Abstract
本发明公开了一种含有石斛多糖的口腔速溶膜剂及其制备方法,包含的组分及各组分的份数如下:芦荟多糖13~16份,铁皮石斛多糖11~14份,成膜材料36~45份,保水剂6~39份,水2~10份。本发明的口腔速溶膜剂将石斛多糖和芦荟多糖复配,所使用的石斛多糖的相对分子量范围为10万~200万,芦荟多糖的相对分子量范围为0.5万~40万,因此口腔速溶膜剂中的多糖的分子量分布范围更广,对免疫的调节效果优于单一的芦荟多糖与石斛多糖。

技术领域
本发明涉及一种保健品,具体涉及一种含有石斛多糖的口腔速溶膜剂及其制备方法。
背景技术
石斛是兰科(Orchidaceae)石斛属(Dendrobium)多年生草本植物,是一种名贵稀有的中药材,素有“人间仙草”之称。医学经典《道藏》中称铁皮石斛为中华九大仙草之首,民间将其称为救命仙草。铁皮石斛Dendrobium officinale,俗称铁皮枫斗,为我国特有的名贵药材。《本草纲目》、《中药大词典》等多部中国古代医学著作中都有铁皮石斛入药的记载。现代生物技术分析表明,石斛多糖和生物碱是石斛兰中最重要的药用有效成分。
石斛多糖具有较好的保健功能。石斛多糖对人体健康有免疫调节作用。人体的免疫调节作用是大多数活性多糖的共同作用,石斛茎中的黏性物质主要是水溶性多糖,它是一类免疫增强剂,临床实验证明石斛多糖对调节机体免疫力有非常显著的作用。石斛多糖对人体健康有降血糖作用。石斛能显著降低机体的血糖值,促进胰岛素分泌和抑制胰高血糖素分泌,且石斛多糖对胰岛细胞具有修复作用;同时石斛多糖还具有胰外降血糖的作用,通过抑制肾上腺素引起的肝糖原分解和促进肝糖原合成,从而可减少血糖的来源,增加血糖的去路,达到降低血糖值的目的。石斛多糖对人体健康有抗氧化、抗衰老作用。石斛活性多糖具有清除自由基、提高抗氧化酶活性和抑制脂质过氧化的活性,起到保护生物膜和延缓衰老的作用。
芦荟提取物是医药、化工、食品方面的重要原料,芦荟多糖是芦荟提取物中主要功效成分之一。芦荟多糖能够提高人体的免疫功能和抗辐射作用,且对于抗癌症、抗艾滋病病毒,有一定的药理治疗作用。
芦荟多糖中主要是乙酰化甘露聚糖,其具有增强免疫力、防治癌症、愈合创伤、抗疾病和抗辐射作用,乙酰化甘露聚糖也称为Acemannan 多糖,乙酰化甘露聚糖是一种白色非晶型粉末,基本上是由线性的β(1-4)-D-甘露糖单元组成的一种长链聚合物,可缓慢地溶解于水形成具有高粘性的胶溶液。
口腔速溶膜剂(也可叫膜片)是一种新型的营养剂形式,从外形看,形状类似于长方形薄片状口香糖,其入口即化,方便携带,入口方式适用于各种人群。
发明内容
本发明所要解决的技术问题是提供一种含有石斛多糖的口腔速溶膜剂及其制备方法。
实现本发明第一目的的技术方案是一种含有石斛多糖的口腔速溶膜剂,包含的组分及各组分的份数如下:芦荟多糖 13~16份,铁皮石斛多糖11~14份,成膜材料36~45份,保水剂6~39份,水2~10份。
口腔速溶膜剂中还包括甜味剂20~25份和/或酸味调节剂1.2~1.5份。
芦荟多糖的相对分子量范围0.5~40万,O-乙酰基含量为20mg/g~50mg/g。
作为优选的,芦荟多糖的相对分子量范围1~30万,O-乙酰基含量为40mg/g~50mg/g。
铁皮石斛多糖的相对分子量范围10万~200万、O-乙酰基含量为10mg/g~40mg/g。
作为优选的,铁皮石斛多糖的相对分子量范围100万~200万、O-乙酰基含量为25mg/g~35mg/g。
所述成膜材料为普鲁兰多糖、甲基纤维素、聚乙二醇、聚乙烯醇、葡聚糖中的一种或一种以上的组合。
