背景技术
癌症占日本人的死亡原因的第1位,一年约35万人死亡,目前仍然为严重的疾病。作为所确立的癌症疗法,主要有外科切除术、抗癌药物治疗及放射线疗法。但是,所述疗法存在复发、生命质量(QOL)降低、以及在为不能接受前面叙述的疗法的中期癌症的情况下没有其他治疗选项等的问题。
长期以来,癌症免疫疗法(癌症疫苗疗法)被期待作为新的疗法,从可以鉴定人类肿瘤抗原中的表位肽的1990年起,在全世界开始了癌症肽疫苗的临床研究。但是,肽单独施用或并用疗法中的临床研究成果的分析结果指出,在1,000例以上的给药例中,成功率仅为2.7%(非专利文献1),难以实用化。
另一方面,在日本还继续进行长期使用特有的癌症肽疫苗的临床研究,其成果逐渐变得显著。近年来,作为以提高治疗成绩为目标的方法之一,还开始采用施用多种癌症肽而不是施用1种癌症肽的战略。例如,通过预先检查患者的HLA分型及特异性免疫应答,实施了选择多种待施用的肽的特制(tailor-made)型的癌症肽疫苗疗法,且安全性及抗肿瘤效果得到确认。具体而言,通过特制型肽疫苗的单独施用或与抗癌剂的并用,在脑肿瘤、宫颈癌、前列腺癌、以及胰腺癌方面实现了优异的临床效果以及安全性(非专利文献2、3、4)。
在癌症肽疫苗疗法中,被认为是主要的效应细胞的表位特异性细胞毒性T淋巴细胞(以下,简称为CTL)引起的细胞性免疫是HLA限制性的,实施了以具有特定的HLA分型、且具体而言仅以具有对象患者数多的HLA-A2或HLA-A24分型的患者为对象的癌症肽疫苗开发。
但是,这2种HLA分型持有比例在日本人中分别为约40%、60%(非专利文献5),存在具有这2种以外的HLA分型的患者不能接受癌症肽疫苗的好处的问题。因此,在治疗开始前需要HLA分型检查成为使治疗的开始时期延迟、使患者负担增大的一个原因。由以上考虑,期望不需要HLA分型检查、可适用于全部癌症患者的癌症肽疫苗的开发、研究。
另外,已知在癌症肽疫苗疗法中,除了作为细胞免疫的CTL的活化之外,通过诱导作为体液免疫的免疫球蛋白产生,有助于延长寿命的效果(非专利文献6)。

发明内容
本发明的目的在于,提供一种癌症抗原肽,所述癌症抗原肽可作为癌症肽疫苗施用于广泛的癌症患者,而不需要HLA分型检查、且不限于具有特定HLA分型的患者。
本发明人等为了解决上述课题进行了潜心研究,结果发现:通过将选自报道具有HLA-A2、HLA-A24、HLA-A26或HLA-A3超型的任一种的限制性CTL的诱导能力或具有上述多种HLA-A限制性的CTL的诱导能力的CTL表位肽中的5个肽经由接头连接而得到的具有5个连接的CTL表位的肽,可以施用于广泛的癌症患者而不需要HLA分型检查、且不限于具有特定的HLA分型的患者,从而施用该具有5个连接的CTL表位的肽,由此,较强地诱导对构成该肽的CTL表位肽特异性的CTL及免疫球蛋白,直至完成了本发明。
即,本发明具有以下特征。

发明效果
根据本发明,可以提供一种癌症抗原肽,其可以作为癌症肽疫苗施用于广泛的癌症患者,而不需要HLA分型检查、且不限于具有特定的HLA分型的患者。
本发明的具有5个连接的CTL表位的肽可以施用于广泛的癌症患者,例如HLA-A2阳性患者群体、HLA-A24阳性患者群体、HLA-A26阳性患者群体、HLA-A3超型阳性患者群体,而不需要HLA分型检查,可以治疗和/或预防该患者中的癌症或起因于其的疾病。另外,由于观察到构成本发明的具有5个连接的CTL表位的肽的肿瘤抗原在多个癌症种类中表达,因此本发明的具有5个连接的CTL表位的肽可以作为用于治疗或预防各种癌症种类的药物组合物(更详细而言为免疫治疗剂)使用。进而,与将所构成的CTL表位肽进行混合而等量施用的情况相比,通过施用本发明的具有5个连接的表位的肽,可以更强地诱导CTL表位肽特异性CTL及免疫球蛋白,并且可以将抗肿瘤免疫更有效地活化。

