Abstract
本发明涉及一种利用蛹虫草发酵液配制保健饮料的方法,该方法包括以下步骤:(1)蛹虫草母种活化;(2)摇瓶液体菌种制备;(3)种子罐液体菌种制备;(4)发酵罐发酵培养;(5)蛹虫草口服液的调配。本发明设计科学合理,配制方法配方简明、工艺简单;得到的产品口感细腻、丰富、纯正,具有扶正益气、补肺益肾、止咳平喘、健胃整肠、调节体内血脂、血糖、提高免疫力等辅助功能;本发明保留了蛹虫草发酵产物中所有功能性物质,提高了功效性成份的利用率,对于充分利用蛹虫草资源、增加蛹虫草产品的种类、提高蛹虫草的经济价值都具有较高的理论意义和市场价值。

技术领域
本发明属于食品研发技术领域,涉及功能饮料,特别涉及一种利用蛹虫草液体发酵饮料的方法。
背景技术
冬虫夏草(Cordyceps sinensis)是我国名贵的中药材,其宿主蝙蝠蛾幼虫对生长环境的要求极为严苛,难以实现规模化饲养。野生冬虫夏草资源紧缺,价格十分昂贵,寻求野生虫草的替代品逐渐成为人们的研究热点。蛹虫草(Cordyceps militaris)为子囊菌亚门,麦角菌目,麦角菌科、虫草属的模式种,其营养价值丰富,是一种药食两用真菌。蛹虫草的应用最早起源于中国,明代李时珍的《本草纲目》中记载,蝉花主治小儿天吊惊痈、夜啼、心悸,其中蝉花即指蛹虫草和蝉花等多种虫草菌;《新华本草纲要》中也有记载,蛹虫草味甘、性平,有益肺补肾、补精髓、止血化疲的功效。中医学认为蛹虫草甘平、入肺肾二经,既有益肾补阳,用于治疗肾阴不足、腰膝酸软、弦晕耳鸣等疾症,又有补肺阴,对肺肾不足、久咳虚弱及劳咳痰血者有较好的疗效。蛹虫草与冬虫夏草的功效成份和药理作用类似,其中多糖类、虫草酸、虫草素、超氧化物歧化酶等功能性物质具有抗菌、抗炎、抗肿瘤、抗氧化、调节免疫系统等功能,对脑血栓、脑出血、心肌梗塞等具有预防与辅助治疗作用。
柠檬是世界上最有药用价值的水果之一,它富含维生素C、维生素B1、维生素B2、维生素B3、钙、磷、铁、奎宁酸、柠檬酸、苹果酸、橙皮苷、柚皮苷、香豆精,高钾低钠,对人体十分有益。柠檬味酸甘、性平、入肝,有化痰止咳,生津健脾的功效,对人体发挥的作用犹如天然抗生素,具有抗菌消炎、抗氧化、增强人体免疫力等多种功能,对促进肌肤的新陈代谢、延缓衰老及抑制色素沉着等十分有效。
金桔皮色金黄、汁多香甜,含有特殊的挥发油、金桔苷等物质,具有令人愉悦的香气,是颇具特色的水果。中医学认为,金桔味甘,人肺、胆经,具有抗炎、去痰、助消食、双向调节血压、增强心脏功能和理气止咳等功效,对支气管炎有较显著疗效。除此之外金桔还具有防止血管破裂,减缓血管硬化,减少毛细血管脆性和通透性等作用。
随着人们生活水平的提高,对养生保健越来越重视,对保健品的需求量也越来越大。我国自古以来有食疗的传统,所以药食两用真菌在保健品领域具有巨大的发展空间。蛹虫草自身具有较高的药用价值,但因其有效成份提取工艺复杂,提取率较低而限制了蛹虫草的开发和应用,因此发明一项能将蛹虫草液体发酵产物完全利用的工艺,开发出一种具有保健功能的复合型饮料,不仅可提高其利用率,还可为蛹虫草资源的科学研究和开发应用提供新思路。
目前所报道的以蛹虫草为发酵菌种制备的功能性饮料,普遍采用微生物培养基或以一些药、食源性提取液进行发酵。然后利用分离提取技术对菌体和菌液进行分离。一种是以发酵液为主体,向其中添加一些功效成分提取物进行复配,另一种是将菌体通过干燥、粉碎等工艺,通过复配制备成悬浮型饮料,如此工艺环节繁琐,耗能量大,操作复杂,不利大规模生产,而且产品在口感、风味、外观等均存在一些不足。因此,蛹虫草饮料开发技术还有待于进一步发展。
