本发明公开一种辣木叶有效成分的提取方法,通过辣木叶总提取物的制备、辣木叶抑菌活性提取物的制备、辣木叶抑菌化活性提取物的制备、辣木叶美白活性提取物的制备、辣木叶多糖提取物的制备。本发明通过一次提取,同时得到不同极性的辣木叶活性提取物,并通过生物活性测定,可应用于不同的食品、保健品和日化领域,将辣木叶的功效成分进行分离,获得高活性、高含量的辣木叶生物活性提取物。

技术领域
本发明涉一种辣木叶有效成分的提取方法,属天然产物提取技术领域。
背景技术
2012年辣木已被中国卫生部批准为中国新资源食品。辣木叶被报道的成分以黄酮类化合物为主,是目前辣木叶开发中研究最多的化合物,其主要活性表现为抗氧化、降血糖、保护肝脏等。如今全球糖尿病患者数量猛增的形势下,对该病的天然防治药物需求大大增加,而辣木叶黄酮正因其具有降血糖效果的同时,营养成分丰富、副作用小、改善糖尿病并发症等优势,且辣木叶黄酮原料具备采集方便、资源丰富,提取工艺简单等特点,使辣木叶黄酮具有了成为防治糖尿病药物的潜力,这可能是辣木叶的进一步发展方向。
糖尿病是一种以高血糖为特征的代谢性疾病。由于经济高速发展和工业化
进程的加速,生活方式的改变和老龄化进程的加速,使糖尿病患病率快速上升。
糖尿病己成为迫害人类健康的慢性非传染性疾病,其危害仅次于继心脑血管疾病
和肿瘤。据国际糖尿病联盟统计,2011年全球糖尿病患者人数己达3.7亿,其中80%在发展中国家,估计到2030年全球将有近5.5亿糖尿病患者。
在印度传统的医学体系以及西非地区,当地人用辣木树籽油以及辣木叶治疗糖尿病已经很多年。全球排名前五的生物医药技术研究所——古巴芬莱研究院,对辣木治理糖尿病的功效进行过深入研究。实验将糖尿病者共分为两组,每组165人,每天为其中一组患者提供辣木茶和辣木叶粉。据两年的数据统计,服用辣木的患者血糖明显稳定并有所降低,减少了对其他药物的使用,对治疗糖尿病有明显的效果。
100克印度传统辣木的新鲜叶片中的维他命E含量约为9毫克,干燥叶片中的含量约为16.2毫克。根据计算,只要三汤匙(约25克)的辣木叶干粉就含有幼儿每日所需270%的维生素A,42%的蛋白质,125%的钙,70%的铁及22%的维生素C。对怀孕和哺乳中的女性而言,辣木叶片和豆荚亦可帮助本人及胎儿或婴儿维持健康,供给大量的铁质、蛋白质、铜、硫和维他命B。不过,由于辣木本身含有植物碱,高血压、糖尿病患者及孕妇等最好经医师指示后酌量食用。
辣木叶片、果荚富含多种矿物质、维生素、20种氨基酸、46种抗氧素和36种自然防炎体和矿物质。每100克的辣木中含有的维生素C是柑橘的7倍,铁是菠菜的3倍,维生素A是胡萝卜的4倍,钙质是牛奶的4倍,钾是香蕉的3倍,蛋白质是酸奶的2倍。
作为蔬菜和食品有增进营养,食疗保健的功能;也广泛应用于医药、保健等方面,被誉为“生命之树”,“植物中的钻石”。如今随着科学的开发和利用、辣木叶粉、辣木叶胶囊、辣木叶茶、以及辣木叶精片为常见形态、其中以辣木叶破壁提取技术制成的精片功效最好最完美。
辣木叶粉有降血糖、降脂、降压、抗肿瘤、抗氧化、通便、利尿、驱虫、改善睡眠等功效,长期食用可增强人体免疫功能,延缓衰老、预防疾病。印度草医学认为辣木可预防300种疾病。特别是近几年辣木受到欧美等发达国家很多研究机构和消费者的关注和热捧。
经常食用辣木叶可增强免疫力、排毒、塑身、抗老化、抗癌、并对多种慢性及重大疾病都有极大的改善功效。
