动物是指陪伴动物,例如犬和猫。
本发明的化合物可用于治疗具有由瘦素调节的病症的陪伴动物。这种病症包括肥胖和肥胖诱导的或肥胖相关的病症。肥胖是一种状况,其中存在与瘦肉组织相比过量的脂肪组织。肥胖和肥胖诱导的或肥胖相关的病症的治疗是指:施用本发明的化合物来减少食物摄入,以优先减少脂肪组织的量,并减少肥胖的陪伴动物的体重。一种治疗结果可以是:相对于在施用本发明化合物之前动物的体重,减少肥胖陪伴动物的体重。治疗可适当地导致动物减少食物或热量摄入,包括减少总食物摄入,和减少有需求的动物的重量。肥胖和肥胖相关的病症的预防是指:施用本发明的多肽来减少食物摄入,以减少体重,或者维持处在肥胖风险中的动物的体重。预防的另一结果可以是预防之前由于饮食、锻炼或药物疗法而减去的体重的体重恢复。预防的另一结果可以是,如果在肥胖开始之前,在处于肥胖风险中的动物中施用治疗的话,则预防肥胖发生。
本发明的化合物可以与在对具有通过犬瘦素调节的病症的动物的疗法中或者用于治疗所述动物的一种或多种其它活性成分组合或者利用所述一种或多种其它活性成分。
本发明的化合物可以并入适合于施用至动物的组合物中。设计这种组合物来适合于选择的施用方式,并且适当地使用可接受的稀释剂、载体、和/或赋形剂例如分散剂、缓冲剂、表面活性剂、防腐剂、增溶剂、等渗剂、稳定剂等。所述组合物可以按照常规技术设计,所述技术公开在例如Remington,TheScienceandPracticeofPharmacy,第19版,Gennaro,编辑,MackPublishingCo.,Easton,PA1995中,其提供了如从业者通常已知的制剂技术的概略。组合物用的合适的载体包括当与本发明的化合物组合时,保持该化合物的活性并且是与动物免疫系统非反应性的任何材料。本发明的组合物包含化合物和一种或多种药学上可接受的载体、稀释剂或赋形剂。
包含本发明化合物的组合物可使用标准的施用技术施用至处于显示如本文中描述的疾病或病症的风险中的动物。本发明的组合物含有有效量的本发明的化合物。有效量是指:实现所需治疗结果(以剂量计和对于时间段和对于施用方式)的量。化合物的有效量可根据例如动物的疾病状态、年龄、性别和体重以及化合物在动物中引起所需反应的能力因素而改变。有效量也是其中治疗有益效果超过化合物的任何毒性或不利影响的量。有效量可以是动物初始体重的大约2%/周,直至动物达到如由兽医和客户确定的所需体重。有效量/剂量可以为每周约0.1至约10mg/kg动物重量,例如通过皮下注射来施用。有效量/剂量可以是通过SC注射每周一次施用的1mg/kg。
实施例1
SEQIDNO:1的多肽基本上按照US2012/0149,636中所述获得的。SEQIDNO:1的多肽在pH4.0下20mMTris;2M尿素;和50mM甲硫氨酸中被配制成4mg/mL。以10∶1的PEG与多肽摩尔比,向该配制的多肽加入30KPEG(α-甲基-ω-氨基乙氧基氨基甲酰基,聚氧乙烯,SunbrightME-300CA,NOF,日本)。将多肽和PEG轻轻混合,并且允许在28℃下进行48-72小时的缀合。然后,通过常规的阳离子交换层析将PEG化的多肽纯化,并在具有4%海藻糖的PBS(磷酸盐缓冲盐水)7.4中配制成1mg/mL。该处理导致形成含有1-2%之间的SEQIDNO:1的混合物,其中第一甲硫氨酸被切割掉。
体外生物数据
利用在STAT3响应元件控制下的萤光素酶报告基因和OB-Rb(瘦素受体)(其被表达在细胞表面上)两者将HEK293细胞稳定地转染。瘦素结合至瘦素受体,并活化STAT3同二聚体和STAT3/STAT1异二聚体,其与STAT3序列响应元件相互作用并与其结合。该相互作用驱动萤光素酶基因的表达,并刺激细胞产生发光。发光的量与化合物的活性成比例。生物活性是基于4-PLS形曲线的EC50。
第一天
将细胞以20,000个细胞/孔接种到96孔板中,并放置在37℃、5%CO2培养箱中24-30小时。
第二天
野生型(WT)犬瘦素和实施例1的PEG化多肽的起始浓度是1200ng/mL。从中,从各个样品取出400μL体积,并且越过稀释块的行进行4x连续稀释。首先,将300μL/孔测定培养基加入至Col2-11。然后,将100μL从Col1转移至Col2,小心地慢慢混合(不起气泡)8-10次,然后继续将100μL从Col2转移至Col3,并重复直至到达Col11。留下Col12为未刺激的对照孔。然后,用连续稀释的野生型(WT)犬瘦素和实施例1的PEG化多肽刺激细胞。将细胞从培养箱取出,并且小心地从各个孔吸出培养基。然后,将100μL/孔的连续稀释的WT和PEG化多肽从深孔稀释块转移至96孔测定板中的细胞。然后,水平越过下一可用行建立所测试的WT和PEG化多肽的一式两份样品。将板在37℃、5%CO2培养箱中孵育16-18小时。以下显示了具有以ng/mL计的最终浓度的板格式。
第三天
在第三天,准备底物,并添加至细胞,然后由光度计测量它们。在读取板之前大约1-2小时,在室温下解冻一组Steady-Glo(Promega)试剂,其包括:每测定板一小瓶的Steady-Glo冻干的萤光素酶测定底物和一小瓶的Steady-Glo萤光素酶测定缓冲液。将萤光素酶测定缓冲液(~10.5mL)的全部内容物移液到冻干的萤光素酶底物中。将100μL/孔的重构的萤光素酶底物添加至测定板的每个孔,并且用铝箔覆盖测定板,并在室温下在设置为450至600rpm的板摇动器上孵育45-60分钟。然后,在TecanGENiosPRO板读取器上用设置为250毫秒的积分时间来读取板。然后,将数据输入到Excel中,在SigmaPlotApplication上进行EC50测定,并且也进行了相对于WT瘦素的倍数损失活性(foldlossactivity)和百分比相对效力测定。将相对于野生型犬瘦素的倍数损失活性测定为[样品的EC50]除以[野生型瘦素的EC50];瘦素蛋白与参考标准品的%相对效力通过用参考标准品的EC50除以瘦素蛋白的EC50值并乘以100来计算。例如,%相对效力计算为[参考标准品的EC50]除以[样品的EC50]×100。下表显示了使用这些方法分析的WT和实施例1的PEG化多肽。

