Abstract

本发明提供了一种多功能抗草甘膦水稻转化载体pCDMAR‑epsps及其构建方法和应用,其特征在于,载体pCDMAR‑epsps的大小为12774bp,序列如SEQ ID NO:1所示;构建方法包括骨架选择、含pCDMAR‑hyg骨架的感受态细胞制备及培养、酶切、连接、pCDMAR‑UbiEP中间产物的获得、带有酶切位点的T‑nos片段的获得、酶切连接。本发明的有益效果在于:本发明的一种多功能抗草甘膦水稻转化载体pCDMAR‑epsps较现有技术具有更好的安全性,而且对外源基因的稳定性和表达水平具有一定的优势,同时单插入位点中的多酶切位点还为实现同时转入2个及2个以上的多基因聚合育种的可能性。

Description

一种多功能抗草甘膦水稻转化载体pCDMAR-epsps及其构建方 法和应用
技术领域
本发明基因工程技术领域,具体涉及一种多功能抗草甘膦水稻转化载体pCDMAR-epsps及其构建方法和应用。
背景技术
杂草每年给农业生产造成的经济损失约占农作物总产值的10%~20%,因而杂草防治在农业生产中尤为重要。相比用机械除草,喷施除草剂具有省时高效的特点。草甘膦(glyphosate)是世界上应用最广泛的非选择性内吸传导性有机膦类除草剂。
但是,这种灭生性除草剂在除杂草的同时也会给农作物带来明显的伤害。目前,还未发现含有抗除草剂(草甘膦)的水稻种质资源,因此无法通过传统育种方式获得抗除草剂的水稻品种。通过基因工程技术将抗除草剂基因导入农作物,已经获得了多个抗除草剂农作物新品种,并且已经在农业生产中广泛应用。
水稻作为主要的粮食作物,对人们生活具有重要的意义,因此,抗除草剂水稻品种的开发,具有一定的需求和巨大的市场潜力。
但是,目前所构建的外源基因插入载体多以潮霉素抗性基因(hpt)作为筛选标记基因,其安全性受到一定质疑,所以开发一种更加安全、高效的多功能抗草甘膦水稻转化载体及其构建方法就显得尤为重要。
发明内容
为了弥补现有技术的不足,本发明提供了一种多功能抗草甘膦水稻转化载体pCDMAR-epsps及其构建方法和应用。本发明的载体利用抗除草剂基因,不仅可以使作物获得除草剂抗性而方便除草,本身也是筛选标记,还可以用于在田间对其它目的基因的跟踪和选择,本身的安全性也得到了广泛认同;而且本发明的转化载体对外源基因的稳定性和表达水平也进行了优化,单插入位点中的多酶切位点还为实现同时转入2个及2个以上的多基因聚合育种提供了可能性。
本发明提供了一种多功能抗草甘膦水稻转化载体pCDMAR-epsps,大小为12774bp,序列如SEQ ID NO:1所示;且所述载体pCDMAR-epsps中包括11个功能元件,具体见下表:

本发明提供了一种多功能抗草甘膦水稻转化载体pCDMAR-epsps的构建方法,包括以下步骤:
(1)骨架选择:选择pCDMAR-hyg作为骨架,且所述pCDMAR-hyg骨架大小为11724bp,序列如SEQ ID NO:14所示;
(2)含pCDMAR-hyg骨架的感受态细胞制备及培养:将pCDMAR-hyg骨架转化到感受态细胞中,并采用常规方法进行培养,获得含pCDMAR-hyg骨架的感受态细胞;
(3)酶切:采用BclI酶酶切所述pCDMAR-hyg骨架,得到片段Ⅰ、片段Ⅱ、片段Ⅲ;且所述片段Ⅰ的大小为6067bp,序列如SEQ ID NO:15所示;片段Ⅱ的大小为2975bp,序列如SEQID NO:16所示;片段Ⅲ的大小为2682bp,序列如SEQ ID NO:17所示;
(4)连接:将所述片段Ⅰ、片段Ⅱ连接得到连接产物,并通过ClaI/EcoRV酶切鉴定所述连接产物;选择ClaI/EcoRV酶切后片段大小为7346bp和1403bp的连接产物,并将其命名为中间质粒pCDMAR;所述中间质粒pCDMAR的大小为8749bp,序列如SEQ ID NO:18所示;
(5)pCDMAR-UbiEP中间产物的获得:利用SbfI/KpnI酶酶切pCUEP102质粒获得酶切产物,并回收其中大小为3783bp的片段,命名为酶切片段Ⅰ,序列如SEQ ID NO:19所示;之后利用SbfI/KpnI酶切所述中间质粒pCDMAR,回收酶切片段,命名为酶切片段Ⅱ;之后将所述酶切片段Ⅰ和酶切片段Ⅱ进行连接,获得连接产物,命名为pCDMAR-UbiEP中间产物;所述pCDMAR-UbiEP中间产物的大小为12505bp,序列如SEQ ID NO:20所示;
(6)带有酶切位点的T-nos片段的获得:设计两端加KpnI和SacI酶切位点的引物,并采用所述引物扩增pCUEP102上T-nos片段,回收大小为269bp的目的条带,且所述目的条带的序列如SEQ ID NO:8所示;
(7)酶切连接:将所述目的条带与所述pCDMAR-UbiEP中间产物通过KpnI/SacI酶进行酶切连接,得到连接产物,即为一种多功能抗草甘膦水稻转化载体pCDMAR-epsps;且所述载体pCDMAR-epsps的大小为12774bp,序列如SEQ ID NO:1所示。
作为一种优选的方案,步骤(2)中所述感受态细胞为Trans110感受态细胞。
更为优选的是,步骤(4)中所述片段Ⅰ、片段Ⅱ两条片段连接过程使用的连接酶为T4连接酶。
更为优选的是,步骤(7)中所述目的条带与所述pCDMAR-UbiEP中间产物通过KpnI/SacI酶进行酶切连接的连接过程使用的连接酶为T4连接酶。
更为优选的是,步骤(6)中所述引物包括上游引物和下游引物,且所述上游引物序列如SEQ ID NO:21所示,所述下游引物序列如SEQ ID NO:22所示。

本发明的有益效果在于:本发明提供的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps及其构建方法和应用具有以下优势:
(1)本发明的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps的构建方法通过改造原有双T-DNA载体,去除其选择标记基因,包括相应左右Border、启动子以及终止子等元件,得到具有双MAR序列(两个MAR序列之间的部分可以形成DNA环,使外源基因在转基因植物染色质中形成独立的区域,提高外源基因的稳定性和表达水平)、双MAR序列中间具有多克隆位点且不含有选择标记的中间载体;并将启动子、EPSPS和T-nos终止子按要求重组到这个中间质粒上,完成构建。
(2)本发明的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps具有单T-DNA、抗除草剂基因两端有双MAR序列、无选择标记基因、EPSPS基因连接Ubiquitin启动子、启动子靠近Border右臂等特点。
(3)本发明的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps较现有技术具有更好的安全性,而且对外源基因的稳定性和表达水平具有一定的优势,同时单插入位点中的多酶切位点还为实现同时转入2个及2个以上的多基因聚合育种的可能性。

