Abstract
本发明提供一种新型五味消毒饮、制备方法及应用,该新型五味消毒饮由如下重量份的原料药组成:紫背天葵子5‑10份,僵蚕5‑9份,桑白皮6‑12份,金银花10‑20份,野菊花10‑12份,蒲公英10‑12份,紫花地丁5‑10份,和甘草3‑10份;上述药物组合使得各药物功效产生协同作用,从而能够有效治疗小儿急性支气管炎引起的风热邪毒,肺热咳嗽,大便干,舌红,苔黄或腻等症状。

技术领域
本发明涉及中医药技术领域,具体涉及一种新型五味消毒饮、制备方法及应用。
背景技术
热毒症是中医病症名,即温毒,是指火热病邪郁结成毒,也是疔疮、丹毒、热疖等急性热病的统称,又称“火毒”,是由火热郁积所成,易导致疔疮痈肿之类的邪气。
临床表现为手足局部冷痛,痛处肤色青紫发凉,得温痛减,遇冷痛剧,或少腹拘急冷痛,或妇女少腹冷痛,月经后期,经色紫暗,夹有瘀块,伴畏寒肢冷,喜温恶寒,舌淡紫苔白滑,脉沉迟或涩。
热毒炽盛临床表现为壮热口渴,烦躁不安,面红目赤,口舌生疮,痘大而密,疹色紫暗,疱浆混浊,口、咽、眼亦可出现疱疹溃破,便秘尿黄,舌苔黄。上火,口角炎、嘴唇起泡、鼻腔内结庎、口腔溃疡。
目前,针对热毒炽盛的西药主要是一些抗生素类,种类很多,但是其副作用大,长期服用会降低人体自身免疫能力,引起体内代谢紊乱;破坏人体正常菌群;加快细菌的进化,出现耐药菌;而传统的中药组方往往存在治疗的范围太宽泛,针对性不强,导致治疗效果不尽人意等缺点。
发明内容
因此,本发明要解决的技术问题在于克服现有技术中中药组方治疗范围太宽泛,针对性不强,治疗效果不尽人意等缺陷,从而提供一种新型五味消毒饮、制备方法及应用,主要治疗小儿急性支气管炎引起的风热邪毒,肺热咳嗽,大便干,舌红,苔黄或腻等症状。
为此,本发明提供了如下技术方案:
一种新型五味消毒饮,由如下重量份的原料组成:紫背天葵子5-10份,僵蚕5-9份,桑白皮6-12份,金银花10-20份,野菊花10-12份,蒲公英10-12份,紫花地丁5-10份,和甘草3-10份。
优选地,该新型五味消毒饮由如下重量份的原料组成:紫背天葵子5-8份,僵蚕7-9份,桑白皮6-10份,金银花15-20份,野菊花10-12份,蒲公英10-12份,紫花地丁8-10份,和甘草6-10份。
进一步优选地,所述新型五味消毒饮由如下重量份的原料组成:紫背天葵子8份,僵蚕7份,桑白皮10份,金银花15份,野菊花11份,蒲公英11份,紫花地丁8份,和甘草6份。
本发明还提供了一种上述新型五味消毒饮的制备方法,包括如下步骤:
(1)按照选定的重量份数称取上述各原料药,加4-20倍的水煎煮二次,每次2小时,滤过,合并滤液;
(2)将滤液浓缩成相对密度为1.18~1.19(25℃)的稠膏,加入相当于稠膏1倍体积量的乙醇,摇匀,静置2-3h使沉淀,滤过,取滤液回收乙醇,浓缩成相对密度为1.18~1.19(25℃)得稠膏,干燥。
将步骤(2)中干燥后的稠膏研成60目细粉,将所得细粉加入常规辅料按照常规工艺制备得到口服制剂。
所述口服制剂为散剂,片剂,颗粒剂,胶囊剂,溶液剂,乳剂,混悬剂中的任意一种。
本发明还提供了上述新型五味消毒饮或上述新型五味消毒饮的制备方法制得的新型五味消毒饮在治疗小儿急性呼吸道感染中的应用。
本发明所述新型五味消毒饮中,各组份原料的药理及药效为:
紫背天葵子:甘、苦,寒。归肝、胃经。功能主治清热解毒,消肿散结。用于痈肿疔疮,乳痈,瘰疠,毒蛇咬伤。
僵蚕:咸、辛,平。归肝、肺、胃经。功能主治祛风定惊,化痰散结。用于惊风抽搐,咽喉肿痛,皮肤瘙痒,颌下淋巴结炎,面神经麻痹。
