Description

Peach gum extract with skin tightening effect, and preparation method and application thereof
Technical Field
The invention relates to the technical field of cosmetics, in particular to a peach gum extract with an instant skin tightening effect, and a preparation method and application thereof.
Background
Skin refers to the tissue of the body surface outside the muscles, which is the largest organ of the human body, and consists of three layers of structures, namely epidermis, dermis and subcutaneous tissue, and comprises four major accessory organs (sweat glands, sebaceous glands, nails, hairs) and the like. The skin covering the whole body has barrier effects including physical barrier, pigment barrier, immune barrier, and nerve barrier. However, the protective action of the skin is limited, and the skin generally begins to age after 25 years of age due to the influence of endogenous factors and external environmental factors. Three major characteristics of skin aging include wrinkle development, skin laxity, pigmentation, with wrinkles being the initial signs of skin aging and also the most important and intuitive manifestations of skin aging. Wrinkle removal means that wrinkles on the skin disappear or become shallow, wherein the wrinkle removal mechanism mainly comprises the following aspects:
1) the theory of genetics: such as angelica polysaccharide and cistanche tubulosa ergosterioside, can delay cell aging by increasing telomerase activity, thereby delaying wrinkle generation.
2) The theory of free radicals: such as superoxide dismutase SOD, vitamin E, pawpaw sulfhydryl enzyme, etc., can effectively remove superoxide radical, hydroxyl radical, etc. in vivo, thereby preventing aging of body cells, delaying aging process of skin, and relieving wrinkles.
3) The theory of matrix metalloproteinases: such as soybean isoflavone, can promote collagen synthesis and relieve wrinkle by reducing matrix metalloproteinase MMP-1 activity.
4) Theory of metabolic disorders: such as puerarin and human acidic fibroblast growth factor aFGF, can promote cell differentiation and proliferation, promote cell metabolism, smooth and fine skin and reduce wrinkles.
5) The theory of photoaging: such as ZnO, TiO2The ultraviolet scattering agent, the ultraviolet absorbent such as methyl aminobenzoate and octyl methoxycinnamate, etc. can isolate UVA and UVB ultraviolet rays, and can relieve the damage of light to the skin, thereby relieving the generation of wrinkles.
Through the anti-aging approach, although the skin aging process can be delayed to a certain extent, the anti-aging method is long in time consumption and slow in effect. Therefore, the development of an anti-aging product capable of firming skin and removing wrinkles in a short time has been desired by consumers and cosmetic practitioners. Firming components are increasingly used in anti-aging skin care products because they have the effect of firming and smoothing skin immediately. The mechanism of the skin-firming cream is mainly characterized in that a layer of protective film with a three-dimensional net structure can be formed on the surface of skin by the skin-firming component, and the protective film shrinks immediately after being dried to stretch the skin and achieve the effects of firming the skin and relieving wrinkles.
Peach gum (also known as peach gum) is a colloidal translucent substance secreted by trunks of plants of the Rosaceae family such as peaches (Prunus persica (L.) Batsch) or Prunus davidiana (Carr.) Franch) after mechanical injury (such as insect bite, incised wound, etc.) or disease. The liquid and viscous substances collected by the trunk are commonly called peach tree oil; the solid material formed by air drying or other dehydration on the trunk is called raw peach gum.
The pharmacological actions of peach gum are recorded in the ancient and modern recorded empirical prescription, the compendium of materia Medica, the miscellaneous records of famous physicians, and the Qianjin prescription, which are called bitter in taste, mild in nature, tonifying qi, harmonizing blood, quenching thirst, and dredging body fluid. The peach gum can clear blood and reduce fat, relieve pressure, resist wrinkles and soften skin, can promote blood circulation to reduce swelling, and treat stranguria and relieve pain, and has good effect on diarrhea, pain, bloody stranguria, urolithiasis, dysentery and other diseases in clinical application. In summary, peach gum is a food material and a traditional Chinese medicine with high medical, medicinal and edible values.
The peach gum is gum material, and contains polysaccharide 90% or more, and small amount of protein and impurities. Generally applied as a moisturizing raw material. For example, patent document CN110464682A discloses a method for preparing peach gum extract, which comprises: 1) mixing the peach gum raw material with water to obtain peach gum mixed solution; 2) heating the peach gum mixed solution to 60-90 ℃, and keeping the temperature for 1-3h for extraction; 3) filtering the mixed solution obtained in the step 2), wherein the filtrate is the peach gum extracting solution, and the polysaccharide yield is high. The peach gum prepared by the method mainly focuses on the timely moisturizing effect, and is shown in that the moisture content of the skin can be increased, namely the moisture of the skin is more sufficient, so that the problems of skin cell peeling and skin roughness caused by dry skin can be solved, namely the skin scale index SEsc and the skin smoothness parameter SEsm are reduced.
