背景技术
治疗剂在患病区域的选择性释放是改善目前疗法的最重要挑战中的一种。在这种环境下,使用纳米颗粒作为治疗剂的载体(纳米载体)潜在地允许避免可能存在于施用部位和最终靶标之间的生物屏障,以及更具体地,以选择性方式在患病区域而不是正常组织处积聚药物。作为基本的先决条件,纳米载体必须能够以有效的方式结合大量的药物。

然而,天然HFt表现出一些缺点。首先,其能够结合某些类型药物,如例如阿霉素(具有广泛抗肿瘤谱的抗肿瘤药物中的一种)的收率较低,并且这可能限制其可能的用途和临床发展。其次,当通过全身性途径注射时,天然HFt具有非常短的血浆半衰期,约2-3小时。最后,其天然的铁氧化酶活性可能抑制人类成骨细胞的发育和成熟,并导致矿化降低、骨质减少和骨质疏松(Zarjou A,Jeney V,Arosio P,Poli M,Zavaczki E,Balla G,BallaJ.Ferritin ferroxidase activity:a potent inhibitor of osteogenesis.J BoneMiner Res.2010,25:164-72)。因此,建议使用不具有抑制作用的通过位点特异性突变(在下文中称为vHFt)获得的缺乏铁氧化酶活性的HFt变体。
发明内容
为了同时地解决以上列出的关于天然HFt的所有缺点,本发明人决定通过直接作用于蛋白质的外表面来从遗传学角度修饰人类铁蛋白重链(HFt)。在这样做的过程中,显著且令人惊奇地提高了将药物阿霉素封装在蛋白质腔内的能力,并且延长了血浆半衰期,而不影响蛋白质识别肿瘤细胞的能力。
通过如所附权利要求1中所定义的融合蛋白实现了以上和其它目的。其它独立权利要求和从属权利要求涉及本发明的其它方面和具体实施方式,其形成说明书的组成部分。

图10示出了在%药物释放随时间在4℃和37℃下的铁蛋白-阿霉素复合物(HFt-DOXO,HFt-MMP-PAS40-DOXO和HFt-MMP-PAS75-DOXO)的稳定性。由于在该专利中描述的HFt上进行修饰,与相应的天然HFt构建体相比,HFt-MMP-PAS40构建体(用HFt-MMP-PAS75也获得相同的结果)在药物结合形式上更稳定(当存储在4或37℃时,随时间释放的药物%更低)。星号(*)表示在天然HFt样本中,释放的药物量不能在超过这段时间测定,由于材料倾向于变得浑浊并沉淀。
图11示出了DOXORUBICIN、HFt-MMP-PAS75和HFt-MMP-PAS75-DOXO对肉瘤HT-1080细胞的抗增殖活性。
图12示出了来自含阿霉素的化合物的药代动力学实验的结果。在健康小鼠中静脉内注射裸露药物阿霉素和INNO-206(一种新的和更有活性的阿霉素制剂)或者封装在基于铁蛋白的化合物中的阿霉素后在不同时间(10、60、180、360和1440分钟)时计算阿霉素血浆浓度:天然HFt、HFt-MMP-PAS40和HFt-MMP-PAS75。
图13示出了用于评估带有人类胰腺肿瘤(异种移植物)的小鼠的治疗效果的典型实验。用一次剂量为5mg/kg的阿霉素治疗小鼠组(一旦肿瘤大小达到约100mm3)(n=6)4次(每4天)。测试的化合物是:INNO-206、HFt-MMP-PAS40-DOXO和HFt-MMP-PAS75-DOXO。在附图中,从治疗开始大约3周后,报道肿瘤的重量(和代表性图像)。
具体实施方式
作为本发明的主题的融合蛋白包含至少三个结构域。
第一结构域包含人类铁蛋白重链的氨基酸序列。这样的氨基酸序列是天然序列,或是具有至少90%序列同一性的天然序列的变体。由于人类铁蛋白重链具有183个氨基酸的长度(SEQ ID NO:1),与天然序列相比,具有至少90%序列同一性的变体包含多达19个氨基酸取代。该定义尤其包括以上提及的缺乏铁氧化酶活性的vHFt变体的氨基酸序列SEQ IDNO:2,其代表可替换的变体。vHFt的氨基酸序列以两个氨基酸取代为特征:赖氨酸代替62位谷氨酸以及甘氨酸代替65位组氨酸。
