本发明属于保健饮料领域,具体涉及一种用于预防和/或治疗痛风的保健饮品组合物及其制备方法与应用。
背景技术
痛风是一种常见且复杂的关节炎类型,各个年龄段均可能罹患本病,造成痛风的本质原因是体内尿酸水平的升高,造成了尿酸盐在关节和肾脏部位的沉积。尿酸是人类嘌呤化合物的终末代谢产物。嘌呤代谢紊乱导致尿酸在血液里大量积累,引发高尿酸血症。高尿酸血症的出现会引发一系列症状,包括急性痛风性关节炎、痛风性结石、慢性痛风关节炎、关节畸形、慢性间质性肾炎、尿酸性尿路结石、还会造成一定程度的肝脏受损。痛风患者的饮食收到严重限制,例如鱼类等高嘌呤食物,导致其体内缺乏鱼类蛋白等营养,因此,开发出一种既能有效预防和/或治疗痛风,又能补充鱼类蛋白营养的保健饮料意义重大。
发明内容
本发明提供一种组合物,其特征在于按重量份计,所述组合物由以下原料制备得到:鱼胶原蛋白3份、木瓜粉5-10份、绿茶1-5份、丝瓜粉5-10份、椰子粉0.5-5份。
本发明的另一实施方案提供一种保健饮品组合物,其特征在于所述保健饮品组合物由鱼胶原蛋白酶解液和天然植物提取液组成,所述鱼胶原蛋白酶解液为鱼胶原蛋白经碱性蛋白酶、风味蛋白酶酶解后收集的分子量小于10000Da的酶解液;所述天然植物提取液是由木瓜粉、绿茶、丝瓜粉、椰子粉经水浸泡、回流提取后收集的分子量小于10000Da的提取液;其中按重量份计,使用鱼胶原蛋白3份、木瓜粉5-10份、绿茶1-5份、丝瓜粉5-10份、椰子粉0.5-5份。
本发明的另一实施方案提供一种保健饮品组合物的制备方法,其特征在于包括如下步骤:
(1)鱼胶原蛋白酶解液的制备:按重量份计,将鱼胶原蛋白3份用缓冲液100份配置成胶原溶液,加入碱性蛋白酶,调pH 8.5,于50℃下酶解2-4小时后,加入风味蛋白酶,调pH7.5,于50℃下酶解1-3小时后,将酶解液离心10min,取上清液、过超滤膜,收集分子量小于10000Da的酶解液,即得鱼胶原蛋白酶解液;
(2)天然植物提取液的制备:按重量份计,将木瓜粉5-10份、绿茶1-5份、丝瓜粉5-10份、椰子粉0.5-5份用水1000份浸泡0.5小时后,加热回流提取1-2小时后,过滤,收集滤液、过超滤膜,收集分子量小于10000Da的提取液,即得天然植物提取液;
(3)将步骤(1)得到的鱼胶原蛋白酶解液和步骤(2)得到的天然植物提取液混合,即得所述保健饮品组合物。
步骤(1)中碱性蛋白酶、风味蛋白酶的用量均为每克鱼胶原蛋白使用5000U。
本发明的另一实施方案提供上述保健饮品组合物在制备预防和/或治疗痛风的保健饮料中的应用。

本发明所述鱼胶原蛋白为市售鱼胶原蛋白(蛋白含量≥98%),可以是淡水鱼胶原蛋白,也可以是海洋鱼胶原蛋白;所述碱性蛋白酶、风味蛋白酶均为市售产品;所述缓冲液为本领域常用缓冲液,优选磷酸盐缓冲液;调pH时,本领域技术人员根据体系酸碱性,选自本领域常用的酸碱调节剂。本发明所述木瓜粉、丝瓜粉、绿茶、椰子粉均为市售产品,其中木瓜粉可以为皱皮木瓜粉、宣木瓜粉、广西青木瓜粉,优选广西青木瓜粉;所述丝瓜粉,可以由丝瓜去皮后冷冻干燥制备得到。
与现有技术相比,本发明的优点在于:(1)本发明通过在植物提取物中添加鱼胶原蛋白酶解液获得一种能调节体内尿酸、肌酐、黄嘌呤氧化酶(XOD)、腺苷脱氨酶(ADA)含量的保健饮品组合物,该保健饮品组合物有望被开发用于预防和/或治疗痛风;(2)本发明保健饮品组合物中各组分缺一不可,尤其是鱼胶原蛋白和丝瓜粉的比例;(3)克服现有技术中的痛风患者不能摄入鱼类的偏见。

