Abstract

本发明公开了一种芍药总苷提取物及其制备方法,该芍药总苷提取物包括芍药甙、芍药多酚和芍药类黄酮。本发明在提取芍药主要活性成分芍药甙的同时获取芍药多酚、芍药类黄酮的方法,提高芍药的综合利用;且芍药多酚、芍药类黄酮回收工艺简单,易于操作,成本低,乙醇可回收,大孔树脂可再生,均可反复利用,产品纯度较较高,溶剂仅为水和乙醇,符合工业绿色化要求,可实现工业化生产。

芍药总苷提取物及其制备方法
技术领域
本发明属于天然产物化学技术领域,尤其涉及一种芍药总苷提取物及其制备方法。
背景技术
芍药总苷作为白芍或赤芍的有效成分之一,现代药理研究表明具有抑制自身免疫反应,抗炎,止痛,保肝,以及增加心排血量,改善心肺功能等药理作用。其主要成分包括芍药苷、白芍药苷、芍药新苷、芍药内酯A、B、C、氧化芍药苷、苯甲酰芍药苷等具有生理功效的有效成分。
已有研究表明芍药多酚和黄酮具有良好的活血化瘀等生物活性。
发明内容
本发明的目的在于提供一种芍药总苷提取物及其制备方法,在提取芍药主要活性成分芍药甙的同时获取芍药多酚、芍药类黄酮的方法,提高芍药的综合利用。
为实现上述目的,本发明采取的技术方案为:
芍药总苷提取物,包括芍药甙、芍药多酚和芍药类黄酮;该芍药总苷提取物通过以下步骤制备:
S1、将洗净的芍药置于闪式提取器内,加入6倍体积的90%的乙醇溶液,处理1-3分钟,固液分离后,回收乙醇溶液,浓缩,使得提取物呈稠膏状,再用乙酸乙酯对提取物萃取3次,回收溶剂,得提取液;
S2、将所得的提取液以3-5BV/h的流速缓慢通过D101大孔树脂,D101大孔树脂用75%的乙醇以3-5BV/h洗脱,洗脱液再以1BV/h依次过732阳离交换柱、707阴离交换柱和聚酰胺柱,聚酰胺柱流出液浓缩、干燥,得到芍药甙;
S3、收集D101树脂的过柱液,将过柱液减压浓缩3.131mg/ml后,取1.5L树脂,以3BV/h的流速缓慢通过预处理好的LSA-21树脂然后依次用蒸馏水、30%乙醇1000mL梯度洗脱,收集30%乙醇洗脱液的中间段,于旋转蒸发仪上旋干所得到的芍药多酚;
S4、将聚酰胺柱依次用水洗、75%乙醇洗涤,收集75%乙醇洗脱液;将75%乙醇洗脱液浓缩、干燥,得到的芍药类黄酮。
本发明具有以下有益效果:
在提取芍药主要活性成分芍药甙的同时获取芍药多酚、芍药类黄酮的方法,提高芍药的综合利用;且芍药多酚、芍药类黄酮回收工艺简单,易于操作,成本低,乙醇可回收,大孔树脂可再生,均可反复利用,产品纯度较较高,溶剂仅为水和乙醇,符合工业绿色化要求,可实现工业化生产。