本发明的融合蛋白的第二结构域包含至少一种基质金属蛋白酶(MMP)切割位点,特别是MMP-2、MMP-3、MMP-7或MMP-9的氨基酸序列。作为非限制性的实例,在下文中列出了几种肽,其模拟胶原链的切割序列,并由MMP-2和MMP-9以特别有效的方式切割:

所有先前提及的氨基酸序列是制备根据本发明的融合蛋白和纳米颗粒的代表性但非限制性的实例。
与N末端连接的本发明的融合蛋白的第三结构域由富含脯氨酸、丝氨酸和丙氨酸的多肽(为了简便起见称为“PAS”)的氨基酸序列组成,具有在药物(特别是用于药物阿霉素)封装过程中提高蛋白质的稳定性,以及增加蛋白质-药物复合物的稳定性的目的。PAS的存在还能够掩蔽蛋白质表面,从而延长其血浆半衰期。
多肽PAS主要由富含Pro、Ala和Ser的氨基酸序列组成,其形成非结构化聚合物,其长度优选低于80个氨基酸残基,更优选在20至80个氨基酸残基之间,更优选在40至75个氨基酸残基之间。在优选的实施方式中,上述多肽PAS的脯氨酸残基占多肽PAS的总氨基酸残基的10-40%。
特别适合用于本发明范围内,因而优选的PAS多肽的实例是40或75个氨基酸残基的PAS多肽,如例如以下:

通过如以上先前提及的包含一个以上金属蛋白酶切割位点的短肽序列,将稳定和掩蔽的PAS多肽添加至HFt的表面,以便提供具有可置换掩蔽的本发明的融合蛋白质。事实上,通过细胞外基质金属蛋白酶(MMP)可以在靶组织上将PAS多肽选择性地除去。特别地,MMP-2和MMP-9显示为在肿瘤微环境中过表达并参与血管生成、侵袭和肿瘤转移的关键金属蛋白酶。
在本发明范围内在铁蛋白的多聚体表面上使用PAS多肽提供了优于现有技术的几个优点。为了将药物或小分子封装在铁蛋白的蛋白质腔内,迄今为止进行的最明显的行动是直接地修饰药物或蛋白质的内腔本身。以这种方式,药物(或小分子)与铁蛋白内腔中的结合位点之间的相互作用是有利的。例如,阿霉素与铜(II)预复合以增加封装收率(包封收率,encapsulation yield)(Zhen Z等人,ACS Nano.2013Jun 25;7(6):4830-7)。通过用金结合肽以遗传的方式修饰铁蛋白的内腔而将金纳米颗粒封装(Zheng B等人Nanotechnology.2010Jan29;21(4):045305)。然而,本发明人已经决定通过使用惰性PAS聚合物直接作用在蛋白质外表面上,以便在药物存在下在通常用于解离(pH 2.0)和重新缔合(pH 7.5)HFt的pH突然变化期间稳定蛋白质。PAS聚合物的存在也可能限制药物在表面本身上的非特异性结合。
此外,存在PAS也可以表示一种延长蛋白质的血浆半衰期的方法,如XL-proteinGmbh公司最近已经提出了大量蛋白质。事实上,被称为“PASylation”的技术能够通过使用天然非结构化氨基酸链作为PEG的生物替代物来延长生物药物的血浆半衰期(SchlapschyM,Binder U,C,Theobald I,Wachinger K,Kisling S,Haller D,SkerraA.PASylation:a biological alternative to PEGylation for extending the plasmahalf-life of pharmaceutically active proteins.Protein Eng Des Sel.2013,26:489-501)。使用该技术,生物活性蛋白质与包含数百个小氨基酸的残基的多肽序列(脯氨酸、丝氨酸和丙氨酸(PAS))以遗传的方式融合。通常,PAS序列由融合至生物活性蛋白质的100-3000个残基(优选400-600个残基)组成。在专利WO 2008/155134中提及的生物活性蛋白质属于以下类别:结合蛋白、抗体片段、细胞因子、生长因子、酶、激素。没有提及的特别是以vHFt形式的人类铁蛋白重链,不属于列出的类别。