辣木叶是纯天然绿色食品,含有人体所需的全部营养物质,它可取代复合维生素、钙片、鱼肝油等。特别对高血压、高血脂、糖尿病、痛风等有很好的效果。辣木叶可协助改善、预防疾病,改善睡眠、增强记忆力、延缓衰老,还可用来治疗肝脏、脾脏、经络等特殊部位的疾病。它还具有治疗口臭和醒酒等作用。
因此,有必要对辣木叶的化学成分进行提取分离,以期获得具有生物活性的单体化合物。
发明内容
为解决上述问题,本发明提供一种辣木叶有效成分的提取方法,以期获得具有不同生物活性的辣木叶有效成分加以进一步的开发和利用。
本发明通过下列技术方案实现:一种辣木叶有效成分的提取方法,包括以下步骤:
步骤一,辣木叶总提取物的制备:
采集新鲜的辣木叶,在40~50℃下干燥2~3h,粉碎至通过20目筛,采用浓度为50~70v/v%的乙醇,按液料比L/L计为20:1,在室温(20~25℃)下浸泡24~48h,再过滤,所得滤渣再次加入浓度为50~70v/v%的乙醇在室温(20~25℃)下浸泡3~5次,合并滤液,滤液经50~60℃的减压浓缩,得到辣木叶总浸膏,相对密度为0.9~1.2g/L;
步骤二,辣木叶抑菌活性提取物的制备:
将步骤一所得辣木叶总浸膏,加入5~6倍体积的蒸馏水中,全部溶解后,加入等体积石油醚进行萃取4~6h,萃取3~5次,收集水层,合并石油醚层萃取液,经50~60℃的减压浓缩,得到辣木叶石油醚层提取物;通过抑菌活性测定表明,辣木叶石油醚层提取物具有较强的抑菌活性,对各种食品、环境中常见细菌如大肠杆菌、伤寒菌和金黄色葡萄球菌均有较强的抑菌活性,即视为辣木叶抑菌活性提取物。
步骤三,辣木叶抑菌化活性提取物的制备:
将步骤二收集的水层用等体积倍量的二氯甲烷进行萃取4~6h,萃取3~5次,收集水层,合并二氯甲烷层萃取液,经50~60℃的减压浓缩,得到辣木叶二氯甲烷层提取物;通过对提取物DPPH自由基清除试验表明,辣木叶石油醚层具有抗氧化活性,并且随着质量浓度的增大,二氯甲烷层对DPPH自由基的清除能力逐渐增强。在浓度为600ug/ml时趋于平缓,清除率达91.1%。因此,辣木叶抗氧化活性提取物即辣木叶二氯甲烷层提取物。
步骤四,辣木叶美白活性提取物的制备:
将步骤三收集的水层,继续用等体积倍量的乙酸乙酯进行萃取4~6h,萃取3~5次,收集水层,合并乙酸乙酯层萃取液,经50~60℃的减压浓缩,得到辣木叶乙酸乙酯层提取物;通过抗酪氨酸酶活性测定,以曲酸为阳性对照组,与辣木叶乙酸乙酯层提取物对酪氨酸酶抑制率相比可知,相同浓度下,辣木叶乙酸乙酯层提取物的IP%为72.8%,低于曲酸对酪氨酸酶的抑制率,但较辣木叶石油醚层提取物较高,因此,认为辣木叶美白活性提取物为辣木叶乙酸乙酯层提取物。
步骤五,辣木叶多糖提取物的制备:
将步骤四收集的水层,经70~80℃的减压浓缩至相对密度为0.5~1.0g/L,通过大孔吸附树脂(D101、AB-8、XAD16中的一种),采用蒸馏水装柱,上样量为柱体积的1/3,流速4~6BV/h,采用3~5倍柱体积的蒸馏水洗脱,再用浓度为30v/v%的乙醇洗脱3~5倍柱体积,收集蒸馏水和乙醇洗脱液,经50~60℃的减压浓缩,得到辣木叶多糖提取物。
所述步骤五的大孔吸附树脂是型号为D101、AB-8或XAD16的大孔吸附树脂。
所得辣木叶多糖提取物在制备化妆品或保健品中的应用。
本发明对辣木叶的化学成分进行分析分离,以期获得具有生物活性的提取物。本发明用活性追踪法对辣木叶有效部位进行生物活性研究。将辣木叶原材料采用50~70%的乙醇冷浸提取,得到初提物,再将初提物用不同极性的溶剂进行萃取得到辣木叶石油醚提取物、二氯甲烷提取物、乙酸乙酯提取物和辣木叶多糖提取物,并对各提取物进行了生物活性测定,包括抗菌活性、抗氧化活性和酪氨酸酶抑制活性等。本发明的主要目的是对辣木叶中的主要化学成分进行研究,并进行生物活性测定,以期获得具有不同生物活性的辣木叶有效成分加以进一步的开发和利用。