具体实施方式
下面,进一步对本发明进行具体说明。
1.具有5个连接的表位的肽
本发明的“具有5个连接的表位的肽”是指:将选自来自相同和/或不同的肿瘤抗原分子的CTL表位肽中的5个肽经由接头以线性连接并设为1分子的肽。
“来自肿瘤抗原分子的CTL表位肽”为肿瘤抗原在肿瘤细胞内分解而产生的肽,其通过与HLA I类分子结合并呈递在细胞表面上,可以被肿瘤特异性CTL识别、和/或可诱导和/或活化肿瘤特异性CTL。“诱导肿瘤特异性CTL”是指:在生物体外或体内,使特异性地识别来自肿瘤抗原分子的CTL表位肽的CTL分化和/或增殖。另外,“活化肿瘤特异性CTL”是指:通过CTL识别由HLA I类分子所呈递的抗原,产生干扰素γ(IFN-γ)。在本说明书中,有时将“来自肿瘤抗原分子的CTL表位肽”简称为“CTL表位肽”。
作为本发明中使用的CTL表位肽,可以使用公知的CTL表位肽,可列举以下的CTL表位肽:

本说明书中,有时分别将序列号1所示的肽称为“PEP1”,序列号2所示的肽称为“PEP2”,序列号3所示的肽称为“PEP3”,序列号4所示的肽称为“PEP4”,序列号5所示的肽称为“PEP5”,序列号6所示的肽称为“PEP6”,序列号7所示的肽称为“PEP7”。
在本发明中,具有以下氨基酸序列且具有与原来的肽同等或比其更高的CTL诱导能力以及免疫球蛋白产生诱导能力的肽也可以作为“CTL表位肽”使用,所述氨基酸序列具有在PEP1、PEP2、PEP3、PEP4、PEP5、PEP6及PEP7的各氨基酸序列中的一个或多个氨基酸的置换、插入、缺失、和/或添加。在此,“多个”为1~3个、优选1~2个。作为这样的肽,可列举例如用具有与原来的氨基酸类似的特性的氨基酸置换(即,通过保守的氨基酸置换而得到的)肽。
PEP1、PEP2、PEP3、PEP4及PEP5被HLA-A2捕获,PEP2、PEP4、PEP5及PEP6被HLA-A24捕获,PEP2、PEP4、PEP5及PEP7被HLA-A3超型捕获,以及PEP2及PEP5被HLA-A26捕获,可以诱导和/或活化CTL。另外,编码这些CTL表位肽的基因被确认在多个癌症种类中表达(Yang D etal.,Cancer Res.1999,59:4056-63、Harashima N et al.,Eur.J.Immunol.2001,31(2),323-32)。
选自PEP1、PEP2、PEP3、PEP4、PEP5、PEP6及PEP7中(可以重复选择)的5个肽经由接头分别以线性连接。
接头只要可以在向生物体施用具有5个连接的表位的肽时被裂解、且被连接的CTL表位肽彼此被分离即可,可列举例如:酯键、醚键、酰胺键、糖链接头、聚乙二醇接头、氨基酸接头等。作为用作氨基酸接头的氨基酸序列的实例可包括精氨酸二聚体、赖氨酸二聚体、甘氨酸二聚体、甘氨酸五聚体、甘氨酸六聚体、丙氨酸-丙氨酸-酪氨酸(AAY)、异亮氨酸-亮氨酸-丙氨酸(ILA)、精氨酸-缬氨酸-赖氨酸-精氨酸(RVKR)等,优选为精氨酸二聚体。
所选择的CTL表位肽及其连接顺序,可以通过以下来确定:将以给定的组合及给定的连接顺序合成的具有5个连接的表位的肽施用于人类HLA-A转基因小鼠并评价各CTL表位肽特异性CTL诱导在生物体内的有无。
优选地,本发明的具有5个连接的表位的肽具有选自以下的(1)~(12)的一个或多个特征:
(1)含有PEP5和PEP2经由接头从N末端侧依次相邻的序列;
(2)含有PEP2和PEP4经由接头从N末端侧依次相邻的序列;