所述保水剂为甘油和/或二甘醇。
芦荟多糖的相对分子量范围10~40万,O-乙酰基含量为40mg/g~50mg/g。
实现本发明第二目的的技术方案是一种含有石斛多糖的口腔速溶膜剂的制备方法,包括以下步骤:
①配制涂膜溶液;涂膜溶液中包含的组分及各组分的份数为芦荟多糖 13~16份,铁皮石斛多糖11~14份,成膜材料36~45份, 保水剂6~39份,水72~88份。
②涂膜;开启涂布机,使涂布刀里充满步骤①配制的涂膜溶液,随着薄膜基材的运动,由涂布刀将涂膜溶液均匀的涂覆在薄膜基材上。
③烘干;涂覆了涂膜溶液的薄膜基材行进至烘干区域,在68℃~77℃下烘干去除水分,得到膜剂,烘干后的膜剂和薄膜基材一起收卷。
④分切包装;将步骤③成卷的膜剂和薄膜基材分切、再放卷切割后包装。
配制涂膜溶液时,将称取的芦荟多糖、铁皮石斛、成膜材料、保水剂溶解于水中后,将得到的混合溶液于3~5℃静置8~12h,取出,放至室温后得到涂膜溶液待用。
所配制的涂膜溶液的特性黏数为30~70mL/g。
步骤②涂膜时的涂膜速度0.16~0.20m/min。
本发明具有积极的效果:
(1)本发明的口腔速溶膜剂将石斛多糖和芦荟多糖复配,所使用的石斛多糖的相对分子量范围为10万~200万,芦荟多糖的相对分子量范围为0.5万~40万,因此口腔速溶膜剂中的多糖的分子量分布范围更广,对免疫的调节效果优于单一的芦荟多糖与石斛多糖。
(2)膜剂中各组分的含量关系到速溶膜剂成品的合格率,成品膜剂软硬适中:硬度太低,与包装隔离纸黏附,食用时不便取出;硬度过高则易折断。本发明在制备含有乙酰化甘露聚的口腔速溶膜剂时,合理配置各组分的含量,从而在涂膜、切割、包装后,成品率达到合理、可接受的范围。
(3)进一步的,不同种芦荟、即便是同一种芦荟,因生长环境、产地的不同,生长时所受的光照、温度以及其他环境影响,其中的多糖的分子量还是会有所差异,导致黏度及表面张力也有所不同,所以配制涂膜溶液时各物料比例要做出相应调整,保证成品率。本发明对石斛多糖和芦荟多糖的指标作了要求,用符合指标的石斛多糖和芦荟多糖,得到了与包材剥离效果好、软硬适中的速溶膜剂。

下面介绍的是本发明的多个可能实施例中的一些,旨在提供对本发明的基本了解,并不旨在确认本发明的关键或决定性的要素或限定所要保护的范围。容易理解,根据本发明的技术方案,在不变更本发明的实质精神下,本领域的一般技术人员可以提出可相互替换的其他实现方式。因此,以下具体实施方式仅是对本发明的技术方案的示例性说明,而不应当视为本发明的全部或者视为对本发明技术方案的限定或限制。
本发明的含有石斛多糖的口腔速溶膜剂包含的组分及各组分的份数如下:芦荟多糖 13~16份, 铁皮石斛多糖11~14份,成膜材料36~45份, 保水剂6~39份,水2~10份。
作为优选的,为了提升口腔速溶膜剂的口感,还可以加入甜味剂20~25份和\或酸味调节剂1.2~1.5份。
所述甜味剂为食品级甜味剂,例如是木糖醇、阿斯巴甜、三氯蔗糖、安赛蜜中的一种或一种以上的组合。除了上述提及的甜味剂,不排他使用其他食用级甜味剂。
所述酸味调节剂为食品级酸味调节剂,例如苹果酸、柠檬酸、酒石酸、烟酸中的一种或一种以上的组合。除了上述例举的酸味调节剂,不排他使用其他食用级酸味调节剂。
其中,膜剂中的芦荟多糖的指标要求为:相对分子量范围0.5~40万,O-乙酰基含量为40mg/g~50mg/g,结构主要为2,3或6位部分乙酰化的β-(1-4)连接的甘露聚糖和β-(1-4)连接的直链葡甘露聚糖。本实施例中,甘露糖残基与葡萄糖残基的摩尔比为8:1。
