本发明涉及体外诊断试剂技术领域,具体地说是一种用于低密度脂蛋白胆固醇测定试剂的稳定剂,其特征在于该稳定剂由明胶、MgSO4、甘油、甜菜碱、海藻糖、聚乙二醇、EDTA组成,用于低密度脂蛋白胆固醇测定试剂的稳定剂,可以显著延长低密度脂蛋白胆固醇测定试剂的有效期至2年,开瓶稳定性达到3个月,且明显降低空白吸光度值,试剂操作简单,结果准确,稳定性好。

技术领域
本发明涉及体外诊断试剂技术领域,具体地说是一种配置简单、试剂有效期增长、开瓶稳定性高的用于低密度脂蛋白胆固醇测定试剂的稳定剂。
背景技术
众所周知,低密度脂蛋白胆固醇用于冠心病和动脉粥样硬化的诊断。动脉粥样硬化(AS)斑块中沉淀的脂质主要是低密度脂蛋白(LDL),在各类脂质中,LDL被认为是主要的致病因素,而高密度脂蛋白(HDL)可能起保护作用,血清总胆固醇(TC)大,致反映LDL胆固醇(LDL-CHO)水平,但也受HDL-C水平的影响,因此在AS脂类危险因素判别中,TC偏高时,测定LDL-C有重要临床意义。
目前,低密度脂蛋白胆固醇检测试剂盒多采用直接法-表面活性剂清除法,由于其高灵敏度,高准确性、高精密性及适用性而在临床上广泛使用,但因为酶的活性会受到PH、温度、化学试剂的干扰,从而导致检测试剂存在稳定性差的问题,影响临床使用,目前市场上的低密度脂蛋白胆固醇检测试剂的保存条件及期限为,未开封试剂避光保存于2~8℃,有效期12个月;试剂开封后避光保存于2~8℃,在无污染情况下有效期1个月。
发明内容
本发明的目的是解决上述现有技术的不足,提供一种配置简单、试剂有效期增长、开瓶稳定性高的用于低密度脂蛋白胆固醇测定试剂的稳定剂。
本发明解决其技术问题所采用的技术方案是:
一种用于低密度脂蛋白胆固醇测定试剂的稳定剂,其特征在于该稳定剂由明胶 、MgSO4 、甘油 、甜菜碱、海藻糖 、聚乙二醇、EDTA 组成,其重量组分为:明胶 1~10ml/L,MgSO4 0.1~5g/L,甘油 1~15ml/L,甜菜碱0.1~1mol/L,海藻糖 0.1g/L~20 g/L,聚乙二醇 0.5~2g/L,EDTA 0.5~2.0g/L。
本发明中用于低密度脂蛋白胆固醇检测试剂,由试剂1和试剂2组成:
R1:PIPES-Na 80mmol/L,胆固醇酯酶 2KU/L,胆固醇氧化酶 2KU/L,过氧化氢酶200LU/L,4-氨基安替比林 200mmol/L,三甲基-β-环糊精 5g/L,环氧乙烷十八烷胺 3g/L,亚铁氰化钾 50μmol/L,氯化钠 25ml/L,Proclin300 0.05%。
R2: PIPES-Na 80mmol/L,过氧化物酶 2KU/L,氯化钠 0.9g/L,BSA 1.5g/L,叠氮钠 2.0g/L,TritonX-100 18g/L。
本发明所述稳定剂的使用量在试剂中的体积为5 ml/L。
本发明所述的稳定剂的PH为6.8-7.0,PH对于酶的活性有很大的影响,影响机制很复杂,主要有下列几个方面:1.pH过小(过酸)、过大(过碱)都能使酶蛋白变性而失活.2.pH的改变能影响酶活性中心上必须基团的解离程度,同时也可以影响底物和辅酶的解离程度,从而影响酶分子对底物分子的结合和催化,只有在特定的pH下,酶、底物和辅酶的解离状态,最适宜它们相互结合,并发生催化作用,从而使酶反应速度达到最大值。这个pH称为酶的最适PH(optimun pH)酶的最适pH不是一个常数,它的大小与底物的种类和浓度、缓冲液的性质和浓度、介质的离子浓度、温度、反应时间有关,在测定某种酶的活力时,采用该酶的最适pH,并用适当的缓冲液维持最适pH。
本发明由于该稳定剂由明胶 、MgSO4 、甘油 、甜菜碱、海藻糖 、聚乙二醇、EDTA组成,用于低密度脂蛋白胆固醇测定试剂的稳定剂,可以显著延长低密度脂蛋白胆固醇测定试剂的有效期至2年,开瓶稳定性达到3个月,且明显降低空白吸光度值,试剂操作简单,结果准确,稳定性好。
具体实施方式
一种用于低密度脂蛋白胆固醇测定试剂的稳定剂,其特征在于该稳定剂由明胶 、MgSO4 、甘油 、甜菜碱、海藻糖 、聚乙二醇、EDTA 组成,其重量组分为:明胶 1~10ml/L,MgSO4 0.1~5g/L,甘油 1~15ml/L,甜菜碱0.1~1mol/L,海藻糖 0.1g/L~20 g/L,聚乙二醇 0.5~2g/L,EDTA 0.5~2.0g/L,所述的用于低密度脂蛋白胆固醇检测试剂,由试剂1和试剂2组成:R1:PIPES-Na 80mmol/L,胆固醇酯酶 2KU/L,胆固醇氧化酶 2KU/L,过氧化氢酶200LU/L,4-氨基安替比林 200mmol/L,三甲基-β-环糊精 5g/L,环氧乙烷十八烷胺 3g/L,亚铁氰化钾 50μmol/L,氯化钠 25ml/L,Proclin300 0.05%,R2: PIPES-Na 80mmol/L,过氧化物酶 2KU/L,氯化钠 0.9g/L,BSA 1.5g/L,叠氮钠 2.0g/L,TritonX-100 18g/L,所述稳定剂的使用量在试剂中的体积为5 ml/L,所述的试剂的PH为6.8-7.0,这个PH值是胆固醇酯酶,胆固醇氧化酶及过氧化氢酶的最适PH值,在此PH值的条件下,可以有效的保护酶不发生变性而失活,以延长酶的活性,从而有效的延长试剂的稳定性。在此PH值范围内,加入样品后,酶可以最大程度的催化反应,从而使酶反应速度达到最大值。
低密度脂蛋白胆固醇检测试剂的反应原理为:
(1)具有特异选择性的缓冲液作用于血清中CM、VLDL、HDL,并在胆固醇酯酶和胆固醇氧化酶的催化下反应生成H2O2,H2O2被过氧化氢酶分解为H2O和O2,此时由于缺少Trinder反应的其中一个色原,测定体系不显色;
(2)在表面活性剂的作用下,胆固醇酯酶和胆固醇氧化酶催化LDL-C反应并显色,用生化分析仪比色定量。
反应一:CM VLDL HDL + 选择性抑制剂 + CHER + CHOD→ 4-胆甾烯酮 + H2O2
H2O2 + CAT→H2O + O2
反应二:LDL+叠氮钠+表面活性剂+ CHER+CHOD→ 4-胆甾烯酮+H2O2
H2O2 + 4-AAP +HRP→苯醌亚胺色素
本发明高/低密度脂蛋白胆固醇检测试剂,适用于各类自动、半自动生化仪,以奥林巴斯AU400全自动生化仪为例,其操作如下:

分析方法:终点法,试剂的用量为R1 225ul,样本量为3ul,将样本加入试剂1于37℃孵育3~5分钟后读取吸光度值A1,然后加入试剂2,加样量为75 ul,于37℃孵育5分钟后读取吸光度值A2,检测波长主波长为546nm,副波长为700nm。
本实验的目的是检测试剂的空白吸光度值、开瓶稳定性、长效稳定性。
操作步骤:使用定标液定标,取三个水平的质控各测定10次,取平均值,计算相对偏差。质控采用朗道质控(CAT.NO.LE2663 LOT.NO.2162CH LOT.NO.2220CHLOT.NO.2141CH),三个水平的靶值范围分别为

结果分析:根据检测数据,计算测定值均值、相对偏差。
表一结果显示,本发明试剂空白本地明显低于对比试剂,表明试剂的准确度高,精密性好,测试误差小,干扰因素少。
表二结果显示,对比试剂仅在一个月内表现出良好的测试结果,后期偏离靶值幅度较大,而本发明试剂在开瓶后3个月,对同一质控测定10次,相对偏差均小于6%,表明试剂在开瓶后非常稳定,准确性良好,为测试人员提供了方便快捷的检测试剂。
表三结果显示,对比试剂仅在一年内内表现出良好的测试结果,后期偏离靶值幅度较大,而本发明试剂在规定的储存条件下放置两年后,试剂依然非常稳定,相对偏差均小于6%,准确性好。显著延长了试剂的使用期限。

本发明试剂中一定比例的MgSO4及过氧化氢酶起到了降低空白本底的作用,其中过氧化氢酶通过催化过氧化氢反应生成水和氧气从而减少因4-氨基安替比林被氧化而产生的颜色反应,达到降低空白本底的效果。 MgSO4的硫酸根离子可以降低4-氨基安替比林被氧化的速度而减少因4-氨基安替比林被氧化而产生的颜色反应,从而降低空白本底。
含有MgSO4及过氧化氢酶与不含有MgSO4及过氧化氢酶的对比试验实验数据:

本发明试剂中一定比例的明胶可以减缓有机试剂对酶的失活作用。甘油及甜菜碱起到抑制酶分子间聚集的作用,维持酶分子的二级结构,增强酶的稳定性。海藻糖即可通过氢键与酶表面分子结合,稳定酶活性,也可抑制酶分子间聚集,有效阻止酶分子变性失活。聚乙二醇具有疏水亲水的双重特性,可以提高酶的稳定性。酶分子易受到蛋白水解酶的影响,一旦条件适宜,就会发生水解反应,EDTA可避免水解现象,维持酶分子的活性。

本发明的用于低密度脂蛋白胆固醇测定试剂的稳定剂,可以显著延长高/低密度脂蛋白胆固醇测定试剂的有效期至2年,开瓶稳定性达到3个月,且明显降低空白吸光度值,试剂操作简单,结果准确,稳定性好,实验采用奥林巴斯AU400全自动生化仪进行,但本发明的试剂不限于上述仪器,还适用于其他半自动或全自动生化分析仪。
现有的低密度脂蛋白胆固醇检测试剂的成分有胆固醇酯酶、胆固醇氧化酶、过氧化物酶、tween-80、曲拉通x-100、4-氨基安替比林、缓冲液,没有稳定剂,所以开瓶稳定性仅一个月,开瓶效期仅一年。

1.一种用于低密度脂蛋白胆固醇测定试剂的稳定剂,其特征在于该稳定剂由明胶 、MgSO4 、甘油 、甜菜碱、海藻糖 、聚乙二醇、EDTA 组成,其重量组分为:明胶 1~10ml/L,MgSO4 0.1~5g/L,甘油 1~15ml/L,甜菜碱0.1~1mol/L,海藻糖 0.1g/L~20 g/L,聚乙二醇 0.5~2g/L,EDTA 0.5~2.0g/L,所述的用于低密度脂蛋白胆固醇检测试剂,由试剂1和试剂2组成:
