技术领域
本发明属于保健品领域,尤其涉及一种眼部保健品及其制备方法。
背景技术
目前,随着互联网技术的不断普及,电子产品的应用越来越广泛,人们眼睛盯着电子屏幕的时间越来越长,眼部疾病特别是近视的出现频率越来越高,并且呈年轻化趋势发展。据统计世界近视眼患者就超过5亿,单纯性近视给患者的生活带来不便,而高度近视,特别是病理性近视则可能伴随各种严重的并发症,甚至致盲。目前近视彻底根治的方法为手术方法,但是,手术方法有一定的风险性,如果不成功有致盲的风险。因而做好眼部保健,预防近视眼就越来越引起人们的关注。
叶黄素酯、β-胡萝卜素与二十二碳六烯酸(DHA)都是比较常见的眼部保健成分,广泛应用于各种眼部保健品之中。其中,叶黄素酯是一种重要的类胡萝卜素脂肪酸酯,它广泛存在于万寿菊花提取物、南瓜、甘蓝、苜蓿等植物体内,其主要结构由两个六元碳环与一个含十八碳原子的共轭双键的长链连接而成,叶黄素酯被摄入体内后,在人体脂肪酶的作用下,水解成游离态的叶黄素,沉积于人眼底黄斑中,可作为近紫外蓝光的吸收剂,通过捕获自由基、防止或降低氧化和自由基对视网膜带来的伤害而起到保护功能。β-胡萝卜素是一种抗氧化剂,具有解毒作用,是维护人体健康不可缺少的营养素,在抗癌、预防心血管疾病、白内障及抗氧化上有显著的功能。而DHA是一种长链多不饱和脂肪酸,是视网膜的重要构成成分,占视网膜脂肪总量的50%,其能够保护视网膜,提高视网膜对光的敏感度,改善视力。
但是,叶黄素酯与DHA的性质相对活泼,比较容易氧化,β-胡萝卜素对光热也比较敏感,不易储存;另外,由叶黄素酯、β-胡萝卜素或DHA制得的保健品中活性成分含量和生物利用度都比较低,特别是叶黄素酯受到胃酸影响较大,不利于保健品充分发挥其效果。
为了解决以上存在的问题,人们一直在寻求一种理想的技术解决方案。
发明内容
本发明的目的是针对现有技术的不足,从而提供一种眼部保健品及其制备方法。
为了实现上述目的,本发明所采用的技术方案是:
一种眼部保健品,包括活性成分和医学上可接受的辅料,以质量份数计,所述活性成分包括组分:二十二碳六烯酸3~7份、叶黄素酯0.15~40份、β-胡萝卜素2~3份、蓝莓提取物0.15~40份和决明子提取物0.5~0.8份与阿胶6~10份;
所述辅料可根据需要选用硬质酸镁、木糖醇、可压性淀粉等常见辅料。
基于上述,以质量份数计,所述活性成分还包括组分:沙棘0.10~0.20份、松花粉0.20~0.30份、枸杞0.10~0.15份、菊花提取物0.20~0.25份、桑叶提取物0.12~0.15份、益智仁0.23~0.27份和酸枣仁0.22~0.28份。
基于上述,以质量份数计,所述活性成分还包括组分:薄荷提取物0.02~0.08份、橘皮提取物0.05~0.07份、茯苓0.02~0.04份、薏苡仁0.03~0.05份、砂仁0.02~0.05份、核桃仁0.01~0.03份、甘草0.50~0.08份和大枣0.02~0.06份。
所述决明子提取物、所述菊花提取物、所述桑叶提取物、所述薄荷提取物的制备可采用水提法。其中,所述决明子提取物/所述菊花提取物的制备可采用以下步骤:
将决明子/桑叶/菊花和水按质量比1:(3~7)混合煎煮提取2~3h,过滤,收集滤液和滤渣,将滤渣重复提取2~3次,合并滤液,进行浓缩、干燥、粉碎处理,得到所述决明子提取物/桑叶/所述菊花提取物。