在本发明的具有5个连接的表位的肽中,对被连接的5种CTL表位肽的优选2种、进一步优选3种、更优选4种、最优选5种CTL表位肽,可以诱导和/或活化各CTL表位肽特异性CTL,以及可以诱导各CTL表位肽特异性免疫球蛋白产生。需要说明的是,在此,“免疫球蛋白”是指IgG、IgM、IgA、IgD。
2.具有5个连接的表位的肽的制造
本发明的具有5个连接的表位的肽可以通过常规的液相合成法、固相合成法等肽合成法、利用自动肽合成仪的肽合成法等来制造(Kelley et al.,Genetics Engineering Principles and Methods,Setlow,J.K.eds.,Plenum PressNY.(1990)Vol.12,p.1-19;Stewart et al.,Solid-Phase Peptide Synthesis(1989)W.H.Freeman Co.;Houghten,Proc.Natl.Acad.Sci.USA(1985)82:p.5132、《新生化学実験講座1タンパク質IV》(1992)日本生化学会编,东京化学同人)。就肽合成而言,准备各氨基酸的待被结合的α-氨基和α-羧基以外的官能团被保护的氨基酸类,以及在各氨基酸的α-氨基和α-羧基之间进行肽键形成反应。通常,将位于肽的C末端的氨基酸残基的羧基经由适当的间隔序列或接头与固相结合。将以上得到的二肽的氨基末端的保护基团选择性地除去,与随后的氨基酸的α-羧基之间形成肽键。连续进行这样的操作,制造具有保护的侧基的肽,最后除去全部的保护基团,并从固相中进行分离。保护基团的种类和保护方法、肽键合法的详细内容,详细地记载于上述的文献。
可选地,可以通过使用编码本发明的具有5个连接的表位的肽的核酸的基因重组法和噬菌体展示法等制造肽。
基因重组法包括:将编码本发明的具有5个连接的表位的肽的DNA插入适当的表达载体中,将载体导入适当的宿主细胞中,将细胞进行培养,从细胞内或从细胞外液回收目标肽。载体没有限定,例如为质粒、噬菌体、粘粒、噬菌粒、病毒等载体。用于导入载体的宿主细胞为大肠杆菌和枯草杆菌等细菌、酵母细胞、昆虫细胞、动物细胞(例如哺乳动物细胞)、植物细胞等,向这些细胞的转化或转染包括例如磷酸钙法、电穿孔法、脂质转染法、粒子枪法、PEG法等。将转化的细胞进行培养的方法按照用于宿主生物培养的常规方法进行。为了使本发明的肽容易回收,优选使通过表达而生成的肽分泌于细胞外。为此,将编码使肽能够从该细胞分泌的肽序列的DNA与编码目标肽的DNA的5’末端侧结合。可选地,也可以回收累积在细胞内的目标肽。该情况下,将细胞物理性或化学性地破坏,并使用蛋白质纯化技术回收目标肽。
所制造的肽可以通过常规方法,例如通过色谱法诸如凝胶过滤色谱法、离子交换柱色谱法、亲和色谱法、反相柱色谱法、HPLC、硫酸铵分级、超滤法、免疫吸附法等进行回收或纯化。
3.细胞毒性T淋巴细胞
使用本发明的具有5个连接的表位的肽,可以在生物体外获得损害癌细胞的CTL表位肽特异性的CTL。用于使用CTL表位肽生物体外诱导CTL的方法为公知的(例如日本特开2006-14637号公报),且在本发明中也可以利用这些方法。例如,将来自健康人或癌症患者的外周血单核细胞(PBMC)中的板粘附细胞在细胞因子诸如GM-CSF、IL-4等的存在下进行培养,以诱导树状细胞(DC)。用本发明的具有5个连接的表位的肽冲击致敏该树状细胞之后,进行X射线照射,以制备抗原递呈细胞(刺激物)。在不能使用DC的情况下,可以使用本发明的具有5个连接的表位的肽冲击致敏来自健康人或相同的癌症患者的外周血单核细胞(PBMC)之后,进行了X射线照射的物质。接着,加入来自健康人的外周血单核细胞(PBMC)或来自癌症患者的外周血单核细胞(PBMC)或相关淋巴节中的淋巴细胞(应答物),在细胞因子诸如IL-2、IL-4、IL-7等存在下进行培养。其后,加入本发明的具有5个连接的表位的肽再次刺激如上述那样经由脉冲致敏而得到的抗原呈递细胞,并在细胞因子诸如IL-2等存在下进一步培养。
