菌合剂中菌种和酶达到以下规格即可:枯草芽孢杆菌≥5.6×107cfu,乳酸菌≥4×106cfu,酿酒酵母3.0×107cfu,纤维素酶200U,木聚糖酶2000U,蛋白酶6000U。
具体步骤如下:
1、将大豆洗净、水泡发后,用磨浆机磨成糊状备用;核桃仁洗净、水泡发后,用磨浆机磨成糊状备用,料水比均优选1:1.5,磨浆机型号不限。
2、将准备好的上述两种原料按90:10斤比例混合后加入玉米粉20斤、蔗糖2斤,再加入15g酶菌合剂,混合均匀。该步骤中,当微生物发酵产热时,复合酶就发生水解作用,蛋白质底物在微生物分泌的酶和加入的酶的共同作用下水解底物,蛋白质在酶的水解作用下发生断裂变成不同分子量的肽。一般27-35℃室温中15天以上即可完成水解。
3、取鲜鸡蛋破壳装入容器中搅拌均匀,以100斤生蛋液为例,再加入玉米粉30斤、蔗糖3斤,最后加入30g酶菌合剂,置室温约30℃中,发酵20天以上。
4、将第2步水解完成得到的产物通过胶体磨磨一次至细度80μm后,与第3步得到的产物按重量比例7:3混合匀质后进行下一步。
5、将第4步得到的混合物釆用低温烘干机50℃烘干,粉碎,过100目筛,得蛋白肽粉。
该工艺流程采用微生物发酵技术和酶技术结合的方法,将高品质的大分子球蛋白分解转化为多肽,小肽和部份氨基酸,从而获得不同氨基酸排序和分子量不同的肽。具体过程是通过微生物的代谢作用和酶的剪切作用,对氨基酸和肽链进行移接和重排,使肽链基团修饰和重组,从而获得不同分子量的肽。其中所含的小肽且能有效透过血脑屏障等人体保护性机制,
(三)富肽复方制剂
取(一)制得的富里酸80g加(二)制得的蛋白肽粉20g,混匀,即能配伍成富(富里酸)肽(蛋白肽粉)复方制剂。
基于以上富肽复方制剂,本领域技术人员可根据不同类型的保健食品需求,制备成富肽保健食品。
需要说明的是,本申请中,如未有特殊说明,所用材料及工艺均可采用现有常规技术手段来实现,申请人对此不做具体限定。
Claims (10)
1.一种富肽复方制剂,包括富里酸及蛋白肽粉。
2.根据权利要求1所述的一种富肽复方制剂,其特征在于,所述富里酸与蛋白肽粉的重量比为4:1。
3.一种富肽保健食品,包括权利要求1或2所述的富肽复方制剂。
4.一种富肽复方制剂的制备方法,包括:
1)制备富里酸,又包括以下步骤:
1-1)将刺梨、复合微生物活菌剂及发酵配料混合均匀后,装入容器中,28-35℃,发酵8-15天;
1-2)固液分离,滤渣挤压出汁;
1-3)将分离液与压出的汁混合后,静置沉淀,取上清液,得富里酸;
2)制备蛋白肽粉,又包括以下步骤:
2-1)分别将大豆和核桃仁洗净、加水泡发后,磨成糊状备用;
2-2)将步骤2-1)准备好的两种糊状原料混合后加入玉米粉、糖和酶菌合剂,混合均匀,置于27-35℃、15天以上,完成水解;
2-3)生鸡蛋去壳后装入容器中搅拌均匀,再加入玉米粉和糖,最后加入酶菌合剂,置于27-35℃中,发酵20天以上;
2-4)将步骤2-2)获得的水解产物通过胶体磨研磨后与步骤2-3)获得的发酵产物混合,均质;
2-5)将步骤2-4)所得产物烘干,粉碎,过筛,得蛋白肽粉;
3)将步骤1)制备的富里酸和步骤2)制备的蛋白肽粉混合,得富肽复方制剂。
5.根据权利要求4所述的一种富肽复方制剂的制备方法,其特征在于,步骤1-1)中,所述发酵配料包括氮原、碳源、无机盐和水;每100公斤水和刺梨中加入1-3g复合微生物活菌剂。
6.根据权利要求4或5所述的一种富肽复方制剂的制备方法,其特征在于,所述复合微生物菌剂包括菌种和玉米芯粉,所述菌种包括乳杆菌、酿酒酵母和枯草芽孢杆菌,所述菌种与玉米芯粉的重量比为30-40:60-70,乳杆菌、酿酒酵母和枯草芽孢杆菌的重量比为1:1:1。
7.根据权利要求4所述的一种富肽复方制剂的制备方法,其特征在于,步骤2-2)中,所述酶菌合剂中,酶包括纤维素酶、木聚糖酶和蛋白酶,菌种包括枯草芽孢杆菌、酿酒酵母和乳酸菌,每1g酶菌合剂中,枯草芽孢杆菌≥5.6×107cfu,乳酸菌≥4×106cfu,酿酒酵母≥3.0×107cfu,纤维素酶200U,木聚糖酶2000U,蛋白酶6000U;大豆糊状原料和核桃仁糊状原料按重量比9:1混合,两种糊状原料混合后的总重量与玉米粉、糖和酶菌合剂的重量比为10kg:2kg:200g:3g。
8.根据权利要求4所述的一种富肽复方制剂的制备方法,其特征在于,步骤2-3)中,生蛋液与玉米粉、糖和酶菌合剂的重量比为5000:1500:150:3。
9.根据权利要求4所述的一种富肽复方制剂的制备方法,其特征在于,步骤2-4)中,通过胶体磨研磨至细度为60-100μm;步骤2-2)获得的水解产物研磨后与步骤2-3)获得的发酵产物混合重量比为7:3。
10.根据权利要求4所述的一种富肽复方制剂的制备方法,其特征在于,步骤2-5)中,釆用低温烘干机50℃烘干,粉碎后过100目筛。
