本发明公开了一种鸡腿菇多糖颗粒剂及其制备方法。采用利用市场上生产出的鸡腿菇,辅以其他辅料制成鸡腿菇多糖颗粒剂,其各成分配比如下:本发明利用市场上的鸡腿菇多糖,开发出鸡腿菇多糖颗粒,使用方便,可冲水服用,生产出的鸡腿菇多糖具有明显的降血糖活性,鸡腿菇多糖成分直接作用于血糖,不会对人的胰岛功能产生影响,所以对需注射胰岛素的1型糖尿病效果不佳,但对2型糖尿病早期患者或血糖持续保持较高的患者在连续食用1至3个月后,降糖效果明显。利用此多糖开发出的产品具有明显的保健功能,极具开发空间。

背景技术
鸡腿菇(Coprinus comatus)学名毛头鬼伞,又名鸡腿蘑,属真菌门、担子菌亚门、层菌纲、伞菌目、鬼伞科、鬼伞属。因其形如鸡腿、肉质肉味似鸡丝而得名,是近年来人工开发生产的一种具有商业潜力的珍惜食用菌类,被世界卫生组织及联合国粮农组织确定为集“天然、营养、保健”三种功能于一体的16种珍稀食用菌之一,并被推荐进行大力推广。由于鸡腿菇具高蛋白、低脂肪,氨基酸种类齐全等特点,被誉为“菌种新秀”。
鸡腿菇中除含有丰富蛋白质外,还有丰富的糖类,据测定,干品鸡腿菇中(水分含量为0.2%)总糖含量为57.65%,还原糖含量为53.54%,果糖含量为4.11%。鸡腿菇多糖具有调节血糖的作用,在深层发酵培养基中添加适量的中药,是鸡腿菇在代谢过程中可能对中药的一些成分进行转化,而中药的某些成分也可能促进鸡腿蘑的生长和活性物质产生,从而进一步增强鸡腿菇降血糖的功效。另外,鸡腿菇多糖能提高机体免疫力,抑制肿瘤的生长,具有预防和治疗癌症的功能。除此之外,现代药理研究表明,鸡腿菇多糖还具有调血脂、抑菌、抗氧化,提高免疫力等多种生理活性。随着其食用价值和药用价值的不断研究与开发,鸡腿菇将会越来越受到人们的青睐,因此对鸡腿菇资源的开发具有重要意义。
目前,鸡腿菇多糖应用的剂型有注射剂、胶囊、滴丸、口服液等。上述鸡腿菇多糖剂型应用时有一定的局限性,如口服液剂型给药繁琐,操作不方便,注射剂型对机体刺激性大等。
发明内容
本发明目的在于提供一种鸡腿菇多糖颗粒剂。这种颗粒剂不易吸潮,不易结块,流动性好,服用方便,可冲入水中饮服,同时药物含量稳定。
本发明另一目的在于提供上述鸡腿菇颗粒剂的制备方法。
本发明还一目的在于提供上述鸡腿菇多糖颗粒剂的用途。
实现本发明的技术方案如下:
本发明一种鸡腿菇颗粒剂,其原料组成成分及其重量配比为:鸡腿菇多糖1%-6%,填充剂70%-85%,粘合剂15%-40%,水0.5%-3%。
上述鸡腿菇多糖颗粒剂中所述的填充剂的重量百分比优选为75%-85%。
上述鸡腿菇多糖颗粒剂中所述的粘合剂的重量百分比优选为20%-30%。
上述鸡腿菇多糖颗粒剂中所述的水的重量百分比优选为0.5%-2%。
上述鸡腿菇多糖颗粒剂中所述的填充剂为:糖粉、乳糖、甘露醇、糊精、β-环糊精、山梨醇、木糖醇、淀粉、预胶化淀粉、碳酸钙、微晶纤维素或无机盐中的一种或多种混合物;优选为糖粉。
上述鸡腿菇多糖颗粒剂中所述的粘合剂为:聚维酮K30、糖浆、胶浆、糊精、甲基纤维素或羟丙甲基纤维素中的一种或多种混合物;优选为聚维酮K30。
上述鸡腿菇多糖、填充剂和粘合剂等原料成分皆可于市场上公开购买得到。
上述鸡腿菇多糖颗粒剂的制备方法,包括如下步骤:
(1)将鸡腿菇多糖、粘合剂、填充剂分别干燥,粉碎;鸡腿菇多糖过100目筛,粘合剂、填充剂过80目筛,备用;
(2)按照比例将过筛后的鸡腿菇多糖、填充剂至于混合机中,再按照比例逐渐加入粘合剂和水,混匀制成软材;
(3)将软材过16目筛制成湿颗粒,然后60℃鼓风烘干后,用20目整理,即得鸡腿菇多糖颗粒剂。
上述鸡腿菇多糖颗粒剂在制备降血糖药物上的应用。
上述鸡腿菇多糖颗粒的使用方法,冲水服用。
与现代技术相比,本发明具有的优点:(1)本发明鸡腿菇多糖颗粒剂使用方便,可直接冲入水中饮服;(2)本发明鸡腿菇颗粒剂的飞散性、附着性、团聚性、吸湿性等均较少,药物含量稳定。利用此多糖开发出的产品具有明显的保健功能,极具开发空间。

