Abstract
本发明涉及一种萝卜硫素微乳速释滴丸的制备方法,该方法包括以下步骤:(1)萝卜硫素分散于二乙二醇单乙基醚中;(2)将单亚油酸甘油酯、聚氧乙烯蓖麻油EL和萝卜硫素的二乙二醇单乙基醚分散液三者混合后,采用均质机进行处理,得到萝卜硫素微乳液;(3)将聚乙二醇与上述萝卜硫素微乳液混合,搅拌均匀并水浴加热至40℃保温,得到目标混合药剂;(4)标混合药剂滴加到液体石蜡中,收集冷凝成型的固体滴丸,即得到萝卜硫素微乳速释滴丸。本发明方法得到的产品具有便于携带、易于保存、使用方便、快速释药等优点,萝卜硫素进行了微乳化,在胃肠道环境中可自发形成粒径在微米级以下的微乳,提高了萝卜硫素的吸收速度和生物利用度。
Description
技术领域
本发明涉及一种天然产物制剂制备方法,尤其是涉及一种萝卜硫素的微乳速释滴丸的制备方法。
背景技术
萝卜硫素,是葡萄糖莱菔子苷(硫代葡萄糖苷的一种)经黑芥子硫酸苷酶酶解或酸水解产生的异硫氰酸盐衍生物,相对分子质量为177.29,分子式为C6H11S2NO。萝卜硫素是迄今为止蔬菜中发现的抗癌活力最强的一类异硫代氰酸盐,是目前公认的防癌和抗癌效果最好的天然产物之一。能够抑制I相酶和诱导Ⅱ相酶的表达,对肝癌、乳腺癌、肺癌、食道癌、前列腺癌、胃癌有明显的阻断作用,并且萝卜硫素还具有保护皮肤、抗菌、提高机体抗氧化能力和免疫力等药理作用,因此萝卜硫素作为药品或食品功能成分,具有广阔的市场前景[食品发酵与工业,2011,37(7):197-200]。
但是萝卜硫素是活性很强的一种物质,容易分解,不易在常温下保存和使用,一般需要保存在-20℃下,在温度高于50℃特别容易分解,另外光照和pH值对萝卜硫素的影响也很大[中国药学杂志,2007,42(3):193-196]。萝卜硫素不易在室温下保存,是制约萝卜硫素应用发展的一个重要的因素。我国对萝卜硫素的提取分离进行了大量的研究,但对如何提高萝卜硫素的稳定性研究的不多。如专利CN201410009814.1报道了制备萝卜硫素的胶囊的方法,但在制备过程中采用了60~70℃真空浓缩,萝卜硫素容易分解,并且只有4℃可长时间保藏。专利CN201410215852.2报道了一种莱菔硫烷(萝卜硫素)胶囊的制备方法,采用抗性糊精、淀粉、羧甲基纤维素钠、微晶纤维素作为辅料,与莱菔硫烷混合,干燥后,经过纳米级的粉碎机粉碎至100nm颗粒粉末,将该粉末罐装成胶囊,得到莱菔硫烷胶囊,目的是为了解决莱菔硫烷胶囊有效成分破碎不充分、不便于人体吸收的问题。
因此,寻找一种通过简单廉价的方法得到可以常温保存的萝卜硫素制剂产品是非常必要,将对开发以萝卜硫素为主要活性成分的药品和保健品的发展具有积极推动作用。
发明内容
本发明的目的就是为了克服上述现有技术存在的缺陷而提供一种萝卜硫素微乳速释滴丸的制备方法。
本发明的目的可以通过以下技术方案来实现:
一种萝卜硫素微乳速释滴丸的制备方法,该方法包括以下步骤:
(1)萝卜硫素分散于二乙二醇单乙基醚中;
(2)将单亚油酸甘油酯、聚氧乙烯蓖麻油EL和所述萝卜硫素的二乙二醇单乙基醚分散液三者混合后,采用均质机进行处理,得到萝卜硫素微乳液;
(3)将聚乙二醇加入到上述萝卜硫素微乳液中混合,搅拌均匀并水浴加热至40℃保温,得到目标混合药剂;
