具体实施方式
本发明基于认识到妥尔油脂肪酸可用于调节动物消化道的微生物种群。
在从同质发酵向异质发酵代谢途径中进行动物消化道的微生物种群的调节,并且在本发明的一个实施方式中,它改善饲料利用度(改善的营养价值)。在本发明的另一实施方式中,调节动物消化道的微生物种群改善饲料转化率。
术语“妥尔油脂肪酸”或“TOFA”应理解为是指由粗妥尔油的蒸馏以及蒸馏的妥尔油的进一步精制而得到的组合物。TOFA通常包括90%-98%(w/w)的脂肪酸。进一步,TOFA可包括1%-10%(w/w)的树脂酸。
在本发明的一个实施方式中,妥尔油脂肪酸包括1%-10%(w/w)的树脂酸。
在本发明的一个实施方式中,TOFA包括2%-9%(w/w)的树脂酸。
在本发明的一个实施方式中,TOFA包括5%-9%(w/w)的树脂酸。

妥尔油脂肪酸(TOFA)通过对蒸馏的妥尔油进行精制来生产。蒸馏的妥尔油(DTO)通过对粗妥尔油进行分馏来生产,其中粗妥尔油作为克拉夫特木浆制造法的副产物而得到。
在本发明的一个实施方式中,TOFA是经干燥的。TOFA可通过喷雾干燥、滚筒干燥或通过其它任何已知的合适的干燥方法进行干燥。

在上下文中,术语“饲料补充剂”应理解为是指在喂养动物过程中可添加到饲料中或可直接使用的组合物。饲料补充剂可包括不同的活性成分。饲料补充剂可以0.0001-5千克/吨饲料总量干重,优选0.005-1千克/吨饲料总量干重的浓度添加。根据本发明的TOFA或包括该TOFA的饲料补充剂可直接添加到饲料或饲料补充剂中,或者一般来讲可根据需要进行进一步加工。
进一步,根据本发明的TOFA或包括该TOFA的饲料补充剂可添加到饲料或饲料补充剂中,或者可单独给动物服用(即不作为任何饲料组合物的一部分)。
在上下文中,术语“饲料组合物”或“饲料”应理解为是指动物膳食的总饲料组合物或其一部分,包括例如补充的饲料、预混合料以及其它饲料组合物。饲料可包括不同的活性成分。
在本发明的一个实施方式中,饲料补充剂包括吸附到适于用作饲料组合物的载体材料例如甜菜渣中的TOFA。
在本发明的一个实施方式中,饲料补充剂包括经干燥的TOFA。
本发明还涉及一种包括饲料补充剂的饲料组合物在调节动物消化道的微生物种群中的用途,所述饲料补充剂包括妥尔油脂肪酸。
在本发明的一个实施方式中,饲料组合物包括以饲料总量干重计,0.00001%-0.5%(w/w)的饲料补充剂。
在本发明的一个实施方式中,饲料组合物包括以饲料总量干重计,0.0005%-0.1%(w/w)的饲料补充剂。
在本发明的一个实施方式中,用于生产妥尔油脂肪酸或饲料补充剂的方法进一步包括干燥步骤。干燥可通过喷雾干燥、滚筒干燥或通过其它任何已知的干燥方法进行。
本发明还涉及一种用于调节动物消化道的微生物种群的方法,该方法包括给动物服用根据本发明的妥尔油脂肪酸的步骤。
在上下文中,术语“有害细菌”应理解为是指能够以包括与宿主动物竞争营养物的不利方式来影响动物的消化道或健康的任何细菌。在上下文中,术语“微生物种群”应理解为是指栖居于消化道中的微生物,包括细菌(Bacteria)域,古菌(Archaea)域和真核(Eukaryote)域的微小成员,以及肠道寄生虫。取决于例如动物的健康和环境因素,微生物种群因动物种类不同而不同。
在上下文中,术语“动物”应理解为是指所有不同种类的动物,例如单胃动物、反刍动物、毛皮动物、宠物和水产养殖动物。不同动物(包括幼崽)的非限制性实例是奶牛、肉牛、猪、家禽、绵羊、山羊、马、狐狸、狗、猫和鱼。
在本发明的一个实施方式中,以有效量给动物服用TOFA。
本发明具有许多优点。TOFA是容易获得的、天然的、低成本和环境友好的材料。进一步,它是无毒和良好耐受的。本发明有效地将动物消化道中微生物群的组成调节为有利于动物性能的方向。随之,本发明的其它益处是例如改善的动物产量,改善的饲料转化率,更高的产品质量、均一性、营养价值、食品和产品安全性,以及更低的每生产单元的成本。本发明使能够以低成本生产饲料组合物和饲料补充剂。