本发明的主要特点在于通过一次提取,同时得到不同极性的辣木叶活性提取物,并通过生物活性测定,可应用于不同的食品、保健品和日化领域,将辣木叶的功效成分进行分离,获得高活性、高含量的辣木叶生物活性提取物。
本发明与其他关于辣木的专利文献相比,通过植物化学科学方法,活性追踪技术手段,直接获得针对不同功效的辣木叶提取物,而不是简单将辣木叶粉碎直接作为添加剂而使用。2015年公开的“一种辣木叶茶的快速加工方法”(CN103234722A)是将辣木叶直接粉碎,添加辅料制备成茶叶饮用,并未将辣木叶活性物质进行分离,加上辣木叶含有大量的草酸和蛋白质,服用过量会导致腹泻,损伤脾胃,因此,辣木叶的活性成分应该进行分离纯化,再分别应用。
与现有技术相比,本发明的有益效果在于:
(1)本发明中采用50~70%乙醇提取辣木叶活性成分,不会提取出过量的辣木叶蛋白和草酸。由于辣木叶蛋白质含量高达13.20mg/g,过多摄入蛋白质会引起身体肥胖,辣木叶所含的草酸对于容易结石的体质并不是好的保健品,还容易引起腹泻,因此,本发明采用的70%乙醇提取辣木叶总提取物,能最大程度保留辣木叶活性成分,而排出其他干扰成分。
(2)本发明采用活性追踪法,比较不同极性辣木叶提取物的生物活性,分别获得辣木叶抗氧化活性提取物、辣木叶抑菌活性提取物、辣木叶美白活性提取物和辣木叶总多糖,2015年公布的发明专利“一种辣木叶总黄酮的提取方法及其用途”CN2015101573236,简单采用超声波辅助提取辣木叶总黄酮,获得的是含有黄酮的辣木叶总提取物,和本发明步骤一类似,并未对其有效成分进行分离纯化,因此有效成分含量过低,导致活性不显著。而本发明通过植物化学手段,根据化学成分的极性不同,对辣木叶总提取物进一步进行分离纯化,收集相同极性的同类物质,以期获得较高的生物活性,活性测定结果表明,辣木叶不同极性提取物的生物活性差异很大,考虑到辣木叶提取物的高活性和高附加值,因此通过本发明的方法,实现对辣木叶有效成分分开利用。
(3)本发明采用适合工业化生产的提取方法对辣木叶生物活性进行分步提取,并未采用复杂不适合工业化大规模生产的操作程序,目的是为了合理有效开发辣木叶资源,方便大生产应用,但依然有别于其他提取方法。提取物厂最常使用的提取方法为水提获得植物总提取物,再使用水提取醇沉淀,获得纯化产物,但本发明与常规提取方法不同之处在于,将冷浸法、分步萃取法和大孔吸附树脂法同时应用于一个体系中,同时依次萃取,直接获得目标产物,生物活性测定结果显示,不同目标产物具有不同的生理活性。

实施例1
步骤一,辣木叶总提取物的制备:
采集新鲜的辣木叶,在45℃下干燥2.5h,粉碎至通过20目筛,采用浓度为60v/v%的乙醇,按液料比L/L计为20:1,在室温(20~25℃)下浸泡36h,再过滤,所得滤渣再次加入浓度为60v/v%的乙醇在室温(20~25℃)下浸泡4次,合并滤液,滤液经55℃的减压浓缩,得到辣木叶总浸膏,相对密度为0.9~1.2g/L;
步骤二,辣木叶抑菌活性提取物的制备:
将步骤一所得辣木叶总浸膏,加入5倍体积的蒸馏水中,全部溶解后,加入等体积石油醚进行萃取5h,萃取4次,收集水层,合并石油醚层萃取液,经55℃的减压浓缩,得到辣木叶石油醚层提取物;通过抑菌活性测定表明,辣木叶石油醚层提取物具有较强的抑菌活性,对各种食品、环境中常见细菌如大肠杆菌、伤寒菌和金黄色葡萄球菌均有较强的抑菌活性,即视为辣木叶抑菌活性提取物。