根据在下文中将进一步详细描述的本发明人进行的测试,对于人类(天然或变体)铁蛋白重链,PAS序列可以比WO 2008/155134中所描述的短得多,例如低于80个氨基酸残基。换句话说,通过利用HFt形成由24个单体组成的纳米颗粒的特定能力,更短的PAS序列证明在延长体内HFt的血浆半衰期上是有效果和有效率的。掩蔽PAS多肽的长度的选择是开发具备有期望特征的HFt的关键步骤。事实上,在一方面,体内半衰期必须足够长以使HFt在期望位置处积聚。在另一方面,HFt必须能够在富含金属蛋白酶(MMP)的环境中从掩蔽聚合物释放其本身。如果PAS序列太短,那么该构建体可能非常快地从机体被清除。如果PAS序列太长,则可能阻碍穿过将构建体与靶组织和细胞(即,血管、紧密连接、肿瘤间质等)分离和/或掩蔽一个以上MMP切割位点的生物屏障。
此外,如在本发明的融合蛋白的情况下,当蛋白质的N末端被修饰时,实现过表达、完全可溶解且适当地折叠和组装的最终蛋白质产物是事先不明显的。事实上,熟知蛋白质的N-末端修饰可能损害它们的表达、溶解性和/或适当的折叠和组装。
所有这些结果,特别是药物阿霉素的封装收率的改善已经由本发明人令人惊讶地和出乎意料地实现,本发明人通过使用重组蛋白的基因融合技术和生产技术两者,构建了基于人类铁蛋白重链(HFt)的以天然形式或以变体形式(优选vHFt)的纳米颗粒。特别地,如将在与实施例相关的部分中详细描述的,制备遗传构建体,其在单个核酸序列(例如DNA)中编码图1所示的三种序列:i)HFt(或vHFt);ii)可由MMP-2/9切割的短肽序列(MMP);iii)富含Pro、Ser和Ala(PAS)的非结构多肽序列,优选具有在20至80个残基之间的长度。将序列ii)和iii)结合至HFt的N末端,用于它们的可逆掩蔽。
在一些实施方式中,本发明的融合蛋白质HFt包含分别将第一结构域连接至第二结构域和/或将第二结构域连接至第三结构域的第一和/或第二接头氨基酸序列。第一和/或第二氨基酸序列可以彼此相同或不同。
如上所述,本发明人获得的HFt融合蛋白自发地形成能够携带治疗性(化学的化合物、单克隆抗体、肽等)和/或诊断性分子的HFt纳米颗粒(图2)。在优选的实施方式中,将至少5种治疗性和/或诊断性分子封装在HFt纳米颗粒的内腔中或共价结合至HFt纳米颗粒的表面。由于PAS多肽的存在,与未修饰的蛋白质相比,结合的药物量和蛋白质-药物复合物本身的稳定性显著地增加。如在本文中所使用的,术语“蛋白质-药物复合物的稳定性”是指HFt融合蛋白将药物保留在腔中并且在复合物的储存期间不随时间释放药物的能力。在其最终使用前从HFt蛋白质中释放药物可以导致不良影响,如沉淀、聚集和最终产品的损失。
用作为用于药物或诊断剂的靶向结合和释放系统的本发明的HFt纳米颗粒将在循环中和在不表达MMP或不充分表达MMP的组织中暂时无活性,并且随后在它们达到富含MMP(MMP-2/9)的靶组织时被活化,在该靶组织中可以释放其负载(图3)。治疗性分子是例如药学的活性成分。如在本文中所使用的,表述“药学的活性成分”或更简单的“活性成分”是指任何药学上活性分子(化学的化合物,单克隆抗体,肽等),例如可用于癌症治疗的分子。用于本发明的优选活性成分是例如但不限于阿霉素、紫杉醇、吉西他滨和铂类活性成分。也可以使用以上列出的活性成分的前体。
诊断性分子是例如成像剂。如在本文中所使用的,术语“成像剂”涉及允许器官、组织或身体系统的可视化的分子。示例性的成像剂包括顺磁剂,光学探针和放射性核素。术语“光学探针”是指可以通过在第一波长处激发并以第二波长读取来探测的荧光化合物。示例性的光学探针包括异硫氰酸荧光素和5-(和6)-羧基四甲基罗丹明、琥珀酰亚胺酯。
在诊断性和治疗性应用中,作为靶向载体系统的本发明的HFt纳米颗粒可以通过任何合适的给药途径施用于受试者或患者,例如口服、不经肠道、静脉内、腹膜内、肌内、作为栓剂、病灶内、经鼻内或皮下、鞘内、淋巴内、通过吸入微滴,或通过植入缓释装置,例如渗透泵。如在本文中所使用的,术语“受试者”涉及动物,如哺乳动物,包括人、牛、绵羊、山羊、马、狗、猫、兔、大鼠、小鼠等。