实施例1
1.实验材料
1.1菌株:E.coli DH 5α(根据《分子克隆》实验室制备)、E.coli Trans110(购于北京全式金生物技术有限公司,目录号:CD311).
1.2所用质粒及中间产物:pCDMAR-hyg(Constructed by our lab)、pCUEP102(Constructed by our lab)、pCDMAR、pCDMAR-UbiEP、pCDMAR-epsps。
1.3培养基:LB medium:Tryptone 10g/L,Yeast extract 5g/L,NaCl 10g/L.Adjust pH to 7.0 with 1M NaOH.For plates,add agar 15g/L.(MolecularCloning)。
1.4重要试剂:质粒小提试剂盒购自promaga公司、凝胶纯化试剂盒购自promaga公司、限制性内切酶及连接酶等购自NEB公司。
1.5重要仪器:凝胶成像仪购自Alpha Innotech公司、紫外分光光度计购自岛津公司、低温冷冻离心机购自Eppendorf公司、PCR仪购自Eppendorf公司UV1000型探照紫外仪购自UVP公司。
2.试验方法
2.1质粒构建:参考《分子克隆》中质粒DNA的制备,利用限制性内切酶消化,连接,筛选,鉴定获得重组转化子。
2.2 PCR检测:参考《分子克隆》,利用Taq酶,标准的链式反应扩增程序进行PCR反应,产物进行1%的琼脂糖电泳观察结果。
2.3大肠杆菌的转化与培养(参考分子克隆III):连接产物冰浴30min后42℃温浴90s,热击入大肠杆菌DH5α感受态细胞,37℃温浴1h后,涂布在含有相应抗性的LB培养基上,37℃倒置培养过夜。
2.4一种多功能抗草甘膦水稻转化载体pCDMAR-epsps的构建方法,包括以下步骤,流程参见图1:
(1)骨架选择:选择pCDMAR-hyg作为骨架,且pCDMAR-hyg骨架大小为11724bp,序列如SEQ ID NO:14所示,质粒结构见示意图2;
(2)含pCDMAR-hyg骨架的感受态细胞制备及培养:将pCDMAR-hyg骨架转化到感受态细胞中,并采用常规方法进行培养,获得含pCDMAR-hyg骨架的感受态细胞;所述感受态细胞为Trans110感受态细胞;
(3)酶切:采用BclI酶酶切pCDMAR-hyg骨架,得到片段Ⅰ、片段Ⅱ、片段Ⅲ(酶切产物电泳图参见图3);且片段Ⅰ的大小为6067bp,序列如SEQ ID NO:15所示;片段Ⅱ的大小为2975bp,序列如SEQ ID NO:16所示;片段Ⅲ的大小为2682bp,序列如SEQ ID NO:17所示;
(4)连接:将片段Ⅰ、片段Ⅱ通过T4连接酶连接得到连接产物,并通过ClaI/EcoRV酶切鉴定连接产物(酶切产物电泳图参见图4);选择ClaI/EcoRV酶切后的片段大小为7346bp和1403bp的连接产物,并将其命名为中间质粒pCDMAR;中间质粒pCDMAR的大小为8749bp,序列如SEQ ID NO:18所示,其结构示意图参见图5;
(5)pCDMAR-UbiEP中间产物的获得:利用SbfI/KpnI酶切pCUEP102质粒(pCUEP102质粒结构参见图6),并回收酶切产物(酶切产物电泳图参见图7)中大小为3783bp的片段,命名为酶切片段Ⅰ,序列如SEQ ID NO:19所示;利用SbfI/KpnI酶切中间质粒pCDMAR,回收酶切片段,命名为酶切片段Ⅱ;之后将酶切片段Ⅰ和酶切片段Ⅱ进行连接,获得连接产物,命名为pCDMAR-UbiEP中间产物;pCDMAR-UbiEP中间产物大小为12505bp,序列如SEQ ID NO:20所示,结构示意图参见图8;
(6)带有酶切位点的T-nos片段的获得:设计两端加KpnI和SacI酶切位点的引物,并采用引物扩增pCUEP102上T-nos片段(PCR扩增产物电泳图参见图9),回收大小为269bp的目的条带,且目的条带的序列如SEQ ID NO:8所示;引物包括上游引物和下游引物,且上游引物序列如SEQ ID NO:21所示,下游引物序列如SEQ ID NO:22所示;
(7)酶切连接(具体流程参见图10):将目的条带与pCDMAR-UbiEP中间产物通过KpnI/SacI酶进行酶切,之后通过T4连接酶连接,得到连接产物即为一种多功能抗草甘膦水稻转化载体pCDMAR-epsps;且载体pCDMAR-epsps的大小为12774bp,序列如SEQ ID NO:1所示,其结构示意图参见图11。