步骤三,辣木叶抑菌化活性提取物的制备:
将步骤二收集的水层用等体积倍量的二氯甲烷进行萃取5h,萃取4次,收集水层,合并二氯甲烷层萃取液,经55℃的减压浓缩,得到辣木叶二氯甲烷层提取物;通过对提取物DPPH自由基清除试验表明,辣木叶石油醚层具有抗氧化活性,并且随着质量浓度的增大,二氯甲烷层对DPPH自由基的清除能力逐渐增强。在浓度为600ug/ml时趋于平缓,清除率达91.1%。因此,辣木叶抗氧化活性提取物即辣木叶二氯甲烷层提取物。
步骤四,辣木叶美白活性提取物的制备:
将步骤三收集的水层,继续用等体积倍量的乙酸乙酯进行萃取5h,萃取4次,收集水层,合并乙酸乙酯层萃取液,经55℃的减压浓缩,得到辣木叶乙酸乙酯层提取物;通过抗酪氨酸酶活性测定,以曲酸为阳性对照组,与辣木叶乙酸乙酯层提取物对酪氨酸酶抑制率相比可知,相同浓度下,辣木叶乙酸乙酯层提取物的IP%为72.8%,低于曲酸对酪氨酸酶的抑制率,但较辣木叶石油醚层提取物较高,因此,认为辣木叶美白活性提取物为辣木叶乙酸乙酯层提取物。
步骤五,辣木叶多糖提取物的制备:
将步骤四收集的水层,经75℃的减压浓缩至相对密度为0.5~1.0g/L,通过大孔吸附树脂(D101),采用蒸馏水装柱,上样量为柱体积的1/3,流速5BV/h,采用4倍柱体积的蒸馏水洗脱,再用浓度为30v/v%的乙醇洗脱4倍柱体积,收集蒸馏水和乙醇洗脱液,经55℃的减压浓缩,得到辣木叶多糖提取物。
实施例2
步骤一,辣木叶总提取物的制备:
采集新鲜的辣木叶,在40℃下干燥3h,粉碎至通过20目筛,采用浓度为50v/v%的乙醇,按液料比L/L计为20:1,在室温(20~25℃)下浸泡48h,再过滤,所得滤渣再次加入浓度为50v/v%的乙醇在室温(20~25℃)下浸泡5次,合并滤液,滤液经50℃的减压浓缩,得到辣木叶总浸膏,相对密度为0.9~1.2g/L;
步骤二,辣木叶抑菌活性提取物的制备:
将步骤一所得辣木叶总浸膏,加入5倍体积的蒸馏水中,全部溶解后,加入等体积石油醚进行萃取6h,萃取3次,收集水层,合并石油醚层萃取液,经50℃的减压浓缩,得到辣木叶石油醚层提取物;通过抑菌活性测定表明,辣木叶石油醚层提取物具有较强的抑菌活性,对各种食品、环境中常见细菌如大肠杆菌、伤寒菌和金黄色葡萄球菌均有较强的抑菌活性,即视为辣木叶抑菌活性提取物。

实施例3
步骤一,辣木叶总提取物的制备:
采集新鲜的辣木叶,在50℃下干燥2h,粉碎至通过20目筛,采用浓度为70v/v%的乙醇,按液料比L/L计为20:1,在室温(20~25℃)下浸泡24h,再过滤,所得滤渣再次加入浓度为70v/v%的乙醇在室温(20~25℃)下浸泡3次,合并滤液,滤液经60℃的减压浓缩,得到辣木叶总浸膏,相对密度为0.9~1.2g/L;
步骤二,辣木叶抑菌活性提取物的制备:
将步骤一所得辣木叶总浸膏,加入6倍体积的蒸馏水中,全部溶解后,加入等体积石油醚进行萃取4h,萃取3次,收集水层,合并石油醚层萃取液,经60℃的减压浓缩,得到辣木叶石油醚层提取物;通过抑菌活性测定表明,辣木叶石油醚层提取物具有较强的抑菌活性,对各种食品、环境中常见细菌如大肠杆菌、伤寒菌和金黄色葡萄球菌均有较强的抑菌活性,即视为辣木叶抑菌活性提取物。

将步骤四收集的水层,经80℃的减压浓缩至相对密度为0.5~1.0g/L,通过大孔吸附树脂(XAD16),采用蒸馏水装柱,上样量为柱体积的1/3,流速6BV/h,采用5倍柱体积的蒸馏水洗脱,再用浓度为30v/v%的乙醇洗脱5倍柱体积,收集蒸馏水和乙醇洗脱液,经60℃的减压浓缩,得到辣木叶多糖提取物。
