Fermenting and extracting according to the preparation method of the embodiment 1, and sequentially and respectively preparing single traditional Chinese medicine fermentation liquor.
Comparative example 9 Chinese medicinal compound extract
According to the dosage of the traditional Chinese medicine compound mixture consistent with the embodiment 1, the traditional Chinese medicine compound mixture is prepared from the following traditional Chinese medicine components in parts by weight: 10 parts of loquat leaves, 15 parts of olive leaves, 10 parts of wild chrysanthemum flowers, 10 parts of cogongrass rhizome, 15 parts of sophora flower buds, 15 parts of sophora flavescens ait, 10 parts of coptis chinensis and 15 parts of liquorice.
The preparation method comprises the following steps: removing impurities from the traditional Chinese medicine raw materials, cleaning, drying, crushing to 100 meshes, adding deionized water according to the liquid-material mass ratio of 8:1, soaking for 2.5h, extracting at normal temperature by using a high-pressure homogenizing extractor under the extraction pressure of 60MPa, centrifugally separating the extract, concentrating by using a membrane, adsorbing the concentrate by using macroporous resin, eluting for multiple times by using ethanol with the volume fraction of 10-95%, collecting the eluent, concentrating by using the membrane to obtain the concentrate with the same weight as the traditional Chinese medicine, adding butanediol with the same weight as the concentrate for dissolving, and filtering to obtain the traditional Chinese medicine compound extract.
Test example 1 evaluation of efficacy in inhibiting Propionibacterium acnes
1. Preparation of Chinese medicine extract sample
The traditional Chinese medicine compound fermentation liquor for cosmetics with the oil-controlling and acne-removing effects prepared in the embodiment 1, the single traditional Chinese medicine fermentation liquor prepared in the comparative examples 1-8 and the traditional Chinese medicine compound extracting solution prepared in the comparative example 9 are prepared into sample aqueous solutions with the mass fractions of 0.5%, 1.0%, 1.5%, 2.0% and 3.0% for later use.
2. Method for testing inhibition capacity of propionibacterium acnes
10 kinds of aqueous solutions 1m L were each added to a sterile glass test tube, and 8m L of a previously prepared modified GAM broth (Nissui, Japan, No. ZZM-05433) was poured, and the resulting mixture was taken out, purified and CO-incubated at 35 deg.C2Culturing clinical Propionibacterium acnes in incubator for 48h, respectively injecting 1m L bacterial suspension into improved GAM broth culture medium test tube containing sample water solution, using a glass test tube containing no bacterial suspension as negative control, sequentially inoculating Propionibacterium acnes in each sample culture medium from low concentration, and placing at constant temperature of 35 deg.C and CO as soon as possible2The incubator is used for 48 hours. In addition, the bacteriostatic effect of the example 1 is obviously better than that of the comparative example 9, which shows that the bacteriostatic effect of the traditional Chinese medicine compound mixture is obviously enhanced after fermentation.
Test example 2 evaluation experiment of oil control efficiency
1. Principle of experiment
The skin was tested for oil content using a grease tester Sebumeter SM 815. Based on the principle of a photometer, a special extinction adhesive tape with the thickness of 0.1mm can be changed into a semitransparent adhesive tape after absorbing grease on human skin, the light transmission quantity of the semitransparent adhesive tape can be changed, the more grease is absorbed, the greater the light transmission quantity is, and the content of the grease on the skin can be measured.

The preparation method comprises the following steps:
(1) mixing and stirring raw materials including deionized water, EDTA-2Na, glycerol and panthenol, heating to 80 ℃, and stirring until the raw materials are transparent and completely dissolved;
(2) mixing the raw materials 1, 3-butanediol and sodium hyaluronate at normal temperature, uniformly stirring, adding the mixture into the solution obtained in the step (1), keeping the temperature at 80 ℃ for 10 minutes, and uniformly stirring;
(3) cooling to 45 deg.C under stirring, adding the rest materials, and stirring;
(4) vacuum defoaming, discharging at 38 deg.C or below, and making the material body be aqueous solution.
Respectively preparing samples #1 to #3 according to a preparation method, wherein the samples #2 to #3 are comparison samples, selecting a cosmetic which has better sales and is declared to have the effects of controlling oil and removing acnes on the market, and filling the cosmetic into a sample bottle, wherein the sample bottle is marked as a sample #4 and is also used as a comparison sample.
4. Test object
1. 180 oily skin volunteers aged 20-40 years old;
2. patients without severe systemic disease, immunodeficiency or autoimmune disease, the subject site has not received skin treatment, cosmetic and other tests that may affect outcome; those with inactive allergic disease; those without constitutional hypersensitivity; hormone drugs and immunosuppressants have not been used in the last month; the test sites were not enrolled in other clinical trials now or in the last three months.
5. Test method
The subjects were randomly divided into 3 groups of 30, and the group numbers were #1 to #2 and # 4. The subjects sit still for 30 minutes in a laboratory with the temperature of 22 +/-3 ℃ and the relative humidity of 50 +/-5 percent, the skin oil content of the left forehead and the right forehead is respectively tested by using an oil tester Sebumeter SM815 and the data is recorded as D0, the samples #3 and the correspondingly numbered samples of each group are distributed to each subject, the subjects are guided to carry out half-face comparison and trial, the two samples are respectively smeared on the left half-face or the right half-face, 0.5g of the sample is smeared once in the morning and at the evening every day and lasts for 28 days, and the skin oil content is respectively tested by returning to the constant temperature laboratory on the 14 th day and the 28 th day and the data is recorded as D14 and D28. Respectively calculating the oil content change rate of D14 and D28 according to the following formula:

(4) The experimental results are as follows:

The result shows that the skin oil content is obviously reduced after 14 days of the experimental sample #1 added with the fermentation liquid prepared in the embodiment 1 of the invention, and the effect is more obvious after the continuous use for 28 days, while the oil control effect is obviously inferior to that of the sample #1 when the experimental sample #2 prepared by the extracting solution containing the comparative example 9 is continuously used, which shows that the acne removing effect of the traditional Chinese medicine compound is obviously enhanced after the fermentation. And the oil control effect of the samples #1 and #2 is much stronger than that of the sample #4, which shows that the traditional Chinese medicine compound of the invention has obvious oil control effect.
Test example 3 irritation evaluation test
Specifically, 500 mu L of the fermentation liquid prepared in example 1 and comparative examples 1 to 8 was placed in a 2m L centrifuge tube, and then 2% rabbit red blood cell (commercially available) suspension 500 mu L was added, the mixture was gently shaken and mixed, incubated in a 37 ℃ incubator for 3 hours, centrifuged at 2000r/min for 3 minutes, the supernatant was extracted, physiological saline was used as a negative control, distilled water was used as a positive control, the absorbance was measured at a wavelength of 414nm, and the hemolysis was calculated according to the formula, and the results are shown in Table 4.

The comparison of the experimental results of the embodiment 1 and the comparative examples 1 to 8 shows that the hemolysis ratio of the fermentation liquid of the traditional Chinese medicine compound is much lower than that of the fermentation liquid of a single traditional Chinese medicine prepared by the comparative examples 1 to 8, and the irritation of the fermentation liquid after the compound is obviously reduced.

Effect example 1 evaluation of acne removing Effect, oil controlling Effect and skin irritation
Evaluation target: 30 young people with acne on the face and the age of 15-30 years are selected, wherein 15 male people and 15 female people are selected.
The evaluation method comprises the following steps: the oil-control acne-removing mask prepared in application example 3 is applied to the face in the morning and at night every day by a double-blind method, and the face is massaged after the mask is taken down until all the mask liquid is absorbed by the skin.
Evaluation course of treatment: the acne removing effect evaluation takes 20 days as a treatment course, the oil control effect evaluation takes 28 days as a treatment course, and the irritation evaluation takes 20 days as a treatment course.
The evaluation standard of the acne removing effect is that ① has obvious effect that acne is removed by more than 70 percent, ② has effective effect that acne is removed by more than 30 percent, and ③ has no effect that acne is removed by less than 30 percent.
Evaluation standard of oil control effect: the test method was the same as that described in test example 2.
Irritation evaluation criteria: the examination was performed according to the criteria of Table 8. The experiment was carried out for a total of 20 days.

Evaluation results of oil control effect: after 28 days, the skin oil reduction rate of the volunteers is 41.6%, which shows that the facial mask for controlling oil and removing acnes prepared in application example 3 has a good oil control effect.
Evaluation results of irritation: no allergic phenomenon was found in all volunteers.

the traditional Chinese medicine compound fermentation liquor for cosmetics with the effects of controlling oil and removing acnes adopts a fermentation extraction method, namely, after the traditional Chinese medicine compound mixture is subjected to fermentation treatment, the traditional Chinese medicine compound fermentation liquor is obtained through filtration, sterilization and centrifugal separation;
the method specifically comprises the following steps:
① the composite bacteria is added in the form of bacteria liquid, the composite bacteria liquid is composed of lactobacillus plantarum and saccharomyces cerevisiae according to the proportion of (1-6): 1, and the concentration of the composite bacteria liquid is 106~108cfu/mL;