(4)将目标混合药剂滴加到液体石蜡中,收集冷凝成型的固体滴丸,即得到萝卜硫素微乳速释滴丸。
步骤(1)中制备萝卜硫素的二乙二醇单乙基醚分散液的方法为:萝卜硫素超声分散于二乙二醇单乙基醚中,其中萝卜硫素与二乙二醇单乙基醚的质量比为1:(3~25)。
步骤(1)中萝卜硫素的纯度为25%~99%。
步骤(2)中萝卜硫素微乳液各物质的配比为:单亚油酸甘油酯、聚氧乙烯蓖麻油EL、萝卜硫素的二乙二醇单乙基醚分散液的体积比为(1~10):(1~10):5。
步骤(3)中混合药剂的制备方法中各物质的配比为:聚乙二醇与萝卜硫素微乳液质量体积比(1.5~5)g:1mL。
步骤(3)中聚乙二醇的分子量为800-4000。
步骤(4)中将混合药剂以每分钟20~90滴的速度,于5~30cm高度滴入2~5℃的液体石蜡中,收集冷凝在底部的成型固体滴丸即得萝卜硫素微乳速释滴丸。
所述的萝卜硫素微乳速释滴丸可以作为保健品、食品或药品,萝卜硫素以微乳的形式存在于单亚油酸甘油酯、聚氧乙烯蓖麻油EL和二乙二醇单乙基醚中,由于隔绝了空气和氧气等,可以在常温下保存,在25℃保存1年,萝卜硫素的含量降低不到2%。
与现有技术相比,本发明工艺简单易行,容易工业化,整个工艺价格便宜,投资少,并且得到的产品具有便于携带、易于保存(可常温保存)、使用方便、快速释药等优点,由于对萝卜硫素进行了微乳化,在胃肠道环境中可自发形成粒径在微米级以下的微乳,提高了萝卜硫素的吸收速度和生物利用度。
具体实施方式
下面结合具体实施例对本发明进行详细说明。
实施例1
(1)将10g萝卜硫素(纯度98%)超声分散于100g二乙二醇单乙基醚;
(2)将50mL步骤(1)制备的萝卜硫素的二乙二醇单乙基醚分散液、100mL单亚油酸甘油酯、100mL聚氧乙烯蓖麻油EL混合后,采用均质机于8000转进行处理,得到萝卜硫素微乳液;
(3)将200g聚乙二醇(分子量1000)加入到100mL步骤(2)制备的萝卜硫素微乳液中混合,搅拌均匀并水浴加热至40℃保温,得到目标混合药剂;
(4)将100mL步骤(3)制备的混合药剂以每分钟60滴的速度,于30cm高度滴入2℃的液体石蜡中,收集冷凝成型的固体滴丸,即得到萝卜硫素微乳速释滴丸。
(5)将步骤(4)制备的萝卜硫素微乳速释滴丸置于室内药品柜,1年后,萝卜硫素降低率为1.6%。
实施例2
(1)将10g萝卜硫素(纯度80%)超声分散于90g二乙二醇单乙基醚;
(2)将50mL步骤(1)制备的萝卜硫素的二乙二醇单乙基醚分散液、50mL单亚油酸甘油酯、50mL聚氧乙烯蓖麻油EL混合后,采用均质机于10000转进行处理,得到萝卜硫素微乳液;
(3)将200g聚乙二醇(分子量1500)加入到100mL步骤(2)制备的萝卜硫素微乳液中混合,搅拌均匀并水浴加热至40℃保温,得到目标混合药剂;
(4)将100mL步骤(3)制备的混合药剂以每分钟30滴的速度,于15cm高度滴入5℃的液体石蜡中,收集冷凝成型的固体滴丸,即得到萝卜硫素微乳速释滴丸。
(5)将步骤(4)制备的萝卜硫素微乳速释滴丸置于室内药品柜,1年后,萝卜硫素降低率为1.9%。
实施例3
(1)将10g萝卜硫素(纯度95%)超声分散于150g二乙二醇单乙基醚;