将上述所得产物进行下列测定:

应用实例:
将上述实施例所得辣木叶抑菌活性提取物应用于制备辣木保湿洁面乳:分别称取氨基酸起泡剂30mL,1%羧甲基纤维素钠水溶液30mL,甘油10mL,依次溶解于去离子水30mL中,共同组成100mL溶液,混合均匀后,加入尼泊金乙酯0.5g,辣木叶抑菌活性提取物5滴,转移至洗面奶瓶中,密封保存。
将上述实施例所得辣木叶抗氧化活性提取物应用于制备保湿化妆水:分别称取质量百分比为0.5%的辣木叶抗氧化活性提取物,1%鳄梨油,0.5%燕麦葡聚糖,5%丙二醇,进行混合在50℃水浴中加热溶解形成溶液A,再分别称取质量百分比为10%丙三醇,1%透明质酸钠,1 %海藻酸钠,1%尼泊金乙酯,80%去离子水,进行混匀,形成溶液B,在70℃水浴加热条件下,将A相全部转移到B相中,边加边搅拌,直至得到一种均匀透明的溶液,根据需要,可添加植物精油或不添加,分装,密封保存。
将上述实施例所得辣木叶美白活性提取物应用于制备辣木美白精华:分别称取质量百分比1%辣木叶美白活性提取物,1%玻尿酸原液,5%卡波姆,1%海藻酸钠,5%甘油,1%尼泊金乙酯,依次加入86%去离子水中,边加边搅拌,直至成为透明均匀的胶体溶液,分装,密封保存。
将上述实施例所得辣木叶多糖提取物应用于制备辣木压片糖果:称取质量百分比为10%的辣木叶多糖提取物,30%乳糖,30%麦芽糊精,20%糖粉,5%硬脂酸镁,2%甘露醇,3%柠檬酸,依次进行混合,将混合后的粉末放入压片机中,一次压制成型,包装,得到辣木叶压片糖果。
Claims (3)
1.一种辣木叶有效成分的提取方法,其特征在于包括以下步骤:
步骤一,辣木叶总提取物的制备:
采集新鲜的辣木叶,在40~50℃下干燥2~3h,粉碎至通过20目筛,采用浓度为50~70v/v%的乙醇,按液料比L/L计为20:1,在室温下浸泡24~48h,再过滤,所得滤渣再次加入浓度为50~70v/v%的乙醇在室温下浸泡3~5次,合并滤液,滤液经50~60℃的减压浓缩,得到辣木叶总浸膏;
步骤二,辣木叶抑菌活性提取物的制备:
将步骤一所得辣木叶总浸膏,加入5~6倍体积的蒸馏水中,全部溶解后,加入等体积石油醚进行萃取4~6h,萃取3~5次,收集水层,合并石油醚层萃取液,经50~60℃的减压浓缩,得到辣木叶石油醚层提取物;
步骤三,辣木叶抑菌化活性提取物的制备:
将步骤二收集的水层用等体积倍量的二氯甲烷进行萃取4~6h,萃取3~5次,收集水层,合并二氯甲烷层萃取液,经50~60℃的减压浓缩,得到辣木叶二氯甲烷层提取物;
步骤四,辣木叶美白活性提取物的制备:
将步骤三收集的水层,继续用等体积倍量的乙酸乙酯进行萃取4~6h,萃取3~5次,收集水层,合并乙酸乙酯层萃取液,经50~60℃的减压浓缩,得到辣木叶乙酸乙酯层提取物;
步骤五,辣木叶多糖提取物的制备:
将步骤四收集的水层,经70~80℃的减压浓缩至相对密度为0.5~1.0g/L,通过大孔吸附树脂,采用蒸馏水装柱,上样量为柱体积的1/3,流速4~6BV/h,采用3~5倍柱体积的蒸馏水洗脱,再用浓度为30v/v%的乙醇洗脱3~5倍柱体积,收集蒸馏水和乙醇洗脱液,经50~60℃的减压浓缩,得到辣木叶多糖提取物。
2.根据权利要求1所述的辣木叶有效成分的提取方法,其特征在于:所述步骤五的大孔吸附树脂是型号为D101、AB-8或XAD16的大孔吸附树脂。
3.权利要求1或2的辣木叶有效成分的提取方法,其所得辣木叶多糖提取物在制备化妆品或保健品中的应用。