本发明利用蛹虫草为菌种,将其发酵产物与其他功能性成分直接进行复配,制备一款营养价值和保健功效俱佳的新型饮料,该工艺简单易行、安全环保,不产生“三废”,不仅有良好的经济效益,且能实现清洁化生产。本发明采取一步发酵成型的工艺,培养基配方利于蛹虫草对各种营养成分的综合利用,可促进代谢,产生风味物质及功效成分,再通过复配,得到的产品风味独特,气味清爽怡人,功效显著。
通过对公开专利文献的检索,并未发现与本专利申请相同的公开专利文献。
发明内容
本发明的目的在于克服现有技术的不足,提供一种利用蛹虫草发酵液配制保健饮料的方法,该方法配方简明、工艺简单;保留了蛹虫草发酵产物中所有功能性物质,提高了功效性成份的利用率,对于充分利用蛹虫草资源、增加蛹虫草产品的种类、提高蛹虫草的经济价值都具有较高的理论意义和市场价值。
本发明解决其技术问题是通过以下技术方案实现的:
一种利用蛹虫草发酵液配制保健饮料的方法,其特征在于:该方法包括如下步骤:
1)蛹虫草母种活化:将蛹虫草菌种接种到PDA培养基中,于22~25℃恒温培养箱中培养7~10天,获得活化的蛹虫草菌种;
2)摇瓶液体菌种制备:摇瓶液体菌种培养基配方:蔗糖40g/L,蛋白胨20g/L,磷酸二氢钾1g/L,硫酸镁1g/L,pH6-7,121℃高压灭菌20min,备用;具体配制步骤为:采用2000mL三角瓶用于液体种子培养,接入2cm2的斜面菌种,摇床转速180r/min,装液量为800/2000mL,温度20~25℃,振荡培养5天,得到摇瓶菌种液;
3)种子罐液体菌种制备:
a)种子罐空消:121℃高压灭菌20min;
b)种子罐发酵培养基配方:蔗糖40g/L,蛋白胨20g/L,磷酸二氢钾1-2g/L,硫酸镁1-2g/L,维生素B110-30 mg/L,维生素B1210-30 mg/L,消泡剂适量,pH6-7,121℃高压灭菌20min,备用;
c)将摇瓶菌种液接种至装有发酵培养基的300L种子罐中,装液量为50%~60%,接种量为10%~20%,接种后于20~25℃,搅拌转速180-220r/min,通气量25~30L/min,发酵培养4~5天,得到种子罐菌种液;
4)发酵罐发酵培养:
a)发酵罐空消:121℃高压灭菌20min;
b)发酵罐培养基:蔗糖40g/L,蛋白胨30g/L,磷酸二氢钾1-2g/L,硫酸镁1-2g/L,维生素B110-30 mg/L,维生素B1210-30 mg/L,消泡剂适量,pH6-7,121℃高压灭菌20min,备用;
c)将种子罐菌种液接种至装有发酵培养基的3T发酵罐中,装液量为50%~60%,接种量为7.5%~10%,接种后于20~25℃,搅拌转速180~220r/min,通气量25~30L/min,发酵培养5~7天,得到蛹虫草发酵物;
5)蛹虫草口服液的调配:蛹虫草口服液配方:水50%~70%,蛹虫草发酵物10%~30%,金桔原汁5%~15%,果葡糖浆3%~5%,白砂糖2%~4%,柠檬原汁2%~4%,一水柠檬酸0.02%~0.05%,苹果酸0.02%~0.05%,柠檬酸钠0.02%~0.05%,羧甲基纤维素钠0.02%~0.05%,黄原胶0.02%~0.05%;具体步骤为:
a)将蛹虫草发酵产物使用胶体磨胶磨至粒度为5μm;
b)新鲜金桔洗净,用刺孔压籽机刺孔压籽后,在0.8%的食盐水中浸泡40min,脱苦,去籽,沥干,打浆,备用;柠檬经清洗、切分、打浆,备用;将得到的原料分别离心,分离原汁中的杂质以及沉淀物等,得到原汁;
c)将胶体磨处理后的蛹虫草发酵物与水、金桔原汁、柠檬原汁、果葡糖浆、白砂糖混匀,然后加入柠檬酸、苹果酸和柠檬酸钠搅拌均匀成混合液;
d)取羧甲基纤维素钠、黄原胶加入适量水加热至完全溶解,加入到步骤(c)混合液中并搅拌均匀成饮料;
e)将步骤(d)调配好的饮料置于20MPa,55℃下均质2次,然后在130℃下瞬时高温灭菌10s;
f)待饮料冷却至90~95℃时,趁热灌装,灌装后在95℃常压下杀菌15min,然后冷却至室温,得到蛹虫草复合饮料。
本发明的优点和有益效果为:
1、本发明的方法配方简明、工艺简单;得到的产品口感细腻、丰富、纯正,具有扶正益气、补肺益肾、止咳平喘、健胃整肠、调节体内血脂、血糖、提高免疫力等辅助功能。
2、本发明的方法保留了蛹虫草发酵产物中所有功能性物质,提高了功效性成份的利用率,对于充分利用蛹虫草资源、增加蛹虫草产品的种类、提高蛹虫草的经济价值都具有较高的理论意义和市场价值。
3、本发明工艺简单易行、安全环保,不产生废气、废水、废渣,不仅有良好的经济效益,能满足保健品的市场需求,且能实现清洁化生产。

具体实施方式
下面通过具体实施例对本发明作进一步详述,以下实施例只是描述性的,不是限定性的,不能以此限定本发明的保护范围。
一种利用蛹虫草发酵液配制保健饮料的方法,其创新之处在于:该方法包括如下步骤:
1)蛹虫草母种活化:将蛹虫草菌种接种到PDA培养基中,于22~25℃恒温培养箱中培养7~10天,获得活化的蛹虫草菌种;
2)摇瓶液体菌种制备:摇瓶液体菌种培养基配方:蔗糖40g/L,蛋白胨20g/L,磷酸二氢钾1g/L,硫酸镁1g/L,pH6-7,121℃高压灭菌20min,备用;具体配制步骤为:采用2000mL三角瓶用于液体种子培养,接入2cm2的斜面菌种,摇床转速180r/min,装液量为800/2000mL,温度20~25℃,振荡培养5天,得到摇瓶菌种液;
3)种子罐液体菌种制备:
a)种子罐空消:121℃高压灭菌20min;
b)种子罐发酵培养基配方:蔗糖40g/L,蛋白胨20g/L,磷酸二氢钾1-2g/L,硫酸镁1-2g/L,维生素B110-30 mg/L,维生素B1210-30 mg/L,消泡剂适量,pH6-7,121℃高压灭菌20min,备用;
c)将摇瓶菌种液接种至装有发酵培养基的300L种子罐中,装液量为50%~60%,接种量为10%~20%,接种后于20~25℃,搅拌转速180-220r/min,通气量25~30L/min,发酵培养4~5天,得到种子罐菌种液;
4)发酵罐发酵培养:
a)发酵罐空消:121℃高压灭菌20min;
b)发酵罐培养基:蔗糖40g/L,蛋白胨30g/L,磷酸二氢钾1-2g/L,硫酸镁1-2g/L,维生素B110-30 mg/L,维生素B1210-30 mg/L,消泡剂适量,pH6-7,121℃高压灭菌20min,备用;