作为用于CTL诱导的细胞培养用培养基,使用T淋巴细胞可以生存的任何培养基即可。例如,可以使用在RHAMα培养基(Kawai,K.,Sasaki,T.,Saijo-Kurita,K.,Akaza,H.,Koiso,K.,and Ohno,T.,Cancer Immunol.Immunother.35,225-229,1992中所记载的LAK medium)、AIMV培养基(GIBCO BRL,Life Technologies,INC.)、或RPMI1640培养基等中添加多种细胞因子诸如IL-2等和胎牛血清(FCS)等的培养基。
培养条件按照本领域技术人员公知的条件即可。例如,将培养温度设为33℃~41℃、优选37℃。另外,可以将含有空气或适当浓度的氧和为了将培养基的pH保持在约7.4而含有适当浓度的二氧化碳(例如5%CO2)的惰性气体作为气相使用。优选培养4~10天,更优选7天。通过进行这样的培养而诱导的CTL含有对构成具有5个连接的表位的肽的5个CTL表位肽中优选2种、进一步优选3种、更优选4种、最优选5种CTL表位肽的各CTL表位肽特异性的CTL,并因此它们可以特异性地损害癌细胞。
4.药物组合物
本发明的具有5个连接的表位的肽可以作为用于癌症的免疫疗法中使用的药物组合物的有效成分利用。
在本发明的药物组合物中,可以含有一种或多种上述的具有5个连接的表位的肽作为有效成分。通过含有多种具有5个连接的表位的肽,可以得到更强的效果。
观察到编码本发明的具有5个连接的表位的肽中所含的PEP1、PEP2、PEP3、PEP4、PEP5、PEP6及PEP7的基因在多个实体癌症及血液癌症中表达。作为实体癌症,可列举例如:脑肿瘤、肺癌、乳腺癌、甲状腺癌、宫颈癌、子宫体癌、卵巢癌、食道癌、胃癌、GIST、胰腺癌、结肠癌、直肠癌、肛门癌、肾癌、肝癌、胆道癌、头颈癌、膀胱癌、前列腺癌、恶性黑色素瘤、皮肤癌、舌癌、骨肉瘤、软骨肉瘤、纤维肉瘤、脂肪肉瘤、血管肉瘤、横纹肌肉瘤、平滑肌肉瘤等。作为血液癌症,可列举例如:白血病、恶性淋巴瘤、骨髓瘤等。因此,本发明的具有5个连接的表位的肽对这些癌症的治疗或预防是有用的。在此,“癌症的治疗或预防”是指预防癌症的发生/复发、抑制癌症的进展/恶化、改善癌症的症状。
本发明的药物组合物可以含有常规的各种有机或无机载体物质等作为药学上可接受的制剂原料。作为可以利用的药物载体,可以例示根据制剂的施用形式而通常使用的稳定剂、抗菌剂、缓冲剂、等渗剂、螯合剂、pH调节剂、表面活性剂(界面活性剤)、填充剂、增稠剂、粘结剂、湿润剂、崩解剂、表面活性剂(表面活性剤)、润滑剂、舒缓剂、稀释剂或赋形剂等,优选制备这些物质作为利用常规方法添加的配合剂。
另外,本发明的药物组合物可以含有已知在疫苗的施用时使用的佐剂。作为佐剂,可以列举:完全弗氏佐剂(CFA)、不完全弗氏佐剂(IFA)、明矾、脂质A、单磷酰脂质A、BCG(Bacillus-Calmette-Guerrin)等细菌制剂、结核菌素等细菌成分制剂、钥孔虫戚血兰素和酵母甘露聚糖等天然高分子物质、胞壁酰三肽或胞壁酰二肽或它们的衍生物、白矾(alum)、非离子性嵌段共聚物等,可以使用这些物质的1种或使用2种以上的组合。佐剂可以与本发明的药物组合物以混合物形式或作为乳液同时进行施用。
