Abstract
本发明公开了一种黑果枸杞天然复合抗氧化剂的制备方法,包括以下步骤:原料的选取和预处理→制备虾青素提取物→粉碎→制备柚子皮提取物→制备黑果枸杞提取物→制备甘草提取物→制备小麦胚芽提取物→取虾青素提取物、柚子皮提取出物、黑果提取物、甘草提出物和小麦胚芽提取物进行复配,然后干燥至恒重,卫生检测后进行包装,本发明提供的天然复合抗氧化剂不仅有效地提高了抗氧化性能,又可起到保鲜和抑菌的作用,同时,还具有其他的保健功效,例如美容肌肤,抗肿瘤、预防心脑血管疾病,降血糖等功效。

技术领域
本发明涉及食品添加剂技术领域,具体为一种黑果枸杞天然复合抗氧化剂的制备方法。
背景技术
随着食品工业的不断进步,可食品抗氧化剂也得到了长足的发展。长期以来,人们一直使用合成抗氧化剂如二丁基羟基甲苯、丁基羟基茴香醚、丁基对苯二酚、没食子酸丙酯等,易分解、功效低且具有刺激性,且动物实验表明合成抗氧化剂还具有毒性和致癌作用。合成抗氧化剂使人们产生了不安的心理。现在人们将研究重点转向了天然抗氧化剂,并取得了许多研究成果。
然而,然而单一的天然抗氧化剂活性低,功效单一。
发明内容
本发明的目的在于提供一种黑果枸杞天然复合抗氧化剂的制备方法以解决上述背景技术中提出的问题。
为实现上述目的,本发明提供如下技术方案:一种黑果枸杞天然复合抗氧化剂的制备方法,包括以下步骤:
步骤一、原料的选取:取黑果枸杞:30-45份;甘草:15-20份;柚子皮:10-16份;小麦胚芽:20-24份;虾青素:5-12份作为原料;
步骤二、原料的预处理:
(1)将柚子去皮,并将柚子皮和和黑果枸杞于太阳下晾晒1-2d;
(2)制备虾青素提取物:取虾青素原料,以食用油作为提取剂,于提取温度为60-90℃;提取时间为0.5-1h的条件下采用油溶法提取虾青素以作备用;
步骤三、粉碎:将步骤二中晾晒后的柚子皮、黑果枸杞和甘草均通过超声波粉碎机分别进行粉碎,制得柚子皮粉、黑果枸杞粉和甘草粉,再将制得柚子皮粉、黑果枸杞粉和甘草粉分别进行脱脂处理;
步骤四、制备柚子皮提取物:取脱脂后的柚子皮粉,以液料重量比为10-12,用浓度为60-65%的乙醇回流提取6-7h,抽滤,所得滤液经蒸发器在温度为70-82℃下浓缩,浓缩液用重蒸水溶解,然后用大孔树脂对其进行精制,精制调件为:上柱液的pH值为6.0-6.5,上柱温度为60-68℃,吸附阶段的流速为1-2ml/min,解吸阶段使用70-75wt%的乙醇溶液,乙醇溶液过柱流速为3-5ml/min;
步骤五、制备黑果枸杞提取物:取脱脂后的黑果枸杞粉,以液料重量比为1-2,用浓度为65-75%的乙醇回流提取8-17h,抽滤,所得滤液经蒸发器在温度为80-86℃下浓缩,浓缩液用重蒸水溶解,然后用大孔树脂对其进行精制,精制调件为:上柱液的pH值为6.1-6.8,上柱温度为56-60℃,吸附阶段的流速为2-4ml/min,解吸阶段使用75-85wt%的乙醇溶液,乙醇溶液过柱流速为2-4ml/min;
步骤六、制备甘草提取物:将脱脂后的甘草粉,以液料重量比为6-8用无水乙醇进行回流提取8-10h,抽滤得到的滤液在75-85℃下的蒸发器中反复浓缩析出,再对析出物进行干燥至恒重即可;
步骤七、制备小麦胚芽提取物:将小麦胚芽进行干燥,并将干燥的小麦胚芽冷压榨成油,然后用90-95%的乙醇将油提取,静置30-45min,并在65-70℃的温度下进行真空蒸发,直至乙醇除去;
步骤八、取虾青素提取物、柚子皮提取出物、黑果提取物、甘草提出物和小麦胚芽提取物进行复配,然后干燥至恒重,卫生检测后进行包装。
优选的,所述原料的重量份数分别为:黑果枸杞:35份;甘草:18份;柚子皮:14份;小麦胚芽:20份;虾青素:7份。
优选的,所述虾青素原料为红蟹壳或龙虾壳。
优选的,所述食用油为豆油,且虾青素原料和食用油的质量比为1:9。
优选的,所述柚子皮粉、黑果枸杞粉和甘草粉均过60-70目筛。
与现有技术相比,本发明的有益效果是:
(1)黑果枸杞含有丰富的原花青素OPC,其OPC含量超过蓝莓,原花青素OPC是最有效的天然水溶性自由基清除剂,其功效是VC的20倍、VE的50倍,因此,具有延缓衰老和美容肌肤的保健功效,另外,黑果枸杞还含有天然的胡萝卜素以及多种维生素,从而能有效预防心脏病和癌症,除此之外,由于黑果枸杞含有胍的衍生物,可以作为糖尿病治疗患者的辅助保健品,能够有效地降低血清胆固醇和三酰甘油的含量,减轻和预防动脉硬化。
(2)小麦胚芽含有小麦胚芽蛋白中还含有一种由谷氨酸、半胱氨酸、甘氨酸三个氨基酸经肽键缩合而成的含硫活性三肽——谷胱甘肽,它具有抗氧化延衰老功能,通过谷胱甘肽催化,可与过氧化物反应,还原氧化物,保护人体细胞免受氧化损害,特别能保护大脑功能,并能传递氨基酸生物功能,促进生长发育。
(3)柚子皮含有类黄酮,有助血气运行,抗衰老和抗发炎等,而且还含有丰富的维生素C以及类胰岛素等成分,故有降血糖、降血脂、减肥、美肤养容等功效。
(4)柚子皮中的柚皮甙能降低毛细血管的通透性,从而有显著的助消炎的作用,醛糖还原酶在哺乳动物体内催化葡萄糖向山梨醇的转化,这是糖尿病后遗症如白内障和神经疾病的主要起因,而柚皮中的柚皮甙有抑制醛糖还原酶的作用,从而抑制糖尿病的并发症,而且柚皮中的柚皮甙对金黄色葡萄球菌、大肠杆菌、痢疾杆菌和伤寒杆菌等有抑制作用,因此,具有杀菌和抑菌的功效。
(5)由(1)、(3)和(4)可得,黑果枸杞和柚子皮的协同作用下,提高了抗氧化剂的抗氧化性能外,同时也提高了降血糖和美容肌肤的功效。
(6)小麦胚芽还含有钙、钾、镁、铁、锌、铬、硒、磷、锰、铜等多种矿物质和微量元素。小麦胚芽中色素的成分是小麦黄酮,它是一种水溶性色素,对心血管疾病具有很好的治疗功能。
(7)本发明中制备的天然复合抗氧化剂中含有天然的虾青素,是一种氧化性极强的类胡萝卜素,具有抗氧化、抗衰老的功效,虾青素作为食品添加剂用于食品的着色、保鲜及营养。虾青素为脂溶性,具艳丽红色和强抗氧化性能,对于食品尤其是含脂类较多的食品,既有着色效果又可起到保鲜作用,同时还具有抗肿瘤、预防心脑血管疾病的作用,因此,虾青素、黑果枸杞、小麦胚芽和柚子皮起到协同的作用,提高了抗氧化性,同时,虾青素、小麦胚芽和黑果枸杞的相互协同的作用下,能够起到很好的预防心脑血管疾病和心血管疾病的功效。
综上,本发明提供的天然复合抗氧化剂不仅有效地提高了抗氧化性能,又可起到保鲜和抑菌的作用,同时,还具有其他的保健功效,例如美容肌肤,抗肿瘤、预防心脑血管疾病,降血糖等功效。
具体实施方式
下面对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例一
一种黑果枸杞天然复合抗氧化剂的制备方法,包括以下步骤:
步骤一、原料的选取:取黑果枸杞:35份;甘草:18份;柚子皮:14份;小麦胚芽:20份;虾青素:7份作为原料;
步骤二、原料的预处理:
(1)将柚子去皮,并将柚子皮和和黑果枸杞于太阳下晾晒1-2d;
(2)制备虾青素提取物:取虾青素原料,以食用油作为提取剂,于提取温度为60-90℃;提取时间为0.5-1h的条件下采用油溶法提取虾青素以作备用;
步骤三、粉碎:将步骤二中晾晒后的柚子皮、黑果枸杞和甘草均通过超声波粉碎机分别进行粉碎,制得柚子皮粉、黑果枸杞粉和甘草粉,再将制得柚子皮粉、黑果枸杞粉和甘草粉分别进行脱脂处理;
步骤四、制备柚子皮提取物:取脱脂后的柚子皮粉,以液料重量比为10-12,用浓度为60-65%的乙醇回流提取6-7h,抽滤,所得滤液经蒸发器在温度为70-82℃下浓缩,浓缩液用重蒸水溶解,然后用大孔树脂对其进行精制,精制调件为:上柱液的pH值为6.0-6.5,上柱温度为60-68℃,吸附阶段的流速为1-2ml/min,解吸阶段使用70-75wt%的乙醇溶液,乙醇溶液过柱流速为3-5ml/min;
步骤五、制备黑果枸杞提取物:取脱脂后的黑果枸杞粉,以液料重量比为1-2,用浓度为65-75%的乙醇回流提取8-17h,抽滤,所得滤液经蒸发器在温度为80-86℃下浓缩,浓缩液用重蒸水溶解,然后用大孔树脂对其进行精制,精制调件为:上柱液的pH值为6.1-6.8,上柱温度为56-60℃,吸附阶段的流速为2-4ml/min,解吸阶段使用75-85wt%的乙醇溶液,乙醇溶液过柱流速为2-4ml/min;
步骤六、制备甘草提取物:将脱脂后的甘草粉,以液料重量比为6-8用无水乙醇进行回流提取8-10h,抽滤得到的滤液在75-85℃下的蒸发器中反复浓缩析出,再对析出物进行干燥至恒重即可;
步骤七、制备小麦胚芽提取物:将小麦胚芽进行干燥,并将干燥的小麦胚芽冷压榨成油,然后用90-95%的乙醇将油提取,静置30-45min,并在65-70℃的温度下进行真空蒸发,直至乙醇除去;
步骤八、取虾青素提取物、柚子皮提取出物、黑果提取物、甘草提出物和小麦胚芽提取物进行复配,然后干燥至恒重,卫生检测后进行包装。
在本实施例中,所述虾青素原料为红蟹壳或龙虾壳。
在本实施例中,所述食用油为豆油,且虾青素原料和食用油的质量比为1:9。
在本实施例中,所述柚子皮粉、黑果枸杞粉和甘草粉均过60-70目筛。

在本实施例中,所述原料的重量份数分别为:黑果枸杞:40份;甘草:15份;柚子皮:10份;小麦胚芽:23份;虾青素:12份。

Claims (5)
1.一种黑果枸杞天然复合抗氧化剂的制备方法,其特征在于:包括以下步骤:
步骤一、原料的选取:取黑果枸杞:30-45份;甘草:15-20份;柚子皮:10-16份;小麦胚芽:20-24份;虾青素:5-12份作为原料;
步骤二、原料的预处理:
(1)将柚子去皮,并将柚子皮和和黑果枸杞于太阳下晾晒1-2d;
(2)制备虾青素提取物:取虾青素原料,以食用油作为提取剂,于提取温度为60-90℃;提取时间为0.5-1h的条件下采用油溶法提取虾青素以作备用;
步骤三、粉碎:将步骤二中晾晒后的柚子皮、黑果枸杞和甘草均通过超声波粉碎机分别进行粉碎,制得柚子皮粉、黑果枸杞粉和甘草粉,再将制得柚子皮粉、黑果枸杞粉和甘草粉分别进行脱脂处理;