c)将种子罐菌种液接种至装有发酵培养基的3T发酵罐中,装液量为50%~60%,接种量为7.5%~10%,接种后于20~25℃,搅拌转速180~220r/min,通气量25~30L/min,发酵培养5~7天,得到蛹虫草发酵物;
5)蛹虫草口服液的调配:蛹虫草口服液配方:水50%~70%,蛹虫草发酵物10%~30%,金桔原汁5%~15%,果葡糖浆3%~5%,白砂糖2%~4%,柠檬原汁2%~4%,一水柠檬酸0.02%~0.05%,苹果酸0.02%~0.05%,柠檬酸钠0.02%~0.05%,羧甲基纤维素钠0.02%~0.05%,黄原胶0.02%~0.05%;具体步骤为:
a)将蛹虫草发酵产物使用胶体磨胶磨至粒度为5μm;

f)待饮料冷却至90~95℃时,趁热灌装,灌装后在95℃常压下杀菌15min,然后冷却至室温,得到蛹虫草复合饮料。

Claims (1)
1.一种利用蛹虫草发酵液配制保健饮料的方法,其特征在于:该方法包括如下步骤:
1)蛹虫草母种活化:将蛹虫草菌种接种到PDA培养基中,于22~25℃恒温培养箱中培养7~10天,获得活化的蛹虫草菌种;
2)摇瓶液体菌种制备:摇瓶液体菌种培养基配方:蔗糖40g/L,蛋白胨20g/L,磷酸二氢钾1g/L,硫酸镁1g/L,pH6~7,121℃高压灭菌20min,备用;具体配制步骤为:采用2000mL三角瓶用于液体种子培养,接入2cm2的斜面菌种,摇床转速180r/min,装液量为800/2000mL,温度20~25℃,振荡培养5天,得到摇瓶菌种液;
3)种子罐液体菌种制备:
a)种子罐空消:121℃高压灭菌20min;
b)种子罐发酵培养基配方:蔗糖40g/L,蛋白胨20g/L,磷酸二氢钾1-2g/L,硫酸镁1-2g/L,维生素B110-30 mg/L,维生素B1210-30 mg/L,消泡剂适量,pH6-7,121℃高压灭菌20min,备用;
c)将摇瓶菌种液接种至装有发酵培养基的300L种子罐中,装液量为50%~60%,接种量为10%~20%,接种后于20~25℃,搅拌转速180-220r/min,通气量25~30L/min,发酵培养4~5天,得到种子罐菌种液;
4)发酵罐发酵培养:
a)发酵罐空消:121℃高压灭菌20min;
b)发酵罐培养基:蔗糖40g/L,蛋白胨30g/L,磷酸二氢钾1~2g/L,硫酸镁1~2g/L,维生素B110~30mg/L,维生素B1210~30mg/L,消泡剂适量,pH6~7,121℃高压灭菌20min,备用;
c)将种子罐菌种液接种至装有发酵培养基的3T发酵罐中,装液量为50%~60%,接种量为7.5%~10%,接种后于20~25℃,搅拌转速180~220r/min,通气量25~30L/min,发酵培养5~7天,得到蛹虫草发酵物;
5)蛹虫草口服液的调配:蛹虫草口服液配方:水50%~70%,蛹虫草发酵物10%~30%,金桔原汁5%~15%,果葡糖浆3%~5%,白砂糖2%~4%,柠檬原汁2%~4%,一水柠檬酸0.02%~0.05%,苹果酸0.02%~0.05%,柠檬酸钠0.02%~0.05%,羧甲基纤维素钠0.02%~0.05%,黄原胶0.02%~0.05%;具体步骤为:
a)将蛹虫草发酵产物使用胶体磨胶磨至粒度为5μm;
b)新鲜金桔洗净,用刺孔压籽机刺孔压籽后,在0.8%的食盐水中浸泡40min,脱苦,去籽,沥干,打浆,备用;柠檬经清洗、切分、打浆,备用;将得到的原料分别离心,分离原汁中的杂质以及沉淀物等,得到原汁;