体内生物数据
这个研究的目的是评价实施例1的PEG化多肽对肥胖的雄性和雌性犬中体重、身体组成和喂养行为的影响。将实施例1的PEG化多肽在有7.4的pH和4%(w/v)海藻糖的磷酸盐缓冲盐水中配制成5.1mg/ml。
使用十八(18)只全部超过一(1)岁龄的犬:九(9)只未阉割的雄性和九(9)只未阉割的雌性小猎犬。犬是肥胖的,重约12至18kg(26.4至39.6lbs)。在第一个四周的适应期期间,(或者直至达到所需的起始重量),犬饲喂实验室高脂肪(约45%)的干燥食物,其满足或超过维持和健康的营养需求。在适应期的这个部分所有的犬都自由饲喂以促进重量增加。在适应期的最后两周期间和该研究的剩余时间,所有的犬均饲喂商品化的正常脂肪(大约12%)的干燥犬粮,以便减少内源性瘦素水平,并恢复瘦素敏感性。治疗组1和2中的犬在该研究的整个剩余时间期间继续自由地提供食物。治疗组3中的动物按照饮食的标签说明每天饲喂两次。通过碗或在各个笼中含有的自动给水系统,允许动物自由地获取水。在研究过程期间,不施用任何其它伴随药物。
这个研究是在每一性别中作为随机区组设计来实施的。犬被随机分配到圈。通过在每一性别中基线(第-14天(Day-14))体重将动物划分区组(blocked)。存在有三只雄性的三个区组和有三只雌性的三个区组。雄性区组一由三只具有最低体重的雄性组成,且雌性区组一将由三只具有最低体重的雌性组成。在每一性别中的第二区组由具有其次最低体重的三只雄性和三只雌性组成。在每一性别中最后一个的区组包含具有最高体重的三只雄性和三只雌性。
下表显示了使用的治疗、剂量方案和动物数量。

将动物分入九只动物的两个性别组中。在每一性别组内,基于第-14天的体重,将动物从最低到最高排名。基于渐增的基线体重,将每一性别组中的动物分入三只动物的区组中。使用由赞助商提供的随机化表格,将动物随机分配至圈位置。
通过皮下注射在第0、7、14、21、28天向对照和治疗组施用。在将动物转换至低脂肪配给之前,在第-14天研究中登记之前,获得血清样品(大约2-3ml),以便使犬适应放血操作,并建立基线内源性瘦素水平。下表提供了在第-14天和第-1天的循环瘦素水平。

在第-1天、第21天和第42天(研究的最后一天),将所有三个治疗组中的动物都进行DEXA(双能X射线吸收法)扫描,以确定瘦肉相比于脂肪组织的百分比。
对于登记在治疗组1和2中的动物,在第-7天至第42天在大约各自相同的时间,记录每天的饲料消耗。
在所有治疗组中的动物在第-7天开始每天24小时录像记录,并持续至研究结束,来观察包括每天进餐次数和每餐饲喂的平均持续时间在内的饲喂行为。
在研究过程中每周各个动物体重中的初级终点(primaryendpoint)将改变,并且这些将被概述并与治疗组比较。次级终点将是:
1)身体组成:在研究过程中将概述各个动物的DEXA扫描结果(包括瘦肉和脂肪组织的百分比);和
2)饲料消耗/饲喂行为:对于每一动物,将概述在治疗1和2中各个动物的平均每天饲料消耗。
将观察记录各个动物饲喂行为的录像来确定在开始治疗之前和在开始治疗之后每一动物消耗的进餐次数和进餐持续时间。
对体重影响的结果提供在下表中。

Claims (9)
1.一种包含SEQIDNO:1的多肽和聚乙二醇(PEG)部分的化合物,其中所述PEG部分共价连接至所述SEQIDNO:1的多肽的对-乙酰-苯丙氨酸(pAF)残基。
2.权利要求1所述的化合物,其中所述PEG部分具有约30kDa的分子量。
3.权利要求2所述的化合物,其中所述PEG部分是α-甲基-ω-氨基乙氧基氨基甲酰基、或聚氧乙烯。
4.一种治疗陪伴动物中肥胖的方法,其包括将有效量的权利要求1-3任一项所述的化合物施用至有需要的动物。
5.一种预防陪伴动物中肥胖的方法,其包括将有效量的权利要求1-3任一项所述的化合物施用至有需要的动物。
6.权利要求1-3任一项所述的化合物,其用于治疗。
7.权利要求1-3任一项所述的化合物,其用于治疗动物中的肥胖。
8.权利要求1-3任一项所述的化合物,其用于预防动物中的肥胖。
9.一种组合物,其包含权利要求1-3任一项所述的化合物和一种或多种载体、稀释剂或赋形剂。