桑白皮:甘,寒。归肺经。功能主治泻肺平喘,利水消肿。用于肺热喘咳,水肿胀满尿少,面目肌肤浮肿。
金银花:性甘、寒。归肺、胃经。清热解毒,消炎退肿。主治外感风热或温病发热,中暑,热毒血痢,痈肿疔疮,喉痹,多种感染性疾病。
野菊花:味苦、辛、凉。清热解毒,疏风散热,散瘀,明目,降血压。防治流行性脑脊髓膜炎,预防流行性感冒、感冒,治疗高血压、肝炎、痢疾、痈疖疗疮都有明显效果。
蒲公英:苦甘,寒。归入肝、胃经。功能主治:清热解毒,利尿散结。治急性乳腺炎,淋巴腺炎,瘰疬,疔毒疮肿,急性结膜炎,感冒发热,急性扁桃体炎,急性支气管炎,胃炎,肝炎,胆囊炎,尿路感染。
紫花地丁:味苦、微辛,性寒。归心、肝经。气微散泄;具有清热解毒,凉血消肿的功效;主治疔疮痈肿、痄腮,瘰疬,丹毒,乳痈,肠痈,湿热与泻痢,黄疸,目赤肿痛,喉痹,毒蛇咬伤。
甘草:性味:甘,平。归心经、肺经、脾经、胃经。益气补中,清热解毒,祛痰止咳,缓急止痛,调和药性。主治脾气虚弱,倦怠乏力,心悸气短,咳嗽痰多,脘腹、四肢挛急疼痛,热毒疮疡,咽喉肿痛,药物、食物中毒;缓和药物烈性、毒性。
本发明技术方案,具有如下优点:
1.本发明提供的新型五味消毒饮,由如下重量份的原料组成:紫背天葵子5-10份,僵蚕5-9份,桑白皮6-12份,金银花10-20份,野菊花10-12份,蒲公英10-12份,紫花地丁5-10份,和甘草3-10份,组方中的紫背天葵子具有清热解毒,止咳,活血,消肿之功效,僵蚕具有祛风定惊,化痰散结的功能,桑白皮具有泻肺平喘之功效,三者共为君药,金银花清热解毒,野菊花疏风清热、消肿解毒,二者为臣药,蒲公英清热解毒,利尿散结,紫花地丁清热解毒,凉血消肿,二者为佐药,甘草益气补中,清热解毒,祛痰止咳,缓急止痛,调和药性,为使药,将上述药物组合使得各药物功效产生协同作用,从而能够有效治疗小儿急性支气管炎引起的风热邪毒,肺热咳嗽,大便干,舌红,苔黄或腻等症状。
2.本发明提供的新型五味消毒饮的制备方法工艺简单,适于工业化生产。
具体实施方式
实施例1
本实施例提供一种新型五味消毒饮及其制备方法
(1)称取紫背天葵子8Kg,僵蚕7Kg,桑白皮10Kg,金银花15Kg,野菊花11Kg,蒲公英11Kg,紫花地丁8Kg,和甘草6Kg,混合,加4倍的水煎煮二次,每次2小时,滤过,合并滤液;
(2)将滤液浓缩成相对密度为1.18~1.19(25℃)的稠膏,加入相当于稠膏1倍体积量的乙醇,摇匀,静置3h使沉淀,滤过,取滤液回收乙醇,浓缩成相对密度为1.18~1.19(25℃)得稠膏,干燥,研成60目细粉;
(3)将步骤(2)所得细粉加入常规辅料按照常规工艺制备得到片剂。
实施例2
本实施例提供一种新型五味消毒饮及其制备方法
(1)称取紫背天葵子10Kg,僵蚕5Kg,桑白皮12Kg,金银花10Kg,野菊花12Kg,蒲公英12Kg,紫花地丁5Kg,和甘草10Kg,加20倍的水煎煮二次,每次2小时,滤过,合并滤液;
(2)将滤液浓缩成相对密度为1.18~1.19(25℃)的稠膏,加入相当于稠膏1倍体积量的乙醇,摇匀,静置2h使沉淀,滤过,取滤液回收乙醇,浓缩成相对密度为1.18~1.19(25℃)得稠膏,干燥,研成60目细粉;
(3)将步骤(2)所得细粉加入常规辅料按照常规工艺制备得到颗粒剂。
实施例3
本实施例提供一种新型五味消毒饮及其制备方法
(1)称取紫背天葵子5Kg,僵蚕9Kg,桑白皮6Kg,金银花20Kg,野菊花10Kg,蒲公英10Kg,紫花地丁10Kg,和甘草3Kg,加10倍的水煎煮二次,每次2小时,滤过,合并滤液;
