本发明公开了一种含N‑糖基化壳聚糖的快速成膜流体敷料及其制备与应用,所述敷料由如下原料制成:N‑糖基化壳聚糖、5mg/mL生理盐水、乳酸、聚乙烯醇、去离子水、橄榄油、薄荷脑、质量浓度30%双氧水和体积浓度95%乙醇水溶液;本发明敷料具有止血、抑菌、镇痛、促进伤口愈合等功效;具有良好的透气性和防水性;不受伤口大小、形状、所在部位的限制,成膜有一定弹性,从而涂覆部位活动自如;良好的生物相容性及生物安全性,无刺激性,不产生过敏等症状;制备工艺简单、成本低廉,且携带和使用均方便。

一种含N-糖基化壳聚糖的快速成膜流体敷料及其制备与应用
(一)技术领域
本发明涉及一种生物医用敷料及其制备方法。
(二)背景技术
目前,在医疗领域中,人们还是习惯采用各种创口贴来进行创伤创口的护理及修复,而传统创口贴是处理伤口的首选产品。传统创口贴由一条长形状的胶布和一小块网状编织的脱脂棉构成,尽管它具有价格低廉、使用便捷等优点,传统创口贴还是具有以下缺点:(1)透气性差,易使伤口周围皮肤泛白变皱,不利于伤口愈合;(2)创口贴通过胶布固定在伤口周边,更换时易撕扯触及创面,以及创面肉芽组织易长入脱脂棉的网眼中,更换时创口贴与创面伤口粘连,造成二次损伤并给患者带来疼痛;(3)不能保持创面湿润,延迟创面愈合。
针对传统创口贴使用中存在的缺陷和问题,近年来出现了一种新型的生物流体敷料膜。生物流体敷料膜主要是将促进创面修复的有效成分与可成膜的高分子材料混合,以涂抹或喷雾的方式,在创口上形成一层透气保湿的隔离薄膜,保护创面并形成湿性环境,此类敷料不需要包扎,覆盖伤口无缝隙,适用于任意形状的伤口,也适合人体腔道等难以包扎部位的创口治疗。
壳聚糖是广泛应用于生物医学领域的一种天然高分子材料,具有良好的生物相容性、生物降解性、抗菌性、促凝血和促愈合等特点。目前,以壳聚糖为基体的抗菌敷料已有多种,但壳聚糖在水中的溶解性极差,大大限制了其应用。
(三)发明内容
本发明目的是提供一种成膜速度较快、具有良好生物相容性、抗菌效果好、促进伤口愈合、使用便携、安全可靠的含N-糖基化壳聚糖的流体敷料及其制备方法。
本发明采用的技术方案是:
本发明提供一种含N-糖基化壳聚糖的快速成膜流体敷料,所述敷料由如下原料制成:N-糖基化壳聚糖、5mg/mL生理盐水、乳酸、聚乙烯醇、去离子水、橄榄油、薄荷脑、质量浓度30%双氧水和体积浓度95%乙醇水溶液;所述N-糖基化壳聚糖与聚乙烯醇和薄荷脑质量比为1:2.5-40:0.5-5;所述生理盐水、乳酸、去离子水、橄榄油、双氧水和乙醇水溶液体积用量以N-糖基化壳聚糖质量计分别为5-75ml/g、0.5-1.5ml/g、25-200ml/g、0.5-3ml/g、1-7.5ml/g、20-110ml/g。
进一步,所述N-糖基化壳聚糖与聚乙烯醇和薄荷脑质量比为1:4-20:0.5-4,更优选所述N-糖基化壳聚糖与聚乙烯醇和薄荷脑质量比为1:5-15:0.5-2.5。
进一步,所述生理盐水、乳酸、去离子水、橄榄油、双氧水和乙醇水溶液体积用量以N-糖基化壳聚糖质量计分别为25-75ml/g、0.75-1.5ml/g、50-200ml/g、1-3ml/g、2.5-7.5ml/g、40-90ml/g。更优选所述生理盐水、乳酸、去离子水、橄榄油、双氧水和乙醇水溶液体积用量以N-糖基化壳聚糖质量计分别为25-50ml/g、0.75-1.5ml/g、50-150ml/g、1-2.5ml/g、2.5-5ml/g、40-75ml/g。