进而,除上述具有5个连接的表位的肽之外,本发明的药物组合物可含有以下的一种或多种组合:公知的来自肿瘤抗原分子的CTL表位肽或含有其的肽、或将它们连接而成的肽(以下,记载为“公知的来自肿瘤抗原分子的CTL表位肽等”)。作为公知的来自肿瘤抗原分子的CTL表位肽,可列举例如:WT-1 p126-134、修饰的(M236Y)WT-1 p235-243、NY-ESO-1p157-165、修饰的(T210M)gp100 p209-217、存活素-2B p80-88、Her-2/neup63-71、VEGFR2 p169-177、MART-1 p26-35、磷脂酰肌醇蛋白聚糖-3p298-306、SPARC p143-151等,但并不限定于这些。该情况下,本发明的具有5个连接的表位的肽和公知的来自肿瘤抗原分子的CTL表位肽等既可以为单一剂型的制剂形式,也可以是,含有本发明的具有5个连接的表位的肽作为有效成分的制剂和含有公知的来自肿瘤抗原分子的CTL表位肽等作为有效成分的制剂为不同的制剂形式。
本发明的药物组合物可以根据施用形式而选择制剂形式。作为代表性的制剂形式,可以制作液体制剂、乳剂、脂质体制剂、脂质乳剂、环糊精等包合物、悬浮剂、软膏剂、霜剂、经皮吸收剂、经粘膜吸收剂、片剂、丸剂、胶囊剂、散剂、粉末剂、颗粒剂、细粒剂、糖浆剂等,但并不限定于这些。这些制剂根据施用途径而进一步分类为经口剂、非经口剂、经鼻剂、经阴道剂、栓剂、舌下剂、吸入剂、点眼剂、点耳剂等,且这些制剂可以分别按照常规的方法进行调制、成形或制备。另外,除可以作为液体制剂使用之外,这些制剂也可以设为可进行冷冻干燥并保存的状态之后,在使用时用含有水和生理盐水等的缓冲液等进行溶解而制备成适当的浓度后使用。
另外,本发明的药物组合物可以含有使用上述本发明的具有5个连接的表位的肽在生物体外诱导的CTL作为有效成分。这样的药物组合物优选为非经口剂的形式。
5.治疗方法
本发明的药物组合物可以施用于广泛的癌症患者、例如选自由HLA-A2、HLA-A24、HLA-A26及HLA-A3超型构成的组中的HLA分型阳性患者群体,可以在治疗开始前不需要HLA分型检查而开始治疗。
本发明的药物组合物通过对癌症患者进行施用,可以诱导和/或活化构成作为有效成分的具有5个连接的表位的肽所含有的2种以上、优选3种以上、更优选4种以上、进一步优选5种的CTL表位肽特异性CTL,以及可以诱导该CTL表位肽特异性免疫球蛋白产生。作为有效成分所含有的具有5个连接的表位的肽的施用引起的免疫球蛋白产生的诱导,与具有5个连接的表位的肽中所含的CTL表位肽没有连接而各自地混合施用时所观察到的免疫球蛋白产生的诱导相比显著地更高,因此,本发明中的具有5个连接的表位的肽可以有效地治疗或预防癌症患者中的癌症。
本发明的药物组合物可以经由经口施用、静脉施用、动脉施用、肌肉内施用、皮下施用、皮内施用、舌下施用、腹腔内施用、直肠内施用、经皮施用、经粘膜施用、经鼻施用、经阴道施用、经眼施用、吸入施用等进行施用。含有多种具有5个连接的表位的肽、或公知的来自肿瘤抗原分子的CTL表位肽等作为有效成分的制剂分别作为不同的药物组合物进行制备的情况下,各药物组合物可以通过相同或不同的施用途径、同时或非同时进行施用。
本发明的药物组合物的施用量可以根据待治疗的癌症的状态/严重度、每个患者的年龄、体重等主要因素而适当调整,但优选对以0.000l mg~1000mg、优选0.00l mg~100mg、更优选0.01mg~50mg的量含有具有5个连接的表位的肽的药物组合物,将其在数日、数周或数个月重复施用1次。另外,在含有使用本发明的具有5个连接的表位的肽在生物体外进行诱导的CTL作为本发明的药物组合物的有效成分的情况下,优选每1天每kg体重以1周~2周的间隔施用2×106~2×108个CTL。