为了使本发明的目的及优点更加清楚明白,以下结合实施例对本发明进行进一步详细说明。应当理解,此处所描述的具体实施例仅仅用以解释本发明,并不用于限定本发明。
实施例1
芍药总苷提取物,包括芍药甙、芍药多酚和芍药类黄酮;该芍药总苷提取物通过以下步骤制备:
S1、将洗净的芍药置于闪式提取器内,加入6倍体积的90%的乙醇溶液,处理1-3分钟,固液分离后,回收乙醇溶液,浓缩,使得提取物呈稠膏状,再用乙酸乙酯对提取物萃取3次,回收溶剂,得提取液;
S2、将所得的提取液以3BV/h的流速缓慢通过D101大孔树脂,D101大孔树脂用75%的乙醇以3BV/h洗脱,洗脱液再以1BV/h依次过732阳离交换柱、707阴离交换柱和聚酰胺柱,聚酰胺柱流出液浓缩、干燥,得到芍药甙;
S3、收集D101树脂的过柱液,将过柱液减压浓缩3.131mg/ml后,取1.5L树脂,以3BV/h的流速缓慢通过预处理好的LSA-21树脂然后依次用蒸馏水、30%乙醇1000mL梯度洗脱,收集30%乙醇洗脱液的中间段,于旋转蒸发仪上旋干所得到的芍药多酚;
S4、将聚酰胺柱依次用水洗、75%乙醇洗涤,收集75%乙醇洗脱液;将75%乙醇洗脱液浓缩、干燥,得到的芍药类黄酮。
实施例2
芍药总苷提取物,包括芍药甙、芍药多酚和芍药类黄酮;该芍药总苷提取物通过以下步骤制备:
S1、将洗净的芍药置于闪式提取器内,加入6倍体积的90%的乙醇溶液,处理1-3分钟,固液分离后,回收乙醇溶液,浓缩,使得提取物呈稠膏状,再用乙酸乙酯对提取物萃取3次,回收溶剂,得提取液;
S2、将所得的提取液以5BV/h的流速缓慢通过D101大孔树脂,D101大孔树脂用75%的乙醇以5BV/h洗脱,洗脱液再以1BV/h依次过732阳离交换柱、707阴离交换柱和聚酰胺柱,聚酰胺柱流出液浓缩、干燥,得到芍药甙;
S3、收集D101树脂的过柱液,将过柱液减压浓缩3.131mg/ml后,取1.5L树脂,以3BV/h的流速缓慢通过预处理好的LSA-21树脂然后依次用蒸馏水、30%乙醇1000mL梯度洗脱,收集30%乙醇洗脱液的中间段,于旋转蒸发仪上旋干所得到的芍药多酚;
S4、将聚酰胺柱依次用水洗、75%乙醇洗涤,收集75%乙醇洗脱液;将75%乙醇洗脱液浓缩、干燥,得到的芍药类黄酮。
实施例3
芍药总苷提取物,包括芍药甙、芍药多酚和芍药类黄酮;该芍药总苷提取物通过以下步骤制备:
S1、将洗净的芍药置于闪式提取器内,加入6倍体积的90%的乙醇溶液,处理1-3分钟,固液分离后,回收乙醇溶液,浓缩,使得提取物呈稠膏状,再用乙酸乙酯对提取物萃取3次,回收溶剂,得提取液;
S2、将所得的提取液以4BV/h的流速缓慢通过D101大孔树脂,D101大孔树脂用75%的乙醇以4BV/h洗脱,洗脱液再以1BV/h依次过732阳离交换柱、707阴离交换柱和聚酰胺柱,聚酰胺柱流出液浓缩、干燥,得到芍药甙;
S3、收集D101树脂的过柱液,将过柱液减压浓缩3.131mg/ml后,取1.5L树脂,以3BV/h的流速缓慢通过预处理好的LSA-21树脂然后依次用蒸馏水、30%乙醇1000mL梯度洗脱,收集30%乙醇洗脱液的中间段,于旋转蒸发仪上旋干所得到的芍药多酚;
S4、将聚酰胺柱依次用水洗、75%乙醇洗涤,收集75%乙醇洗脱液;将75%乙醇洗脱液浓缩、干燥,得到的芍药类黄酮。

1.芍药总苷提取物,其特征在于:包括芍药甙、芍药多酚和芍药类黄酮。
2.芍药总苷提取物的制备方法,其特征在于:包括如下步骤:
S1、将洗净的芍药置于闪式提取器内,加入6倍体积的90%的乙醇溶液,处理1-3分钟,固液分离后,回收乙醇溶液,浓缩,使得提取物呈稠膏状,再用乙酸乙酯对提取物萃取3次,回收溶剂,得提取液;
S2、将所得的提取液以3-5BV/h的流速缓慢通过D101大孔树脂,D101大孔树脂用75%的乙醇以3-5BV/h洗脱,洗脱液再以1BV/h依次过732阳离交换柱、707阴离交换柱和聚酰胺柱,聚酰胺柱流出液浓缩、干燥,得到芍药甙;
S3、收集D101树脂的过柱液,将过柱液减压浓缩3.131mg/ml后,取1.5L树脂,以3BV/h的流速缓慢通过预处理好的LSA-21树脂然后依次用蒸馏水、30%乙醇1000mL梯度洗脱,收集30%乙醇洗脱液的中间段,于旋转蒸发仪上旋干所得到的芍药多酚;
S4、将聚酰胺柱依次用水洗、75%乙醇洗涤,收集75%乙醇洗脱液;将75%乙醇洗脱液浓缩、干燥,得到的芍药类黄酮。
