Background
In recent years, with the rapid development of global industrialization and urbanization, environmental pollution, particularly air pollution, is more and more serious, and skin is used as a physiological barrier of human body and is the first defense line for resisting various pollution sources. Not only can the skin be damaged by the source of respiratory inhalation, but many topically applied substances can be absorbed through the skin into cells or tissues in the body, disrupting the skin barrier function.
At present, air pollutants harmful to human health include suspended dust, particulates (PM10, PM2.5), Polycyclic Aromatic Hydrocarbons (PAHs), ozone, nitrogen oxides, sulfur oxides, carbon oxides, and the like, wherein PM2.5 is a non-negligible skin aggressor, has a large surface area, strong activity, a fine porous structure, and is more likely to be accompanied by toxic and harmful substances, such as: PAHs, heavy metals, microorganisms and the like, and PM2.5 cannot penetrate through the skin, is easy to adhere to the surface of the skin and continuously releases adsorbed toxic and harmful substances, and the harmful substances can enter the deep layer of the skin through pores to damage healthy cells of the skin, cause skin aging, and cause a series of skin problems such as allergy, dermatitis, color spots and the like. Besides air pollution, ultraviolet pollution can also cause skin damage, UVA has strong penetrating power and can directly reach the dermis layer of the skin, elastic fibers and collagen fibers are damaged, the skin is tanned, and the skin is most harmful; UVB is absorbed by ozone in the stratosphere, causing sunburn and redness, swelling, heat, pain in the skin, and in severe cases, blistering or peeling. The long-term exposure to UV radiation can cause skin aging, skin dryness, pachylosis, wrinkle formation, loss of elasticity, melanin precipitation, dermatitis, and skin cancer.
Therefore, the anti-pollution cosmetics firstly isolate the adhesion of pollutants such as PM2.5, dust, polycyclic aromatic hydrocarbon and the like to the skin, remove pollution particles on the skin, protect the skin from being damaged by the pollutants, consider skin invasion caused by light pollution, and cause the problems of skin barrier damage, skin sensitivity aggravation, skin inflammation initiation and the like when the skin is in a polluted environment for a long time. However, the current pollution-resistant cosmetics cannot effectively resist pollution sources from multiple aspects, or can not well resist the pollution, resist inflammation, relieve and repair skin barriers, and along with the gradual improvement of the awareness of people on the pollution prevention of the skin, the demand of protective cosmetics is continuously expanded, so that the development of a cosmetic additive capable of effectively preventing pollution is very important for the cosmetics.

The preparation method of the cosmetic additive comprises the following steps: weighing the components according to any one group of the weight in the embodiments 1-16 and the comparative examples 1-6, and stirring and mixing the components uniformly at normal temperature to obtain the nano-silver paste.
The application method of any one group of cosmetic additives in examples 1 to 16 and comparative examples 1 to 6 is to add any one group of cosmetic additives into cosmetics, wherein the cosmetic additives with the anti-pollution effect account for 1 to 10 percent of the total mass of the cosmetics.
The cosmetic additive with the anti-pollution effect is added into cosmetics, and can be prepared into skin care cosmetics such as emulsion, essence, cream, lotion, makeup removing cream, gel, facial mask and the like according to the conventional process and auxiliary materials for preparing the cosmetics. The selection of the other ingredients in the cosmetic product at the time of manufacture may be made according to conventional procedures in the art.
In order to better illustrate the advantages of the present invention, the experimental results of the technical solution provided by the present invention are given below:
1. dust adsorption test
1) Test samples: the test samples prepared by adding the cosmetic additives of examples 1 to 16 and comparative examples 1 to 6 to a common emulsion external substrate, wherein the cosmetic additive having an anti-pollution effect accounts for 5% of the total mass of the emulsion, and the emulsions of comparative examples 7 and 8.
2) The test method comprises the following steps: 115 road commuters were selected and randomly divided into 23 groups (5 per group), hands were cleaned with a hand lotion and wiped dry with paper towels. Each group applied 0.2g of the emulsion base of comparative example 8 to an area of approximately 2 square centimeters on the left forearm and 0.2g of each of the other groups of test samples to an area of approximately 2 square centimeters on the right forearm. Let them work on the road for 4 hours, then stick the dust off the forearms with 3M tape, weigh and average. The test results are shown in Table 1.