本发明的上述实施方式可以彼此任意组合的方式使用。若干实施方式可以组合在一起而形成本发明进一步的实施方式。本发明涉及的产品、方法或用途可包括至少一个上述本发明的实施方式。
实施例
下面将对本发明进行更详细的描述。
实施例1
进行这个实验以研究具有或没有甜菜渣(SBP)载体的具有5%树脂酸的TOFA在体外对肉鸡回肠的微生物种群和发酵的影响。
实验
40天龄肉鸡的回肠内容物用作模拟模型中的模拟介质和接种菌。由一批TOFA准备试验处理。
具有5%树脂酸的TOFA的制备方法如下:
1、具有20%干物质含量的TOFA
将等分量的TOFA加热至90℃,与精磨的SBP粉末混合,并干燥至每千克含375克干TOFA。
2、消化的TOFA
TOFA的胃肠消化:部分液体TOFA和载体吸附的TOFA在稀释系列中用胃蛋白酶-HCl-处理(pH2.25)以及随后的胰酶-胆汁酸-NaOH处理(pH6.2)进行消化。进行该消化是为了评估产品在进入具有更高微生物活性的远端肠之前是否耐受上胃肠道的条件。
模拟在总共160个2毫升塑料离心小瓶中、1.5毫升体积、10小时模拟时间的条件下进行。在以下四个TOFA的干物质浓度下对样品进行了试验:0%、0.005%、0.01%、0.01%和1%。
对所有模拟样品的短链脂肪酸和微生物总数量进行了分析。此外,通过定量实时PCR(qPCR)对挑选的样品的数种微生物种类或群进行了分析。对回肠模拟样品的乳酸杆菌(lactobacilli)、肠球菌(enterococci)和链球菌(streptococci)进行了分析。
结果
结果表明,在回肠模拟模型中,1千克/吨含量的TOFA增加了乙酸和丙酸的浓度而降低了乳酸的浓度。这表明从同质发酵向异质发酵代谢途径的微生物代谢的调节,其可看作是改善饲料转化率的一个非常积极的变化。将TOFA的量改为0.1千克/吨不利地影响了乳酸杆菌、肠球菌和链球菌的种群数量,所有这些菌都是乳酸产生者。回肠中总细菌数量不受TOFA的影响,这表明响应于TOFA量的改变其它细菌种群增加了。预消化的TOFA影响了许多研究的参数,而甜菜渣载体几乎不影响发酵。
实施例2
进行这个实验以研究具有5%树脂酸的TOFA对饲料的营养价值和饲料转化率的影响。
实验
240只刚孵出的雄性罗斯508肉鸡分配在40个开放式围栏中,每个围栏中6只肉鸡,并且每种喂养处理方式重复8个围栏。
具有5%树脂酸含量的TOFA吸附到研磨的甜菜渣(SBP)载体上,并添加到饲料中。该饲料是小麦-大豆类初始配方。膳食处理方式:
1、对照,没有TOFA
2、对照+TOFA0.1%(1千克/吨)
3、对照+TOFA0.05%(500克/吨)
4、对照+TOFA0.01%(100克/吨)
在第1、11、14和17天对小鸡进行称重。在相同的时期,测量饲料消耗量并计算饲料转化率(FCR)。记录每日死亡率。
17天之后,用气相色谱法对105个回肠食糜样品和105个盲肠食糜样品中的短链脂肪酸(SCFA)进行了分析并且通过qPCR对数种微生物种类或群进行了分析。
结果
结果表明,具有5%树脂酸的膳食TOFA,以0.1-1千克/吨的水平喂养,在第8、11和14天剂量依赖性地增加了肉鸡的体重。0.1-1千克/吨的TOFA有利地且剂量依赖性地调节了从同质发酵向异质发酵方向的小肠微生物发酵。回肠和盲肠的产气荚膜梭菌的数量没有显著受到膳食TOFA量的改变的影响。0.5千克/吨的TOFA降低了具有大于1×109个肠球菌细胞或链球菌细胞,或大于1×1012个乳酸杆菌细胞的样品的频率。1千克/吨的TOFA降低了具有高计数的链球菌或乳杆菌的样品的频率。
结果表明,如果添加在饲料中,TOFA能调节肉鸡或其它种类家禽的消化道的微生物种群,并改善饲料转化率。
实施例3