本发明的药物组合物也可以与通常用于癌症化疗的药品并用而施用于癌症患者。可列举例如:环磷酰胺、替莫唑胺、苯达莫司汀等烷基化剂,替加氟-尿嘧啶配合剂、替加氟-吉莫斯特-氧嗪酸钾配合剂、甲氨蝶呤、吉西他滨等代谢拮抗剂,顺铂、奥沙利铂等含铂制剂,依立替康、艾日布林、紫杉醇、多西他塞、长春新碱等植物生物碱制剂,阿霉素、博来霉素、放射菌素D等抗癌抗生素,伊马替尼、舒尼替尼、吉非替尼、索拉非尼、依维莫司、曲妥珠单抗、贝伐单抗、利妥昔单抗、西妥昔单抗、帕尼单抗、モガムリズマブ等分子靶向治疗剂,比卡鲁胺、雌莫司汀、依西美坦等激素治疗剂等。这些药品可以通过与本发明的药物组合物相同或不同的施用途径、同时或非同时进行施用。
以下,通过实施例对本发明进行具体说明,但本发明并不限定于这些实施例。
实施例
实施例1肽的合成、纯化
CTL表位肽及具有5个连接的表位的肽使用市售的肽合成仪Prelude(Protein Technologies,Inc.),通过固相合成法(Fmoc法)进行合成。将得到的各种合成肽用YMC-Pack Pro C18柱(YMC Co.,Ltd.)及HPLC系统(Gilson)进行纯化,在冷冻干燥后冷温暗处保存,供于以下所示的实施例。
将合成的CTL表位肽的氨基酸序列示于表1,将具有5个连接的表位的肽的氨基酸序列示于表2。
需要说明的是,分别合成与HLA-A2结合的作为对照肽的WT1(序列号8)、与HLA-A24结合的作为对照肽的Her2(序列号9)。

实施例2小鼠模型中的表位肽特异性CTL的诱导以及利用ELISPOT法的CTL的检测
将具有5个连接的表位的肽用大冢蒸馏水(大冢制药工场)制备成2mg/mL或4mg/mL,填充于B Braun Injekt注射器。在其它注射器中填充等量的不完全弗氏佐剂(IFA)之后,将两注射器用GP注射器连接器连接,通过使具有5个连接的表位的肽溶液和IFA充分混合而制备乳液。将其在小鼠(HLA-A2.1转基因的,HLA-A24转基因的(Taconic))的尾根部周围每周1次各100μL、总计施用2次。末次施用1周后,从小鼠中回收腹股沟淋巴节。将淋巴节细胞悬浮液以完全培养基(RPMI-1640,10%热灭活的FBS,100U/mL青霉素,100μg/mL链霉素,50μM 2-巯基乙醇)制备成5×106个细胞/mL,分别加入靶标CTL表位肽(最终浓度10μg/mL)、重组小鼠IL-15(最终浓度100ng/mL)、重组小鼠IL-21(最终浓度100ng/mL),以1mL/孔接种于24孔板之后,在37℃、5%CO2培养箱内培养8天。8天后,回收细胞,在包括在鼠IFN-γELISpot试剂盒(GEN-PROBE)中的抗IFN-γ抗体固定化板上以1×105细胞/孔接种。接着,将由同系小鼠脾制备并照射有30Gy的X射线的脾细胞以1×105细胞/孔接种于相同的孔作为抗原呈递细胞后,加入靶标CTL表位肽、或阴性对照肽(最终浓度:10μg/mL),在37℃、5%CO2培养箱内培养一夜。第二天,按照试剂盒中包含的说明使产生IFN-γ的细胞的斑点显色。产生IFN-γ的细胞的斑点数用ELISPOT分析仪(Immunospot S6,Cellular Technology Ltd.)进行定量。就CTL表位肽特异性CTL的诱导而言,通过该实验得到的靶标CTL表位肽添加孔的产生IFN-γ的细胞的斑点数与阴性对照肽添加孔的产生IFN-γ的细胞的斑点数相比显著地高(Student’s t-test,p<0.05)的情况下,判断为阳性。需要说明的是,作为阴性对照肽,使用WT1或Her2。进而,为了将CTL诱导强度进行可视化,设为(靶标CTL表位肽添加孔的产生IFN-γ的细胞斑点的平均数)-(阴性对照肽添加孔的产生IFN-γ的细胞斑点的平均数)=Δ时,将10≤Δ<100的情况作为阳性表示,将100≤Δ<200的情况作为中阳性表示,将200≤Δ的情况作为强阳性表示。
