The color of the plant composite enzyme is yellow and semitransparent, and the dry matter content of the plant composite enzyme is determined to be 0.52%.
Example 2 preparation of plant combination enzymes
Mixing herba Schizonepetae, bamboo, semen Apii Graveolentis, and pericarpium Citri Junoris, wherein the mixture accounts for 60g, 20g, 10g, and 10g of 100g of herba Schizonepetae, bamboo, semen Apii Graveolentis, and pericarpium Citri Junoris; 1kg of deionized water was added; then ultrasonic oscillation is carried out, the ultrasonic power is 300W, the temperature is 70 ℃, and the ultrasonic oscillation time is 60 min; after ultrasonic oscillation is finished, transferring all materials into a water bath heating device, heating in a water bath until the system keeps a slightly boiling state, continuously extracting for 2 hours, standing after extraction is finished, and cooling to 35 ℃; adding sucrose accounting for 15 percent of the mass of the extracting solution and mixing evenly; then adding biological compound strains with the mass ratio of 5 percent of the extract to start fermentation, and maintaining the temperature at 35 ℃ under the condition of keeping out of the sun and carrying out sealed fermentation for 7 days. After the fermentation process is finished, centrifuging the mixed fermentation liquor, and taking the supernatant to store at 4 ℃; namely the plant composite ferment of the invention. The color of the plant composite enzyme is yellow and semitransparent, and the dry matter content of the plant composite enzyme is determined to be 0.94%.
Example 3 preparation of plant combination enzymes

After the fermentation process is finished, centrifuging the mixed fermentation liquor, and taking the supernatant to store at 4 ℃; namely the plant composite ferment of the invention. The color of the plant composite enzyme is yellow and transparent, and the dry matter content is measured to be 0.36 percent.

Experimental mites: the method for obtaining the human demodex by adopting a transparent adhesive tape pasting overnight method comprises the following specific steps: the examinee is ordered to clean the face thoroughly before sleeping, and then the transparent adhesive tapes with the length of 6cm and the width of 1.5cm are respectively stuck to the forehead, the cheek, the nose, the chin and the like of the examinee. And taking off the film the next day, adhering the film on a glass slide, and performing microscopic examination.

1.2 Experimental methods
1.2.1 experimental groups: the number of the liquid to be detected is 1-6, and meanwhile, a 2% metronidazole solution is used as a positive control group, and liquid paraffin is used as a blank control group. Each group of 10 demodex mites tested. The experiment was repeated 3 times for each concentration of the drug solution and the results averaged.
1.2.2 methods of observation: sucking 200 μ L of the liquid medicine with a micropipette, dropping onto a carrier, spreading the liquid medicine with a push sheet, and sticking the adhesive tape for detecting Demodex mites on the glass slide to ensure that the liquid medicine is fully contacted with the worm body. 1.2.3 criteria for mortality: continuously observing under 400 times microscope for 1min, determining death of mite with proclarm or immobile leg or paw as initial judgment, and determining death of mite with continuous observation after 30 min.

As can be seen from the results of Table 1 and FIG. 1, the plant combined enzyme has a remarkable killing effect on human demodex, the lethality is 60-70%, and is remarkably higher than that of a positive control group by 2% metronidazole; the plant combined water extract without biological fermentation also has a certain killing effect on human demodex, and the fatality rate is 30-40% and is lower than that of a positive control group. Therefore, the biological fermentation process promotes further dissolution and transformation of effective substances in the plant formula, and the acaricidal effect is obviously improved.
Example 5 inhibition of tyrosinase Activity by plant combination enzyme and plant combination aqueous extract
Tyrosinase (Tyrosinase, TYR), also known as polyphenol oxidase, participates in the first two steps of melanin synthesis, and the activity of Tyrosinase is related to the amount of melanin synthesis. In human body, tyrosinase is mainly present in melanocytes of epidermal cells of skin, and has an important relationship with the occurrence of some common skin diseases such as dyschromatosis and malignant melanoma. Therefore, the tyrosinase inhibitor is also used as an additive with the effects of whitening and lightening spots and added into cosmetics to improve the tyrosinase metabolism of pigment cells in skin and prevent pigmentation, so that the problem of the acne spots after healing of the acne spots can be improved, and lightening of the acne spots is promoted.
1 materials and methods
1.1 Experimental materials
Preparation of 1.1.1L-tyrosine solution (7.5mmol/L)
0.2721g of L-tyrosine is accurately weighed, a plurality of drops of concentrated hydrochloric acid are firstly added, about 50mL of deionized water is added, after the concentrated hydrochloric acid and the deionized water are completely dissolved by slight heating, the pH value is adjusted to 7.0 by using sodium hydroxide solution, and the deionized water is added to a constant volume of 200 mL.
1.1.2 preparation of tyrosinase liquid
Preparing tyrosinase liquid from fresh and intact potatoes, which comprises the following specific operations: cleaning potato, and precooling at 4 deg.C for about 4 h. Peeled, cut into pieces of about 1X 1cm3, and frozen at-20 ℃ overnight. Weighing, adding 4 deg.C pre-cooled sodium phosphate buffer solution at a ratio of 1: 1(W: V), homogenizing with tissue triturator, filtering with 3 layers of gauze, centrifuging the filtrate at 4000r/min for 10min to obtain supernatant as obtained tyrosinase crude enzyme solution, storing at 4 deg.C, and using up in 2 hr.
1.1.3 preparation of test solutions
Taking 1mL of the plant combination enzyme and the plant combination water extract prepared in the example 1 and the comparative example 1 respectively, and diluting to 3mL for later use, wherein the samples are No. 1 and No. 2; taking 1mL of the plant combination enzyme and the plant combination water extract prepared in the example 2 and the comparative example 2 respectively, and diluting to 6mL for later use, wherein the samples are No. 3 and No. 4; the plant combination enzyme and the plant combination water extract prepared in example 3 and comparative example 3 were each 1mL diluted to 2mL for use, samples nos. 5 and 6. The positive control is a 1% arbutin solution.
1.2 Experimental methods
The total reaction system was 5 mL. The specific design is shown in table 2:

Note: when the absorbance is measured by a spectrophotometer, the test solution, the standard control and the positive control are respectively zeroed by the negative control 1, the negative control 2 and the negative control 3.
In the experiment, phosphate buffer solution, test solution (including positive control) and enzyme solution are sequentially added into a test tube, and water bath is carried out at 30 ℃ for 10 min. The substrate L-tyrosine was then added and the timer was started immediately. The absorbance at a wavelength of 475nm at 20min of the reaction was determined. The inhibition rate of the test solution (including the positive control) on the tyrosinase is calculated by using the following formula with the corresponding negative control as a reference during the determination.

Wherein "A" is the absorbance of the standard control and "B" is the absorbance of the test solution (or positive control).
Each experiment was done in 3 replicates. The high inhibition rate indicates that the inhibition intensity of the compound on tyrosinase activity is high.

The results in table 2 and fig. 2 show that: the fermented liquid obtained by fermenting the plant combined water extract, namely the plant combined enzyme prepared in the embodiments 1-3, has the inhibition capability on tyrosinase, which is greatly improved compared with that of the unfermented water extract, and is also greatly superior to that of arbutin with the concentration of 1% of a positive control (in whitening daily chemical products, the common addition concentration of arbutin is 1%). The biological fermentation process promotes further dissolution and transformation of effective substances in the plant formula, and the application value of the plant combined enzyme can be greatly improved.
Example 6 preparation of a mite and acne removing skin care lotion containing phytolacca de-acne phytolacca composition enzyme as a main functional component a formula of the mite and acne removing skin care lotion containing the phytolacca de-acne phytolacca composition enzyme is shown in table 3.