如在本文中所使用的,术语“治疗(treating)”或“治疗(treatment)”是指在治疗某种疾病、病变、病症或症状上的成功或改善,或者在某些情况下预防症状或病症发作的证据。
在治疗性应用中,本发明的HFt纳米颗粒用于施用治疗有效剂量的药学活性成分。“治疗有效剂量”旨在表示对于所施用的产生治疗效果的剂量。确切剂量将取决于许多因素,包括治疗目标、受试者、待治疗的疾病等等,并且可以由本领域普通技术人员通过使用本身已知的方法容易地确定(参见,例如,Lieberman,Pharmaceutical Dosage Forms(第1-3卷,1992);Lloyd,The Art,Science and Technology of Pharmaceutical Compounding(1999);Pickar,Dosage Calculations(1999);和Remington:The Science and Practiceof Pharmacy,2003年第20版,Gennaro编辑,Lippincott,Williams&Wilkins)。
本发明的HFt纳米颗粒可用于治疗需要施用药学成分的任何疾病,例如通过在纳米颗粒的腔内螯合活性成分或者通过将其共价结合至纳米颗粒表面。纳米颗粒也可以用于诊断,更具体地用于疾病成像,通过在纳米颗粒的腔内螯合成像剂或者通过将其共价结合至纳米颗粒表面。

实施例
实施例1
构建HFt-MMP-PAS40和HFt-MMP-PAS75融合蛋白的表达载体
作为设计HFt-MMP-PAS(或vHFt-MMP-PAS)构建体的第一步,基于通过X射线晶体学以实验方式确定的且可以以代码3AJO在蛋白质数据库(PDB)获得的HFt的三维结构,构建了几种分子模型。为了以计算的方式修改该结构,使用了InsightII分子建模软件(AccelrysInc.)。
i)为了构建Vannucci等人(2012)描述的具有合适的流体动力学体积的HFt变体的模型,使用含有110个PEG单元的结合至在HFt表面上显示的-SH基团的聚合物。将PEG聚合物模拟成蛋白质表面上的紧密折叠的构象,以反映针对持续循环PEG化的(PEGylated)HFt(PEG 5kDa)的已经以实验的方式测量的流体动力学体积(参见Vannucci等人,2012)。
ii)为了构建新的HFt的模型,将选择的MMP切割序列(SEQ ID NO:5)结合至每个HFt亚基(SEQ ID NO:1)的暴露的N末端,并且构建多个不同长度的PAS聚合物。通过三个甘氨酸残基将这些PAS聚合物连接至MMP切割序列上,并将其体积与PEG化的HFt的体积进行比较。最初地,选择两种PAS长度,因为它们反映了两种不同的情况,即伸展构象(PAS40)和更多折叠的构象(PAS75),考虑到由于存在肽键和脯氨酸残基,因而PAS聚合物与高度柔性的PEG相比具有显著降低的自由度。
通过将三种不同序列组合成一个单一序列来获得HFt-MMP-PAS40基因:HFt(SEQID NO:1)、MMP(SEQ ID NO:5)和PAS(SEQ ID NO:9)。将由三个甘氨酸残基组成的接头插入至HFt和MMP之间以及MMP和PAS之间。
通过将三种不同的序列组合成一个单一序列:HFt(SEQ ID NO:1)、MMP(SEQ IDNO:5)和PAS(SEQ ID NO:10)来实现HFt-MMP-PAS75基因。将由三个甘氨酸残基组成的接头插入到HFt和MMP之间以及MMP和PAS之间。
通过使用GENEART AG(Germania)合成含有HFt-MMP-PAS40基因或HFt-MMP-PAS75基因的pET-11a表达载体。考虑到用于在大肠杆菌(Escherichia coli)中高水平表达的密码子优化进行基因合成。