所述薄荷提取物的制备可采用以下步骤:
称取新鲜薄荷叶片洗净,先研磨为泥浆糊状,再按照质量比为1:(2~4)加入水,常温下提取0.5~1h,固液分离,过滤,收集滤液和滤渣,将滤渣重复提取2~3次,合并滤液,进行浓缩、冷冻干燥、粉碎处理,得到所述薄荷提取物。
所述橘皮提取物的制备采用溶剂提取法,具体的,所述橘皮提取物的制备步骤包括:
将阴干的橘皮粉碎后和无水乙醇按质量比1:(2~5)中混合,常温下超声提取30~50min,过滤,滤液经浓缩、干燥、粉碎处理后制得所述橘皮提取物。
基于上述,所述蓝莓提取物的制备步骤包括:
将蓝莓搅碎加入生物酶进行酶解,得到酶解产物;对所述酶解产物进行离心分离,得到上清液;对所述上清液进行浓缩,得到所述蓝莓提取物;其中,所述复合生物酶包括酸性纤维素酶、酸性蔗糖酶和单宁酶。
一种所述眼部保健品的制备方法,其包括以下步骤:
先将阿胶加热熔化,然后将阿胶、明胶和水按质量比1:(1~1.5):(5~10)混合均匀,制得阿胶溶液;
在8000~10000r/min搅拌条件下,在所述阿胶溶液中依次缓慢加入乳化剂、叶黄素酯、β-胡萝卜素和二十二碳六烯酸,15~30min加完,继续搅拌15~30min,得到初乳液;其中,所述乳化剂可选用吐温-80、硬脂酸或蓖麻油;
在压力为50~70MPa对所述初乳液进行高压均质,然后在2000~3000r/min转速下继续搅拌2~3h,得到均质液;转速下调至500~600r/min,在所述均质液中加入凝聚剂,继续搅拌形成微囊,形成的微囊倒入凝聚剂稀释液,静置分层,去除上清液之后,在沉积层中加入固化剂,静置1~2h,过滤,冷冻干燥,得到复合微囊;
其中,所述凝聚剂与阿胶的质量比为(10~20):1,所述凝聚剂可采用质量分数为22%~25%的硫酸钠溶液,所述凝聚剂稀释液与阿胶的质量比为(20~30):1,所述凝聚剂稀释液可采用质量分数为10%~15%的硫酸钠溶液;所述固化剂与阿胶的质量比为(5~7):1,所述固化剂可采用甲醛或戊二醛;
将所述蓝莓提取物和环糊精按质量比1:(5~7)在10~25℃混合,得到环糊精饱和液,对所述环糊精饱和液进行冷冻干燥,得到环糊精包合粉;
将所述复合微囊、所述环糊精包合粉、所述决明子提取物和润湿剂混合,依次制粒、干燥、压片处理,得到所述眼部保健品。
基于上述,以质量份数计,所述活性成分还包括组分:沙棘0.10~0.20份、松花粉0.20~0.30份、枸杞0.10~0.15份、菊花提取物0.20~0.25份、桑叶提取物0.12~0.15份、益智仁0.23~0.27份和酸枣仁0.22~0.28份;
得到所述眼部保健品的步骤包括:将沙棘、松花粉、枸杞、益智仁、酸枣仁、进行超微粉碎至粒径为800~900目,得到原料微粉;将所述复合微囊、所述环糊精包合粉、所述决明子提取物、所述桑叶提取物、所述菊花提取物所述原料微粉和润湿剂混合,依次制粒、干燥、压片处理,得到所述眼部保健品。其中,所述润湿剂可以采用水、乙醇或其他润湿用辅料。
基于上述,所述活性成分还包括组分:薄荷提取物0.02~0.08份、橘皮提取物0.05~0.07份、茯苓0.02~0.04份、薏苡仁0.03~0.05份、砂仁0.02~0.05份、核桃仁0.01~0.03份、甘草0.05~0.08份和大枣0.02~0.06份;
