From the experimental results in table 4, it can be seen that the skin moisture content increase rate can reach more than 46% after 1 hour by using the repair essence prepared by the invention, and the skin moisture content increase rate can be maintained at more than 39% after 8 hours, which indicates that the repair essence has better moisturizing effect on skin and can achieve long-acting water-locking and moisturizing effects, while the skin moisture content increase rate is smaller and the moisturizing effect is poorer in the comparative example, which indicates that each component in the repair essence provided by the invention has synergistic effect, has good moisturizing effect, enables skin to be moisturized and smooth, and achieves the effects of lasting moisturizing and moistening.
3. Skin barrier function test
The skin moisture loss TEWL is an important parameter for evaluating the function of a skin moisture protective layer and can be used for evaluating the strength of the skin barrier function. The better the skin protection layer, the higher the moisture content and the lower the value of the skin moisture loss TEWL, which has the unit: g/hm2
The test method comprises the following steps: selecting 50 volunteers of 20-45 years old as trial objects, randomly dividing the 50 volunteers into 10 groups (5 in each group), and enabling the skin of the test subjects to be healthy and accord with the selection standard. Each subject was uniformly applied the repair essence of examples 1 to 5 and comparative examples 1 to 5 on the face after cleansing in the morning, the TEWL values before application of the cosmetics and after application of 1 hour and 8 hours were respectively measured using a Tewameter TM300, the forehead region of the subject was selected as the test area, and the variation rate of the TEWL value was calculated. The change rate (%) (water loss after use-initial water loss)/initial water loss, and the average change rate (%) (sum of change rates of TEWL values per group/number of persons in the group, the smaller this value, the smaller the water loss, the stronger the skin barrier function of the test sample. During the experiment, the subject was not able to apply any other cosmetic product at the experimental site. The results of the skin TEWL measurements are shown in Table 5.
Table 5 skin barrier function test results
Figure GDA0003200880770000102
Figure GDA0003200880770000111
The experimental results in table 5 show that the repair essence of the examples has a good skin barrier function, the mean change rate of the skin TEWL value after 1 hour is more than 30%, and the mean change rate of the skin TEWL value after 8 hours is more than 25%, while the change rate of the skin TEWL value is smaller when the comparative repair essence is used, which indicates that the repair essence prepared by the invention can enhance the skin barrier function and resist the damage of the external environment to the skin.
4. Antiallergic efficacy test
The test principle is as follows: when the allergen invades the organism for the first time, the organism generates antibody IgE, and when the allergen invades the organism again, the antibody is combined with IgE on the surface of a sensitized cell and FceRI crosslinking occurs, so that an activation signal is started, signal conduction is triggered, and various enzymes including intracellular hyaluronidase are activated, and the process is called as anaphylactic reaction. Therefore, the anti-allergy activity can be judged by the hyaluronidase activity inhibition rate, and the higher the hyaluronidase activity inhibition rate is, the stronger the anti-allergy effect of the component is reflected, and the weaker the anti-allergy effect is, on the contrary, the component is.
The test method comprises the following steps: 0.1mL of 0.2 was added to the tube5mmo1/LCaC12Incubating the solution and 0.5mL of hyaluronidase solution (500U/mL) at 37 ℃ for 20min, adding 0.5mL of the repair essence of examples 1, 3 and 5 and comparative examples 1, 3 and 5, respectively, and incubating at 37 ℃ for 20 min; adding 0.5mL of sodium hyaluronate solution, and keeping the temperature at 37 ℃ for 30 min; then standing for 5min at normal temperature, adding 0.1mL of 0.4mo1/LNaOH solution and 0.5mL of phthalein acetone solution, heating in a boiling water bath for reaction for 15min, and immediately carrying out ice bath for 5 min; then adding 1.0mL of Ellisib reagent, diluting with 3.0mL of absolute ethanol, standing for 20min, developing, measuring the absorbance value at 530nm with a spectrophotometer, and substituting into the following formula to calculate the inhibition rate. The results are shown in Table 6.
Hyaluronidase inhibition (%) - (A-B) - (C-D) ]/(A-B) × 100%
In the formula:
a-absorbance value of control solution (sample solution replaced by acetic acid buffer solution)
B-blank control solution absorbance value (sample solution and enzyme solution were replaced by acetic acid buffer solution)
C-absorbance value of test sample solution
D-absorbance of sample blank solution (replacing enzyme solution with acetic acid buffer solution)
Evaluation criteria: in order to facilitate the evaluation of the anti-allergic activity of the sample, the anti-allergic effect is classified into the following 4 grades according to international standards: the inhibition rate is more than or equal to 70 percent, and the strong antiallergic effect is achieved; the inhibition rate is more than or equal to 50 percent, and the medium antiallergic effect is achieved; the inhibition rate is more than or equal to 30 percent and less than 50 percent, and the weak antiallergic effect is achieved; an inhibition rate of less than 30% is no antiallergic effect.
TABLE 6 anti-allergic efficacy test results
Sample (I) Example 1 Example 3 Example 5 Comparative example 1 Comparative example 3 Comparative example 5
Inhibition ratio (%) 72.30% 75.48% 80.12% 47.81% 43.57% 58.66%
From the experimental results in table 6, it can be seen that the repair essence in the examples can significantly inhibit the activity of hyaluronidase, the inhibition rates are all greater than 70%, indicating that the repair essence has strong antiallergic effect, while the comparative examples show weak antiallergic effect, indicating that the repair essence of the invention has soothing and antiallergic effects.