实施例2
HFt-MMP-PAS40和HFt-MMP-PAS75融合蛋白的细菌表达和纯化
将来自实施例1的含有HFt-MMP-PAS40或HFt-MMP-PAS75的pET-11a表达载体插入大肠杆菌BL21(DE3)中,并通过双脱氧测序方法测序以证实存在正确的基因。将含有重组质粒的大肠杆菌BL21(DE3)细胞在37℃下在1L的含有氨苄青霉素的Terrific Broth(TB)液体培养基(23.6g/L酵母提取物,11.8g/L胰蛋白胨,9.4g/L K2HPO4,2.2g/L KH2PO4)中培养直至OD600为0.6。通过添加0.5mM的异丙基硫代-β-D-半乳糖苷(IPTG)诱导基因表达,并将培养物进一步孵育3小时。通过离心(15000rpm持续20分钟)收集细胞,悬浮在50mM Tris-HCl(pH7.5)、0.5mM二硫苏糖醇、1mM EDTA和300mM NaCl中,通过超声处理破坏。以15000rpm将裂解物离心40分钟,并且将含有可溶性成分的上清液在37℃下用0.1mg/mL的富含5mMMgCl2的DNA酶处理30分钟,加热至75℃持续10分钟,在冰上冷却,然后离心除去变性的蛋白质。通过使用浓度为65%的饱和(w/v)硫酸铵将回收的上清液沉淀。将沉淀物重新悬浮并相对30mM Tris-HCl、pH 7.5、2.5M NaCl透析过夜,然后装载在预先用磷酸盐缓冲盐水(PBS)平衡的Superdex 200hiload 26/600柱(GE Healthcare)上。将各成分合并、浓缩、无菌过滤并储存于4℃。通过利用对在SDS存在下跑出的15%PAGE凝胶进行考马斯亮蓝染色评估所有制剂的纯度。使用ProtParam软件获得的摩尔消光系数,在280nm下用分光光度法测定蛋白质浓度(www.expasy.org)。
HFt-MMP-PAS75构建体的细菌表达谱如图4所示。
实施例3
制备PEG化型(PEGylated version)的HFt蛋白
用PEG 5kDa将天然HFt PEG化并用作为尺寸排阻色谱实验(SEC)中的参考。在pH7.0的PBS中并在室温下,将溶液(2mg/mL)与1.0mM甲氧基聚乙二醇马来酰亚胺5KDa(Sigma-Aldrich)一起孵育约2小时,伴有搅拌。随后,用来自Amicon Ultra-15(MilliporeCorporate)的30kDa自旋过滤装置将样品过滤并用H2Odd进行5次交换,以除去过量的试剂。将PEG化的样品(HFt-PEG5K)无菌过滤并储存于4℃。
实施例4
通过尺寸排阻色谱法(SEC)评估HFt、HFt-MMP-PAS40、HFt-MMP-PAS75和HFt-PEG5K的流体动力学体积
通过使用以pH 7.5的磷酸盐缓冲盐水(PBS)平衡的凝胶过滤Superose 6柱进行SEC实验。以过滤的PBS将所有样品制备为1mg/mL。
用Origin 8.0(Originlab Corporation,Northampton,MA)分析所有SEC洗脱曲线。HFt及其融合蛋白或官能化型(functionalised version)的洗脱曲线如图5所示。这些结果表明,两种PAS化型都具有比天然蛋白更高的流体动力学体积,并且与HFt的PEG化型(PEG 5kDa)相当,其中HFt-MMP-PAS75略高而HFt-MMP-PAS40略低。此外,与PEG化型相比,两种PAS化型都具有更加均匀且单分散的曲线。出于本发明人的目的,这些流体动力学体积被认为是适当的值,并且将HFt-MMP-PAS75和HFt-MMP-PAS40融合蛋白两者进行进一步地表征,并用于封装药物以及用作为癌症治疗的纳米载体。
实施例5
在体外蛋白酶MMP2/9存在下从HFt-MMP-PAS75融合蛋白中除去保护性PAS
