Abstract

In a second aspect, the present invention provides a process for the preparation of an emulsion comprising an anti-premature composition according to the first aspect, said emulsion comprising the steps of:
(1) mixing the humectant and water to obtain a water-phase mixed solution; mixing the emulsifier with an oil phase to obtain an oil phase mixed solution;
(2) mixing the water phase mixed liquor and the oil phase mixed liquor obtained in the step (1) and homogenizing to obtain homogenized liquor;
(3) and (3) mixing and stirring the homogenized liquid obtained in the step (2), the anti-aging composition and the skin conditioner to obtain the emulsion containing the anti-aging composition.
Preferably, the temperature of the aqueous phase mixing in step (1) is 80-85 ℃, for example 80 ℃, 81 ℃, 82 ℃, 83 ℃, 84 ℃, 85 ℃ and the like, and the time of the aqueous phase mixing is 5-10min, for example 5min, 6min, 7min, 8min, 9min, 10min and the like.
Preferably, the temperature of the oil phase mixing in step (1) is 75-80 deg.C, such as 75 deg.C, 76 deg.C, 77 deg.C, 78 deg.C, 79 deg.C, 80 deg.C, etc., and the time of the oil phase mixing is 5-10min, such as 5min, 6min, 7min, 8min, 9min, 10min, etc.

Detailed Description
The technical solution of the present invention is further explained by the following embodiments. It should be understood by those skilled in the art that the examples are only for the understanding of the present invention and should not be construed as the specific limitations of the present invention.
Each of the components in the following preparation examples is commercially available.
Preparation example 1
The preparation example provides a curcuma alismatifolia root extract, which is prepared by the following preparation method:
(1) freezing root of Zingiber officinale Roscoe at-15 deg.C for 18h, pulverizing at-2 deg.C, and sieving to obtain 150 mesh Zingiber officinale Roscoe root powder;
(2) mixing the ginger flower root powder obtained in the step (1) with n-butanol, leaching for 2 hours at 25 ℃, and concentrating the solution obtained by leaching to obtain a concentrated solution; mixing the concentrated solution with ethanol, leaching at 40 deg.C for 1 hr, and filtering to obtain crude extract of rhizoma Zingiberis recens;
wherein the mass ratio of the ginger flower root powder to the n-butanol to the ethanol is 1:10: 8;
(3) and (3) passing the crude extract of the ginger flower roots obtained in the step (2) through macroporous adsorption resin AB-8, washing with deionized water, removing water solution, washing with 60 wt% ethanol water solution, collecting the washed ethanol water solution part, and concentrating to obtain the ginger flower root extract.
Preparation example 2
The preparation example provides a radix scutellariae extract, which is prepared by the following preparation method:
(a) pulverizing radix Scutellariae, and sieving to obtain 200 mesh radix Scutellariae powder;
(b) mixing the radix scutellariae powder obtained in the step (a) with an ethanol water solution with the concentration of 60 wt% according to the mass ratio of 1:10, performing microwave extraction for 8min at the power of 600W, and concentrating to obtain a radix scutellariae crude extract;
(c) dissolving the crude extract of radix scutellariae obtained in the step (b) by using 10 wt% of ethanol water solution, purifying by using a macroporous resin gel column D101, eluting by using 60 wt% of ethanol water solution, and concentrating to obtain the radix scutellariae extract.
Preparation example 3
The present preparation example provides a cocoa seed extract prepared by the following preparation method:
(A) pulverizing cacao seed to particle size of 100 mesh, mixing with n-hexane at a mass ratio of 1:10, soaking at 30 deg.C for 3 hr, filtering, and drying to obtain defatted cacao seed powder;
(B) mixing the defatted cocoa seed powder obtained in the step (A) with water, heating to 45 ℃, mixing with biological enzyme and acid, wherein the mass ratio of the defatted cocoa seed powder to the water to the cellulase is 1:15:0.006, carrying out enzymolysis reaction for 1.5h at 45 ℃ and pH of 5, and then inactivating to obtain a cocoa seed enzymolysis product;
(C) mixing the cocoa seed enzymolysis product obtained in the step (B) with 60 wt% ethanol water solution according to the mass ratio of 1:10, performing ultrasonic extraction for 50min at the power of 300W, filtering, and concentrating to obtain the crude extract of cocoa seeds.