(2)将滤液浓缩成相对密度为1.18~1.19(25℃)的稠膏,加入相当于稠膏1倍体积量的乙醇,摇匀,静置2.5h使沉淀,滤过,取滤液回收乙醇,浓缩成相对密度为1.18~1.19(25℃)得稠膏,干燥,研成60目细粉;
(3)将步骤(2)所得细粉加入常规辅料按照常规工艺制备得到散剂。
对比例
本对比例提供一种宣肺平喘的中药组合物及其制备方法
(1)称取僵蚕7Kg,桑白皮10Kg,和甘草6Kg,混合,加4倍的水煎煮二次,每次2小时,滤过,合并滤液;
(2)将滤液浓缩成相对密度为1.18~1.19(25℃)的稠膏,加入相当于稠膏1倍体积量的乙醇,摇匀,静置3h使沉淀,滤过,取滤液回收乙醇,浓缩成相对密度为1.18~1.19(25℃)得稠膏,干燥,研成60目细粉;
(3)将步骤(2)所得细粉加入常规辅料按照常规工艺制备得到片剂。
实验例本发明的新型五味消毒饮在治疗急性支气管炎中的考察
1.材料和方法
1.1实验动物
清洁级昆明种小鼠,4周龄,体重18-22g,雌雄各半。小鼠适应性喂养1周后进行实验。实验前禁食过夜,自由饮水。
1.2药物
实施例1中制备的新型五味消毒饮;中国专利CN108714177A中实施例1制备的中药组合物。
1.3试剂盒
小鼠IL-2ELISA试剂盒(上海仁捷生物科技有限公司),小鼠IL-8ELISA试剂盒(上海颖心实验室设备有限公司)。
1.4实验方法
1.4.1小鼠急性支气管炎模型模型构建
将上述小鼠随机分成2大组,正常组10只,模型组50只,除正常组(空白对照组)外,其余小鼠放于烟熏箱内,均以刨花、烟叶各50g置于熏炉内引燃,烟雾通过管道引流入烟熏箱,刺激烟熏箱内小鼠呼吸道,每次30min,每日两次,一周后结束。
观察造模后的小鼠,是否出现咳嗽、流涕、精神萎靡、消瘦等征象。取模型组小鼠肺组织,做病理切片,观察光镜下模型组小鼠是否出现气管、支气管粘膜充血、肿胀,浅层纤毛上皮细胞受损,粘液腺体肥大,粘膜下层中性粒细胞浸润,有大量分泌物等情况。观察正常组小鼠,是否出现健康活泼,毛发光泽,无咳嗽、流涕等征象。观察其光镜下病理切片是否显示肺内各支气管结构正常,柱状上皮细胞整齐排列,无明显损伤,纤毛无缺失,支气管粘膜无充血,肺泡结构清晰,肺泡壁毛细血管无充血,未见炎症细胞浸润等情况。依据以上综合分析构建的小鼠模型,以模型组小鼠典型病理改变为依据,证实急性支气管炎模型小鼠造模成功。
1.4.2实验分组
将上述造模成功的小鼠随机分成6组,每组10只;正常组为空白对照组,具体分组如下:
①空白对照组:给予同体积生理盐水灌胃给药,每日一次。
②模型对照组:给予同体积生理盐水灌胃给药,每日一次。
③阳性对照组:给予中国专利CN108714177A中实施例1制备的中药组合物,以100mg/kg以等体积灌胃给药,每日一次。
④低剂量组:取实施例1制备得到新型五味消毒饮,以50mg/kg以等体积灌胃给药,每日一次。
⑤中剂量组:取实施例1制备得到的新型五味消毒饮,以100mg/kg以等体积灌胃给药,每日一次。
⑥高剂量组:取实施例1制备得到的新型五味消毒饮,以150mg/kg以等体积灌胃给药,每日一次。
⑦对比例组:取对比例制备得到的宣肺平喘的中药组合物,以100mg/kg以等体积灌胃给药,每日一次。
2.检测指标、数据处理及结果
2.1检测指标
各组按照上述剂量灌胃治疗7天后,各组小鼠禁食12小时(可自由饮水),称重后摘眼球取血,用无菌干燥管收集标本,室温下3000rpm离心5min,取上清液。将上清液分别按照小鼠IL-2ELISA试剂盒和小鼠IL-8ELISA试剂盒中的说明书操作,分别检测IL-2和IL-8的含量。

3.1IL-2检测结果