Comparative example 6 0.0069 0.0036 47.34%
Comparative example 7 0.0055 0.0034 38.90%
From the experimental results in table 1, it can be seen that the dust amount of the right forearm is significantly reduced after the right forearm is applied with the emulsion containing the cosmetic additive of the embodiment of the present invention, wherein the dust amount is reduced by more than 70% in the embodiments 13 to 16, while the dust amount is reduced by only 38.90% after the emulsion of the comparative example 7 is applied with a certain commercially available anti-pollution emulsion, which indicates that the cosmetic additive of the present invention can significantly reduce the adsorption of pollutants such as dust on the skin. After the emulsions of comparative examples 1 to 6 were used, the amount of dust reduction was relatively lower than that of the emulsions prepared by using the cosmetic additives of the examples, which shows that the effect of adding several extracts of the cosmetic additives of the present invention to the emulsions alone was significantly inferior to the effect of adding all nine ingredients of the cosmetic additives of the present invention to the emulsions, and the effect of reducing the adhesion of dust pollutants to the skin was gradually enhanced with the increase in the types of the extracts of the present invention, and thus, it can be known that the extracts provided by the present invention have a synergistic effect and can better resist pollutant particles such as PM2.5, dust, and the like.
2. Skin barrier function test
1) The test principle is as follows: normal skin has a strong barrier function and can prevent external harmful substances from entering the skin. When barrier function is compromised, pathways are formed between keratinocytes from which water is lost. Thus, the transdermal loss of water increases when the skin barrier function is diminished and vice versa decreases. The better the skin protection layer, the higher the moisture content and the lower the value of the skin moisture loss TEWL. The test judges the strength of the skin barrier function by the percutaneous water loss (TEWL), thereby evaluating the anti-pollution efficacy of the cosmetic additive of the present invention.
2) Test samples: the cosmetic additives of examples 2, 4, 6, 8, 10, 12, 14, 16, comparative examples 1, 3, 5 were added to a common emulsion external base to prepare test samples in which the cosmetic additive having anti-contamination efficacy accounted for 5% of the total mass of the emulsion, and the emulsion of comparative example 7.
3) The test method comprises the following steps: 130 female subjects with dry and water-deficient facial skin are selected, the female subjects are 20-45 years old and randomly divided into 13 groups, 10 persons in each group are detected for three days before the detection, the face does not use any cosmetics, 0.2g of emulsion is respectively taken by each group of subjects to be coated on the facial skin, the emulsion is respectively used once every morning and evening, the samples are continuously used for 28 days, the samples are stopped at 29 days and 30 days, the TEWL values of the face are respectively measured by a Tewameter after the samples are used for 1 day and 31 days, and the TEWL values are respectively repeated for 5 times to obtain the average value. The test results are shown in Table 2.
Table 2 skin barrier function test
Figure BDA0002000112470000091
Figure BDA0002000112470000101
As can be seen from the experimental results in Table 2, the change rate of the TEWL value of the skin water loss after the emulsion added with the cosmetic additive of the embodiment of the invention is more than 60%, which is higher than that of a certain commercially available anti-pollution emulsion of the comparative example 7, and the effect of the cosmetic additive of the invention on enhancing the skin barrier function is demonstrated. The change rate of TEWL values after using the emulsions of comparative examples 1, 3 and 5 was relatively lower than that of the emulsions prepared using the cosmetic additives of the examples, indicating that the effects of enhancing the skin barrier function and reducing the loss of skin moisture are gradually enhanced as the kinds of the extracts of the present invention are increased, and thus it is understood that the extracts provided by the present invention have a synergistic effect.
3. Anti-inflammatory efficacy test
1) The test principle is as follows: the mouse auricle swelling experiment is one of non-specific inflammation models, mainly uses chemical substances as inflammatory agents to induce the mouse ear swelling, and is usually used for evaluating the anti-inflammatory activity of some synthetic compounds and plant active ingredients.
2) Test samples: the cosmetic additives of examples 1, 3, 5, 7, 9, 11, 13, 15 and comparative examples 2, 4, 6 were added to a common emulsion external base to prepare test samples in which the cosmetic additive having an anti-contamination effect was 5% by mass of the total mass of the emulsion, and the emulsion of comparative example 7.
3) The test method comprises the following steps: taking 130 male mice (female mice can generate more estrogen which has certain anti-inflammatory effect and is not suitable for anti-inflammatory experiments), randomly dividing into 13 groups, each group comprises 10 female mice, one group is blank control (applied with normal saline with equal dosage), the other groups are applied with test samples, each test sample is applied in advance for 30d, 1g is applied each time, 30 microliter of dimethylbenzene is applied to the auricle of the right ear after 30min of last administration to cause an acute inflammation model, and the left ear is not applied as a normal ear control. After 1h of xylene coating, blood was taken and sacrificed, ear pieces were punched out at the same positions of the left and right ears by a punch having a diameter of 6mm, the masses were weighed on an analytical balance, and the swelling degree and the swelling inhibition ratio were calculated by taking the average value. The results are shown in Table 3.

