Abstract
The invention provides an agar gel germination bed, a hole maker and a seed germination method, and belongs to the technical field of seed germination, wherein the agar gel germination bed takes agar gel as a germination bed matrix, 40-60 germination holes are formed in the agar gel, and the concentration of the agar gel is 0.3-3%; the thickness of the agar gel is 1-1.5 cm, and the depth of the germination holes is 0.8-1.2 cm. The seed germination method comprises the following steps: 1) injecting the agar solution into a container after hot melting, wherein the thickness of the agar solution is 1-1.5 cm; 2) vertically inserting the hole making device into the agar solution, and pulling out the hole making device after the agar solution is solidified to obtain an agar gel germination bed; 3) and placing the seeds in the germinating holes for culturing. The seed germination method can ensure that the wheat quickly germinates, has high germination rate and can carry out uniform and stable batch production of wheat seedlings.
Description
Technical Field
The invention belongs to the technical field of seed germination, and particularly relates to an agar gel germination bed, a hole maker and a seed germination method.
Background
The germination beds used in the traditional germination method mainly comprise three types, namely a paper germination bed, a sand germination bed and a soil germination bed, and the paper germination bed is mainly selected for wheat seed germination. However, in practice, paper beds have been found to suffer from a number of disadvantages. For example, a paper germination bed has poor water retention performance, and water needs to be constantly supplemented in the germination process of seeds in order to ensure that the seeds can normally germinate; if the water is supplemented too much at one time, the mixture is mixed into a whole, and the counting is difficult. Secondly, the paper germinating bed has poor adsorptivity and difficult dibbling, seeds are easy to roll in the dibbling process, and moreover, the paper germinating bed has poor supporting performance and is easy to damage, thus influencing the germinating effect.
Disclosure of Invention
In view of the above, the present invention aims to provide an agar gel germination bed, a perforator and a seed germination method; the seed germination method can ensure that the wheat quickly germinates, has high germination rate and can carry out uniform and stable batch production of wheat seedlings.
The invention provides an agar gel germinating bed, which takes agar gel as a germinating bed matrix, wherein 40-60 germinating holes are formed in the agar gel, and the concentration of the agar gel is 0.3% -3%; the thickness of the agar gel is 1-1.5 cm, and the depth of the germination holes is 0.8-1.2 cm.

The invention provides a hole making device which comprises a bottom plate and a plurality of hole making columns fixedly connected to the bottom plate, wherein the hole making columns are vertical to the bottom plate; the diameter of the hole making column is 0.3-0.4 cm, and the height of the hole making column is 0.8-1.2 cm; the distance between two adjacent hole making columns is 1.0-1.2 cm.
The invention provides a seed germination method, which comprises the following steps:
1) injecting the agar solution into a container after hot melting, wherein the thickness of the agar solution is 1-1.5 cm;
2) vertically inserting the hole making device into the agar solution, and pulling out the hole making device after the agar solution is solidified to obtain an agar gel germination bed;
3) and placing the seeds in the germinating holes for culturing.
Preferably, the seeds are wheat seeds.
Preferably, the temperature of the culture in the step 3) is 27-29 ℃, and the humidity of the culture is more than or equal to 40%.
Preferably, water is not supplemented during the culturing process.

The invention has the beneficial effects that: the agar gel germinating bed provided by the invention takes the agar gel as the substrate, so that the problem that the paper germinating bed needs frequent water replenishing is solved, and compared with the paper germinating bed, the paper germinating bed is easier to sow and count; the wheat seeds can be fixed in the germination holes, water is not required to be supplemented, and the problem of seed accumulation caused by water addition is avoided; the method of the invention can ensure that the wheat germinates rapidly, has high germination rate and can carry out uniform and stable batch production of wheat seedlings. The preparation method of the agar gel germinating bed provided by the invention is simple and easy to operate, can greatly reduce the labor and has low cost.