Except that the ginger flower root extract, the scutellaria root extract and the cacao seed extract were prepared by the above-mentioned preparation methods, the remaining components in the following examples and comparative examples were all commercially available.

Test example 1
Safety performance testing
The safety performance tests were performed on the emulsions provided in application examples 1-9 and on the emulsions provided in comparative application examples 1-6 by the following method:
(1) haemolysis test of erythrocytes
Preparation of erythrocyte suspension: selecting healthy rabbits, taking 9mL of blood from heart, adding 1mL of 2% potassium oxalate solution, centrifuging, discarding supernatant, diluting the precipitate to 20mL with 20mmol/L PBS solution, and storing at 4 ℃ for later use. The samples to be tested with a final volume fraction of 1% were added separately, and the untreated group was used as a cell control group, with 2 duplicate wells per group. After 48h of culture, the cells were washed 1 time with PBS, 100. mu.L of lysate was added to each well, the cells were scraped off and collected, and the supernatant was centrifuged. 50 μ L of cell supernatant was applied to a 96-well plate, 50 μ L of 1% L-dopa solution was added, incubation was performed at 37 ℃ for 1h, and absorbance was read at 475nm using M3 plate reader.
Relative tyrosinase activity (%) - (assay well absorbance-blank absorbance)/(cell control absorbance-blank absorbance) × 100%.
(2) Experiment for inhibiting melanin synthesis
Mouse melanoma B16 cells in logarithmic growth phase were inoculated into T25 cell culture flasks and cultured overnight. The samples to be tested were added to the test cells in a final volume fraction of 1%, and the untreated group was used as a cell control group. After 48h of culture, washing with PBS 1 time, adding 1mL of 1mol/L NaOH solution, scraping and collecting cells, placing in a water bath at 80 ℃ for 30min, adding the supernatant into a 96-well plate, and reading the absorbance value at 475nm by using an M3 plate reader.
Relative melanin content (%) — x 100% (assay well absorbance value-blank control absorbance value)/(cell control absorbance value-blank control absorbance value), wherein the tyrosinase activity of the control was 100% and the melanin content of the control was 100%.
(3) Human body evaluation of whitening effect
150 volunteers 25-50 years old are selected, male and female are not limited, the volunteers are randomly divided into 15 groups, 10 persons in each group are respectively smeared on the arm of each sample to be tested, the sample to be tested is respectively used for 1 time in the morning and at night every day, the smearing amount of each time is 2mL, and the total using time is 4 weeks. The skin melanin content change before and after use of the subject was evaluated using a skin red melanin tester (HexameterMX 18). The measurement range of the used instrument is 0-999, and the higher the measurement value, the higher the melanin content in the skin is.
The specific test results are shown in table 3:

The test results in table 3 show that the relative tyrosinase activity of the emulsion provided by the invention is 17-18%, and the inhibition effect on tyrosinase is good; meanwhile, the synthesis of melanin in cells can be obviously inhibited, and the relative melanin content is 8-12%; and the human body evaluation result shows that the emulsion provided by the invention can obviously reduce the melanin content of the subject after being used for 4 weeks, and the reduction of the melanin of the subject reaches more than 60%. The anti-aging composition can be matched with each component in the skin conditioner of the emulsion to realize synergistic interaction, so that the anti-aging composition can effectively inhibit the synthesis and transfer of melanin, promote the decomposition and elimination of the melanin, inhibit the activity of tyrosinase, promote the metabolism of pigment, help the catabolic impurities, prevent pigmentation, and make the skin smooth, delicate, white and glossy after the composition is used.
Test example 3
Histamine inhibition test
Histamine inhibition tests were performed on the emulsions provided in application examples 1-9 and on the emulsions provided in comparative application examples 1-6 by the following method:
histamine is a chemical transmission medium in organisms, is present in cytoplasmic granules of basophils and mast cells of human bodies, and has positive correlation between skin allergy, inflammation and pruritus and histamine release in mast cells. Research proves that the anti-allergy and relieving effects can be achieved by inhibiting histamine release; experiments mast cell degranulation agent (Compound48/80) was used to induce histamine release from guinea pig isolated mast cells, which was then inhibited by the relaxation efficacy test using the emulsions provided in application examples 1-9 and the emulsions provided in comparative application examples 1-6. The content of histamine release from guinea pig mast cells was determined by HPLC, and the rate of inhibition of histamine release was measured for each sample.