膜剂中的铁皮石斛多糖的指标要求为:相对分子量范围10万~200万、O-乙酰基含量为30mg/g~40mg/g,主要以1,4-链接为主,分支结构较少的长链乙酰化葡甘露聚糖。本实施例中,甘露糖残基与葡萄糖残基的摩尔比为 5:1。
其中,成膜材料为普鲁兰多糖、甲基纤维素、聚乙二醇、聚乙烯醇、葡聚糖中的一种或一种以上的组合。
其中,保水剂为甘油或二甘醇,或者是甘油和二甘醇。
(实施例1)
本实施例的含有石斛多糖的口腔速溶膜剂为厚度50μm的长方形膜剂,长30mm,宽20mm。入口即化。厚度、长、宽根据要求摄入的剂量调节。
含有石斛多糖的口腔速溶膜剂包含的组分及各组分的份数如下:芦荟多糖 13份,铁皮石斛多糖11.5份,普鲁兰多糖36份,甘油6.5份,水5份。
本实施例的口腔速溶膜剂含水率6.9%,能方便地与食品包装隔离纸剥离。
作为成膜材料的普鲁兰多糖可以由改性纤维素、聚乙二醇、聚乙烯醇、葡聚糖中的一种或一种以上的组合取代,也可以和改性纤维素、聚乙二醇、聚乙烯醇、葡聚糖中的一种或一种以上复配使用。所述改性纤维素为甲基纤维素、乙基纤维素、羟丙基甲基纤维素、羟乙基纤维素中的一种或一种以上的组合。
作为保水剂的甘油可由其他种类保水剂,例如二甘醇取代,也可以和二甘醇复配使用。
(实施例2)
本实施例的含有石斛多糖的口腔速溶膜剂其余与实施例1相同,不同之处在于:还包括木糖醇20.4份,苹果酸1.2份;其中水6份。
木糖醇作为甜味剂、苹果酸作为酸味调节剂,可以调节膜剂的口感,使其酸甜可口。
本实施例的口腔速溶膜剂含水率6.3%,能方便地与食品包装隔离纸剥离。
(实施例3)
本实施例的含有石斛多糖的口腔速溶膜剂包含的组分及各组分的份数如下:
芦荟多糖 15份, 铁皮石斛多糖12份,甲基纤维素40份,甘油12份,木糖醇21份,苹果酸1.2份,水7份。
本实施例的口腔速溶膜剂含水率6.7%,能方便地与食品包装隔离纸剥离。
(实施例4)
本实施例的含有石斛多糖的口腔速溶膜剂包含的组分及各组分的份数如下:
芦荟多糖 15份, 铁皮石斛多糖14份,聚乙二醇45份,甘油20份,木糖醇25份,苹果酸1.5份,水8份。
本实施例的口腔速溶膜剂含水率6.7%,能方便地与食品包装隔离纸剥离。
(实施例5)
本实施例的含有石斛多糖的口腔速溶膜剂包含的组分及各组分的份数如下:
芦荟多糖 15份,铁皮石斛多糖14份,聚乙烯醇45份,二甘醇20份,木糖醇25份,苹果酸1.5份,水8份。
本实施例的口腔速溶膜剂含水率6.7%,能方便地与食品包装隔离纸很容易剥离。
(实施例6)
本实施例的含有石斛多糖的口腔速溶膜剂包含的组分及各组分的份数如下:
芦荟多糖 15份,铁皮石斛多糖14份,葡聚糖45份,甘油10份,二甘醇10份,木糖醇22份,苹果酸1.3份,水8份。
本实施例的口腔速溶膜剂含水率6.4%,能方便地与食品包装隔离纸剥离。
(实施例7)
以实施例2的口腔速溶膜剂为例,介绍本发明制备含有石斛多糖的口腔速溶膜剂的方法。
本实施例制备实施例2的含有石斛多糖的口腔速溶膜剂,包括以下步骤:
①配制涂膜溶液。
称取芦荟多糖44g,铁皮石斛39g,普鲁兰多糖122g,木糖醇69g,甘油22g,苹果酸4g,将上述称取的原料溶解于1222g水中,得到均匀的混合溶液,将溶液于3~5℃(本实施例中为4℃)静置过夜(8~12h),取出,放至室温后得到涂膜溶液待用。
按照《中国药典2015年版四部<通则0633>,黏度测定法》检测,特性黏数为51 mL/g。
②涂膜。
开启涂布机电源。
调节涂膜速度0.16~0.20m/min、烘干温度68℃~77℃。本实施例中调节涂膜速度0.18m/min、温度73℃。