Detailed Description
The invention provides an agar gel germinating bed, which takes agar gel as a germinating bed matrix, wherein 40-60 germinating holes are formed in the agar gel, and the concentration of the agar gel is 0.3% -3%; the thickness of the agar gel is 1-1.5 cm, and the depth of the germination holes is 0.8-1.2 cm.
In the invention, the diameter of the germinating hole is preferably 0.3-0.4 cm. In the present invention, the agar gel germination bed is preferably fixed in a container, and the present invention has no special requirement for the shape and material of the container, and can be carried out by using a container conventional in the art. In the specific implementation process of the invention, the container can be selected as a culture dish; the diameter of the culture dish is preferably 10 cm.
In the present invention, when the seed to be germinated is a naked seed, the concentration of the agar gel is preferably 1.5% to 3%, more preferably 1.5% to 2.1%. When the seeds to be germinated are coated seeds, the concentration of the agar gel is preferably 0.3 to 1.5%, more preferably 0.4 to 0.9%. In the invention, the concentration of the agar gel specifically refers to the gram of agar powder used in each 100ml of buffer solution; when the concentration of the agar gel is 0.3%, it means that 0.3g of agar powder is contained in 100ml of the buffer solution; in the present invention, the buffer is preferably tap water and agar gel melting buffer.
The invention provides a hole making device which comprises a bottom plate and a plurality of hole making columns fixedly connected to the bottom plate, wherein the hole making columns are vertical to the bottom plate; the diameter of the hole making column is 0.3-0.4 cm, and the height of the hole making column is 0.8-1.2 cm; the distance between two adjacent hole making columns is 1.0-1.2 cm.
The shape and material of the bottom plate are not particularly limited, and the shape and material can be conventional in the field. In the present invention, the shape of the bottom plate preferably corresponds to the shape of the agar gel germination bed. In the present invention, it is preferable that the bottom plate and the perforated columns are made of a wooden material.
The invention provides a seed germination method, which comprises the following steps: 1) injecting the agar solution into a container after hot melting, wherein the thickness of the agar solution is 1-1.5 cm; 2) vertically inserting the hole making device into the agar solution, and pulling out the hole making device after the agar solution is solidified to obtain an agar gel germination bed; 3) and placing the seeds in the germinating holes for culturing.
In the invention, the agar solution is injected into a container after being melted, and the thickness of the agar solution is 1-1.5 cm. In the invention, the temperature of the agar solution is preferably 110-125 ℃, and in the invention, the thickness of the agar solution is preferably 1.1-1.4 cm.
In the invention, the hole making device is vertically inserted into the agar solution, and the agar solution is pulled out after being solidified to obtain the agar gel germinating bed. In the present invention, the perforator is preferably inserted immediately after the agar solution is injected into the container.
In the present invention, seeds are placed in the germination wells for cultivation. In the present invention, the seed is preferably a wheat seed; preferably, one seed is placed in each germination hole. In the invention, the temperature of the culture is 27-29 ℃, more preferably 28 ℃, and the humidity of the culture is preferably more than or equal to 40%. In the invention, the culture is preferably carried out in a constant temperature incubator, and the illumination condition in the culture process is 12h illumination and 12h darkness. The present invention preferably does not employ water supplementation during the culturing process.
The technical solutions provided by the present invention are described in detail below with reference to examples, but they should not be construed as limiting the scope of the present invention.
The wheat seeds in the following examples are all provided by crops of the academy of sciences of the inner Mongolia farming-animal husbandry.
Agar powder produced by Shanghai Gaokeng company with gel strength of 900g/cm2The total ash content is less than or equal to 6.5 percent, the heavy metal content is less than 40ppm, and the lead content is less than 10 ppm.
The seed cultivation temperature is 28 ℃, the relative humidity is more than 40%, and the seeds are cultivated in an artificial climate incubator (RZH850A) with 12h alternating illumination and darkness, and the length of radicle and seeds is taken as the germination standard.
The germination of naked seeds is strictly performed according to the national standard GB/T17317-1998, and the coating of seeds is performed according to GB 15671-1995.
The germination potential is the percentage of the number of the germinated seeds in the seed number of the tested variety when the number of the germinated seeds reaches the maximum peak in the germination process.
The germination percentage is the percentage of the number of normal germinated seeds to the number of test seeds within a predetermined number of days (at the time of initial counting) at the initial stage of the germination test.
Analysis of variance was performed using SPSS16.0 and the results were the average of four replicates.
The culture dish has a diameter of 10cm at the bottom, a height of 2.0cm and a thickness of 0.2cm at the bottom.
Example 1
1. Preparation of a puncher: on a square wood board with the side length of 8cm, 7 rows of cylinders with the diameter of 0.3cm and the length of more than 1.0cm are fixed, except 8 holes in the middle row, 7 cylinders are fixed in each row, and the maximum distance between every two adjacent rows and columns is 1.2 cm.
2. Dissolving agar powder by adopting an SX-500 autoclave to obtain agar solution with required concentration, injecting the obtained agar solution into the culture dish in the step 2 to ensure that the thickness of the liquid agar is more than 1.0cm, then immediately downwards inserting the cylinder on the puncher into the culture dish, after solidification, slightly and vertically pulling out the puncher, and leaving 50 seeding holes on an agar bed.
Preparing agar gel germination beds with agar gel concentrations of 0.3%, 0.6%, 0.9%, 1.2%, 1.5%, 1.8%, 2.1%, 2.4%, 2.7%, and 3.0% according to the above method; 3 parts of each concentration is prepared, and then, naked seeds of Nomame No. 2, Nomame No. 4, Yongliang No. 4, Ningchun 50 and Longmai 33 and coated seeds of Nomame No. 2 are sown in the holes on the agar bed. The following operations are carried out:
① spreading 2 pieces of filter paper in the culture dish, and wetting the filter paper with distilled water to obtain a paper bed as a control, wherein the number and spacing of the seeds are the same as those of the agar bed.
And (3) counting the germination rate, the germination potential and the germination index of the seeds in the germination bed, wherein the specific data are shown in tables 1, 2 and 3.