The preparation process comprises the following steps: heating cetostearyl alcohol, squalane, caprylic capric triglyceride, white oil, and stearyl alcohol polyoxyethylene 2 ether, stirring to 75 deg.C to dissolve completely to obtain phase A; adding appropriate amount of deionized water into plant composite enzyme, Hamamelis mollis extractive solution and carbomer U20, heating and stirring to 75 deg.C for dissolving completely, and keeping the temperature for 20-30 min; then adding glycerol, propylene glycol, butanediol, vitamin E and essence, stirring and dissolving uniformly under the condition of heat preservation, and complementing the balance of deionized water to obtain phase B; adding phase A into phase B, stirring under continuously maintaining temperature, dissolving, cooling to about 45 deg.C, adding TEA and phenoxyethanol, stirring, standing, and cooling to obtain the final product.
The mite-removing acne-removing skin-care emulsion is represented by an embodiment of the plant combined enzyme prepared by the invention in a mite-removing acne-removing daily chemical product, and can also be prepared into daily chemical products with mite-removing and acne-removing effects, such as water aqua, gel, cream, powder, facial mask, manual soap, spray, washing and care products and the like according to a common formula in the daily chemical industry.
Example 7 evaluation of Effect of mite-removing, acne-removing and skin-caring emulsion
Evaluation target: 44 young people with acne on face and 15-30 years of age were selected, 24 male and 20 female.
The evaluation method comprises the following steps: the mite-removing and acne-removing skin care lotion prepared in example 6 is smeared three times in the morning, noon and evening by a double-blind method, and 28 days are a treatment course.
Evaluation criteria: the effect is remarkable: acne is faded by more than 70 percent; ② has certain effect: acne is faded by more than 30 percent; ③ invalid: the acne is faded by less than 30 percent.
Evaluation results were as follows: 29 cases of obvious effect account for 65.9 percent; 10 effective cases account for 22.7 percent; and 5 invalid cases account for 11.4 percent. The total effective rate reaches 88.6 percent, and no adverse reaction such as skin allergy, red swelling and the like is reported. The mite and acne removing plant combined enzyme has a good acne removing effect.
The above embodiments are preferred embodiments of the present invention, but the present invention is not limited to the above embodiments, and any other changes, modifications, substitutions, combinations, and simplifications which do not depart from the spirit and principle of the present invention should be construed as equivalents thereof, and all such changes, modifications, substitutions, combinations, and simplifications are intended to be included in the scope of the present invention.

Claims (10)

1. The mite and acne removing plant combined enzyme is characterized by being prepared by carrying out water extraction and fermentation on at least one of schizonepeta, bamboo, celery seeds and orange peels.

The mite-removing and acne-removing plant combined enzyme according to claim 1, wherein 30-60 parts of herba pseudoschizonepetae, bamboo, celery seeds and orange peels are taken according to the mass component ratio: 20-40 parts of bamboo: 10-30 parts of celery seeds: 10-30 parts of orange peel; the herba schizonepetae is at least one of stems, leaves and flowers of the herba schizonepetae; the bamboo is at least one of green bamboo, tabasheer and bamboo leaf.
3. The mite-removing and acne-removing plant combined enzyme according to claim 2, wherein the mass component ratio of the nepeta cataria, the bamboo, the celery seeds and the orange peels is 40 parts: 30 parts of: 15 parts of: 15 parts.
4. The mite-removing and acne-removing plant combined enzyme according to claim 2, wherein the mass component ratio of the nepeta cataria, the bamboo, the celery seeds and the orange peels is 60 parts: 20 parts of: 10 parts of: 10 parts.

The preparation method of the plant combined ferment for removing mites and acnes as claimed in claim 1, which comprises the following steps: a. mixing herba Schizonepetae, bamboo, semen Apii Graveolentis, and pericarpium Citri Junoris, and adding 60% ethanol solution deionized water 30 times of the total mass of the mixture; b. performing ultrasonic oscillation with ultrasonic power of 200W, temperature of the mixed solution of 50 ℃ and ultrasonic oscillation time of 120 min; c. after ultrasonic oscillation is finished, transferring all materials into a water bath heating device, and heating in a water bath until the system keeps a micro-boiling state for 4 hours for continuous extraction; d. after extraction, standing, cooling to 25 ℃, adding sucrose accounting for 5% of the mass of the extracting solution, and uniformly mixing; e. then adding biological compound strains with the mass proportion of 5 percent of the extract to start fermentation, maintaining the temperature at 25 ℃ in a dark condition, sealing and fermenting for 15d, centrifuging the mixed fermentation liquor after the fermentation process is finished, and taking supernatant to be stored at 4 ℃; the plant combined enzyme for removing mites and acnes is prepared.
7. The plant-combined ferment for removing mites and acnes as claimed in claim 1, wherein the bio-coincidence bacterial species is a combination of any four to six species selected from the group consisting of Nocardia actinomycetes, Bacillus mucilaginosus, Bacillus subtilis, Beauveria bassiana, Trichoderma reesei and Candida.
8. The application of the mite and acne removing plant combined enzyme according to claim 1 in preparation of daily chemical products.
9. The application of the mite and acne removing plant combined enzyme in preparing daily chemical products according to claim 8, wherein the daily chemical products comprise water aqua, essence, emulsion, cream, gel, powder, facial mask, handmade soap, spray and wash care products.

The application of the plant combination enzyme for removing mites and removing acnes in the preparation of daily chemical products according to claim 8, wherein the addition amount of the plant combination enzyme for removing mites and removing acnes in the daily chemical products is 0.5-50% by mass.