得到所述眼部保健品的步骤包括:将沙棘、松花粉、枸杞、益智仁、酸枣仁、茯苓、薏苡仁、砂仁、核桃仁、甘草和大枣进行超微粉碎至粒径为800~900目,得到原料微粉;将所述复合微囊、所述环糊精包合粉、所述决明子提取物、所述桑叶提取物、所述菊花提取物、薄荷提取物、橘皮提取物、所述原料微粉和润湿剂、蜂蜜、木糖醇混合,依次制粒、干燥、压片处理,得到所述眼部保健品。
与现有技术相比,本发明具体突出的实质性特点与显著进步。具体的,本发明提供的眼部保健品,以二十二碳六烯酸、叶黄素酯、β-胡萝卜素、蓝莓提取物和决明子提取物与阿胶为主要活性成分,活性成分之间相互结合有效提高生物利用度,促进该眼部保健品保健效果的发挥;具体的,阿胶中含有棕榈酸甲酯、硬脂酸甲酯、油酸甲酯、亚油酸甲酯与亚麻酸甲酯等多种脂肪酸,在自身起到补血滋阴效果的同时,还可以有效促进人体对于二十二碳六烯酸、叶黄素酯、β-胡萝卜素、蓝莓提取物等脂溶性营养物质的吸收,而决明子具有清热明目的功效,可以降低肝肾负担,调节人体的消化系统,其与阿胶相互配合共同促进人体对于活性成分的吸收利用;另外,叶黄素酯和β-胡萝卜素相互配合还可以起到增益效果。
进一步,本发明还提供该眼部保健品的制备方法,采用阿胶和明胶作为微囊的壁材,结合脂溶性的二十二碳六烯酸、叶黄素酯、β-胡萝卜素制备肠溶微囊,有效促进吸收;同时,在制备过程中,先乳化制备初乳液,然后进行高压均质,使乳化体系的稳定性进一步增加,避免破乳,降低粒径分布范围,有利于在下一步骤中形成均一的微囊,可以有效避免胃酸影响,提高生物利用度。另外,该方法简单、易操作,便于工业应用,具有广阔的应用前景。

下面通过具体实施方式,对本发明的技术方案做进一步的详细描述。
实施例1
本实施提供一种眼部保健品,包括活性成分和医学上可接受的辅料,以质量份数计,所述活性成分包括组分:二十二碳六烯酸5份、叶黄素酯3份、β-胡萝卜素2份、蓝莓提取物2份和决明子提取物0.6份与阿胶8份;其中,二十二碳六烯酸、叶黄素酯和β-胡萝卜素源于微囊粉。
进一步,为了提高活性成分的稳定性,二十二碳六烯酸、叶黄素酯和β-胡萝卜素可以选用市售微囊粉;例如:嘉兴市维它食品添加剂有限公司销售的藻油二十二碳六烯酸(DHA)微囊粉、西安通泽生物科技有限公司销售的叶黄素酯微囊粉、济南安姆生物技术有限公司销售的β-胡萝卜素微囊粉。
进一步,所述蓝莓提取物的主要成分为花青素,其制备步骤包括:
将蓝莓搅碎加入生物酶进行酶解,得到酶解产物;对所述酶解产物进行离心分离,得到上清液;对所述上清液进行浓缩干燥,得到所述蓝莓提取物;其中,所述复合生物酶包括酸性纤维素酶、酸性蔗糖酶和单宁酶且各个酶活均大于100IU,以便充分降解蓝莓的细胞壁,使花青素充分溶液之中。
进一步,所述决明子提取物的制备步骤包括:
将决明子和水按质量比1:5混合煎煮提取2.5h,过滤,收集滤液和滤渣,将滤渣重复提取2次,合并滤液,进行浓缩、干燥、粉碎处理,得到所述决明子提取物。在其他的实施例中,决明子和水的比例、提取时间以及滤渣的重复提取次数可根据需要进行调整。
进一步,所述辅料可根据需要选用水、乙醇、硬质酸镁、木糖醇、蜂蜜、可压性淀粉等常见辅料。
