背景技术
动物消化道中微生物种群的不平衡和有害细菌的生长可导致动物生长和产量的显著损失。这些不平衡表现为肠道紊乱,例如腹泻。虽然使用例如防止微生物生长的抗生素和其它试剂可防止动物的微生物感染,但预期对它们的使用会有更加严格的规定。反刍动物能够利用对像人类的单胃动物很少或根本没有营养价值的富含纤维的原料。然而,反刍动物的饲料转化效率相对较低,并且它们的甲烷生成占全球温室气体排放量的份额显著。随着食品的需求增加,有必要改善反刍动物的饲料转化效率并降低它们的甲烷生成。总的来讲,越来越需要在动物喂养中使用既可以调节动物消化道中的微生物种群,同时又容易获得、具有良好耐受性和环境友好性的成分。
作为克拉夫特(Kraft)木浆制造法的副产物而得到的粗妥尔油,其分馏产生蒸馏的妥尔油(DTO),其通常包括大于10%的树脂酸和小于90%的脂肪酸。蒸馏的妥尔油的进一步精制产生妥尔油脂肪酸(TOFA),其可提供为具有不同的树脂酸和脂肪酸含量的多种组合物。因为TOFA是脂肪酸的廉价来源,它先前已用于动物营养品中作为能量来源。例如,GB955316公开了妥尔油脂肪酸的碱金属盐在改善反刍动物的体重增加和氮贮留中的用途。

具体实施方式
本发明是基于这样的认知,妥尔油脂肪酸能被用于在增强瘤胃发酵和/或降低瘤胃甲烷生成。
在发酵纤维中,特别是纤维素和半纤维素主要分解成三种挥发性脂肪酸(VFA):乙酸、丙酸和β-羟基丁酸。蛋白和非结构性碳水化合物(果胶、糖、淀粉)也被发酵。
术语“妥尔油脂肪酸”或“TOFA”应理解为是指由粗妥尔油的蒸馏以及对蒸馏的妥尔油的进一步精制而得到的组合物。TOFA通常包括90~98%(w/w)的脂肪酸。进一步,TOFA可包括1~10%(w/w)的树脂酸。

实施例
下面将对本发明进行更详细的描述。
实施例1
甲烷抑制试验
按照以下方式制备TOGA:通过添加足够的水以将混合物的总干物质(TOFA)百分比调节至18~20%,将混合物加热至+90℃,在+90℃保持温度120分钟,在此期间以15分钟为间隔轻轻地搅拌该混合物。
用瘤胃瘘管的奶牛进行甲烷抑制试验是为了研究TOFA在瘤胃中降低甲烷生成速率的趋势。测定瘤胃液样本的产甲烷菌数,由于它们是产甲烷的生物。测量样本的短链脂肪酸谱,包括支链脂肪酸的浓度,由于它们表明TOFA对瘤胃发酵是否有影响。
将6.5克的干TOFA/头/天分四份喂给三头瘤胃瘘管的泌乳奶牛达21天。TOFA首先在甜菜渣上干燥,然后混入配合饲料。在饮食干预前、在TOFA给料期间每周一次,以及在两周的洗脱期后,对瘤胃样本进行取样。对十五个试验样本通过气相色谱进行分析短链脂肪酸(SCFA),并且通过qPCR进行分析产甲烷菌数、原生动物数和总细菌数。
结果
结果表明,在TOFA给料期内,甲烷生成菌数的数值下降,而原生动物和细菌总数并没有受到产品影响。在TOFA给料期内,乳酸、丙酸和戊酸以及总短链脂肪酸的水平趋于在瘤胃液中下降。TOFA趋于提高丁酸的相对比例。响应于饮食TOFA变化,支链脂肪酸的浓度和相对比例趋于下降。
实施例2
瘤胃体外发酵
进行这个实验以研究具有5%树脂酸的TOFA在体外对瘤胃发酵的影响。由一批有代表性的TOFA产品准备所述处理。将等分量的TOFA加热至90℃,与精磨的甜菜渣(SBP)混合,并干燥至每千克含375克的干TOFA。在发酵容器中,以三种剂量(2毫克/克,10毫克/克和50毫克/克)对SBP担载的TOFA进行测试。对照处理含有等量SBP而没有TOFA。
在模拟中饲料的总量为每发酵容器1克的干物质。饲料含有50%牧草青贮和50%配合饲料发酵容器用通过清除O2的热铜催化剂用CO2进行冲洗,并且用厚丁基橡胶瓶塞进行密封。在无氧的CO2流下,将总共38ml的厌氧还原的温度调节(+38℃)缓冲液(参见1970年USDA出版的《农业手册》,第379卷)引入各个每个模拟容器中。发酵培养基的初始pH被设为6.8,并且它用磷酸盐和碳酸盐进行缓冲。每种处理引入5个重复容器中。
从每天喂食8千克配合饲料和约40千克牧草青贮(约13千克干物质)的瘤胃瘘管的奶牛取样瘤胃液。瘤胃液立即从农场被输送到预热的热水瓶中,并用于在两小时内接种。过滤的瘤胃液被以5%的最终体积添加到血清瓶中,然后该容器用丁基橡胶瓶塞进行密封。在+38℃,下模拟继续进行12小时。
在12小时发酵期间,对总气体生成和pH进行测定,以了解瘤胃微生物的一般代谢活性和所述产品的效果。此外,总微生物数由定量实时PCR进行分析。
使用双尾t检验对数据进行分析,双尾t检验包括比较喂食处理与对照处理。
结果
具有最高剂量(50毫克/克)的气体生成显著降低了气体生成。
相比于对照,所有剂量的TOFA提高了细菌的总数(图1)。
两个最高TOFA剂量显著减慢了瘤胃液中的pH值从初始pH值6.8开始的下降(图2)。
结果表明,TOFA导致瘤胃微生物群的变化(shift)。在适度剂量下,它抑制了具有最高气体生成能力的微生物,但刺激了细菌的整体增长。TOFA控制pH值降低的事实表明,它对瘤胃的酸中毒具有缓和效果。
对于本领域技术人员来讲显而易见的是,随着技术的进步,本发明的基本思想可以以多种方式实施。因此,本发明及其实施方式不限于上述实施例,而是它们可以在权利要求书的范围内变化。