将CTL表位肽特异性CTL诱导的研究结果示于图1。如图1所示,具有5个连接的表位的肽中任一部分的序列为PEP5-RR-PEP2、或PEP4-RR-PEP2、或C末端为PEP3的具有5个连接的表位的肽显示至少3种以上的CTL诱导。另一方面,在TPV18那样的连接顺序的5连接体中,显示不多于2种的CTL诱导。
由以上的结果来看,对于已知CTL表位肽,即使将经由精氨酸二聚体而连接的具有5个连接的表位的肽施用于小鼠,也显然不一定能够诱导全部的CTL表位肽特异性的表位特异性CTL。
实施例3CTL表位肽固定化的珠的制备
按以下的步骤将肽固定化在xMAP Multi-Analyte COOH Microspheres(Luminex corporation、以下称为串珠)上。用MES缓冲液(0.1M MES-NaOH,pH7.0)清洗珠之后,通过离心分离除去上清液。将其重复两次之后,将珠悬浮在75μL的MES缓冲液中。此时,加入制备的1mg/mL的CTL表位肽溶液100μL,进一步加入10mg/mL EDC(1-乙基-3-(3-二甲氨基丙基)碳二亚胺盐酸盐)5μL并充分混合之后,在30℃、暗处反应一夜。第二天,通过离心分离而除去上清液后,加入1M Tris-HCl 125μL,在30℃、暗处孵育30分钟。除去上清液后,用漂洗缓冲液(PBS(-),0.05%Tween20)清洗两次,其后悬浮于免疫封闭剂(DS Pharma Biomedical),由此完成CTL表位肽的固定化。
实施例4CTL表位特异性IgG抗体滴度测定
用大冢蒸馏水(大冢制药工场)以2mg/mL制备本发明的具有5个连接的表位的肽,填充于B Braun Injekt注射器。在其它注射器中填充等量的IFA之后,将两注射器用GP注射器连接器连接,使该具有5个连接的表位的肽溶液和IFA充分混合,由此制备乳液。将其在CBF1小鼠(C57BL/6×Balb/c F1)尾根部周围每周1次各100μL、总计施用3次。末次施用1周后,从小鼠采血,得到血清样品。作为对照组,使用PEP2、PEP3、PEP4、PEP5、PEP7的各肽混合物(各1mg/mL)制备乳液,按与具有连接的5个表位的肽相同的日程进行施用(混合物施用组)。
在96孔过滤板中分配以免疫封闭剂稀释的CTL表位肽固定化的珠,用漂洗缓冲液清洗后,以100μL/孔添加通过免疫封闭剂稀释了200倍的小鼠血清样品。将板在30℃培养器中以600rpm搅拌,培养90分钟。用漂洗缓冲液清洗3次后,以100μL/孔添加通过免疫封闭剂稀释了500倍的生物素化的抗小鼠IgG(H+L)(Vector Labolatories),在30℃、600rpm搅拌条件下培养60分钟。用漂洗缓冲液清洗3次后,以100μL/孔添加通过免疫封闭剂稀释了500倍的链霉亲和素-R-藻红蛋白缀合物(Invitrogen),在30℃、600rpm搅拌条件下培养30分钟。用漂洗缓冲液清洗3次后,以100μL/孔添加漂洗缓冲液并将珠悬浮后,用Bio-Plex Suspension Array System(BIO-RAD)测定各珠特异性结合的PE色素的平均荧光强度。
CTL表位特异性IgG抗体滴度测定结果如图2所示。需要说明的是,图2表示:通过将等量的IFA和大冢蒸馏水(大冢制药工场)混合制备的乳液,按与CTL表位肽混合物以及具有5个连接的表位的肽相同的日程施用于小鼠的阴性对照组(以下,简称为IFA组)的血清中CTL表位特异性IgG抗体滴度测定结果的比较,诱导了几倍的该IgG抗体的产生。
