Abstract

本发明涉及一种心包胶原复合材料及其制备方法和用途。该材料包括壳聚糖、透明质酸、硫酸软骨素等水溶性多糖或羟基磷灰石、BMP、PDGF等骨替代材料及骨生长因子的至少一种。心包胶原复合材料的制备方法为:取猪心包去除大块脂肪,然后浸水、脱脂、浸碱、脱碱、酶解、复合、交联、消毒。该心包胶原复合材料可用作牙种植引导骨再生屏障隔离膜、口腔修复膜、骨修复材料,其具有良好的生物相容性及致密性、高机械强度和优良的可控降解时间。

一种心包胶原复合材料及其制备方法和用途
技术领域
本发明涉及化学化工、生物工程、医学、材料学领域,尤其涉及一种心包胶原复合材料及其制备方法和用途。
背景技术
引导骨再生膜分为不可吸收膜与可吸收膜两大类,其中不可吸收膜有聚四氟乙烯膜(expanded polytetrafluoroethylene,e-PTFE)、钛增强聚四氟乙烯膜(Ti-e-PTFE)、肽膜,这些高聚物材料具有良好的理化、热及机械性能,但由于其惰性原因致使体内不可降解,患者需进行二次手术将其取出,因此造成额外的手术费用及痛苦,并且延长了患者的愈合周期。可吸收膜克服了这一缺点得到广泛应用,但目前临床广泛应用的Bi-Gide胶原膜、海奥口腔修复膜、博特医用胶原膜等引导骨再生膜均存在机械性能差、降解时间短及骨诱导特性不理想等缺陷。
临床应用骨修复材料主要有自体骨、同种异体骨及人工骨材料三大类,自体骨作为骨缺损修复的最佳选择,成为骨移植的金标准,但来源有限且取骨区可能产生并发症,取骨又造成第二术区的创伤,因而临床应用受限。同种异体骨的来源比自体骨多,但与宿主细胞间的免疫排斥反应明显,并有感染疾病及肝炎病毒的可能。人工骨材料近几年成型迅速,能更好的合成与缺损骨形态接近的骨修复替代材料,但它的骨诱导性和仿生物性骨结构方面还不够理想。
发明内容
一种心包胶原复合材料,包括猪心包,其特征是:猪心包脱细胞后复合水溶性多糖、骨替代材料或骨生长因子的一种,其中水溶性壳聚糖有壳聚糖、透明质酸、硫酸软骨素,骨替代材料为羟基磷灰石,骨生长因子为骨形成蛋白,复合水溶性多糖可用作牙种植引导骨再生屏障隔离膜以及颌面部修复、腭裂修复,复合骨替代材料及骨诱导生长因子可用作骨修复材料。
本发明的目的在于提供一种心包胶原复合材料,它具有良好的生物相容性、可控生物降解性以及高机械强度,克服现有技术不足。
本发明是通过以下技术方案实现的:
上述心包胶原复合材料的制备工艺流程包括:去脂肪→水洗→脱脂→碱处理→脱碱→软化→复合→交联→PBS清洗→灭菌
具体流程步骤如下:
1.将市场上购来的心包以玻璃板或钢板去除脂肪,并放于冰箱中冷冻24h;
2.将去除脂肪的心包放入烧杯内以去离子水反复冲洗3次,每次持续5分钟,此时若再有脂肪组织出现,以剪刀和镊子锐性剔除;
3.脱脂:料液比3:1,温度40℃,加入猪心包质量比0.6%的脱脂剂,作用1.5h。料液比4:1,水洗2次;
4.碱处理:料液比3:1,温度28℃,加入猪心包质量比0.8%的氢氧化钠,脱脂剂0.6%,转30min,每隔50min转10min,总共16h。料液比4:1,水洗2次;
5.脱碱:料液比2.5:1,温度30℃,加入猪心包质量比3.0%的氯化铵,脱脂剂0.6%,pH值控制在8-8.5,作用1.5h,每隔30min测pH值。料液比3:1,水洗2次;
6.软化:料液比1.5-2.5,温度35-40℃,先加入猪心包质量比0.3%-0.5%的胰酶作用20-40min,再加入猪心包质量比0.3%-0.6%的中性蛋白酶作用90-150min,充分除去细胞外基质;料液比3:1,水洗3次;
7.复合:料液比2.0-3.0,温度35-40℃,加入猪心包质量比0.5%-2.0%的水溶性多糖、骨替代材料及骨生长因子的一种,复合90-120min;
8.交联:料液比2.0-3.0,温度28-32℃,加入猪心包质量比0.2-0.4%的戊二醛交联30-60min;
9.PBS清洗:料液比3:1,温度25℃,洗3次;
10.消毒:钴60辐照灭菌消毒。
本发明与现有技术及产品相比,具有以下优点:
1.运用此种脱细胞方法脱除细胞后的猪心包细胞脱除更彻底,免疫性更低,生物相容性优良。
2.经过复合后的心包胶原材料具有充足的空间维持能力,致密性更好,阻挡成纤维细胞的透过能力更强。
3.交联后的心包胶原材料机械强度提高,生物降解时间延长,更能满足引导骨再生材料及骨修复材料的临床要求。
4.通过复合不同材料的方法可根据不同临床要求制备成引导骨再生膜、口腔修复膜及骨修复材料。
具体实施方式:
下面通过实例对本发明进行具体的描述
实施例1
本实施例提供一种含多糖的心包复合材料,采用以下步骤制备:
1.将市场上购来的心包以玻璃板或钢板去除脂肪,并放于冰箱中冷冻24h;
2.将去除脂肪的心包放入烧杯内以去离子水反复冲洗3次,每次持续5分钟,此时若再有脂肪组织出现,以剪刀和镊子锐性剔除;
3.脱脂:料液比3:1,温度40℃,加入猪心包质量比0.6%的脱脂剂,作用1.5h。料液比4:1,水洗2次;
4.碱处理:料液比3:1,温度28℃,加入猪心包质量比0.8%的氢氧化钠,脱脂剂0.6%,转30min,每隔50min转10min,总共16h。料液比4:1,水洗2次;
5.脱碱:料液比2.5:1,温度30℃,加入猪心包质量比3.0%的氯化铵,脱脂剂0.6%,pH值控制在8-8.5,作用1.5h,每隔30min测pH值。料液比3:1,水洗2次;
6.软化:料液比2.0,温度37℃,先加入猪心包质量比0.4%的胰酶作用40min,再加入猪心包质量比0.6%的中性蛋白酶作用90min,充分除去细胞外基质;料液比3:1,水洗3次;
7.复合:料液比3.0,温度35℃,加入猪心包质量比1.0%的壳聚糖,复合90min;
8.交联:料液比2.0,温度30℃,加入猪心包质量比0.2%的戊二醛交联60min;
9.PBS清洗:料液比3:1,温度25℃,洗3次;
10.消毒:钴60辐照灭菌消毒。
通过上述方法制备的心包胶原复合材料可用作牙种植引导骨再生屏障隔离膜、口腔修复膜。
新制备的心包胶原复合材料主要技术性能指标如下:

实施例2
本实施例提供一种含骨替代材料或生长因子的心包复合材料,采用以下步骤制备:
按例1操作
7.复合:液比2.5:1,温度35℃,pH值6.4-6.8,羟基磷灰石2.0%,反应90分钟,PBS清洗;
8.交联:液比2.0:1,温度30℃,戊二醛0.2%,分次加入,调节pH值8,反应60分钟,水洗至pH值为中性;
9.PBS清洗:液比3:1,温度25℃,洗3次。
10.消毒:钴60辐照灭菌消毒。
通过上述方法制备的心包胶原复合材料可用作骨修复材料。

Claims (4)

1.一种心包胶原复合材料的制备方法为:选取猪心包为原料,去表面脂肪后水洗,然后脱脂、碱处理、脱碱、软化、复合、交联、PBS清洗、灭菌,其特征在于:
1)将市场上购来的心包以玻璃板或钢板去除脂肪,并放于冰箱中冷冻24h;
2)将去除脂肪的心包放入烧杯内以去离子水反复冲洗3次,每次持续5分钟,此时若再有脂肪组织出现,以剪刀和镊子锐性剔除;
3)脱脂:料液比3:1,温度40℃,加入猪心包质量比0.6%的脱脂剂,作用1.5h;料液比4:1,水洗2次;
4)碱处理:料液比3:1,温度28℃,加入猪心包质量比0.8%的氢氧化钠,脱脂剂0.6%,转30min,每隔50min转10min,总共16h;料液比4:1,水洗2次;

8)交联:料液比2.0-3.0,温度28-32℃,加入猪心包质量比0.2-0.4%的戊二醛交联30-60min;
9)PBS清洗:料液比3:1,温度25℃,洗3次;
10)灭菌:钴60辐照灭菌消毒。
2.权利要求1所述的制备方法制备的心包胶原复合材料,其特征在于:包括猪心包,其特征是:猪心包脱细胞后复合水溶性多糖、骨替代材料或骨生长因子的一种;
所述水溶性多糖、骨替代材料或骨生长因子中的一种与猪心包的重量比为0.5%-2.0%:1;
水溶性多糖有壳聚糖、透明质酸、硫酸软骨素;
骨替代材料为羟基磷灰石,骨生长因子为骨形成蛋白。
3.权利要求书2所述心包胶原复合材料的用途,其特征是该心包胶原复合水溶性多糖可用作牙种植引导骨再生屏障隔离膜以及颌面部修复、腭裂修复。
4.权利要求书2所述心包胶原复合材料的用途,其特征是该心包胶原复合骨替代材料及骨诱导生长因子可用作骨修复材料。
