Abstract

本发明公开了一种含紫草素的创可贴及其制备方法,创可贴由平布胶布、吸水垫和离型纸组成,紫草素混合物均匀涂布于吸水垫上,紫草素混合物具体由提纯分离于紫草中的紫草素用γ‑环糊精包埋处理制得,本发明制备的创可贴除了具备一般创可贴的功能外,还具备紫草素特有的药理作用,增强了抗菌、消炎、止痛、促进伤口愈合、减少疤痕形成等作用。且制备方法操作简单,对人体无刺激性、无明显毒性,可广泛应用于小伤口的处理。

Description

一种含紫草素的创可贴及其制备方法
技术领域
本发明涉及创口医用敷料领域,尤其涉及一种含紫草素的创可贴及其制备方法。
背景技术
创可贴,是人们生活中最常用的一种外科用药。创可贴,又名“止血膏药”,具有止血,护创作用。它是由一条长形的胶布,中间附以一小块浸过药物的纱条构成。由于它的结构限制,创可贴只能用于小块的创伤应急治疗,从而起到暂时的止血,保护创面的作用。目前市场上创可贴种类繁多,但大多止血护疮功效不甚明显,难以使愈合伤口快速愈合,反因长期使用创可贴,创可贴外层的胶布不透气,就会使伤口和伤口周围的皮肤发白,变软导致细菌的继发的感染;反而导致伤口更加的恶化。
目前市场上创可贴大多难以达到快速愈合伤口的要求。如中国专利CN105126150A公开了一种含壳聚糖混合物的创可贴,但壳聚糖混合物见效慢,需长时间使用,在一定程度上会造成伤口恶化;中国专利号CN103041170A公开了一种中草药创可贴,但中草药发挥功效向来缓慢,同样需长时间使用。
紫草素(shikonin),又名5,8-二羟基-2-[(1R)-1-羟基-4-甲基戊-3-烯基]萘-1,4-二酮,是紫草中特有的一种脂溶性萘醌色素类化合物,作为天然中药活性成分,具有良好的抗肿瘤和抗氧化、神经保护、抗病毒、抗炎等多重作用,已成为国内外研究的热点。但紫草中含有称为吡咯里西啶类生物碱类的化学成分,简称PAs,毒性集中于肝脏,又称为肝毒吡咯里西啶生物碱。早在2001年7月6日,美国药品监督管理局FDA就在其官方网站上发出警告,由于紫草这种成分会对人的健康造成危害,甚至损害肝脏,要求厂家停止销售内服的含紫草成分的营养补充剂,外用时也不能用于开放式的伤口。而在部分欧盟国家,就连这类外用药膏也不允许出售。由于制备高纯度紫草素的关键技术尚未产业化,目前国内以紫草素为原料的新药产品暂未正式进入市场,2010版中国药典仍然收载了一些含有PAs成分但缺少安全限量规定的品种,其中也包括紫草。
本发明采用提纯后的紫草素作为创可贴的功效性成分,避免了吡咯里西啶类生物碱的安全性隐患,具有抗炎、抑菌、止痒及促进烧伤创口愈合等多重功效,且安全,无副作用。
发明内容
发明目的:为了克服现有技术中存在的问题,本发明提出了一种对皮肤外伤、红肿疼痛等小伤口具有显著疗效、抗菌消炎,止痛见效快、促使伤口快速愈合的含紫草素的创可贴及其制备方法。
技术方案:为了解决上述技术问题,本发明所采用的技术方案为:一种含紫草素的创可贴,由平布胶布、吸水垫、离型纸组成,所述吸水垫位于平布胶布的中段,所述吸水垫上均匀涂布有紫草素混合物,所述离型纸覆盖于干燥挥发后的所述紫草素混合物上;所述紫草素混合物由γ-环糊精和紫草素组成;所述紫草素提纯分离于紫草。
更为优选的,所述紫草素混合物呈长方形状涂布于所述吸水垫表面。
更为优选的,所有述吸水垫为医用即溶止血纱布、全棉水刺无纺布、医用胶原蛋白海绵中的一种。
更为优选的,所述紫草素混合物采用将紫草素用γ-环糊精包埋处理的方法制备而成,具体的将紫草素用1-5mL乙酸乙酯溶解,与3-10gγ-环糊精混合,减压浓缩干燥成粉末,加水调成糊状,制得所述紫草素混合物,其中紫草素的质量浓度为0.01%-0.15%。
更为优选的,参考本公司另一已公开中国发明专利CN105949043A中相关方法,所述紫草素提纯分离于紫草的方法为:A)酶解:将紫草粉碎,分散于水中,加入复合酶制剂进行酶解;以质量百分比计,所述复合酶制剂的组成为:纤维素酶,50~80%;果胶酶,15~30%;其余为木聚糖酶;所述复合酶制剂的平均酶活为10000~50000单位;所述复合酶制剂的添加量为紫草质量的0.2~0.8%;B)碱水提取:酶解后向体系中加入氢氧化钠进行提取,得紫草萃取物;C)分离纯化:从所述的紫草萃取物中分离纯化得所述的紫草素。
本发明还公开了上述含紫草素的创可贴的制备方法,包括如下步骤:a)首先对紫草中的紫草素进行分离提纯;b)将提纯得到的紫草素与γ-环糊精用包埋处理的方法制备成紫草素混合物,增加紫草素的溶解性,提高药效;c)将紫草素混合物均匀涂布于平布胶布中段的吸水垫上,经干燥挥发,粘于平布胶布上;d)将离型纸覆盖于干燥挥发后的所述紫草素混合物上,即得所需创可贴。
有益效果:本发明提供的一种含紫草素的创可贴及其制备方法,创可贴由平布胶布、吸水垫和离型纸组成,紫草素混合物均匀涂布于吸水垫上,紫草素混合物具体由提纯分离于紫草中的紫草素用γ-环糊精包埋处理制得,本发明制备的创可贴除了具备一般创可贴的功能外,还具备紫草素特有的药理作用,增强了抗菌、消炎、止痛、促进伤口愈合、减少疤痕形成等作用。且制备方法操作简单,对人体无刺激性、无明显毒性,可广泛应用于小伤口的处理。