具体实施方式
实施例1
(1)鱼胶原蛋白酶解液的制备:按重量份计,将鱼胶原蛋白3g用(磷酸)缓冲液100g配置成胶原溶液,按5000U/g的量加入碱性蛋白酶,调pH 8.5,于50℃下酶解2小时后,按5000U/g的量加入风味蛋白酶,调pH 7.5,于50℃下酶解1小时后,将酶解液离心10min,取上清液、过超滤膜,收集分子量小于10000Da的酶解液,即得鱼胶原蛋白酶解液;
(2)天然植物提取液的制备:按重量份计,将木瓜粉5g、绿茶1g、丝瓜粉5g、椰子粉0.5g用水1.0L浸泡0.5小时后,加热回流提取1小时后,过滤,收集滤液、过超滤膜,收集分子量小于10000Da的提取液,即得天然植物提取液;
(3)将步骤(1)得到的鱼胶原蛋白酶解液和步骤(2)得到的天然植物提取液混合,即得所述保健饮品组合物(以下简称产品A)。
实施例2
(1)鱼胶原蛋白酶解液的制备:按重量份计,将鱼胶原蛋白3g用(磷酸)缓冲液100g配置成胶原溶液,按5000U/g的量加入碱性蛋白酶,调pH 8.5,于50℃下酶解4小时后,按5000U/g的量加入风味蛋白酶,调pH 7.5,于50℃下酶解3小时后,将酶解液离心10min,取上清液、过超滤膜,收集分子量小于10000Da的酶解液,即得鱼胶原蛋白酶解液;
(2)天然植物提取液的制备:按重量份计,将木瓜粉10g、绿茶5g、丝瓜粉10g、椰子粉5g用水1.0L浸泡0.5小时后,加热回流提取2小时后,过滤,收集滤液、过超滤膜,收集分子量小于10000Da的提取液,即得天然植物提取液;
(3)将步骤(1)得到的鱼胶原蛋白酶解液和步骤(2)得到的天然植物提取液混合,即得所述保健饮品组合物(以下简称产品B)。
实施例3
(1)天然植物提取液的制备:按重量份计,将木瓜粉5g、绿茶1g、椰子粉0.5g用水1.0L浸泡0.5小时后,加热回流提取1小时后,过滤,收集滤液、过超滤膜,收集分子量小于10000Da的提取液,即得天然植物提取液;
(2)将实施例1步骤(1)得到的鱼胶原蛋白酶解液和本实施例步骤(1)得到的天然植物提取液混合,即得保健饮品组合物(以下简称产品C)。
实施例4动物实验
本实验选择使用雄性昆明小鼠,体重30g左右,6-8周龄,建模使用灌胃酵母膏和氧嗪酸钾联合造模法,使小鼠机体的尿酸大量积累,罹患高尿酸血症。测定指标分别是尿酸,肌酐(因痛风引起的肾脏病变导致的血液肌酐升高),腺苷脱氨酶(ADA),黄嘌呤氧化酶(XOD)(它们是尿酸代谢的关键酶,因灌胃酵母膏以及氧嗪酸钾的缘故致使代谢紊乱,ADA和XOD活性会出现升高)。
55只昆明小鼠分成五大组,分别是空白组、模型对照组、阳性药物组、给药治疗低剂量组(产品A)、给药治疗高剂量组(2倍浓度产品A,通过浓缩产品A得到)。每组11只小鼠。前7天小鼠自由摄食,自由摄水,适应环境。第8天开始建模,建模成功(第14天,建模共7天)后开始给药(15-28天,给药共14天),具体方案见表1。

给药14天后取血清测定尿酸和肌酐含量,解剖小鼠取肝脏,分别测定黄嘌呤氧化酶(XOD)和腺苷脱氨酶(ADA)活性,结果见下图(图1-5)。图1试验结果表明,建模7天后,小鼠痛风模型建模成功。图2表明,随着给药,阳性药物三天开始有效果,给药饮料组(低)7天开始有效果,并且效果与阳性药物达到一致,直到14天给药饮料组(高、低)能够与阳性药物维持相同的效果。图3-图5表明小鼠的黄嘌呤氧化酶、腺苷脱氨酶和肌苷含量,给药14天后,给药饮料组都能够使各项指标恢复空白组状态,能够达到和阳性药物类似的效果。但是不存在剂量依赖。
