本发明公开了一种火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,其中火棘果提取物是由以下方法制得:S1、净制;S2、组织破碎;S3、煎煮;S4、酸化处理;S5、大孔吸附树脂柱层析纯化;S6、醇提:加入乙醇冷浸,提取液滤过,合并滤液;S7、真空减压浓缩得到火棘果提取物。本发明的有益效果为:控制用于制作保健食品或药品中火棘果中的红色素、总酚、总黄酮类化学成分的含量,将其制成口服固体制剂等,可起到提高视觉敏感度、防近视、防远视、防夜盲等症的作用。本发明拓宽了火棘果的利用范围,提高了火棘果的开发利用价值,具有广阔的市场前景。
Description
技术领域
本发明涉及火棘果的新用途,尤其是火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途。
背景技术
火棘为蔷薇科火棘Pyracantha属火棘P.fortuneana植物的果实,又名救命粮、救兵粮、火把果、赤阳子等,最早收载于本草学专著《滇南本草》(明·兰茂著)。味甘、酸。治胸中痞块、食积、消虫、明目,泻肝经之火,止妇人崩漏皆效。
火棘果具有很高的食用价值和药用价值,如火棘果可做成果冻、果酱、调味汁、保健酒、果汁、果醋等食品,这主要是因为火棘果富含维生素、人体必需的微量元素、粗蛋白、人体必需氨基酸、果胶等多种营养成分。其营养成分齐全,含量丰富、比例协调,相较苹果、猕猴桃等水果更具特色;现代研究发现火棘果中的联苯苷与二苯基呋喃苷类具有酪氨酸酶抑制活性,是美白的药效物质,因此,现已有将火棘提取物开发作为化妆品的专利与应用。另外,研究还证实火棘果的水、醇提取物具有乙酰胆碱酯酶、α-葡萄糖苷酶、α-淀粉酶的抑制活性。以上研究揭示火棘果是一个具有抗Ⅱ型糖尿病、阿尔兹海默症、皮肤保养等多重功能的药物。
但在现代火棘果的研究与应用之中,还无火棘果具有治疗近视、远视、夜盲、提高视觉敏感度作用的研究报道。查询历代本草,发现仅有最早记载火棘的《滇南本草》[兰茂撰.滇南本草.北京:中国中医药出版社,2013.01.]记载火棘具有明目功效。但从中医理论分析,其明目是因火棘能泻肝经之火,而肝开窍于目,故能清肝明目,其适用范围是因肝经火旺而致的暴发火眼之症。从现有的文献可查询的例证有二:(1)火棘根、叶、果可治火眼(李耕冬,贺廷超编著.彝医植物药续集.成都:四川民族出版社,1992.01.);(2)火棘叶、三颗针各15g,水煎洗,可治红眼病(杨卫平,夏同珩主编.常用中草药图谱及配方4.贵阳:贵州科技出版社,2012.09.)。也就是说,火棘在《滇南本草》中的明目作用与本专利所提出的火棘提取物具有的提高视觉敏感度、防治近视、远视、夜盲等症并不是同一种作用。
现代研究表明,火棘果中含有丰富的酚类、黄酮、红色素、鞣质及多糖等类成分。据此,我们认为火棘果中的红色素、黄酮及酚类成分是本品提高视觉敏感度,防治近视、夜盲、远视等症的药效成分,原因如下:(1)火棘红色素由两类成分组成:一类是亲脂性的红色色素,主要含有:α-胡萝卜素,β-胡萝卜素,γ-胡萝卜素与多顺式番茄红素,其含量为0.01-0.07%(干果),另一类是亲水性的花色苷类成分,如矢车菊-3-葡萄糖苷氯化物、牵牛花素、飞燕草素、锦葵素等,此两类成分均是提高视觉敏感度、防治夜盲、近视的药效成分,其中胡萝卜素类成分可在人体内转化为维生素A,防治夜盲症;花色苷类成分也具有多方面的作用,如具有抗氧化、抗突变、抗癌、抗炎、促进视红素再生等作用,其中如花青素-3-阿拉伯糖苷、花青素-3-乳糖苷对人的视觉有保护作用。同时,火棘果中还含有丰富的黄酮等酚酸类成分,如(−)-表儿茶素,(+)-儿茶素,芹菜素,苯甲酸,咖啡酸,绿原酸,阿魏酸,没食子酸,陈皮苷,山萘酚,柚皮素,柚皮苷,对香豆酸,对羟基苯甲酸,原儿茶酸,迷迭香酸,芦丁,紫丁香酸,反式桂皮酸,香草醛,此类成分具有较强的Vp样作用,有强力持续毛细血管浸透作用;血管保护作用;抗凝血;抗病毒、抗菌;免疫调节;改善微循环等作用,此也可对防治近视、提高视觉敏感度起到辅助作用。现代研究火棘果多糖还具有抗疲劳、肝保护、提高免疫功能等作用,此也可对防治近视起到辅助作用。目前还未公开火棘果在防治近视、夜盲、提高视觉敏感度等方面的用途,因此,在现有技术水平上,火棘果提取物在制备防近视、夜盲、提高视觉敏感度等方面的保健食品与药品中有着十分巨大的开发潜能。
发明内容
本发明的目的在于克服现有技术的不足,提供一种火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,能够提高火棘果的开发利用效率。
本发明的目的是通过以下技术方案来实现的:
火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,所述的火棘果提取物是由以下方法制得:
S1、净制:挑选色泽饱满、无虫蛀的新鲜火棘果,并除去杂质,将所选火棘果用水漂洗干净;
S2、组织破碎:将净制后的新鲜火棘果加入到匀浆机中,并加入新鲜火棘果重量1-1.2倍的水,搅拌捣碎2-5min,得到火棘果浆;
S3、煎煮:在火棘果浆中加入其重量3-5倍的水煎煮1.5-3h,用纱布过滤,得到火棘果液;
S4、酸化处理:在火棘果液中加入2-5%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到1-4;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:6-8的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用2-3BV的蒸馏水洗脱,后用4-6BV的70-80%的乙醇洗脱,收集洗脱液;
所述的大孔吸附树脂柱的类型为HP、SP系列树脂柱或XDA系列树脂柱,作为优选的,大孔吸附树脂柱选用XDA-8、HP-30或SP-207。
S6、醇提:在洗脱液中加入3-5倍质量分数为95%的乙醇在室温冷浸3-5次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.2-1.3的火棘果提取物,其中,提取物中红色素含量以番茄红素计不少于10.0μg/g,总黄酮含量以芦丁计不少于5.0%,总酚含量不少于30.0%。
火棘果中果胶(多糖)含量较高,是其他有效含量的数倍乃至数十倍,因此在醇提的过程之前,申请人考虑对果胶进行酶解,盐酸乙醇溶液的加入给果胶酶提供了良好的酶解环境,酶解后的果胶含量达到符合制备防近视保健食品与药品的条件,同时,盐酸乙醇有利于花色素苷类的提取,其主要是因为酸的加入,可使花色苷处于烊盐状态,从而提高了它们的亲水性,使其在乙醇中的溶解度增大。同时,酸的加入还可以破坏酚类物质与蛋白质、多糖、以及自身之间的氢键和疏水键作用,从而提高花色素苷类的提取。
步骤S5层析纯化可以有效地降低火棘果中果胶的含量,天然果胶类物质以原果胶、果胶、果胶酸的形态广泛存在于植物的果实、根、茎、叶中,是细胞壁的一种组成成分,它们伴随纤维素而存在,构成相邻细胞中间层粘结物,使植物组织细胞紧紧黏结在一起。原果胶是不溶于水的物质,但可在酸、碱、盐等化学试剂及酶的作用下,加水分解转变成水溶性果胶。果胶本质上是一种线形的多糖聚合物,分子量大,因此,通过大孔吸附树脂的分子筛作用,可将果胶随药液流出而去除,而有效成分吸附在柱上。最后,再使用高浓度乙醇将有效成分洗脱,即可提高火棘果提取物中有效成分的含量。
将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成制剂。
所述制剂为保健食品、药用口服溶液剂、口服固体制剂。
所述的口服固体制剂口含片由下述重量份的成分组成:火棘果提取物1-1.5份,湿润剂重量份的95%乙醇,乳糖粉5-7份,薄荷油0.01-0.05份,滑石粉0.01-0.05份,硬脂酸镁0.01-0.05份。
所述的口服固体制剂的制备方法为:在上述重量份的乳糖粉中加入湿润剂量的乙醇和上述重量份的火棘果提取物搅拌5-10min,制成软材,然后用16-20目筛制粒,湿颗粒在55-60℃下低温干燥2-4h,然后用16-20目筛整粒,再加入上述重量份的薄荷油、滑石粉和硬脂酸镁,混匀,压片,即得。其中,乙醇作为湿润剂,用于将乳糖粉、薄荷油、滑石粉以及硬脂酸镁湿润后混合到火棘果提取物中,乙醇的用量标准为混合物中无明显的粉粒即可。作为优选的,95%乙醇的用量为乳糖粉、滑石粉、硬脂酸镁重量总和的5-10%。
所述的流出液经过减压浓缩,在浓缩液中加入2倍量的乙醇,静置,滤取沉淀,然后将沉淀溶于水中,用Sevage法除去蛋白质后,加入2倍量的乙醇,滤取沉淀,依次用乙醇、无水乙醇、丙酮、乙醚洗涤,除去脂溶性成分和水分,减压低温干燥可得火棘果果胶提取物,可以作为火棘果副产品的二次开发,经过大孔吸附树脂柱的流出液中含有较高含量的水溶性果胶,可用于果酱、果冻的制造;防止糕点硬化;改进干酪质量;制造果汁粉等。
本发明的有益效果是:
(1)利用乙醇作溶剂,提取火棘果中有效成分,控制用于制作保健食品或药品中火棘果中的红色素、总黄酮、总酚含量,将其制成保健食品、药用口服溶液剂、口服固体制剂,可起到直接与间接的防近视、防夜盲等症的作用,提高了火棘果的开发利用效率;
(2)提取物中红色素含量以番茄红素计不少于10.0μg/g,总黄酮含量以芦丁计不少于5.0%,总酚含量不少于30.0%,同时以乳糖粉作为辅料并配以薄荷油,改善了保健食品和药品的外观与口感,长期食用可以有效预防近视、夜盲等眼疾。

实施例1
火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,所述的火棘果提取物是由以下方法制得:
S1、挑选色泽饱满、无虫蛀的新鲜火棘果,将所选新鲜窄叶火棘Pyracantha angustifolia果放入水中清洗,洗净与除杂;
S2、组织破碎:将洗净后的新鲜火棘果加入到匀浆机中,并加入新鲜火棘果重量1倍的水,搅拌捣碎3min,得到火棘果液,静置待用;
S3、煎煮:在火棘果浆中加入其重量3倍的水煎煮1.5h,用纱布过滤,得到火棘果液;
S4、酸化处理:在火棘果液中加入2%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到1;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:6的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用2BV的蒸馏水洗脱,后用5BV的70%的乙醇洗脱,收集洗脱液;
S6、醇提:在洗脱液中加入5倍质量分数为95%的乙醇在25℃下冷浸3次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.2的火棘果提取物,其中,提取物中红色素含量以番茄红素计为13.2μg/g,总黄酮含量以芦丁计为5.3%,总酚含量为35.1%。
将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成口服固体制剂。
所述的口服固体制剂口含片由下述重量份的成分组成:火棘果提取物1份,湿润剂重量份的95%乙醇,乳糖粉6份,薄荷油0.03份,滑石粉0.04份,硬脂酸镁0.03份,在上述重量份的乳糖粉中加入湿润剂量的乙醇和上述重量份的火棘果提取物搅拌8min,制成软材,然后用16目筛制粒,湿颗粒在58℃下低温干燥2h,然后用16目筛整粒,再加入上述重量份的薄荷油、滑石粉和硬脂酸镁,混匀,压片,即得。
实施例2
火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,所述的火棘果提取物是由以下方法制得:
S1、挑选色泽饱满、无虫蛀的新鲜火棘果,将所选新鲜窄叶火棘Pyracantha angustifolia果放入水中清洗,除杂;
S2、组织破碎:将洗净后的新鲜火棘果加入到匀浆机中,并加入新鲜火棘果重量1.2倍的水,搅拌捣碎2min,得到火棘果液,静置待用;
S3、煎煮:在火棘果浆中加入其重量4倍的水煎煮2h,用纱布过滤,得到火棘果液;
S4、酸化处理:在火棘果液中加入4%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到2;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:7的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用3BV的蒸馏水洗脱,后用4BV的80%的乙醇洗脱,收集洗脱液;
S6、醇提:在洗脱液中加入3倍质量分数为95%的乙醇在35℃下冷浸3次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.3的火棘果提取物,其中,提取物中红色素含量以番茄红素计为11.8μg/g,总黄酮含量以芦丁计为5.8%,总酚含量为33.4%。
将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成口服固体制剂。
所述的口服固体制剂由下述重量份的成分组成:火棘果提取物1.5份,95%乙醇适量,蔗糖粉5份,薄荷油0.03份,滑石粉0.05份,硬脂酸镁0.01份,在上述重量份的乳糖粉中加入湿润剂量的乙醇和上述重量份的火棘果提取物搅拌5min,制成软材,然后用16目筛制粒,湿颗粒在56℃下低温干燥3h,然后用16目筛整粒,再加入上述重量份的薄荷油、滑石粉和硬脂酸镁,混匀,压片,即得。
实施例3
火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,所述的火棘果提取物是由以下方法制得:
S1、挑选色泽饱满、无虫蛀的新鲜火棘果,将所选新鲜窄叶火棘Pyracantha angustifolia果放入水中清洗,除杂;
S2、组织破碎:将洗净后的新鲜火棘果加入到匀浆机中,并加入新鲜火棘果重量1.1倍的蒸馏水,搅拌捣碎5min,得到火棘果液,静置待用;
S3、煎煮:在火棘果浆中加入其重量5倍的水煎煮3h,用纱布过滤,得到火棘果液;
S4、酸化处理:在火棘果液中加入3%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到2;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:8的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用2BV的蒸馏水洗脱,后用6BV的75%的乙醇洗脱,收集洗脱液;
S6、醇提:在洗脱液中加入4倍质量分数为95%的乙醇在30℃下冷浸4次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.25的火棘果提取物,其中,提取物中红色素含量以番茄红素计为15.6μg/g,总黄酮含量以芦丁计为5.1%;总酚含量为32.4%。
将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成口服固体制剂。
所述的口服固体制剂口含片由下述重量份的成分组成:火棘果提取物1.2份,湿润剂重量份的95%乙醇,乳糖粉7份,薄荷油0.05份,滑石粉0.01份,硬脂酸镁0.05份,在上述重量份的乳糖粉中加入湿润剂量的乙醇和上述重量份的火棘果提取物搅拌10min,制成软材,然后用20目筛制粒,湿颗粒在58℃下低温干燥2.5h,然后用20目筛整粒,再加入上述重量份的薄荷油、滑石粉和硬脂酸镁,混匀,压片,即得。
实施例4
火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,所述的火棘果提取物是由以下方法制得:
S1、挑选色泽饱满、无虫蛀的新鲜火棘果,将所选新鲜火棘Pyracantha fortuneana果放入水中清洗,除杂、洗净;
S2、组织破碎:将洗净后的新鲜火棘果加入到匀浆机中,并加入新鲜火棘果重量1倍的蒸馏水,搅拌捣碎4min,得到火棘果液,静置待用;
S3、煎煮:在火棘果浆中加入其重量3.5倍的水煎煮2h,用纱布过滤,得到火棘果液;
S4、酸化处理:在火棘果液中加入5%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到2;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:7的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用2BV的蒸馏水洗脱,后用6BV的78%的乙醇洗脱,收集洗脱液;
S6、醇提:在洗脱液中加入5倍质量分数为95%的乙醇在32℃下冷浸5次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.2的火棘果提取物,其中,提取物中红色素含量以番茄红素计为12.5μg/g,总黄酮含量以芦丁计为5.2%,总酚含量为34.2%。
将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成口服固体制剂。
所述的口服固体制剂口含片由下述重量份的成分组成:火棘果提取物1.4份,湿润剂重量份的95%乙醇,乳糖粉5.5份,薄荷油0.01份,滑石粉0.03份,硬脂酸镁0.04份,在上述重量份的乳糖粉中加入湿润剂量的乙醇和上述重量份的火棘果提取物搅拌7min,制成软材,然后用16目筛制粒,湿颗粒在60℃下低温干燥4h,然后用18目筛整粒,再加入上述重量份的薄荷油、滑石粉和硬脂酸镁,混匀,压片,即得。
实施例5
火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,所述的火棘果提取物是由以下方法制得:
S1、挑选色泽饱满、无虫蛀的新鲜火棘果,将所选新鲜火棘Pyracantha fortuneana果放入水中清洗,除杂、洗净;
S2、组织破碎:将洗净后的新鲜火棘果加入到匀浆机中,加入新鲜火棘果重量1.2倍的蒸馏水,搅拌捣碎3min,得到火棘果液,静置待用;
S3、煎煮:在火棘果浆中加入其重量4倍的水煎煮2.5h,用纱布过滤,得到火棘果液;
S4、酸化处理:在火棘果液中加入4%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到1;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:6的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用3BV的蒸馏水洗脱,后用4BV的72%的乙醇洗脱,收集洗脱液;
S6、醇提:在洗脱液中加入4.5倍质量分数为95%的乙醇在33℃下冷浸4次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.2的火棘果提取物,其中,提取物中红色素含量以番茄红素计为15.5μg/g,总黄酮含量以芦丁计为5.8%,总酚含量为41.9%。
将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成口服固体制剂。
所述的口服固体制剂由下述重量份的成分组成:火棘果提取物1份,湿润剂重量份的95%乙醇,乳糖粉6.5份,薄荷油0.02份,滑石粉0.02份,硬脂酸镁0.02份,在上述重量份的乳糖粉中加入湿润剂量的乙醇和上述重量份的火棘果提取物搅拌10min,制成软材,然后用20目筛制粒,湿颗粒在55℃下低温干燥3h,然后用20目筛整粒,再加入上述重量份的薄荷油、滑石粉和硬脂酸镁,混匀,压片,即得。
实施例6
火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,所述的火棘果提取物是由以下方法制得:
S1、挑选色泽饱满、无虫蛀的新鲜火棘果,将所选新鲜火棘Pyracantha fortuneana果放入水中清洗,除杂、洗净;

S4、酸化处理:在火棘果液中加入4%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到3;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:8的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用3BV的蒸馏水洗脱,后用5BV的75%的乙醇洗脱,收集洗脱液;
S6、醇提:在洗脱液中加入3.5倍质量分数为95%的乙醇在34℃下冷浸4次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.3的火棘果提取物,其中,提取物中红色素含量以番茄红素计为14.1μg/g,总黄酮含量以芦丁计为5.3%,总酚含量为36.1%。
将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成保健食品。
实施例7
火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,所述的火棘果提取物是由以下方法制得:
S1、挑选色泽饱满、无虫蛀的新鲜火棘果,将所选新鲜火棘Pyracantha fortuneana果放入水中清洗,除杂、洗净;
S2、组织破碎:将洗净后的新鲜火棘果加入到匀浆机中,并加入新鲜火棘果重量1.1倍的蒸馏水,搅拌捣碎4min,得到火棘果液,静置待用;
S3、煎煮:在火棘果浆中加入其重量4.5倍的水煎煮3h,用纱布过滤,得到火棘果液;
S4、酸化处理:在火棘果液中加入3%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到4;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:6的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用2BV的蒸馏水洗脱,后用6BV的78%的乙醇洗脱,收集洗脱液;
S6、醇提:在洗脱液中加入4.5倍质量分数为95%的乙醇在28℃下冷浸5次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.2-1.3的火棘果提取物,其中,提取物中红色素含量以番茄红素计为15.2μg/g,总黄酮含量以芦丁计为5.3%,总酚含量为34.5%。
将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成药用口服溶液剂。

S2、组织破碎:将洗净后的新鲜火棘果加入到匀浆机中,并加入新鲜火棘果重量1.2倍的蒸馏水,搅拌捣碎3min,得到火棘果液,静置待用;
S3、煎煮:在火棘果浆中加入其重量3.5倍的水煎煮2h,用纱布过滤,得到火棘果液;
S4、酸化处理:在火棘果液中加入5%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到2.5;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:7的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用3BV的蒸馏水洗脱,后用5BV的75%的乙醇洗脱,收集洗脱液;