注:与模型对照组相比,*P<0.05,**P<0.01;与阳性对照组相比,#P<0.05,##P<0.01;与对比例组相比,$P<0.05,$$P<0.01。
由表1结果可知,各给药组均能提升模型组小鼠血清中抗炎因子IL-2水平(P<0.05),IL-2是T细胞生长因子,其水平的提升,能够增强机体的免疫功能,从而达到抑制炎症的作用。本发明药物组合物的高、中、低剂量组与模型对照组相比差异性显著,P<0.05,其中高剂量组、中剂量组差异性更显著(P<0.01),说明本发明的药物组合物随着给药剂量的增加,其在治疗急性呼吸道感染方面效果越显著。本发明药物组合物的高、中剂量组分别与阳性对照组、对比例组相比差异性显著,P<0.05,其中高剂量组差异性更显著(P<0.01),说明其在治疗急性呼吸道感染方面效果要优于阳性对照组和对比例组。
3.2IL-8检测结果
注:与模型对照组相比,*P<0.05,**P<0.01;与阳性对照组相比,#P<0.05,##P<0.01;与对比例组相比,$P<0.05,$$P<0.01。
由表2结果可知,各给药组可在降低各模型组小鼠血清IL-8水平,IL-8是重要的促炎因子,在气道内聚、活化中性粒细胞,诱导释放溶酶体酶,是炎症反应的重要介质,降低IL-8水平,达到抑制炎症的作用。本发明药物组合物的高、中、低剂量组与模型对照组相比差异性显著,P<0.05,其中高剂量组差异性更显著(P<0.01),说明本发明的药物组合物随着给药剂量的增加,其在治疗急性呼吸道感染方面效果越显著。本发明药物组合物的高、中剂量组与阳性对照组相比差异性显著,P<0.05,其中高剂量组差异性更显著(P<0.01),本发明药物组合物的高、中、低剂量组与对比例组相比差异性显著,P<0.01,综上,说明本发明药物组合物在治疗急性呼吸道感染方面效果要优于阳性对照组和对比例组。

1.一种新型五味消毒饮,其特征在于,由如下重量份的原料组成:紫背天葵子5-10份,僵蚕5-9份,桑白皮6-12份,金银花10-20份,野菊花10-12份,蒲公英10-12份,紫花地丁5-10份,和甘草3-10份。
2.根据权利要求1所述一种新型五味消毒饮,其特征在于,由如下重量份的原料组成:紫背天葵子5-8份,僵蚕7-9份,桑白皮6-10份,金银花15-20份,野菊花10-12份,蒲公英10-12份,紫花地丁8-10份,和甘草6-10份。
3.根据权利要求1或2所述一种新型五味消毒饮,其特征在于,由如下重量份的原料组成:紫背天葵子8份,僵蚕7份,桑白皮10份,金银花15份,野菊花11份,蒲公英11份,紫花地丁8份,和甘草6份。
4.一种制备如权利要求1-3中任一项所述一种新型五味消毒饮的方法,其特征在于,该方法包括如下步骤:
(1)按照选定的重量份数称取上述各原料药,加4-20倍的水煎煮二次,每次2小时,滤过,合并滤液;
(2)将滤液浓缩成相对密度为1.18~1.19(25℃)的稠膏,加入相当于稠膏1倍体积量的乙醇,摇匀,静置2-3h使沉淀,滤过,取滤液回收乙醇,浓缩成相对密度为1.18~1.19(25℃)得稠膏,干燥。
5.根据权利要求4所述一种新型五味消毒饮的制备方法,其特征在于,将步骤(2)中干燥后的稠膏研成60目细粉,将所得细粉加入常规辅料按照常规工艺制备得到口服制剂。
6.根据权利要求5所述一种新型五味消毒饮的制备方法,其特征在于,所述口服制剂为散剂,片剂,颗粒剂,胶囊剂,溶液剂,乳剂,混悬剂中任意一种。
7.一种如权利要求1-3任一项所述新型五味消毒饮或权利要求4-6任一项所述新型五味消毒饮的制备方法制得的新型五味消毒饮在制备治疗小儿急性支气管炎药物中的应用。