The specific test results are shown in table 4:

From the test results in table 4, it can be seen that the emulsion provided by the present invention has an inhibition rate of over 77% for histamine release. The anti-aging composition can be matched with each component in the skin conditioner of the emulsion, has synergistic effect, and can achieve the anti-allergy and relieving effects by inhibiting the release of histamine.
Test example 4
Functional assessment of soothing sensitive stimulus effects
(1) Test samples: the emulsions provided in application examples 1-9 and the emulsions provided in comparative application examples 1-6;
(2) the test method comprises the following steps: selecting 150 sensitive skin crowds (with the accumulated stabbing pain feeling more than or equal to 3 points) with the age of 25-50 years, randomly dividing the crowds into 15 groups of 10 people each; selecting a subject to perform a nasal sulcus test, wherein the tested part of the subject cannot use any cosmetics, external medicines or internal health care products 15 days before the test, and before the test, the subject is required to clean the face and sit still for 30min in a constant-temperature and constant-humidity room with the temperature of 22 +/-1 ℃ and the humidity of 50 +/-5% within 2 hours after the cleaning and is kept in a relaxed state;
dropping 50 μ L of 10% lactic acid solution on a single-layer filter paper with a diameter of 0.8c m, inquiring the testee about stabbing pain at 3min, 4min and 5min respectively, scoring according to a 4-point method (0 is no stabbing pain, 1 is mild stabbing pain, 2 is moderate stabbing pain and 3 is severe stabbing pain), recording the score of the testee as the initial value of the side lactic acid stimulation score (D0), carrying out the experiment for 7 days totally, using the product by the testee for 1 time each morning and evening according to the requirement, avoiding cross contamination, and carrying out the revisit by the testee on the 7 th day (D7); during testing, the lighting condition is kept consistent, the bed inclination angle and the bed placing position are one, the same testee is tested by the same measurer, and the change rate of the lactic acid stimulation result is calculated;
the specific test results are shown in table 5:

The test data in table 5 show that the change rate of the lactic acid stimulation result reaches below-28% after 7 days of the emulsion provided by the invention, which indicates that the anti-aging composition provided by the invention can be matched with each component in the skin conditioner of the emulsion, has synergistic effect, can better repair the skin to reach the optimal physiological state by adjusting the physiological function of the skin aiming at the sensitive skin, has very obvious repairing and relieving effects on the sensitive skin, and is more consistent with the physiological requirements of the skin.
The applicant states that the invention is illustrated by the above examples of emulsions containing an anti-premature composition according to the invention and the process for their preparation, but the invention is not limited to the above examples, i.e. it is not meant to imply that the invention must be practiced in the light of the above examples. It should be understood by those skilled in the art that any modification of the present invention, equivalent substitutions of the raw materials of the product of the present invention, addition of auxiliary components, selection of specific modes, etc., are within the scope and disclosure of the present invention.
Claims (10)
1. An emulsion containing an anti-aging composition, wherein the emulsion containing the anti-aging composition is prepared from the following raw materials: anti-aging compositions, skin conditioners, moisturizers and water;
the anti-premature composition comprises: ginger root extract, scutellaria root extract and cacao seed extract;
the skin conditioning agent comprises: hydrolyzed red algae extract, ginseng extract, and edelweiss extract.
2. The emulsion containing an anti-aging composition according to claim 1, wherein the anti-aging composition is characterized in that the mass ratio of the ginger flower root extract, the scutellaria root extract and the cacao seed extract is (3-8): (5-15): (2-7);
preferably, in the skin conditioner, the mass ratio of the hydrolyzed red algae extract, the ginseng extract and the edelweiss extract is (1-5): (0.1-0.5).
3. The emulsion containing the anti-aging composition according to claim 1 or 2, wherein the raw materials for preparing the emulsion containing the anti-aging composition comprise, based on 100% by mass of the raw materials for preparing the emulsion containing the anti-aging composition, by mass percent: 0.01-5% of anti-aging composition, 0.01-10% of skin conditioner, 1-2% of emulsifier, 5-25% of humectant and the balance of water.
4. An emulsion containing an anti-aging composition according to any of claims 1 to 3, characterized in that the skin conditioner further comprises: one or more of radix Sophorae Flavescentis extract, radix Glycyrrhizae Uralensis extract, nicotinamide, yeast/fructus oryzae fermentation product filtrate, cracked yeast fermentation product lysate, animal umbilical cord extract, animal placenta protein or animal placenta enzyme;
preferably, the skin conditioning agent comprises, based on 100% by mass of the starting materials for the preparation of the emulsion containing the anti-aging composition: 1-5% of hydrolyzed red algae extract, 0.1-0.5% of ginseng extract, 0.1-0.5% of alpine edelweiss extract, 0.01-0.1% of sophora flavescens root extract, 0.01-0.1% of glycyrrhiza inflate root extract, 0.1-1% of nicotinamide, 0.01-0.1% of yeast/Szechwan grain seed fermentation product filtrate, 0.01-0.3% of schizosaccharomyces cerevisiae fermentation product lysate, 0.01-0.1% of animal umbilical cord extract, 0.01-0.1% of animal placental protein and 0.01-0.1% of animal placental enzyme.
5. An emulsion containing an anti-aging composition according to any of claims 1 to 4, characterized in that the emulsifier comprises: c20-22 alcohol and/or C20-22 alcohol phosphate;
preferably, the emulsifier comprises, based on 100% by mass of the raw materials for preparing the emulsion containing the anti-aging composition: 0.5-1% of C20-22 alcohol and 0.5-1% of C20-22 alcohol phosphate.
6. The emulsion of any of claims 1-5, wherein the humectant comprises: any one or the combination of at least two of butanediol, glycerol polyether-26, trehalose, sodium hyaluronate or 1, 2-hexanediol;
preferably, the humectant comprises, based on 100% by mass of a raw material for preparing the emulsion containing the anti-aging composition: 5-10% of butanediol, 262-5% of glycerol polyether, 2-4% of trehalose, 0.15-0.25% of sodium hyaluronate and 0.1-1% of 1, 2-hexanediol.
7. The emulsion comprising an anti-aging composition according to any of claims 1 to 6, wherein the emulsion is prepared from the following raw materials: any one or a combination of at least two of an emollient, a thickener, a pH adjuster, a chelating agent, an antioxidant, or a fragrance;
preferably, the preparation raw materials of the emulsion containing the anti-aging composition further comprise, based on 100% by mass of the preparation raw materials of the emulsion: emollient 5-10%, thickener 0.2-0.5%, pH regulator 0.16-0.35%, chelating agent 0.01-0.05%, antioxidant 0.1-1%, and aromatic 0.01-0.05%;
preferably, the emollient comprises any one or combination of at least two of squalane, isononyl isononanoate, or jojoba oil;
preferably, the thickener comprises an ammonium acryloyldimethyltaurate/VP copolymer and/or a carbomer;
preferably, the pH adjusting agent is arginine;
preferably, the chelating agent is disodium EDTA;
preferably, the antioxidant is p-hydroxyacetophenone;
preferably, the fragrance is a perfume.
8. Method for the preparation of an emulsion containing an anti-aging composition according to any of claims 1 to 7, characterized in that the method for the preparation of the emulsion comprises the steps of:
(1) mixing the humectant and water to obtain a water-phase mixed solution; mixing the emulsifier with an oil phase to obtain an oil phase mixed solution;
(2) mixing the water phase mixed liquor and the oil phase mixed liquor obtained in the step (1) and homogenizing to obtain homogenized liquor;
(3) and (3) mixing and stirring the homogenized liquid obtained in the step (2), the anti-aging composition and the skin conditioner to obtain the emulsion containing the anti-aging composition.
9. The method for preparing an emulsion containing an anti-aging composition according to claim 8, wherein in the step (1), the temperature for mixing the aqueous phase is 80 to 85 ℃ and the time for mixing the aqueous phase is 5 to 10 min;
preferably, in the step (1), the temperature of the oil phase mixing is 75-80 ℃, and the time of the oil phase mixing is 5-10 min;
preferably, in step (1), the aqueous phase is mixed with a thickening agent;
preferably, in the step (1), an emollient is also added to the oil phase mixture;
preferably, in the step (2), the temperature for mixing and homogenizing is 80-82 ℃, and the time for mixing and homogenizing is 5-10 min.
10. The method for preparing an emulsion containing an anti-aging composition according to claim 8 or 9, wherein in the step (3), the temperature of the mixing and stirring is 40 to 50 ℃, and the time of the mixing and stirring is 5 to 10 min;
preferably, in the step (3), any one or a combination of at least two of a pH regulator, a chelating agent, an antioxidant or a fragrance is further added to the mixing and stirring.
