Abstract

苦瓜素的提取工艺,本发明属于植物胰岛素的提取方法。采用已有的提取工艺提取出的苦瓜素纯度不高,故将其用于治疗糖尿病的效果就不佳。用本发明的提取工艺提取出的苦瓜素纯度可达7%,用于治疗糖尿病的有效率达82%,治愈率为40%,而且无毒副作用,可达到持久稳定的降低血糖的效果。其主要步骤是匀浆、搅拌、抽滤、调pH值、冷藏、沉淀、静置、析出无色针状结晶,用硅胶G进行薄层层析、洗脱、过滤、真空干燥、得无色结晶、再进行回流加热、过滤干燥,最后进行纸层析即得纯结晶苦瓜素。

Description

苦瓜素的提取工艺
技术领域:本发明属于植物胰岛素的提取方法。
背景技术:苦瓜为葫芦科苦瓜属植物,性味苦寒。它具有清热解凉,消渴引饮之功效。但直接食用苦瓜其需用量较大,而且达不到理想的治疗糖尿病的效果。近年来人们采取多种办法将苦瓜素从苦瓜中提取出来,并应用于临床治疗糖尿病。但都因提取的纯度不高,使其疗效不佳。
发明内容:本发明的目的是研制一种苦瓜素的提取工艺,该工艺提取的苦瓜素的纯度较高,所以用于临床治疗糖尿病其疗效也就很好。本发明的工艺流程为:取新鲜苦瓜100g,加入蒸馏水10ml、95%浓度的乙醇45ml、浓硫酸3.6ml,在25~28℃下匀浆10~15分钟。再加入60ml水、95%浓度乙醇250ml,搅拌10~15分钟后再抽滤。向滤液内加入浓度为2.8%的NH4OH,逐渐调至PH值达到3为止。再向溶液内加入冷无水乙醇1.5L、乙醚21L,将溶液放入0.5℃的冰箱中冷藏12小时。将上清液回收后得到白色絮状沉淀物,将沉淀物溶于无水乙醇∶NH4OH为1∶2的溶液中,再向溶液内加入微量锌,在室温下静置18小时。析出无色针状结晶,将粗结晶溶于乙醇∶NH4OH为1∶2的溶液中。用硅胶G进行薄层层析,在100℃下层析0.5小时。再加入展开剂,展开剂由正丁醇、水、醋酸组成,正丁醇∶水∶醋酸=(10~12)∶(4~5)∶(1~2),使迁移率达到0.19~0.24为止。再以乙醇∶NH4OH=1∶(2~3)的溶液进行洗脱,再过滤进行真空干燥,得纯无色结晶1g。将无色结晶1g与6NHCl进行回流加热20小时。再将水鲜液过滤干燥,然后再将干燥物溶于50%浓度的乙醇中,再进行纸层析即得纯结晶苦瓜素。纸层析时加入的展开剂为正丁醇∶水∶醋酸=(55~60)∶(16~20)∶(14~20)的溶液。本发明从苦瓜中提取的苦瓜素纯度可达到7%左右,用于临床治疗糖尿病100例有效率达82%,治愈率为40%。而现有其它方法提取的苦瓜素的纯度一般只能达到2%左右,用于临床治疗糖尿病的有效率为50%左右,治愈率为20%左右。本发明的工艺具有操作简单、成本低、一致性好的优点。本发明提取的苦瓜素可制成注射剂或口服剂,经动物的药理、毒理实验和临床试验,它无毒副作用,可减轻糖尿病的并发症,可有效地降低血糖,解决了植物人工胰岛素不能口服的难题,消除了应用西药等降糖药物久用易产生耐药性及毒副作用的危害,达到了持久稳定的降低血糖的效果。
具体实施方式:取新鲜苦瓜100g,加入蒸馏水10ml、95%浓度的乙醇45ml、浓硫酸3.6ml,在26℃下匀浆13分钟。再加入60ml水、95%浓度乙醇250ml,搅拌13分钟后再抽滤。再向溶液内加入冷无水乙醇1.5L、乙醚2L,将溶液放入0.5℃的冰箱中冷藏12小时。将上清液回收后得到白色絮状沉淀物,将沉淀物溶于无水乙醇∶NH4OH为1∶2的溶液240ml溶液中,再向溶液内加入5~10ml的锌,在室温下静置18小时。析出无色针状结晶,将粗结晶溶于乙醇∶NH4OH为1∶2的溶液240ml中。用硅胶G进行薄层层析,在100℃下层析0.5小时。再加入展开剂,展开剂由正丁醇、水、醋酸组成,取正丁醇∶水∶醋酸=12∶5∶2比例的溶液1520ml,使迁移率达到0.21为止。再以乙醇∶NH4OH为1∶2的溶液240ml进行洗脱,再过滤。将溶液进行真空干燥,得纯无色结晶1g。将无色结晶1g与6NHCl 80ml进行回流加热20小时。再将水鲜液过滤干燥,然后再将干燥物溶于50%浓渡的乙醇240ml中,再进行纸层析,即得纯结晶苦瓜素。滤纸采用WhatmanNO1型号,纸层析时加入的展开剂为正丁醇、水、醋酸混合液,正丁醇∶水∶醋酸=58∶18∶18,溶量为7920ml。纸层析结束时用显色剂进行检测,显色剂为0.25%浓度的卯三酮丙酮溶液。得到的苦瓜素结晶的熔点为232~234℃,而标准胰岛素的熔点为233℃。

Claims (2)

1、苦瓜素的提取工艺,取新鲜苦瓜100g,加入蒸馏水10ml、95%浓度的乙醇45ml、浓硫酸3.6ml,在25~28℃下匀浆10~15分钟;再加入60ml水、95%浓度乙醇250ml,搅拌10~15分钟后再抽滤;向滤液内加入浓度为2.8%的NH4OH,逐渐调至PH值达到3为止;再向溶液内加入冷无水乙醇1.5L、乙醚21L,将溶液放入0.5℃的冰箱中冷藏12小时;将上清液回收后得到白色絮状沉淀物,将沉淀物溶于无水乙醇∶NH4OH为1∶2的溶液中,再向溶液内加入微量锌,在室温下静置18小时;析出无色针状结晶,将粗结晶溶于乙醇∶NH4OH为1∶2的溶液中;其特征在于用硅胶G进行薄层层析,在100℃下层析0.5小时;再加入展开剂,展开剂由正丁醇、水、醋酸组成,正丁醇∶水∶醋酸∶(10~12)∶(4~5)∶(1~2),使迁移率达到0.19~0.24为止;再以乙醇∶NH4OH=1∶(2~3)的溶液进行洗脱,再过滤进行真空干燥,得纯无色结晶1g,将无色结晶1g与6NHCl进行回流加热20小时;再将水鲜液过滤干燥,然后再将干燥物溶于50%浓度的乙醇中,再进行纸层析即得纯结晶苦瓜素。
2、根据权利要求1所述的苦瓜素的提取工艺,其特征在于纸层析时加入的展开剂为正丁醇∶水∶醋酸=(55~60)∶(16~20)∶(14~20)的溶液。