As can be seen from the experimental results of Table 3, the swelling inhibition ratio of the cosmetic additive of the present invention was higher than that of one of the commercially available anti-pollution emulsions of comparative example 7, indicating that the cosmetic additive of the present invention can significantly reduce the inflammatory swelling of mouse auricle caused by xylene. The swelling inhibition ratio using the cosmetic additives of comparative examples 2, 4, and 6 was relatively lower than that of the examples, confirming that the cosmetic additives of the present invention are reasonably compounded to have superior anti-inflammatory effects.
4. Ultraviolet resistance test
1) The test principle is as follows: the sun protection factor SPF is an important parameter for measuring the ultraviolet resistance of cosmetics, and the higher the SPF value is, the stronger the ultraviolet resistance is.
2) Test samples: samples were selected for use in the skin barrier function test.
3) The test method comprises the following steps: the SPF value of each sample is measured by a sun protection index analyzer of Labsphere company, 1g of the sample is uniformly coated on a skin-like transparent PMMA plate of 5cm multiplied by 5cm, then the plate is placed in a dark place for standing for 15 minutes, the plate is placed in a UV-2000S type ultraviolet transmittance analyzer to measure the sun protection performance, 10 different parts are taken for measurement, and the average value is taken. The test results are shown in Table 4.

Figure BDA0002000112470000121
From the experimental results of Table 4, it can be seen that the SPF value of the cosmetic additive of the present invention is higher than that of a commercially available anti-pollution emulsion of comparative example 7, indicating that the cosmetic additive of the present invention has UV resistance. The SPF value of the emulsion of the comparative example is relatively lower than that of the emulsion of the example, which shows that the compounding and the cooperation of the cosmetic additive of the invention have better ultraviolet resistance and slow down the problems of skin aging, melanin precipitation, color spots and the like caused by ultraviolet rays.
5. Benzopyrene damage test on cellular DNA
1) The test principle is as follows: the comet assay can effectively detect and quantitatively analyze the damage degree of single-strand and double-strand gaps of DNA in cells, the more the DNA is damaged, the more chain breaks and fragments are generated, the longer the length is, the more the amount of DNA is migrated under the same electrophoresis condition, the longer the migration distance is, and the damage degree of the DNA of a single cell can be determined by determining the optical density or migration length of a DNA migration part.
2) Test samples: samples were selected for use in the skin barrier function test.
3) The test method comprises the following steps: the experiment takes human keratinocytes as objects, the human keratinocytes are cultured in vitro, each component is added with 2g of test samples respectively, then cultured for 2h, then 10% benzopyrene solution is added, and a comet assay is carried out after 24h to obtain the percentage of DNA damage, so that the effect of the cosmetic additive on reducing the damage of benzopyrene to cells is evaluated. The test results are shown in Table 5.
TABLE 5 results of the benzopyrene damage test on cellular DNA
Figure BDA0002000112470000122
Figure BDA0002000112470000131
As can be seen from the experimental results of Table 5, the percentage of DNA damage of human keratinocytes to which the cosmetic additive of the present invention was added was less than that of comparative example 7, indicating that the cosmetic additive of the present invention was effective against the invasion of skin cells by benzopyrene. The DNA damage percentage of the human keratinocytes added with the cosmetic additives of comparative examples 1, 3 and 5 is higher than that of the examples, which shows that the harm of pollutants such as polycyclic aromatic hydrocarbon and the like to the skin can be better reduced by compounding the cosmetic additives of the invention, and the effect of protecting the skin cells is achieved.
In conclusion, the skin care product can effectively reduce the attachment of pollutants to the skin and resist the invasion of external pollutants through the synergistic effect of the natural active substances of the water shield leaf extract, the quercus robur root extract, the moringa oleifera seed extract, the carex glauca linn extract, the aureobasidium pullulans fermentation product, the alpine edelweiss extract, the mirabilis jalapa extract, the scutellaria alpina flower/leaf/stem extract and the ginkgo leaf extract, and has the effects of relieving, resisting inflammation, repairing and the like.
The above embodiments are preferred embodiments of the present invention, but the present invention is not limited to the above embodiments, and any other changes, modifications, substitutions, combinations, and simplifications which do not depart from the spirit and principle of the present invention should be construed as equivalents thereof, and all such changes, modifications, substitutions, combinations, and simplifications are intended to be included in the scope of the present invention.