请参照图1,一种含紫草素的创可贴,由平布胶布1、吸水垫2、离型纸3组成,所述吸水垫2位于平布胶布1的中段,所述吸水垫2上均匀涂布有呈长方形状的紫草素混合物,所述离型纸3覆盖于干燥挥发后的所述紫草素混合物上;
所述紫草素混合物由γ-环糊精和紫草素组成;具体采用将0.01g紫草素用γ-环糊精包埋处理的方法制备而成,具体的将紫草素用1-2mL乙酸乙酯溶解,与3-5gγ-环糊精混合,减压浓缩干燥成粉末,加100mL水调成糊状,制得所述紫草素混合物,其中紫草素的质量浓度为0.01%。
所述紫草素提纯分离于紫草,具体参考本公司另一已公开中国发明专利CN105949043A中相关方法,所述紫草素提纯分离于紫草的方法为:A)酶解:将紫草粉碎,分散于水中,加入复合酶制剂进行酶解;以质量百分比计,所述复合酶制剂的组成为:纤维素酶,50~80%;果胶酶,15~30%;其余为木聚糖酶;所述复合酶制剂的平均酶活为10000~50000单位;所述复合酶制剂的添加量为紫草质量的0.2~0.8%;B)碱水提取:酶解后向体系中加入氢氧化钠进行提取,得紫草萃取物;C)分离纯化:从所述的紫草萃取物中分离纯化得所述的紫草素。

从表1-表4所显示的临床观察结果数据进行分析,治疗组的完全治愈案例明显优于对照组,可见本发明一种含紫草素的创可贴在止血止痛,消炎抗菌以及伤口愈合速度上来说都明显优于现有创可贴,增强了抗菌、消炎、止痛、促进伤口愈合、减少疤痕形成等作用。

Claims (6)

1.一种含紫草素的创可贴,其特征在于由平布胶布(1)、吸水垫(2)、离型纸(3)组成,所述吸水垫(2)位于平布胶布(1)的中段,所述吸水垫(2)上均匀涂布有紫草素混合物,所述离型纸(3)覆盖于干燥挥发后的所述紫草素混合物上;所述紫草素混合物由γ-环糊精和紫草素组成;所述紫草素提纯分离于紫草。
2.根据权利要求1所述的含紫草素的创可贴,其特征在于:所述紫草素混合物呈长方形状涂布于所述吸水垫(2)表面。
3.根据权利要求1所述的含紫草素的创可贴,其特征在于:所述吸水垫(2)为医用即溶止血纱布、全棉水刺无纺布、医用胶原蛋白海绵中的一种。
4.根据权利要求1所述的含紫草素的创可贴,其特征在于所述紫草素混合物采用将紫草素用γ-环糊精包埋处理的方法制备而成,具体的将紫草素用1-5mL乙酸乙酯溶解,与3-10gγ-环糊精混合,减压浓缩干燥成粉末,加水调成糊状,制得所述紫草素混合物,其中紫草素的质量浓度为0.01%-0.15%。

6.如权利要求1所述的含紫草素的创可贴的制备方法,其特征在于包括如下步骤:a)首先对紫草中的紫草素进行分离提纯;b)将提纯得到的紫草素与γ-环糊精用包埋处理的方法制备成紫草素混合物;c)将紫草素混合物均匀涂布于平布胶布(1)中段的吸水垫(2)上,经干燥挥发,粘于平布胶布(1)上;d)将离型纸(3)覆盖于干燥挥发后的所述紫草素混合物上,即得所需创可贴。
