本发明公开了一种治疗冠心病瓜蒌提取物的提取方法,包括以下步骤:(1)取新鲜瓜蒌去籽,利用高速粉碎机粉碎,用9‑12倍量75%乙醇在室温下浸泡后回流提取1‑4次,每次提取3‑5h,过滤,得乙醇提取液和滤渣,滤渣再用5‑8倍量40‑75%乙醇在室温下浸提1‑4次,每次提取3‑5h,过滤,得乙醇提取液和滤渣,合并乙醇提取液,静置12‑48h,滤去沉淀,回收乙醇溶剂得粗提物;(2)将粗提物用蒸馏水混悬均匀,上大孔吸附树脂柱,水洗脱糖后,用50‑100%乙醇水溶液洗脱,收集乙醇洗脱液,回收溶剂得提取物;(3)将提取物用蒸馏水混悬均匀,用氯仿萃取4‑7次,合并萃取液,减压浓缩后得萃余物,萃余物再用乙酸乙酯萃取3‑5次至萃余物不能再被溶解,合并萃取液,减压浓缩得瓜蒌提取物。

技术领域
本发明涉及瓜蒌生产技术领域,具体涉及一种治疗冠心病瓜蒌提取物的提取方法。
背景技术
瓜蒌为葫芦科属多年生攀缘草本,长可达10米。根状茎肥厚,圆柱状,外皮黄色。茎多分枝,无毛;叶互生,近圆形或心形,雌雄异株;雄花数朵总状花序,少有单生,花冠裂片倒卵形,雌花单生,子房卵形,果实近球形,熟时橙红色,花果期7~11月。瓜蒌有解热止渴、利尿、镇咳祛痰等作用。果实含三萜皂苷、有机酸、树脂、糖类、色素;根含蛋白质、皂苷、酸类。
瓜蒌作为中医抗癌中药已得到广泛应用,特别是近年来在临床上对治疗冠心病等表现出的良好疗效而备受关注。但就当前瓜蒌的开发利用来看,临床上主要以中药复方的形式应用,其有效成分及药理作用尚不十分明确,严重限制了瓜蒌的深度开发及市场潜能的发挥。
发明内容
本发明旨在提供了一种治疗冠心病瓜蒌提取物的提取方法。
本发明提供如下技术方案:
一种治疗冠心病瓜蒌提取物的提取方法,包括以下步骤:
(1)取新鲜瓜蒌去籽,利用高速粉碎机粉碎,用9-12倍量75%乙醇在室温下浸泡后回流提取1-4次,每次提取3-5h,过滤,得乙醇提取液和滤渣,滤渣再用5-8倍量40-75%乙醇在室温下浸提1-4次,每次提取3-5h,过滤,得乙醇提取液和滤渣,合并乙醇提取液,静置12-48h,滤去沉淀,回收乙醇溶剂得粗提物;
(2)将粗提物用蒸馏水混悬均匀,上大孔吸附树脂柱,水洗脱糖后,用50-100%乙醇水溶液洗脱,收集乙醇洗脱液,回收溶剂得提取物;
(3)将提取物用蒸馏水混悬均匀,用氯仿萃取4-7次,合并萃取液,减压浓缩后得萃余物,萃余物再用乙酸乙酯萃取3-5次至萃余物不能再被溶解,合并萃取液,减压浓缩得瓜蒌提取物。

(2)将粗提物用蒸馏水混悬均匀,上大孔吸附树脂柱,水洗脱糖后,用75%乙醇水溶液洗脱,收集乙醇洗脱液,回收溶剂得提取物;
(3)将提取物用蒸馏水混悬均匀,用氯仿萃取6次,合并萃取液,减压浓缩后得萃余物,萃余物再用乙酸乙酯萃取4次至萃余物不能再被溶解,合并萃取液,减压浓缩得瓜蒌提取物。
与现有技术相比,本发明的有益效果是:本发明得到的瓜蒌提取物对冠心病等疾病有较好的治疗效果,可利用瓜蒌提取物制备冠心病等疾病药物,疗效好,同时无副作用。

下面将结合本发明实施例,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
一种治疗冠心病瓜蒌提取物的提取方法,包括以下步骤:
(1)取新鲜瓜蒌去籽,利用高速粉碎机粉碎,用9-12倍量75%乙醇在室温下浸泡后回流提取1-4次,每次提取3-5h,过滤,得乙醇提取液和滤渣,滤渣再用5-8倍量40-75%乙醇在室温下浸提1-4次,每次提取3-5h,过滤,得乙醇提取液和滤渣,合并乙醇提取液,静置12-48h,滤去沉淀,回收乙醇溶剂得粗提物;

(3)将提取物用蒸馏水混悬均匀,用氯仿萃取6次,合并萃取液,减压浓缩后得萃余物,萃余物再用乙酸乙酯萃取4次至萃余物不能再被溶解,合并萃取液,减压浓缩得瓜蒌提取物。

1.一种治疗冠心病瓜蒌提取物的提取方法,其特征在于,包括以下步骤:
(1)取新鲜瓜蒌去籽,利用高速粉碎机粉碎,用9-12倍量75%乙醇在室温下浸泡后回流提取1-4次,每次提取3-5h,过滤,得乙醇提取液和滤渣,滤渣再用5-8倍量40-75%乙醇在室温下浸提1-4次,每次提取3-5h,过滤,得乙醇提取液和滤渣,合并乙醇提取液,静置12-48h,滤去沉淀,回收乙醇溶剂得粗提物;
(2)将粗提物用蒸馏水混悬均匀,上大孔吸附树脂柱,水洗脱糖后,用50-100%乙醇水溶液洗脱,收集乙醇洗脱液,回收溶剂得提取物;
(3)将提取物用蒸馏水混悬均匀,用氯仿萃取4-7次,合并萃取液,减压浓缩后得萃余物,萃余物再用乙酸乙酯萃取3-5次至萃余物不能再被溶解,合并萃取液,减压浓缩得瓜蒌提取物。
2.根据权利要求1所述的一种治疗冠心病瓜蒌提取物的提取方法,其特征在于,包括以下步骤:
(1)取新鲜瓜蒌去籽,利用高速粉碎机粉碎,用10倍量75%乙醇在室温下浸泡后回流提取4次,每次提取3h,过滤,得乙醇提取液和滤渣,滤渣再用8倍量75%乙醇在室温下浸提4次,每次提取5h,过滤,得乙醇提取液和滤渣,合并乙醇提取液,静置48h,滤去沉淀,回收乙醇溶剂得粗提物;
(2)将粗提物用蒸馏水混悬均匀,上大孔吸附树脂柱,水洗脱糖后,用75%乙醇水溶液洗脱,收集乙醇洗脱液,回收溶剂得提取物;
(3)将提取物用蒸馏水混悬均匀,用氯仿萃取6次,合并萃取液,减压浓缩后得萃余物,萃余物再用乙酸乙酯萃取4次至萃余物不能再被溶解,合并萃取液,减压浓缩得瓜蒌提取物。