The existing peach gum extraction method mainly comprises a heat extraction method, an alkali extraction method and an acid extraction method. (1) The heat extraction method generally needs long extraction time at the temperature of 100 ℃ under normal pressure, has low yield and excessive product impurities, and is not suitable for industrial production. Therefore, in order to shorten the extraction time, pressurization and temperature rise are needed, namely the peach gum is extracted by heating and pressurizing through a jacketed reaction pot. The temperature is controlled at 160 deg.C, and extraction time is 5-6 h. However, during the process of raising the temperature, some peach gum polysaccharide molecules are hydrolyzed and broken, so that the molecular weight of the peach gum polysaccharide is reduced, and non-sugar byproducts are formed. (2) The alkali extraction method mainly comprises adjusting pH to 9-11 with alkali, and heating to extract peach gum. Compared with a thermal extraction method, the alkali extraction method has the advantages of low temperature requirement, short extraction time and low energy consumption, but the peach gum can be partially hydrolyzed in the extraction process, the reaction process is complex, the filtration is too time-consuming, and the production process is accompanied with environmental pollution. (3) The acid extraction method has the advantage of rapid extraction by adding acid as a catalyst to hydrolyze peach gum, but the peach gum is extremely easy to be hydrolyzed to form oligosaccharide or monosaccharide without adhesiveness, impurities in the raw materials are decomposed under the action of acid to deepen the color of the peach gum, and the product quality is influenced by increasing the content of inorganic salt during neutralization.
In addition, because the peach gum contains natural pigment, the protein contained in the peach gum generates Maillard reaction due to heating in the extraction process to generate non-enzymatic browning phenomenon, so that the color of the peach gum extracting solution is deepened. Therefore, peach gum products need to be decolorized in industrial production. At present, the peach gum extracting solution decoloration treatment method mainly adopts chemical reagent bleaching or adsorption decoloration. Chemical agent bleaching usually adopts chemical agents with strong oxidizing property, such as hydrogen peroxide, sodium hypochlorite and the like, and the method is easy to cause chemical agent residues, thereby influencing the quality of peach gum products. The adsorption decoloration relates to active carbon and anion exchange resin, and has the characteristics of low price, no influence on biological activity, no toxicity, no harm and the like.
Disclosure of Invention
In order to solve the defects of the prior art, the invention provides a peach gum extract with the effect of tightening skin instantly, and a preparation method and application thereof. The obtained peach gum extract has effects of instantly tightening and smoothing skin, and relieving wrinkle, and is green and safe.
The purpose of the invention is realized by the following technical scheme:
the invention provides a peach gum extract with an instant skin tightening effect, which comprises peach gum polysaccharide, wherein the peach gum polysaccharide comprises the following components in percentage by weight: 150-90% of low molecular weight peach gum polysaccharide S and 210-50% of high molecular weight peach gum polysaccharide S; the molecular weight of S1 is less than 20kDa, and the molecular weight of S2 is more than 200 kDa.
Preferably, the peach gum polysaccharide comprises the following monosaccharides in percentage by mass: 35-70:15-35:0.1-10:0.1-10:0.1-5:0.1-5 of arabinose, galactose, ribose, glucose, rhamnose and mannose.
According to the invention, the high molecular weight peach gum polysaccharide forms a film with a three-dimensional net structure on the skin surface, the low molecular weight peach gum polysaccharide plays a filling effect, and the skin firming effect is obtained only by matching the high molecular weight peach gum polysaccharide and the low molecular weight peach gum polysaccharide in a proper proportion.
The invention also provides a preparation method of the peach gum extract with the effect of tightening skin immediately, which comprises the following steps:
A. crushing: pulverizing peach gum and sieving to obtain peach gum powder;
B. swelling: mixing the peach gum powder with water, stirring at room temperature and swelling to obtain a feed liquid 1;
C. low-temperature extraction: heating the feed liquid 1 to 40-70 ℃, and stirring and leaching to obtain a feed liquid 2;
D. high-temperature extraction: heating the feed liquid 2 to 85-100 ℃, and stirring and leaching to obtain feed liquid 3;
E. grinding and extracting: pulping the feed liquid 1 in a colloid mill to obtain feed liquid 4;
F. and (3) filtering: filtering the feed liquid 3 and the feed liquid 4 by using gauze to obtain a peach gum crude extract;
G. and (3) purification: removing impurities from the crude peach gum extract by using activated carbon and a clarifying agent, centrifuging, and filtering supernatant by using a membrane to obtain a feed liquid 5;
H. concentration: concentrating the feed liquid 5 under reduced pressure to obtain concentrated solution;
I. standing at a low temperature: standing the concentrated solution at low temperature, and membrane filtering to obtain peach gum extract.
Preferably, in the step F-I, the subsequent steps of filtering, purifying, concentrating and standing at low temperature are carried out after the feed liquid 3 and the feed liquid 4 are mixed.
The invention can avoid the problems of sharp reduction of the molecular weight of the peach gum polysaccharide and reduction of the adhesiveness caused by long-term high-temperature extraction by variable-temperature extraction (low-temperature extraction and high-temperature extraction), and can effectively save energy consumption to a certain extent. And the extraction of the peach gum polysaccharide with different molecular weight ranges can be realized through the extraction at different temperatures (the peach gum polysaccharide with higher molecular weight can be obtained through low-temperature extraction, and the peach gum polysaccharide with smaller molecular weight can be obtained through high-temperature extraction).
The invention further adopts grinding extraction to keep the integrity of the peach gum polysaccharide, so as to obtain the high molecular weight peach gum polysaccharide and reduce the generation of byproducts.
Preferably, in the step A, the particle size of the peach gum powder is 20-80 meshes; according to the invention, after the crushing operation is carried out on the dry peach gum, the material body is in a loose fine powder shape (crushing is carried out before use, and the problem of high viscosity is avoided if the storage time is too long is avoided as much as possible); in the case where a small amount of the crushed material is adhered to the inside of the crushing apparatus, the crushed material may be washed after soaking. And the time required by the short peach gum polysaccharide extraction process is greatly shortened (about 40 percent of extraction time is saved) through crushing, the energy consumption is effectively saved, and the product quality is not influenced. The too large particle size of the peach gum can increase the extraction time of the peach gum, and the too small particle size can improve the efficiency, but can increase the risk of dust suction of operators.
In the step B, the dosage ratio of the peach gum powder to the water is 1: 20-200 parts of; the swelling time is 1-5 hours under the condition of stirring at room temperature.
Preferably, in the low-temperature extraction step, the stirring and leaching time is 1-5 hours;
preferably, in the high-temperature extraction step, the stirring extraction time is 5 to 30 hours.
Preferably, in step F, the mixing ratio of the feed liquid 3 to the feed liquid 4 is 10: 1-100 (the efficacy evaluation proves that the instant firming effect is best in the range of the mixing ratio); the aperture of the gauze is 50-400 meshes.
Preferably, in the step G, the using amount of the activated carbon is 0.05-3% of the mass of the peach gum crude extract, and the using amount of the clarifying agent is 1-10% of the mass of the peach gum crude extract;
the clarifying agent comprises one or more of ZTC-II type, ZTC-III type and 101 type.
The method adopts the activated carbon and the clarifying agent to purify the peach gum crude extract, achieves the aims of effectively removing impurities and decoloring, and has the advantages of low price, no residue and easy industrial production.
Preferably, in step H, the temperature for concentration under reduced pressure is 50-70 ℃, and the ratio of concentration under reduced pressure is 2-10: 1.
the peach gum extract is obtained by the steps of variable temperature extraction, colloid mill grinding extraction, activated carbon and clarifying agent purification, concentration, low temperature standing and the like. The peach gum extract composition comprises low molecular weight peach gum polysaccharide S1 (< 20kDa) and high molecular weight peach gum polysaccharide S2 (> 200 kDa). The peach gum extract disclosed by the invention has the effects of instantly tightening skin and removing wrinkles, and can be applied to preparation of skin care cosmetics, especially anti-aging products.
The invention also provides an application of the peach gum extract with the effect of tightening skin immediately in skin care cosmetics, wherein the skin care cosmetics comprise cream, lotion, astringent, gel, mask and ointment.
The peach gum polysaccharide in peach gum is extracted and purified, and the peach gum polysaccharide with different molecular weight distributions is applied to an anti-aging product, so that the peach gum polysaccharide has the effects of tightening skin and removing wrinkles instantly. The raw material has wide application prospect in the industry, and can be used for skin care cosmetics, especially anti-aging products.
Compared with the prior art, the invention has the following beneficial effects:

Effect verification:
experimental method 1:
after cleaning hands and faces of a subject, wiping skin moisture with a paper towel, and after the subject enters a testing environment (constant temperature and humidity environment: temperature 22 +/-2 ℃ and ambient humidity 50 +/-5% RH), the test can be carried out at least by waiting for 10min after the subject has rested.
