Examples of the experiments
Experimental example 1 determination of skin toxicity of Hot spring Water of Nanjing Tangshan
The Real-Time label-free cell analysis technology (RTCA Real Time cellular analysis) is adopted for the hot spring water of Nanjing Tangshan, and the RTCA system is used for detecting the influence of different concentrations of metasilicic acid in the mineral-rich hot spring water on the growth of Hs27 (skin fibroblast), KB (human oral epithelial cell) and Hela (cervical carcinoma cell).
And (3) experimental operation:
sucking out the old culture solution in the culture dish in an ultra-clean workbench under the aseptic condition; washing with PBS for 1 time, adding 0.5ml (T25 culture bottle) trypsin solution containing EDTA into the culture dish, covering, incubating in 37 deg.C incubator for 2-6min, and terminating digestion; adding the culture medium into a 4 ml/culture flask to form cell suspension, uniformly blowing the cells by using a pipette, and counting the concentration of the cell suspension by using a counting plate to prepare the cell concentration required by the experiment: 50000 cells/ml.
Experimental method-the use of RTCA system to detect the effect of hot spring water on cell growth:
add 50. mu.l of medium to the wells of E-Plate 96;
place E-Plate96 on RTCAStation;
the RTCA system automatically scans and detects;
taking out the E-Plate96, and adding 100 μ l of the uniformly mixed cell suspension into each well to make the number of cells in each well be 5 k/well; (Note: after addition of E-Plate96 to the cells, it is not necessary to mix the cells with the original medium in the well)
Placing E-Plate96 in a clean bench at room temperature for 30 min;
E-Plate96 was placed on the RTCASTATION in the incubator;
after the system automatically scans, real-time dynamic detection of cell proliferation is carried out;
the cells were treated with the drug at about 24h of growth, i.e., in hot spring water at a concentration gradient of 100. mu.g/ml, 50. mu.g/ml, 25. mu.g/ml, 12.5. mu.g/ml, 6.25. mu.g/ml, 3.12. mu.g/ml, 1.562. mu.g/ml metasilicate.
From the above experiments, it was confirmed that the hot spring water containing metasilicic acid at a high concentration of 25g/ml or more shows a weak effect of inhibiting the growth of cells in three, and that the hot spring water hardly has toxicity to the skin.
Experimental example 2 evaluation of radical scavenging Performance of mask by DPPH method
DPPH is a relatively stable lipid radical, and the ethanol solution of DPPH is purple and has a maximum absorption peak at 517 nm. After the antioxidant is added, DPPH captures an electron to be paired with a free electron, fades purple to be a colorless substance, the absorption at 517nm disappears, and the fading degree is in quantitative relation with the number of the accepted electrons.
The change of absorbance (Abs) after DPPH free radicals react with the sample solution was measured by an ultraviolet-visible spectrophotometer, and the ability of the sample to provide hydrogen atoms and scavenge free radicals against oxidation was determined, with the results shown in table 1 below.
The antioxidant of the vitamin C with different known standard concentrations is measured by DPPH spectrophotometry by taking an antioxidant vitamin C as a reference system, and the antioxidant is obtained through research, along with the increase of the concentration of the vitamin C, the amount of the antioxidant in the thermal spring water mask is increased, the capability of removing DPPH free radicals is enhanced, the intensity of an absorption peak of DPPH at 517nm is reduced, namely, the larger the change rate of absorbance value is, the stronger the capability of removing free radicals for resisting oxidation is.

Experimental example 4 whitening and skin-weighing essence whitening efficacy evaluation (animal skin tissue test)
The sample used in this example was the sample prepared in example 1.
The specific operation method comprises the following steps:
(1) common grade healthy adult brown-yellow Hartley guinea pigs, female, 40, body weights (250 + -20) g, purchased from the Guangdong province medical animal center.
(2) Randomly selecting 10 guinea pigs, making model, collecting brown skin part, removing coat, irradiating with UVB ultraviolet ray at wavelength of 310nm, and accumulating total irradiation amount of 5000mJ/cm2The irradiation is carried out 1 time per day for 1h for 7 days, and UVB ultraviolet irradiation is carried out every day.
Covering the non-irradiated part with a spot tester, detecting MI and EI indexes of the skin at regular time, stopping irradiation for 7d, continuously dripping 2 ml/piece of original whitening skin-balancing mask liquid every day, and euthanizing the animal to take skin tissues.
(3) Selecting 10 guinea pigs at random, making model, removing coat from brown skin part, irradiating with UVB ultraviolet ray at wavelength of 310nm, and accumulating total irradiation amount of 5000mJ/cm2Irradiating for 1 time every day, 1h every time, continuing for 7d, dripping 2 ml/piece of original whitening skin-balancing facial mask solution on an irradiated part after UVB ultraviolet irradiation experiment every day, and euthanizing animals to take skin tissues.
(4)20 animals were randomly divided into 2 groups, 10 whitening and skin balancing mask stock solution groups, and 10 blank control groups. A whitening and skin balancing facial mask stock solution group is characterized in that 4 relatively independent areas on the back of each guinea pig are taken as administration areas, a closed spot tester is taken, about (0.8 +/-0.2) mL of a test object is placed in the spot tester and is in direct contact with the skin, the spot tester is fixed for 6h, and after the test is finished, the residual test object is removed by warm water.
Model control areas were each smeared with 1mL of 0.9% saline for 5 weeks and skin tissue was harvested after euthanasia.
(5) The obtained skin tissue is fixed by 10% paraformaldehyde solution, and is sliced by conventional paraffin, and the following histochemical staining is respectively carried out:
and L-dopa staining, namely dewaxing the section, soaking the section in 0.1 percent L-dopa solution at 37 ℃ for 4 hours, observing staining change under a mirror, washing the section by PBS, washing the section by distilled water, and performing HE counterstaining.
Fontana-Masson silver ammonia staining, namely dewaxing the slices, washing the slices with water, placing the slices in a silver ammonia solution, placing the slices in a water bath at 60 ℃ for 60min, staining the slices with 0.1% gold chloride for 10min, washing the slices with deionized water, and then using 5% Na2SO3Soaking in the solution for 5min, washing with tap water and deionized water, respectively, dyeing with 0.2% nuclear fast red for 5min, and washing with deionized waterAnd (6) washing. Conventional dehydration, xylene clarity and neutral gum blocking.
(6) Microscopic photographing is carried out on the stained histological sections, the Image is analyzed by Image-ProPlus6.0 analysis software, the results of skin color measurement, dopa staining and ammonia silver staining before and after ultraviolet irradiation are shown in the following table 3,
TABLE 3
(7) The experimental results show that after the brown skin is irradiated by UVB, MI is obviously increased, and EI index is obviously reduced. Histochemical staining showed that the melanin content was all significantly increased, and the melanin granule staining was graded close to grade 5. The values of L/M, L/K and L/E were significantly different from those before and after irradiation.
Indicating that the total amount of the additive is 5000mJ/cm2The skin of the guinea pig can be blackened by the irradiation of the left and right UVB, and after the original solution of the whitening and skin balancing facial mask is dripped, the MI value is obviously lower than that of an unused control group, and the EI value is relatively high; after the skin tissue of guinea pigs in the non-irradiated group is dripped with the original whitening and skin balancing mask solution, the MI value is reduced, but the change value is smaller than the change value range after UVB ultraviolet irradiation.
(8) The experimental result shows that the whitening and skin balancing mask has a whitening effect on animal epidermal tissues, and the whitening and repairing effect after being damaged by ultraviolet irradiation is more obvious.