本发明还提供一种所述含N-糖基化壳聚糖的快速成膜流体敷料的制备方法,所述方法为:
(1)底液制备:按配方量,称取N-糖基化壳聚糖,溶于5mg/mL生理盐水中,再加入乳酸,搅拌溶解,制得底液;
(2)膜液制备:按配方量,称取聚乙烯醇加入到去离子水中,在95℃下加热搅拌2~3h至完全溶解,即得膜液;
(3)将步骤(2)制得的膜液加入到步骤(1)底液中,混合,再依次加入配方量的橄榄油、薄荷脑、质量浓度30%双氧水、体积浓度95%乙醇水溶液,搅拌混合均匀,即得含N-糖基化壳聚糖的快速成膜流体敷料。
本发明还提供一种所述含N-糖基化壳聚糖的快速成膜流体敷料在制备创伤面愈合药物中的应用。
本发明N-糖基化壳聚糖制备步骤如下:称取壳聚糖,加入体积浓度1%乙酸水溶液,室温下搅拌0.5h,待其充分溶解,再加入4.0壳聚糖氨基当量的果糖,30~35℃下磁力搅拌反应7h后,加入硼氢化钠于室温下还原24h,最后用体积浓度95%乙醇水溶液终止反应,将析出产物用无水乙醇洗涤3~4次,离心得沉淀,40℃真空烘干,最后得到N-糖基化壳聚糖,于干燥器内保存备用;所述乙酸水溶液体积用量以壳聚糖重量计为50mL/g,所述果糖与壳聚糖氨基当量比为4:1,所述硼氢化钠与壳聚糖质量比为1.7:4。
本发明制备方法简单,添加的各组分都具备一定的功效:
N-糖基化壳聚糖,克服了现有技术中壳聚糖水溶性较差的缺点,且选取脱乙酰度在60%-70%之间的N-糖基化壳聚糖,更能促进红细胞凝集,从而起到更佳的止血作用;
生理盐水取代乙酸溶液,作为溶解N-糖基化壳聚糖的溶剂,一方面生理盐水能用于消毒伤口,另一方面生理盐水的pH值接近7,改性后的壳聚糖更容易溶于中性溶液中,加速了壳聚糖的溶解速率;
加入乳酸,可以调节壳聚糖生物成膜胶的pH值,并且酸性条件有利于乳酸菌的繁殖生长,而对大肠杆菌等有害微生物产生抑制作用;
聚乙烯醇,是较为常见的水溶性的高分子成膜材料,成膜之后有一定的强度和柔韧性,对皮肤不会产生刺激,还具有很好的吸湿性,同时也具有一定的防水作用;
橄榄油,可作为分散剂,增加膜的柔韧性和可拉伸性,对伤口具有保湿作用,可防止皮肤皱裂及因皮肤分泌过少引起的瘙痒,可减轻伤口疼痛,促进愈合,愈后不留下疤痕。美国、德国等西方国家早已把橄榄油载入药典;
薄荷脑,常作为赋香剂使用,具有清凉感,同时可消炎镇痛止痒,减轻浮肿,添加在敷料中易于被患者接受;
30%双氧水,常用于医用消毒杀菌,其强氧化性可以破坏细菌的蛋白质,使之死亡,另一方面又可以作为致孔剂,双氧水歧化释放氧气而使抗菌膜致孔透气,加上歧化反应会放热,能加速抗菌膜固化速度。通常医用上使用3%双氧水,本实验采用的双氧水微量,且有添加其他试剂稀释,最终得到的双氧水浓度不会对皮肤造成刺激伤害;
乙醇溶剂,主要利用乙醇的迅速挥发特性,使本发明产品能够在创面上快速成膜,有效保护创面。
本发明提供的一种含N-糖基化壳聚糖的快速成膜流体敷料,它采用的是改性之后的N-糖基化壳聚糖,大大提高了壳聚糖的水溶性,此敷料主要由底液和膜液组成,底液为N-糖基化壳聚糖的生理盐水溶液,其中N-糖基化壳聚糖在底液中的浓度为10~50mg/mL,所述膜液为质量分数10%的聚乙烯醇溶液。
与现有的伤口敷料相比,本发明的有益效果主要体现在:
(1)具有止血、抑菌、镇痛、促进伤口愈合等功效;
(2)具有良好的透气性和防水性;
(3)不受伤口大小、形状、所在部位的限制,成膜有一定弹性,从而涂覆部位活动自如;
(4)良好的生物相容性及生物安全性,无刺激性,不产生过敏等症状;