本实施例中该眼部保健品为压片,其制备步骤包括:将二十二碳六烯酸5份、叶黄素酯3份、β-胡萝卜素2份、蓝莓提取物2份和决明子提取物0.6份、阿胶8份混合均匀,加入所述辅料,然后进行制粒、干燥、压片处理,得到所述眼部保健品的压片。其中,所述辅料的用量不高于制得所述保健品总质量的50%;该压片的片重为0.3g。
在其他的实施例中,该眼部保健品还可采用现有技术中的常规方法制备为压片、胶囊、糖果等形式的保健品。
在其他的实施例中,以质量份数计,所述活性成分包括组分:二十二碳六烯酸3份、叶黄素酯0.15份、β-胡萝卜素3份、蓝莓提取物40份和决明子提取物0.8份与阿胶10份;或是二十二碳六烯酸7份、叶黄素酯40份、β-胡萝卜素2.5份、蓝莓提取物0.15份和决明子提取物0.5份与阿胶6份;或是二十二碳六烯酸3~7份、叶黄素酯0.15~40份、β-胡萝卜素2~3份、蓝莓提取物0.15~40份和决明子提取物0.5~0.8份与阿胶6~10份之间的其他配比。
实施例2
本实施例与实施例1基本相同,其区别在于,为了提高该眼部保健品的保健效果,以质量份数计,所述活性成分还包括组分:沙棘0.15份、松花粉0.25份、枸杞0.12份、菊花提取物0.23份、桑叶提取物0.14份、益智仁0.25份和酸枣仁0.25份;所述菊花提取物/所述桑叶提取物的制备步骤包括:将菊花/桑叶和水按质量比1:5混合煎煮提取2.5h,过滤,收集滤液和滤渣,将滤渣重复提取3次,合并滤液,进行浓缩、干燥、粉碎处理,得到所述菊花提取物/所述桑叶提取物。在其他的实施例中,菊花/桑叶和水的比例、提取时间以及滤渣的重复提取次数可根据需要进行调整。

在其他的实施例中,所述活性成分还包括组分:薄荷提取物0.02份、橘皮提取物0.05份、茯苓0.02份、薏苡仁0.03份、砂仁0.02份、核桃仁0.01份、甘草0.50份和大枣0.02份;或是薄荷提取物0.08份、橘皮提取物0.07份、茯苓0.04份、薏苡仁0.05份、砂仁0.05份、核桃仁0.03份、甘草0.08份和大枣0.06份;或是薄荷提取物0.02~0.08份、橘皮提取物0.05~0.07份、茯苓0.02~0.04份、薏苡仁0.03~0.05份、砂仁0.02~0.05份、核桃仁0.01~0.03份、甘草0.50~0.08份和大枣0.02~0.06份之间的其他配比。
实施例4
本实施例与实施例1基本相同,区别在于:本实施例眼部保健品与实施1的制备方法不同。
本实施例提供一种眼部保健品的制备方法,其包括以下步骤:
先将阿胶加热熔化,然后将阿胶、明胶和水按质量比1:1.3:7混合均匀,制得阿胶溶液;
在8000~10000r/min搅拌条件下,在所述阿胶溶液中依次缓慢加入乳化剂、叶黄素酯、β-胡萝卜素和二十二碳六烯酸,15~30min加完,继续搅拌15~30min,得到初乳液;其中,所述乳化剂可选用磷脂、吐温-80、硬脂酸或蓖麻油;本实施例选用吐温-80;
在压力为50~70MPa对所述初乳液进行高压均质,然后在2000~3000r/min转速下继续搅拌2~3h,得到均质液;转速下调至500~600r/min,在所述均质液中加入凝聚剂,继续搅拌形成微囊,形成的微囊倒入凝聚剂稀释液,静置分层,去除上清液之后,在沉积层中加入固化剂,静置1~2h,过滤,冷冻干燥,得到所述微囊粉;