为了分析MMP-敏感性缀合物的酶切,在MMP-2/9存在下研究了HFt-MMP-PAS75的切割。将HFt-MMP-PAS75的溶液与胶原酶IV(含有MMP2和9)混合,并在37℃下孵育2小时。然后,在使用以pH 7.5的磷酸盐缓冲盐水(PBS)平衡的凝胶过滤Superose 6柱的SEC实验中(图6)以及通过SDS凝胶色谱(图7)测试样品。
实施例6
制备携带化学治疗剂的HFt-MMP-PAS40和HFt-MMP-PAS75
作为化学治疗剂,本发明人报道了使用药物阿霉素(DOXO)的实例。通过利用作为pH的函数的蛋白质解偶联-偶联过程,将DOXO封装在两种融合蛋白的蛋白质腔内。通过使用1.5mM DOXO的优选浓度和5μM蛋白质的优选浓度进行反应。通过在酸性条件下,在1.8至2.5之间、更优选2.0的pH下,在黑暗中温和地混合DOXO和蛋白质,进行封装反应。反应温度通常包括在+10℃至+40℃之间(优选+25℃),持续5至30分钟(优选10分钟)的时间段。随后,使用浓缩的NaOH以包括在6.5至9.0之间,更优选pH 7.5的pH快速地调节溶液,并且保持搅拌额外的30分钟。然后,在4℃下以15,000rpm将溶液离心30分钟,并且在黑暗处在4℃下,在pH7.4的磷酸盐缓冲盐水(PBS)中将上清液透析10至16小时之间的时间段。作为最后一步,用30kDa的自旋装置Amicon Ultra-15(Millipore Corporate)将溶液调节至期望浓度。将含有DOXO(HFt-MMP-PAS40-DOXO和HFt-MMP-PAS75-DOXO)的HFt纳米颗粒(NP)无菌过滤并在黑暗中储存于4℃。
实施例7
测试药物阿霉素的封装收率
测试了两种建立的构建体(HFt-MMP-PAS40和HFt-MMP-PAS75)封装阿霉素的能力,并将其与天然HFt蛋白质封装阿霉素的能力以及文献中的其它数据进行比较。样品与酸性异丙醇(2N HCl)在25℃下孵育30分钟的时间段后测定阿霉素的量。通过使用UV-可见的分光光度计进行评估,在485nm处读数,并且DOXO的消光系数为9250M-1cm-1。
然后以%蛋白质回收率和封装的阿霉素分子数量报道了相对产率(图8)。可以看出,与天然HFt相比,本专利的构建体主题已经令人惊讶地和出人意料地获得了至少6倍的更佳的封装药物阿霉素的能力。
实施例8
测试铁蛋白-阿霉素复合物的稳定性
在4℃和37℃下以随时间的%药物释放测试了铁蛋白-阿霉素复合物的稳定性。通过使用以pH 7.5的磷酸盐缓冲盐水(PBS)平衡的凝胶过滤Superose 6柱由尺寸排阻色谱法(SEC)评估释放的阿霉素的百分比。按照在280和485nm处的吸光度,以过滤的PBS将所有样品制备为1mg/mL。HFt-MMP-PAS40-DOXO的典型洗脱曲线如图9所示。
如图10所示,由于在该专利中描述的HFt上进行修饰,与相应的天然HFt构建体相比,HFt-MMP-PAS40构建体(用HFt-MMP-PAS75也获得相同的结果)在其药物结合形式上是更稳定的。
实施例9
HFt-MMP-PAS75-DOXO在体外的抗增殖作用
为了测试增殖,将人类肉瘤细胞(HT-1080)在37℃下以约5×103/孔接种在96孔板上的200μl完全培养基中。第二天,该孔接收PBS、HFt-MMP-PAS75、阿霉素或HFt-MMP-PAS75-DOXO,以阿霉素的不同浓度一式三份,将细胞培养72小时。在培养的最后4小时内,加入[3H]-胸苷(1μCi/孔;1mCi=37MBq),并且通过裂解洗涤的细胞并计数TCA可沉淀的放射性来评估这种掺入。
HFt-MMP-PAS75-DOXO对培养的癌细胞的抗增殖作用如图11所示。结果表明,HFt-MMP-PAS75-DOXO以浓度依赖的方式有效地抑制肉瘤细胞体外生长,与裸露(nude)阿霉素相比,IC50值相同或甚至更低。鉴于潜在的治疗性应用,这些结果是非常重要的。
实施例10
含阿霉素的化合物的药代动力学实验