(5)制备工艺简单、成本低廉,且携带和使用均方便。

N-糖基化壳聚糖的制备:称取壳聚糖4.0g,加入体积浓度1%乙酸水溶液200mL,室温(25-30℃)下搅拌0.5h,待其充分溶解,再加入4.0壳聚糖氨基当量的果糖,30~35℃下磁力搅拌反应7h后,加入1.7g硼氢化钠于室温下还原24h,最后用体积浓度95%乙醇水溶液终止反应,将析出产物用无水乙醇洗涤3~4次,离心得沉淀,40℃真空烘干,获得N-糖基化壳聚糖7.5g,于干燥器内保存备用。
实施例2
含N-糖基化壳聚糖的快速成膜流体敷料,由如下配比的原料制成:
其制备的具体步骤是:
(1)底液制备:称取N-糖基化壳聚糖0.2g,溶于5mL的5mg/mL生理盐水中,再加入0.15mL乳酸,搅拌溶解,制得浓度为40mg/mL的底液;
(2)膜液制备:称取聚乙烯醇(聚合度1750±50)1g,加入到10mL去离子水中,在95℃下加热搅拌2~3h至完全溶解,即得质量浓度10%聚乙烯醇膜液;
(3)将步骤(2)制得的膜液加入到步骤(1)底液中,两者混合均匀,再依次加入橄榄油0.2mL、薄荷脑0.1g、质量浓度30%双氧水0.5mL、体积浓度95%乙醇水溶液8mL,搅拌速度为500r/min,搅拌混合均匀即得含N-糖基化壳聚糖的快速成膜流体敷料22.5mL。
实施例3
含N-糖基化壳聚糖的快速成膜流体敷料,其原料组成和配比为:
其制备方法是:
(1)底液制备:称取N-糖基化壳聚糖0.2g,溶于5mL的5mg/mL生理盐水中,再加入0.2mL乳酸,搅拌溶解,制得浓度为40mg/mL的底液;
(2)膜液制备:称取聚乙烯醇1g,加入到10mL去离子水中,在95℃下加热搅拌2~3h至完全溶解,即得质量浓度10%聚乙烯醇膜液;
(3)将步骤(2)制得的膜液加入到步骤(1)底液中,两者混合均匀,依次加入橄榄油0.3mL、薄荷脑0.5g、质量浓度30%双氧水1mL、体积浓度95%乙醇水溶液10mL,搅拌速度为500r/min,搅拌混合均匀即得含N-糖基化壳聚糖的快速成膜流体敷料25.4mL。
实施例4
含N-糖基化壳聚糖的快速成膜流体敷料,其原料组成和配比为:
其制备方法是:
(1)底液制备:称取N-糖基化壳聚糖0.2g,溶于10mL的5mg/mL生理盐水中,再加入0.2mL乳酸,搅拌溶解,制得浓度为20mg/mL的底液;
(2)膜液制备:称取聚乙烯醇2g,加入到20mL去离子水中,在95℃下加热搅拌2~3h至完全溶解,即得质量浓度10%聚乙烯醇膜液;
(3)将步骤(2)制得的膜液加入到步骤(1)底液中,两者混合均匀,依次加入橄榄油0.4mL、薄荷脑0.5g、质量浓度30%双氧水0.9mL、体积浓度95%乙醇水溶液15mL,搅拌速度为500r/min,搅拌混合均匀即得含N-糖基化壳聚糖的快速成膜流体敷料54.6mL。
实施例5
含N-糖基化壳聚糖的快速成膜流体敷料,其原料组成和配比为:
其制备方法是:
(1)底液制备:称取N-糖基化壳聚糖0.2g,溶于10mL的5mg/mL生理盐水中,再加入0.2mL乳酸,搅拌溶解,制得浓度为20mg/mL的底液;
(2)膜液制备:称取聚乙烯醇3g,加入到30mL去离子水中,在95℃下加热搅拌2~3h至完全溶解,即得质量浓度10%聚乙烯醇膜液;
(3)将步骤(2)制得的膜液加入到步骤(1)底液中,两者混合均匀,依次加入橄榄油0.5mL、薄荷脑0.4g、质量浓度30%双氧水1mL、体积浓度95%乙醇水溶液12mL,搅拌速度为500r/min,搅拌混合均匀即得。