实施例1鸡腿菇多糖颗粒剂的制备
按照如下方法制备:
(1)鸡腿菇多糖(购自杭州众菇善年生物技术有限公司,规格10g/袋,以下同)、糖粉(购自天津博迪化工股份有限公司,规格为25kg/袋,以下同)、羟丙甲基纤维素(购自安惠山和药用辅料有限公司,规格为25kg/袋)等分别干燥,粉碎;鸡腿菇多糖过100目筛,糖粉、羟丙甲基纤维素过80目筛备用。
(2)将鸡腿菇多糖60g、糖粉720g置于混合机中,然后逐渐加入羟丙甲基纤维素360g、水60g,混匀制成软材;将软材过16目筛制成湿粒,然后60℃鼓风烘干后,用20目整粒,然后分装,密封,包装即得鸡腿菇多糖颗粒剂。
实施例2鸡腿菇多糖颗粒剂的制备
按照如下方法制备:
(1)将鸡腿菇多糖、甘露醇(购自河南华森生物科技有限公司,规格为25kg/袋)、糊精(购自山东西王糖业有限公司,规格为25kg/袋)等分别干燥,粉碎;鸡腿菇多糖过100目筛,糊精、甘露醇过80目筛备用;
(2)将鸡腿菇多糖50g、甘露醇1250g置于混合机中,逐渐加入糊精300g和水40g,混匀制成软材;将歉材过16目筛制成湿粒,然后60℃鼓风烘干后,用20目整粒,然后分装,密封,包装即得鸡腿菇多糖颗粒剂。
实施例3鸡腿菇多糖颗粒剂的制备
(1)将鸡腿菇多糖、糖粉、聚维酮K30(购自西安悦来医药科技有限公司,规格为25kg/桶)等分别干燥,粉碎;鸡腿菇多糖过100目筛,糖粉、聚维酮K30过80目筛备用;
(2)将鸡腿菇多糖30g、糖粉420g置于混合机中,逐渐加入聚维酮K30120g、水30g,混匀制成软材;将软材过16目筛制成湿粒,然后60℃鼓风烘干后,用20目整粒,然后分装,密封,包装即得鸡腿菇多糖颗粒剂。
实施例4鸡腿菇多糖颗粒剂的药效对比试验
按照如下方法进行:

1.1.1实验动物:昆明种小鼠50只,清洁级,雌雄各半,体重28+3g,购于厦门大学实验动物中心。
1.1.2材料与试剂:以上实施例所制备的鸡腿菇多糖颗粒剂,四氧嘧啶:Sigma公司产品,血糖测试仪:台欣生物科技研发股份有限公司
1.2方法
1.2.1糖尿病模型建立:实验动物饲喂普通饲料1w适应环境,小鼠禁食12h,正常饮水,基础称重,腹腔注射1%四氧嘧啶260mg/kg bw,72h后,尾尖采血,用血糖仪测空腹血糖值,选择血糖值>11.1mmol/L的小鼠,确定为糖尿病小鼠。
1.2.2动物分组处理:取正常小鼠8只,雌雄各半,设为正常组,饲喂普通饲料。将造模成功的16只糖尿病小鼠,随机分成2组,每组8只,雌雄各半。糖尿病模型对照组,饲喂普通颗粒,治疗组饲喂鸡腿菇多糖颗粒。饲养过程中,每只小鼠平均喂食量为6-7g/d,自由饮水,每天换水,隔日换垫料。整个实验期为21d。

2结果与分析
2.1鸡腿菇多糖颗粒对糖尿病小鼠体重和血糖的影响
注:a,p<0.05,与正常组相比;b,p<0.05,与糖尿病组相比。
2.2结果分析糖尿病组小鼠体重明显减轻,血糖浓度明显升高,与正常组比有显著性差异。与糖尿病组比较,经过饲喂鸡腿菇多糖颗粒的糖尿病小鼠,体重均有一定程度的恢复,血糖浓度明显降低。因此,鸡腿菇多糖颗粒可作为一种辅助降血糖药物或保健食品,有较高的研究价值。
Claims (1)
1.一种降糖鸡腿菇多糖颗粒的生产方法,其特征在于其各成分配比如下:
鸡腿菇多糖 30g
糖粉 420g
聚维酮K30 120g
水 30g,
制备方法如下:
(1)、将鸡腿菇多糖、糖粉、聚维酮K30分别干燥,粉碎;鸡腿菇多糖过100目筛,糖粉、聚维酮K30过80目筛,备用;
(2)、按照比例将过筛后的鸡腿菇多糖、糖粉至于混合机中,再按照比例逐渐加入聚维酮K30和水,混匀制成软材;
(3)、将软材过16目筛制成湿颗粒,然后60℃鼓风烘干后,用20目整粒,然后分装,密封,包装即得鸡腿菇多糖颗粒剂。