分别以产品B、C、实施例1制备的鱼胶原蛋白酶解液、实施例1制备的天然植物提取液替换产品A重复上述动物实验,结果表明,产品B实验结果与产品A一致;产品C(高、低)10天开始有效果,但是对尿酸、肌酐、黄嘌呤氧化酶(XOD)和腺苷脱氨酶(ADA)的调节作用远低于产品A和B;实施例1制备的鱼胶原蛋白酶解液和实施例1制备的天然植物提取液分别是第10天和第12天开始对尿酸、黄嘌呤氧化酶(XOD)、腺苷脱氨酶(ADA)产生调节作用,但是其效果低于产品C,给药期间未发现二者对肌酐能产生明显的调节作用。
Claims (8)
1.一种组合物,其特征在于按重量份计,所述组合物由以下原料制备得到:鱼胶原蛋白3份、木瓜粉5-10份、绿茶1-5份、丝瓜粉5-10份、椰子粉0.5-5份。
2.一种保健饮品组合物,其特征在于所述保健饮品组合物由鱼胶原蛋白酶解液和天然植物提取液组成,所述鱼胶原蛋白酶解液为鱼胶原蛋白经碱性蛋白酶、风味蛋白酶酶解后收集的分子量小于10000Da的酶解液;所述天然植物提取液是由木瓜粉、绿茶、丝瓜粉、椰子粉经水浸泡、回流提取后收集的分子量小于10000Da的提取液;其中按重量份计,使用鱼胶原蛋白3份、木瓜粉5-10份、绿茶1-5份、丝瓜粉5-10份、椰子粉0.5-5份。
3.一种保健饮品组合物的制备方法,其特征在于包括如下步骤:
(1)鱼胶原蛋白酶解液的制备:按重量份计,将鱼胶原蛋白3份用缓冲液100份配置成胶原溶液,加入碱性蛋白酶,调pH 8.5,于50℃下酶解2-4小时后,加入风味蛋白酶,调pH 7.5,于50℃下酶解1-3小时后,将酶解液离心10min,取上清液、过超滤膜,收集分子量小于10000Da的酶解液,即得鱼胶原蛋白酶解液;
(2)天然植物提取液的制备:按重量份计,将木瓜粉5-10份、绿茶1-5份、丝瓜粉5-10份、椰子粉0.5-5份用水1000份浸泡0.5小时后,加热回流提取1-2小时后,过滤,收集滤液、过超滤膜,收集分子量小于10000Da的提取液,即得天然植物提取液;
(3)将步骤(1)得到的鱼胶原蛋白酶解液和步骤(2)得到的天然植物提取液混合,即得所述保健饮品组合物。
4.权利要求3所述的制备方法,其特征在于步骤(1)中碱性蛋白酶、风味蛋白酶的用量均为每克鱼胶原蛋白使用5000U。
5.权利要求1所述的组合物、权利要求2所述的保健饮品组合物、权利要求3-4所述方法制备的保健饮品组合物在制备预防和/或治疗痛风的保健饮料中的应用。
6.权利要求1所述的组合物、权利要求2所述的保健饮品组合物、权利要求3-4所述方法制备的保健饮品组合物在制备预防和/或治疗尿酸、肌酐、黄嘌呤氧化酶(XOD)和腺苷脱氨酶(ADA)中一种或几种异常引起的疾病的保健饮料中的应用。
7.权利要求1所述的组合物、权利要求2所述的保健饮品组合物、权利要求3-4所述方法制备的保健饮品组合物在制备预防和/或治疗尿酸、肌酐、黄嘌呤氧化酶(XOD)和腺苷脱氨酶(ADA)中一种或几种升高引起的疾病的保健饮料中的应用。
8.一种保健饮料,其特征在于所述保健饮料包含权利要求1所述的组合物、权利要求2所述的保健饮品组合物或权利要求3-4所述方法制备的保健饮品组合物和任选包括饮品中常用辅料。