实施例6
对实施例2~5得到的含N-糖基化壳聚糖的快速成膜流体敷料分别进行了性能和药理测试,实验结果如下:
测试1:产品性能参数检测
对本发明制备的含N-糖基化壳聚糖的快速成膜流体敷料性能参数进行检测,实施例2~5的产品均符合以下要求:
(1)性状:为乳白色的半流动性液体,质地均匀。
(2)pH:取实施例2~5的样品各少许,用pH计测得pH都在4.0~6.0之间。
(3)成膜时间:在室温25±1℃,相对湿度60%~70%的条件下,取实施例2~5的含N-糖基化壳聚糖的快速成膜流体敷料,将其涂抹在干燥的平板玻璃上,待其自然干燥,观察可在4~7min之间成膜。
测试2:流体敷料样品抑菌效果
按《药品微生物检验手册》第18章第1节管碟法进行试验,测定结果参见表1:

从表1的结果显示,本发明的产品均有明显抑菌圈,说明对大肠杆菌、金色葡萄球菌、白色念珠菌、铜绿假单胞菌的生长均有明显的抑制作用。
测试3:流体敷料样品皮肤刺激指数(PII)测定
取家兔6只(购于浙江中医药大学动物实验中心),于背部脊柱两侧将脱毛剂均匀涂上,设置5个试验区域,每个区域面积约为1cm×1cm,洗净脱毛剂,归笼观察24h。24h之后,在脱毛区1~4号分别涂抹本发明实施例2~5生产的流体敷料,分别涂抹0.1mL,5号脱毛区不作处理,作为自身对照。持续24h后除去药物,肉眼观察用药皮肤局部有无红斑及水肿、色素沉着、出血点、皮肤粗糙或皮肤菲薄等,结束后用温水清洗给药部位。皮肤刺激试验反应评分和刺激强度评价见表2、表3。

从表4结果表明,本发明制备的液体创伤敷料均无皮肤致敏反应。
测试4:创伤伤口愈合的对比试验
供试品:实施例2~5制备的含N-糖基化壳聚糖的快速成膜流体敷料
对照品:云南白药止血粉
实验动物采用普通家兔(购于浙江中医药大学动物实验中心),10%水合氯醛麻醉后用4%硫化钠脱毛,取俯卧位。常规消毒皮肤后,于每只兔子距脊柱两侧1.0cm处切割长1.5cm,宽1.0cm的伤口4个,深度从皮肤到肌层,伤口的厚度为0.3cm。将4个伤口分别标记为A、B、C、D号,用生理盐水冲洗伤口,备用。A号伤口以0.9%生理盐水上药,每次0.1mL,为阴性对照;B号伤口以云南白药止血粉为阳性对照,每次取药0.1g,均匀涂抹至完全覆盖创口,每12h更换一次;C和D号伤口依次使用本发明的实施例2~5流体敷料(每只兔子伤口上均同时设置两组实验组,即C和D),每次0.1mL,均匀涂抹至完全覆盖创口,每12h更换一次,连续用药15天(“天”以“d”表示)。每个实施组实验动物8只,雌雄各半,体量约为2.0-2.5kg。观察伤口愈合情况,记录伤口愈合时间,记录时间点分别为3d、6d、9d、12d、15d,同时计算创面愈合率。
计算创面愈合率的方法如下:先将创面标记在透明膜上,再以此为模板将质地均匀的硬纸片剪成同样大小,然后用天平称重,以硬纸片重量间接地表示创面大小。按下试计算创面愈合率:
创面愈合率(%)=(原始创面面积-未愈合创面面积)/原始创面面积。
表5各组别伤口愈合率比较
由表5所知,使用本发明的流体敷料后,各创面平均愈合率均高于阴对照组和阳性对照组,且15天时创伤平均愈合率为99%以上,高于阴性对照组20%以上,高于阳性对照组10%以上,说明本发明的含N-糖基化壳聚糖的快速成膜流体敷料促进伤口愈合作用非常好。同时,观察各组伤口痴皮自然脱落后,形成的疤痕有较大差别:阴性对照组形成疤痕凸出皮肤明显,用手触摸可感受到硬结;阳性对照组形成明显疤痕,中间皮肤略微凸出;各实施组形成疤痕凸出皮肤不明显,用手触摸无硬结,且新生皮肤与原皮肤接近,较粉嫩。说明本发明的含N-糖基化壳聚糖的快速成膜流体敷料还可以抑制伤口组织疤痕的异常增生,具有抑制瘫痕形成的作用。