(2)将50mL步骤(1)制备的萝卜硫素的二乙二醇单乙基醚分散液、100mL单亚油酸甘油酯、100mL聚氧乙烯蓖麻油EL混合后,采用均质机于6000转进行处理,得到萝卜硫素微乳液;
(3)将300g聚乙二醇(分子量2000)加入到200mL步骤(2)制备的萝卜硫素微乳液中混合,搅拌均匀并水浴加热至40℃保温,得到目标混合药剂;
(4)将100mL步骤(3)制备的混合药剂以每分钟40滴的速度,于20cm高度滴入3℃的液体石蜡中,收集冷凝成型的固体滴丸,即得到萝卜硫素微乳速释滴丸。
(5)将步骤(4)制备的萝卜硫素微乳速释滴丸置于室内药品柜,1年后,萝卜硫素降低率为1.4%。
上述的对实施例的描述是为便于该技术领域的普通技术人员能理解和使用发明。熟悉本领域技术的人员显然可以容易地对这些实施例做出各种修改,并把在此说明的一般原理应用到其他实施例中而不必经过创造性的劳动。因此,本发明不限于上述实施例,本领域技术人员根据本发明的揭示,不脱离本发明范畴所做出的改进和修改都应该在本发明的保护范围之内。
Claims (9)
1.一种萝卜硫素微乳速释滴丸的制备方法,其特征在于,该方法包括以下步骤:
(1)萝卜硫素分散于二乙二醇单乙基醚中;
(2)将单亚油酸甘油酯、聚氧乙烯蓖麻油EL和所述萝卜硫素的二乙二醇单乙基醚分散液三者混合后,采用均质机进行处理,得到萝卜硫素微乳液;
(3)将聚乙二醇加入到上述萝卜硫素微乳液中混合,搅拌均匀并水浴加热至40℃保温,得到目标混合药剂;
(4)将目标混合药剂滴加到液体石蜡中,收集冷凝成型的固体滴丸,即得到萝卜硫素微乳速释滴丸。
2.根据权利要求1所述的一种萝卜硫素微乳速释滴丸的制备方法,其特征在于,步骤(1)中制备萝卜硫素的二乙二醇单乙基醚分散液的方法为:萝卜硫素超声分散于二乙二醇单乙基醚中,其中萝卜硫素与二乙二醇单乙基醚的质量比为1:(3~25)。
3.根据权利要求1所述的一种萝卜硫素微乳速释滴丸的制备方法,其特征在于,步骤(1)中萝卜硫素的纯度为25%~99%。
4.根据权利要求1所述的一种萝卜硫素微乳速释滴丸的制备方法,其特征在于,步骤(2)中萝卜硫素微乳液各物质的配比为:单亚油酸甘油酯、聚氧乙烯蓖麻油EL、萝卜硫素的二乙二醇单乙基醚分散液的体积比为(1~10):(1~10):5。
5.根据权利要求1所述的一种萝卜硫素微乳速释滴丸的制备方法,其特征在于,步骤(3)中混合药剂的制备方法中各物质的配比为:聚乙二醇与萝卜硫素微乳液质量体积比(1.5~5)g:1mL。
6.根据权利要求1所述的一种萝卜硫素微乳速释滴丸的制备方法,其特征在于,步骤(3)中聚乙二醇的分子量为800-4000。
7.根据权利要求1所述的一种萝卜硫素微乳速释滴丸的制备方法,其特征在于,步骤(4)中将混合药剂以每分钟20~90滴的速度,于5~30cm高度滴入2~5℃的液体石蜡中,收集冷凝在底部的成型固体滴丸即得萝卜硫素微乳速释滴丸。
8.一种采用权利要求1-7中任一项所述的制备方法制得的萝卜硫素微乳速释滴丸。
9.一种如权利要求8所述的萝卜硫素微乳速释滴丸的应用,其特征在于,所述的萝卜硫素微乳速释滴丸作为保健品、食品或药品,在常温下保存,在25℃保存1年,萝卜硫素的含量降低不到2%。