进行这个实验以研究具有9%树脂酸的TOFA在体外对猪小肠的微生物种群和发酵的影响。
实验
由具有9%树脂酸含量的TOFA油准备试验处理。为了模仿活仔猪胃和十二指肠中的主体条件,在将试验产品引入到模拟容器之前,在37℃下首先用胃蛋白酶-HCl(pH3-4)处理试验产品1小时,然后用胆汁酸+胰酶+NaOH(pH6.8-7.2)处理试验产品3小时。在两个TOFA的干物质浓度下对样品进行了试验:0千克/吨、1.5千克/吨和3.0千克/吨,即0%、0.15%和0.3%)。
对于小肠模拟的真实厌氧菌生长介质,回收并合并了5只仔猪(25-30千克)的远端回肠食糜。随后,对食糜进行离心以去除固体颗粒,然后与缓冲液(pH6.5)结合。制备的最终生长介质保持无氧并且用两个剂量的TOFA对其进行处理。
在厌氧手套箱中将来自两只仔猪的新鲜合并的回肠食糜的接种体引入到模拟容器中。接种后,容器用厚丁基橡胶塞子密封,转移到37℃下,并以100rpm在转动摇床中连续地混合。对于每个TOFA浓度,模拟具有5个重复的容器,并且以随机的顺序进行接种以避免任何潜在的系统偏移(systematicshift)。在从容器中取样以进行微生物代谢分析之前孵育10小时。
模拟在总共25个20毫升的玻璃小瓶中、10毫升体积、10小时模拟时间的条件下进行。
对所有模拟样品的总短链脂肪酸以及乙酸、丙酸和乳酸进行了分析。
结果
结果示于图1-图4中。
在回肠模拟模型中,观察到了用TOFA处理抑制了总短链脂肪酸(SCFA)的形成(图1)。抑制的程度相对于对照为-11%至-14%。
与对照处理相比,TOFA以剂量依赖性的方式增加了乙酸的浓度(+23%至+31%)和丙酸的浓度(+94%至+113%)而降低了乳酸的浓度(-64%至-82%)(图2至图4)。这表明调节了从同质发酵向异质发酵代谢途径的微生物代谢,其可看作是非常积极的变化。
对于本领域技术人员来讲显而易见的是,随着技术的进步,本发明的基本思想可以以多种方式实施。因此,本发明及其实施方式不限于上述实施例,而是它们可以在权利要求书的范围内变化。
Claims (15)
1.一种妥尔油脂肪酸在调节动物消化道的微生物种群中的用途。
2.根据权利要求1所述的用途,所述用途是改善饲料利用度。
3.根据权利要求1所述的用途,所述用途是改善饲料转化率。
4.根据前述权利要求1至3中任一项所述的用途,其特征在于,所述妥尔油脂肪酸包括1%-10%(w/w)的树脂酸。
5.根据前述权利要求1至4中任一项所述的用途,其特征在于,所述妥尔油脂肪酸包括2%-9%(w/w)的树脂酸。
6.根据前述权利要求1至5中任一项所述的用途,其特征在于,所述妥尔油脂肪酸包括5%-9%(w/w)的树脂酸。
7.根据前述权利要求1至6中任一项所述的用途,其特征在于,所述妥尔油脂肪酸包括90%-98%(w/w)的脂肪酸。
8.根据前述权利要求1至7中任一项所述的用途,其特征在于,所述妥尔油脂肪酸是经干燥的。
9.一种包括妥尔油脂肪酸的饲料补充剂在调节动物消化道的微生物种群中的用途。
10.根据权利要求9所述的用途,其特征在于,所述妥尔油脂肪酸包括1%-10%(w/w),优选2%-9%(w/w),最优选5%-9%(w/w)的树脂酸。
11.根据前述权利要求9至10中任一项所述的用途,其特征在于,所述妥尔油脂肪酸是经干燥的。
12.根据前述权利要求9至11中任一项所述的用途,其特征在于,所述妥尔油脂肪酸被吸附到载体材料上。
13.一种包括饲料补充剂的饲料组合物在调节动物消化道的微生物种群中的用途,所述饲料补充剂包括妥尔油脂肪酸。
14.根据权利要求13所述的用途,其特征在于,所述饲料组合物包括以饲料总量干重计0.00001%-0.5%(w/w)的饲料补充剂。
15.根据权利要求13或14所述的用途,其特征在于,所述饲料组合物包括以饲料总量干重计0.0005%-0.1%(w/w)的饲料补充剂。