其中,所述凝聚剂与阿胶的质量比为15:1,所述凝聚剂可采用质量分数为23%的硫酸钠溶液,所述凝聚剂与阿胶的质量比为25:1,所述凝聚剂稀释液可采用质量分数为12%的硫酸钠溶液;所述固化剂与阿胶的质量比为6:1,所述固化剂可采用戊二醛;
将所述蓝莓提取物和环糊精按质量比1:6在25℃混合,得到环糊精饱和液,对所述环糊精饱和液进行冷冻干燥,得到环糊精包合粉;
将所述微囊粉、所述环糊精包合粉、所述决明子提取物和所述辅料混合,依次制粒、干燥、压片处理,得到所述眼部保健品。
在其他的实施例中,阿胶、明胶和水按质量比可以为1:1:5、1:1.5:10或是1:(1~1.5):(5~10)范围内的其他值;所述凝聚剂与阿胶的质量比为10:1、20:1或是(10~20):1范围内的其他值;所述凝聚剂可采用质量分数为22%、25%或是22%~25%范围内其他质量分数的硫酸钠溶液,所述凝聚剂稀释液与阿胶的质量比可以为20:1、30:1或是(20~30):1范围内的其他值,所述凝聚剂稀释液可采用质量分数为10%、15%或是10%~15%范围内其他质量分数的硫酸钠溶液;所述固化剂与阿胶的质量比可以为5:1、7:1或是(5~7):1范围中的其他值,所述固化剂可采用甲醛。
所述复合微囊的耐酸实验:将所述复合微囊置于37℃pH=1.2的盐酸溶液中2h后,发现微囊完整未破裂。
所述复合微囊的肠溶性实验:将所述复合微囊分别置于人工肠液和人工胃液中1h后,在人工肠液中,微囊破裂,在人工胃液中,微囊完整无明显变化。
性能验证
对嘉兴市维它食品添加剂有限公司销售的藻油二十二碳六烯酸(DHA)微囊粉、西安通泽生物科技有限公司销售的叶黄素酯微囊粉、济南安姆生物技术有限公司销售的β-胡萝卜素微囊粉、实施例1和实施例4提供的眼部保健品进行灌胃实验,检测血液中二十二碳六烯酸、叶黄素和β-胡萝卜素的浓度,计算生物利用度。
具体方案:选用雄性SD大鼠50只,实测体重为350±2.5g,随机分为5组(每组10只),每只灌胃1mL灌胃液。试验前预先禁食12h后,分别灌胃给予上述样品。然后密集测定血液中二十二碳六烯酸、叶黄素和β-胡萝卜素的含量,至达峰时间后半小时,检测血液药峰浓度(Cmax)。以此作为参比数据计算二十二碳六烯酸、叶黄素酯和β-胡萝卜素的生物利用度,结果见表1。由表1可知,本发明提供的眼部保健品的组分配比及制备方法均可以在一定程度上提高二十二碳六烯酸、叶黄素酯和β-胡萝卜素的生物利用度。

最后应当说明的是:以上实施例仅用以说明本发明的技术方案而非对其限制;尽管参照较佳实施例对本发明进行了详细的说明,所属领域的普通技术人员应当理解:依然可以对本发明的具体实施方式进行修改或者对部分技术特征进行等同替换;而不脱离本发明技术方案的精神,其均应涵盖在本发明请求保护的技术方案范围当中。
Claims (7)
1.一种眼部保健品,包括活性成分和医学上可接受的辅料,其特征在于,以质量份数计,所述活性成分包括组分:二十二碳六烯酸3~7份、叶黄素酯0.15~40份、β-胡萝卜素2~3份、蓝莓提取物0.15~40份和决明子提取物0.5~0.8份与阿胶6~10份。
2.根据权利要求1所述的眼部保健品,其特征在于,以质量份数计,所述活性成分还包括组分:沙棘0.10~0.20份、松花粉0.20~0.30份、枸杞0.10~0.15份、菊花提取物0.20~0.25份、桑叶提取物0.12~0.15份、益智仁0.23~0.27份和酸枣仁0.22~0.28份。
3.根据权利要求1或2所述的眼部保健品,其特征在于,以质量份数计,所述活性成分还包括组分:薄荷提取物0.02~0.08份、橘皮提取物0.05~0.07份、茯苓0.02~0.04份、薏苡仁0.03~0.05份、砂仁0.02~0.05份、核桃仁0.01~0.03份、甘草0.05~0.08份和大枣0.02~0.06份。
4.根据权利要求1所述的眼部保健品,其特征在于,所述蓝莓提取物的制备步骤包括:
将蓝莓搅碎加入生物酶进行酶解,得到酶解产物;对所述酶解产物进行离心分离,得到上清液;对所述上清液进行浓缩,得到所述蓝莓提取物;其中,所述复合生物酶包括酸性纤维素酶、酸性蔗糖酶和单宁酶。
5.一种权利要求1所述的眼部保健品的制备方法,其包括以下步骤:
先将阿胶加热熔化,然后将阿胶、明胶和水按质量比1:(1~1.5):(5~10)混合均匀,制得阿胶溶液;
在8000~10000 r/min搅拌条件下,在所述阿胶溶液中依次缓慢加入乳化剂、叶黄素酯、β-胡萝卜素和二十二碳六烯酸,15~30 min加完,继续搅拌15~30 min,得到初乳液;
在压力为50~70 MPa对所述初乳液进行高压均质,然后在2000~3000r/min转速下继续搅拌2~3 h,得到均质液;转速下调至500~600r/min,在所述均质液中加入凝聚剂,继续搅拌形成微囊,形成的微囊倒入凝聚剂稀释液,静置分层,去除上清液之后,在沉积层中加入固化剂,静置1~2 h,过滤,冷冻干燥,得到复合微囊;
将所述蓝莓提取物和环糊精按质量比1:(5~7)在10~25℃混合,得到环糊精饱和液,对所述环糊精饱和液进行冷冻干燥,得到环糊精包合粉;
将所述复合微囊、所述环糊精包合粉、所述决明子提取物和润湿剂混合,依次制粒、干燥、压片处理,得到所述眼部保健品。
6.根据权利要求5所述的眼部保健品的制备方法,其特征在于,
以质量份数计,所述活性成分还包括组分:沙棘0.10~0.20份、松花粉0.20~0.30份、枸杞0.10~0.15份、菊花提取物0.20~0.25份、桑叶提取物0.12~0.15份、益智仁0.23~0.27份和酸枣仁0.22~0.28份;
得到所述眼部保健品的步骤包括:将沙棘、松花粉、枸杞、益智仁、酸枣仁、进行超微粉碎至粒径为800~900目,得到原料微粉;将所述复合微囊、所述环糊精包合粉、所述决明子提取物、所述桑叶提取物、所述菊花提取物所述原料微粉和润湿剂混合,依次制粒、干燥、压片处理,得到所述眼部保健品。
7.根据权利要求6所述的眼部保健品的制备方法,其特征在于,所述活性成分还包括组分:薄荷提取物0.02~0.08份、橘皮提取物0.05~0.07份、茯苓0.02~0.04份、薏苡仁0.03~0.05份、砂仁0.02~0.05份、核桃仁0.01~0.03份、甘草0.50~0.08份和大枣0.02~0.06份;
得到所述眼部保健品的步骤包括:将沙棘、松花粉、枸杞、益智仁、酸枣仁、茯苓、薏苡仁、砂仁、核桃仁、甘草和大枣进行超微粉碎至粒径为800~900目,得到原料微粉;将所述复合微囊、所述环糊精包合粉、所述决明子提取物、所述桑叶提取物、所述菊花提取物、薄荷提取物、橘皮提取物、所述原料微粉和润湿剂、蜂蜜、木糖醇混合,依次制粒、干燥、压片处理,得到所述眼部保健品。