The skin elasticity tester MPA580 is adopted to test the R2 value of the cheekbones at the left and right sides of a healthy person (R2 is Ua/Uf, the ratio of the rebound quantity of the skin without negative pressure to the maximum stretching quantity with negative pressure is closer to 1, which indicates that the skin elasticity is better), and the T0 data at the beginning are recorded. Then, 100. mu.L of a sample to be tested (a basic gel containing 10% of the peach gum extract prepared in each example and comparative example) was applied to the cheekbones, and the applied part was left to stand completely. After drying for 5-10min, collecting R2 values after 5min, 30min, 1h, 2h, 4h, 6h and 8h respectively, calculating the skin elasticity increase amount delta R2 under corresponding time, and analyzing statistical difference (n is 6).
The formulation of the base gel is as follows:
peach gum extracts prepared in each example and comparative example, 10%; disodium EDTA, 0.02%; carbomer (Carbopol940Polymer), 0.5%; raysoncare BHC (caprylhydroxamic acid, 1, 2-hexanediol, butanediol), 0.8%; NaOH, proper amount; water is added to 100 percent.
The experimental results are as follows:
as shown in fig. 1, when a basic gel containing 10% of the peach gum extract (example 1) is applied to cheekbones of a healthy person, the change of skin elasticity Δ R2 shows a trend of increasing and then decreasing with time, which indicates that the peach gum extract has the effect of firming skin.
When the basic gel containing 10% of the peach gum extract (examples 2-5) is applied to cheekbones of healthy people, the skin elasticity change amount delta R2 shows a trend of increasing and then decreasing with time, and the effect is close to that of example 1, which shows that the peach gum extracts prepared by the examples 2-5 also have the effect of tightening the skin.
The results using the base gel containing 10% of the peach gum extract (comparative examples 1,2 and 3) showed that the skin elasticity variation Δ R2 was nearly zero, indicating that none of the peach gum extracts prepared in comparative examples 1,2 and 3 had the effect of tightening the skin.
Experimental method 2:
after cleaning hands and faces of a subject, wiping skin moisture with a paper towel, waiting for 5-10min in a test environment (constant temperature and humidity environment: temperature 22 +/-2 ℃, and ambient humidity 50 +/-5% RH), and entering a test when the skin is dry to a normal state.
The upper left and lower right parts of the canthus were first marked with small black marks (for positioning), photographed using a skin microscope and active skin analysis system SELS VC98 (3 picture repeat), and the T0 data was recorded. Then, 100. mu.L of a sample to be tested (a basic gel containing 5% of the peach gum extract prepared in each example and comparative example, the formula of the basic gel is the same as that in test method 1) was placed on the back of the hand, the hand was applied to the eye with fingers, the skin was uniformly applied from inside to outside for 10 seconds in a clockwise direction, and the applied part was kept completely still. After the canthus is dried for 5-10min, the canthus images of the testees are respectively collected after 5min, 30min, 1h, 2h, 4h, 6h and 8h, the size and the position of the photographing area of each tester are adjusted according to black marks, a VC98 system is used for calculating the wrinkle parameter change rate, and statistical differences are analyzed (n is 11). Among them, SEr is a skin roughness parameter (the smaller the value, the better the skin, the smaller the skin roughness), SEw is a skin wrinkle parameter (the smaller the value, the better the skin).
The experimental results are as follows:
as shown in fig. 2, when the base gel containing 5% of the peach gum extract (example 1) was applied to the corners of healthy human eyes, the change rate of the wrinkle parameters SEr and SEw showed a tendency of increasing and then decreasing with time. And the ordinate values in fig. 2 are SEr and SEw change rates, taking SEr as an example, the change rate (%) is (initial SEr value-test time SEr value)/initial SEr value, the change rate is positive, namely SEr is reduced, and the change rate of SEw is also positive, namely SEw is also reduced, thereby demonstrating that the peach gum extract has the effect of firming skin.
When the basic gel containing 5% of the peach gum extract (examples 2-5) is smeared on the canthus of healthy people, the change rate of wrinkle parameters SEr and SEw changes along with time, the trend of increasing and then decreasing is shown, the effect is similar to that of example 1, and the peach gum extracts prepared in examples 2-5 also have the effect of firming the skin.
The results of the application of the base gel containing 5% of the peach gum extract (comparative examples 1,2 and 3) showed that the change rate of the wrinkle parameters SEr and SEw was close to zero, indicating that none of the peach gum extracts prepared in comparative examples 1,2 and 3 had the effect of tightening the skin.