Claims (8)

1.一种多功能抗草甘膦水稻转化载体pCDMAR-epsps,其特征在于,所述载体pCDMAR-epsps的大小为12774bp,序列如SEQ ID NO:1所示;且所述载体pCDMAR-epsps中包括11个功能元件,具体见下表:

2.一种多功能抗草甘膦水稻转化载体pCDMAR-epsps的构建方法,其特征在于,所述构建方法包括以下步骤:
(1)骨架选择:选择pCDMAR-hyg作为骨架,且所述pCDMAR-hyg骨架大小为11724bp,序列如SEQ ID NO:14所示;
(2)含pCDMAR-hyg骨架的感受态细胞制备及培养:将pCDMAR-hyg骨架转化到感受态细胞中,并采用常规方法进行培养,获得含pCDMAR-hyg骨架的感受态细胞;
(3)酶切:采用BclI酶酶切所述pCDMAR-hyg骨架,得到片段Ⅰ、片段Ⅱ、片段Ⅲ;且所述片段Ⅰ的大小为6067bp,序列如SEQ ID NO:15所示;片段Ⅱ的大小为2975bp,序列如SEQ IDNO:16所示;片段Ⅲ的大小为2682bp,序列如SEQ ID NO:17所示;
(4)连接:将所述片段Ⅰ、片段Ⅱ连接得到连接产物,并通过ClaI/EcoRV酶切鉴定所述连接产物;选择ClaI/EcoRV酶切后片段大小为7346bp和1403bp的连接产物,并将其命名为中间质粒pCDMAR;所述中间质粒pCDMAR的大小为8749bp,序列如SEQ ID NO:18所示;
(5)pCDMAR-UbiEP中间产物的获得:利用SbfI/KpnI酶酶切pCUEP102质粒获得酶切产物,并回收其中大小为3783bp的片段,命名为酶切片段Ⅰ,序列如SEQ ID NO:19所示;之后利用SbfI/KpnI酶切所述中间质粒pCDMAR,回收酶切片段,命名为酶切片段Ⅱ;之后将所述酶切片段Ⅰ和酶切片段Ⅱ进行连接,获得连接产物,命名为pCDMAR-UbiEP中间产物;所述pCDMAR-UbiEP中间产物的大小为12505bp,序列如SEQ ID NO:20所示;
(6)带有酶切位点的T-nos片段的获得:设计两端加KpnI和SacI酶切位点的引物,并采用所述引物扩增pCUEP102上T-nos片段,回收大小为269bp的目的条带,且所述目的条带的序列如SEQ ID NO:8所示;
(7)酶切连接:将所述目的条带与所述pCDMAR-UbiEP中间产物通过KpnI/SacI酶进行酶切连接,得到连接产物,即为一种多功能抗草甘膦水稻转化载体pCDMAR-epsps;且所述载体pCDMAR-epsps的大小为12774bp,序列如SEQ ID NO:1所示。
3.根据权利要求2所述的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps的构建方法,其特征在于,步骤(2)中所述感受态细胞为Trans110感受态细胞。
4.根据权利要求2所述的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps的构建方法,其特征在于,步骤(4)中所述片段Ⅰ、片段Ⅱ两条片段连接过程使用的连接酶为T4连接酶。
5.根据权利要求2所述的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps的构建方法,其特征在于,步骤(7)中所述目的条带与所述pCDMAR-UbiEP中间产物通过KpnI/SacI酶进行酶切连接的连接过程使用的连接酶为T4连接酶。
6.根据权利要求2所述的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps的构建方法,其特征在于,步骤(6)中所述引物包括上游引物和下游引物,且所述上游引物序列如SEQID NO:21所示,所述下游引物序列如SEQ ID NO:22所示。
7.根据权利要求1所述的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps在获得抗除草剂的水稻品种的应用。
8.根据权利要求1所述的一种多功能抗草甘膦水稻转化载体pCDMAR-epsps在农业领域的应用。