As can be seen from Table 1, the naked and coated wheat seeds of NommaMc2 have obvious difference in germination rate on agar bed and paper bed, and the concentration of agar germination bed has obvious influence on the germination rate of seeds. On a paper bed, the germination rate of the naked seeds is 92 percent and is obviously higher than the germination rate of the coated seeds by 79 percent. The germination rates of the wheat naked seeds and the coated seeds with different agar concentrations are different on the agar bed. The germination rates of the naked wheat seeds and the coated wheat seeds reach the maximum values by the fourth day when the agar concentration is 0.60 percent, and are respectively 97 percent and 95 percent.

As can be seen from Table 2, different wheat varieties have distinct differences in germination vigor on the agar bed and the paper bed, and the concentration of the agar bed also has an influence on the germination vigor of wheat seeds. When the concentration of the wheat variety in an agar bed is 0.3%, the germination potential reaches the maximum value, wherein the germination potential of Ningchun 50 is 99% at most, and the germination potential of Longmai 33 is 88% at least.

As can be seen from Table 3, different wheat varieties have distinct differences in germination potential on the agar bed and the paper bed, and the concentration of the agar bed also has an effect on the germination index of wheat seeds.
According to the embodiment, the agar gel germinating bed provided by the invention takes the agar gel as the substrate, so that the problem that the paper germinating bed needs to be frequently replenished with water is solved, compared with the paper germinating bed, the seeding and counting are easier, and the wheat germination rate is high.

1) injecting the agar solution into a container after hot melting, wherein the thickness of the agar solution is 1-1.5 cm;
2) vertically inserting the perforator of claim 5 into the agar solution, and pulling out the agar solution after the agar solution is solidified to obtain an agar gel germination bed;
3) and placing the seeds in the germinating holes for culturing.
7. The germination method according to claim 6, wherein the seed is a wheat seed.
8. The germination method according to claim 7, wherein the temperature of the cultivation in step 3) is 27 to 29 ℃ and the humidity of the cultivation is not less than 40%.