按要求装上涂布刀和PVC管,注意PVC管跟仪器的接口处要紧密,不留缝隙。
将聚酯薄膜基材安装在机器衬套拆卷机轴上,穿过涂布锟和干燥区域与叠层复卷机轴上的薄膜基材相连接。
使涂布刀里充满步骤①配制的涂膜溶液,随着薄膜基材的运动,由涂布刀将涂膜溶液均匀的涂在薄膜基材上。
③烘干。
涂覆了涂膜溶液的薄膜基材行进至烘干装置内的干燥区域,在68℃~77℃下烘干去除水分,得到一定厚度(50μm)的膜剂,烘干后的膜剂和薄膜基材一起经叠层复卷机轴收紧,收下卷好的膜剂和薄膜基材装入自封袋内,贴好相应标签。
本实施例中膜布在干燥区域内停留9.5min,干燥后的膜剂含水率6.7%。
④分切包装。
取卷好的膜剂于分切设备上分切为宽度为2cm的长卷膜剂半成品,将膜剂半成品于包装设备上进行放卷分段切割然后包装,每段长度2~3cm。
包装时,速溶膜剂封装于包材中,将包材撕开后,速溶膜剂很容易与包材表面脱离,且速溶膜剂柔软无脆性。
按上述配方及工艺,一批配料为1522g(按照涂膜溶液计算),可制成半成品膜剂320g(生产过程中有损耗),按每片30mg计算,理论生产膜剂8500片,实际可生产膜剂 8200片。
其余实施例速溶膜剂的制备方法同上。
Claims (10)
1.一种含有石斛多糖的口腔速溶膜剂,其特征在于包含的组分及各组分的份数如下:芦荟多糖 13~16份,铁皮石斛多糖11~14份,成膜材料36~45份, 保水剂6~39份,水2~10份。
2.根据权利要求1所述的含有石斛多糖的口腔速溶膜剂,其特征在于:口腔速溶膜剂中还包括甜味剂20~25份和/或酸味调节剂1.2~1.5份。
3.根据权利要求1所述的含有石斛多糖的口腔速溶膜剂,其特征在于:芦荟多糖的相对分子量范围0.5~40万,O-乙酰基含量为20mg/g~50mg/g。
4.根据权利要求1所述的含有石斛多糖的口腔速溶膜剂,其特征在于:铁皮石斛多糖的相对分子量范围10万~200万、O-乙酰基含量为10mg/g~40mg/g。
5.根据权利要求1所述的含有石斛多糖的口腔速溶膜剂,其特征在于:所述成膜材料为普鲁兰多糖、甲基纤维素、聚乙二醇、聚乙烯醇、葡聚糖中的一种或一种以上的组合。
6.根据权利要求1所述的含有石斛多糖的口腔速溶膜剂,其特征在于:所述保水剂为甘油和/或二甘醇。
7.一种如权利要求1所述的含有石斛多糖的口腔速溶膜剂的制备方法,其特征在于包括以下步骤:
①配制涂膜溶液;涂膜溶液中包含的组分及各组分的份数为芦荟多糖 13~16份,铁皮石斛多糖11~14份,成膜材料36~45份, 保水剂6~39份,水72~88份;
②涂膜;开启涂布机,使涂布刀里充满步骤①配制的涂膜溶液,随着薄膜基材的运动,由涂布刀将涂膜溶液均匀的涂覆在薄膜基材上;
③烘干;涂覆了涂膜溶液的薄膜基材行进至烘干区域,在68℃~77℃下烘干去除水分,得到膜剂,烘干后的膜剂和薄膜基材一起收卷;
④分切包装;将步骤③成卷的膜剂和薄膜基材分切、再放卷切割后包装。
8.根据权利要求7所述的含有石斛多糖的口腔速溶膜剂的制备方法,其特征在于配制涂膜溶液时,将称取的芦荟多糖、铁皮石斛、成膜材料、保水剂溶解于水中后,将得到的混合溶液于3~5℃静置8~12h,取出,放至室温后得到涂膜溶液待用。
9.根据权利要求8所述的含有石斛多糖的口腔速溶膜剂的制备方法,其特征在于:所配制的涂膜溶液的特性黏数为30~70mL/g。
10.根据权利要求7所述的含有石斛多糖的口腔速溶膜剂的制备方法,其特征在于:步骤②涂膜时的涂膜速度0.16~0.20m/min。