如图2所示,与IFA组相比,在混合物施用组中没有观察到CTL表位特异性IgG抗体产生的诱导。另一方面,确定了,施用具有5个连接的表位的肽的情况下,以高频率观察到强的CTL表位特异性IgG抗体的产生的诱导,进而,该IgG产生的量通过所施用的具有5个连接的表位的肽而显著地诱导数十倍至数百倍以上。另一方面,在TPV17那样的连接顺序的5连接体中,没有观察到多个CTL表位特异性IgG抗体产生的诱导。
以上的结果显示:与将构成具有5个连接的表位的肽的CTL表位肽进行混合而施用相比,通过施用该具有5个连接的表位的肽将抗肿瘤免疫较强地活化。
接受了癌症肽疫苗治疗的癌症患者中的CTL表位肽特异性IgG产生的诱导与表位肽特异性CTL的诱导都有助于延长寿命的效果。因此,本发明与由已知的CTL表位肽和其混合物构成的癌症肽疫苗疗法相比,可以产生优异的延长寿命的效果。因此,本发明的具有5个连接的表位的肽可以作为癌症或起因于癌症的疾病的治疗剂和/或预防剂、癌症肽疫苗优选使用,其与由已知的CTL表位肽和其混合物构成的癌症肽疫苗疗法相比,可以提高治疗成绩。

Claims (10)
1.一种具有5个连接的表位的肽,其从由以下的CTL表位肽:序列号1所示的肽(PEP1)、序列号2所示的肽(PEP2)、序列号3所示的肽(PEP3)、序列号4所示的肽(PEP4)、序列号5所示的肽(PEP5)、序列号6所示的肽(PEP6)、及序列号7所示的肽(PEP7)构成的组中可以重复选择的5个肽经由接头分别连接而成,并且
所述具有5个连接的表位的肽具有选自以下的(1)~(12)的一个或多个特征:
(1)含有PEP5和PEP2经由接头从N末端侧依次相邻的序列;
(2)含有PEP2和PEP4经由接头从N末端侧依次相邻的序列;
(3)含有PEP4和PEP6经由接头从N末端侧依次相邻的序列;
(4)含有PEP6和PEP3经由接头从N末端侧依次相邻的序列;
(5)含有PEP4和PEP1经由接头从N末端侧依次相邻的序列;
(6)含有PEP1和PEP3经由接头从N末端侧依次相邻的序列;
(7)含有PEP4和PEP2经由接头从N末端侧依次相邻的序列;
(8)含有PEP1和PEP4经由接头从N末端侧依次相邻的序列;
(9)含有PEP5和PEP1经由接头从N末端侧依次相邻的序列;
(10)含有PEP6和PEP5经由接头从N末端侧依次相邻的序列;
(11)在C末端含有PEP2;以及
(12)在C末端含有PEP3。
2.根据权利要求1所述的具有5个连接的表位的肽,其包含在N末端含有PEP5和PEP2、PEP6和PEP5、或PEP4和PEP6经由接头从N末端侧依次相邻的序列和/或在C末端含有PEP7和PEP3、PEP4和PEP3、PEP6和PEP3、PEP1和PEP3、PEP5和PEP2、PEP4和PEP2、或PEP5和PEP3经由接头从N末端侧依次相邻的序列。
3.根据权利要求2所述的具有5个连接的表位的肽,其含有选自以下的(a)~(p)中的序列:

7.一种用于治疗或预防癌症的药物组合物,其含有根据权利要求1~5中任一项所述的具有5个连接的表位的肽、或根据权利要求6所述的CTL作为有效成分。
8.根据权利要求7所述的药物组合物,其为免疫治疗剂。
9.一种癌症的治疗方法,其包括:将根据权利要求1~5中任一项所述的具有5个连接的表位的肽、或根据权利要求6所述的CTL施用于癌症患者。
10.一种针对癌症免疫受试者的方法,其包括:将根据权利要求1~5中任一项所述的具有5个连接的表位的肽、或根据权利要求6所述的CTL施用于受试者。