上述实施例为本发明较佳的实现方案,除此之外,本发明还可以其它方式实现,在不脱离本发明构思的前提下任何显而易见的替换,均在本发明的保护范围之内。

Claims (8)

1.一种含N-糖基化壳聚糖的快速成膜流体敷料,其特征在于所述敷料由如下原料制成:N-糖基化壳聚糖、5mg/mL生理盐水、乳酸、聚乙烯醇、去离子水、橄榄油、薄荷脑、质量浓度30%双氧水和体积浓度95%乙醇水溶液;所述N-糖基化壳聚糖与聚乙烯醇和薄荷脑质量比为1:2.5-40:0.5-5;所述生理盐水、乳酸、去离子水、橄榄油、双氧水和乙醇水溶液体积用量以N-糖基化壳聚糖质量计分别为5-75ml/g、0.5-1.5ml/g、25-200ml/g、0.5-3ml/g、1-7.5ml/g、20-110ml/g。
2.如权利要求1所述含N-糖基化壳聚糖的快速成膜流体敷料,其特征在于所述N-糖基化壳聚糖与聚乙烯醇和薄荷脑质量比为1:4-20:0.5-4。
3.如权利要求1所述含N-糖基化壳聚糖的快速成膜流体敷料,其特征在于所述N-糖基化壳聚糖与聚乙烯醇和薄荷脑质量比为1:5-15:0.5-2.5。
4.如权利要求1所述含N-糖基化壳聚糖的快速成膜流体敷料,其特征在于所述生理盐水、乳酸、去离子水、橄榄油、双氧水和乙醇水溶液体积用量以N-糖基化壳聚糖质量计分别为25-75ml/g、0.75-1.5ml/g、50-200ml/g、1-3ml/g、2.5-7.5ml/g、40-90ml/g。
5.如权利要求1所述含N-糖基化壳聚糖的快速成膜流体敷料,其特征在于所述生理盐水、乳酸、去离子水、橄榄油、双氧水和乙醇水溶液体积用量以N-糖基化壳聚糖质量计分别为25-50ml/g、0.75-1.5ml/g、50-150ml/g、1-2.5ml/g、2.5-5ml/g、40-75ml/g。
6.如权利要求1-5之一所述含N-糖基化壳聚糖的快速成膜流体敷料,其特征在于所述N-糖基化壳聚糖制备步骤如下:取壳聚糖,加入体积浓度1%乙酸水溶液,室温下搅拌0.5h,待其充分溶解,再加入4.0壳聚糖氨基当量的果糖,30~35℃下磁力搅拌反应7h后,加入硼氢化钠于室温下还原24h,最后用体积浓度95%乙醇水溶液终止反应,将析出产物用无水乙醇洗涤3~4次,离心得沉淀,40℃真空烘干,得到N-糖基化壳聚糖;所述乙酸水溶液体积用量以壳聚糖重量计为50mL/g,所述果糖与壳聚糖氨基当量比为4:1,所述硼氢化钠与壳聚糖质量比为1.7:4。
7.一种权利要求1所述含N-糖基化壳聚糖的快速成膜流体敷料的制备方法,其特征在于所述方法为:
(1)底液制备:按配方量,称取N-糖基化壳聚糖,溶于5mg/mL生理盐水中,再加入乳酸,搅拌溶解,制得底液;
(2)膜液制备:按配方量,称取聚乙烯醇加入到去离子水中,在95℃下加热搅拌2~3h至完全溶解,即得膜液;
(3)将步骤(2)制得的膜液加入到步骤(1)底液中,混合,再依次加入配方量的橄榄油、薄荷脑、质量浓度30%双氧水、体积浓度95%乙醇水溶液,搅拌混合均匀,即得含N-糖基化壳聚糖的快速成膜流体敷料。
8.一种权利要求1所述含N-糖基化壳聚糖的快速成膜流体敷料在制备创伤面愈合药物中的应用。