为了评估血浆稳定性和药代动力学,将含阿霉素的化合物静脉内注射到健康小鼠中。然后,在不同时间(10、60、180、360和1440分钟)抽取血样,用酸性异丙醇(0.75N HCl)1:10稀释,且-20℃冷冻。第二天,使用多模式扫描板读数器,通过测量在485nm激发和在590nm发射下的荧光来量化提取的阿霉素。评估以下样品:阿霉素、INNO-206(新的和更有活性的阿霉素制剂)、HFt-DOXO、HFt-MMP-PAS40-DOXO和HFt-MMP-PAS75-DOXO。结果示于图12中。
实施例11
评估含阿霉素的化合物在动物模型中的治疗效果
在带有人类胰腺肿瘤(异种移植物)的小鼠中进行用于评估含阿霉素的化合物的治疗效果的典型实验。以一次剂量为5mg/kg的阿霉素治疗小鼠组(一旦肿瘤大小达到约100mm3)(n=6)4次(每4天)。测试的化合物是:INNO-206、HFt-MMP-PAS40-DOXO和HFt-MMP-PAS75-DOXO。在图13中报道了从治疗开始约3周后的肿瘤重量(和代表性图像)的典型实验的结果。如图所示,由本发明人建立的含阿霉素的构建体与药物阿霉素的新制剂(INNO-206)相比在所测试的剂量下具有更高的治疗活性。
Claims (16)
1.一种包含至少三个结构域的融合蛋白,其中:
(a)第一结构域包含天然人类铁蛋白重链的氨基酸序列或它们的与所述天然人类铁蛋白重链的所述氨基酸序列具有至少90%同一性的变体;
(b)第二结构域包含基质金属蛋白酶(MMP)切割位点的氨基酸序列;以及
(c)第三N末端结构域由至少20个氨基酸残基的多肽的氨基酸序列组成,并且其基本上由脯氨酸、丝氨酸和丙氨酸(PAS)组成。
2.根据权利要求1所述的融合蛋白,其中,所述PAS具有低于80个氨基酸残基、优选在20至80个氨基酸残基之间、更优选在40至75个氨基酸残基之间的长度。
3.根据权利要求1或2所述的融合蛋白,其中,所述第一结构域包含天然人类铁蛋白重链的氨基酸序列SEQ ID NO:1或人类铁蛋白重链变体的氨基酸序列SEQ ID NO:2。
4.根据权利要求1至3中任一项所述的融合蛋白,其中,所述第二结构域包含选自由MMP-2、MMP-3、MMP-7和MMP-9所组成的组中的基质金属蛋白酶(MMP)切割位点的氨基酸序列。
5.根据权利要求4所述的融合蛋白,其中,所述基质金属蛋白酶(MMP)切割位点的所述氨基酸序列选自由SEQ ID NO:3-8所组成的组。
6.根据权利要求1至5中任一项所述的融合蛋白,其中,前述PAS多肽的脯氨酸残基占它们的总氨基酸残基的10-40%。
7.根据权利要求1至6中任一项所述的融合蛋白,其中,所述PAS多肽的氨基酸序列选自SEQ ID NO:9和SEQ ID NO:10。
8.根据权利要求1至7中任一项所述的融合蛋白,包含分别将所述第一结构域连接至所述第二结构域和/或将所述第二结构域连接至所述第三结构域的第一和/或第二接头氨基酸序列,其中第一和第二氨基酸序列彼此相同或不同。
9.根据权利要求1至8中任一项所述的融合蛋白,其连接至活性成分和/或成像剂。
10.一种包含多个根据权利要求1至9中任一项所述的融合蛋白的单体,优选24个单体的纳米颗粒。
11.一种编码根据权利要求1至9中任一项所述的融合蛋白或根据权利要求10所述的纳米颗粒的分离的核酸。
12.一种包含根据权利要求11所述的核酸的载体。
13.一种包含根据权利要求11所述的核酸或根据权利要求12所述的载体的宿主细胞。
14.一种药物组合物,包含与至少一种药学上可接受的赋形剂、载体或稀释剂组合的根据权利要求1至9中任一项所述的融合蛋白、根据权利要求10所述的纳米颗粒、根据权利要求11所述的分离的核酸、根据权利要求12所述的载体或根据权利要求13所述的宿主细胞。
15.根据权利要求14所述的药物组合物,用作药剂。
16.根据权利要求14所述的药物组合物,用于肿瘤的治疗性处理和/或诊断。