S6、醇提:在洗脱液中加入4倍质量分数为95%的乙醇在30℃下冷浸3次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.2的火棘果提取物,其中,提取物中红色素含量以番茄红素计为12.4μg/g,总黄酮含量以芦丁计为5.5%,总酚含量为33.5%。
将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成药用口服溶液剂。
以上所述仅是本发明的优选实施方式,应当理解本发明并非局限于本文所披露的形式,不应看作是对其他实施例的排除,而可用于各种其他组合、修改和环境,并能够在本文所述构想范围内,通过上述教导或相关领域的技术或知识进行改动。而本领域人员所进行的改动和变化不脱离本发明的精神和范围,则都应在本发明所附权利要求的保护范围内。
Claims (8)
1.火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途。
2.如权利要求1所述的火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,其特征在于,所述的火棘果提取物是由以下方法制得:
S1、净制:挑选色泽饱满、无虫蛀的新鲜火棘果,并除去杂质,将所选火棘果用水漂洗干净;
S2、组织破碎:将净制后的新鲜火棘果加入到匀浆机中,并加入新鲜火棘果重量1-1.2倍的水,搅拌捣碎2-5min,得到火棘果浆;
S3、煎煮:在火棘果浆中加入其重量3-5倍的水煎煮1.5-3h,用纱布过滤,得到火棘果液;
S4、酸化处理:在火棘果液中加入2-5%v/v的盐酸乙醇溶液,直至火棘果液的pH值达到1-4;
S5、大孔吸附树脂柱层析纯化:将酸化后的火棘果液转入径高比为1:6-8的大孔吸附树脂柱层析纯化,收集流出液,保存另用,然后对大孔吸附树脂柱先用2-3BV的蒸馏水洗脱,后用4-6BV的70-80%的乙醇洗脱,收集洗脱液;
S6、醇提:在洗脱液中加入3-5倍质量分数为95%的乙醇在室温冷浸3-5次,提取液滤过,合并滤液;
S7、真空减压浓缩得到相对密度1.2-1.3的火棘果提取物,其中,提取物中红色素含量以番茄红素计不少于10.0μg/g,总黄酮含量以芦丁计不少于5.0%,总酚含量不少于30.0%。
3.根据权利要求2所述的火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,其特征在于:步骤S1所述的火棘果为蔷薇科火棘属Pyracantha植物火棘P.fortuneana、窄叶火棘P.angustifolia的果实。
4.根据权利要求2所述的火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,其特征在于:将步骤S7所述火棘果提取物加入药学上可接受的辅料,按照常规方法制备成制剂。
5.根据权利要求4所述的火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,其特征在于:所述制剂为保健食品、药用口服溶液剂、药用口服固体制剂。
6.根据权利要求5所述的火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,其特征在于,所述口服固体制剂为口含片,该口含片由下述重量份的成分组成:火棘果提取物1-1.5份,湿润剂重量份的95%乙醇,乳糖粉5-7份,薄荷油0.01-0.05份,滑石粉0.01-0.05份,硬脂酸镁0.01-0.05份。
7.根据权利要求6所述的火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,其特征在于,所述的口含片的制备方法为:在上述重量份的乳糖粉中加入湿润剂量的乙醇和上述重量份的火棘果提取物搅拌5-10min,制成软材,然后用16-20目筛制粒,湿颗粒在55-60℃下低温干燥2-4h,然后用16-20目筛整粒,再加入上述重量份的薄荷油、滑石粉和硬脂酸镁,混匀,压片,即得。
8.根据权利要求2所述的火棘果提取物在制备提高视觉敏感度保健食品与药品中的用途,其特征在于,所述的流出液经过减压浓缩,在浓缩液中加入2倍量的乙醇,静置,滤取沉淀,然后将沉淀溶于水中,用Sevage法除去蛋白质后,加入2倍量的乙醇,滤取沉淀,依次用乙醇、无水乙醇、丙酮、乙醚洗涤,除去脂溶性成分和水分,减压低温干燥可得火棘果果胶提取物。
