本专利为一株产巴卡亭III的红豆杉内生真菌,种类是Diaporthe?phaseolorum(Cooke&Ellis)Sacc.用液体PDA培养基在室温条件下按照每分钟80-100转的震荡培养方式培养这种菌8-10天后,包含培养液和菌丝的培养物中巴卡亭III(baccatin?III)含量达到0.8mg/L以上。该菌株液体培养产生的巴卡亭III能够用于人工合成紫杉醇。

Description

一株产巴卡亭III的红豆杉内生真菌
技术领域:本专利属于生物工程技术领域,具体地说属于一个真菌菌株,可用于生产巴卡亭III,进而由此人工合成紫杉醇。
背景技术:紫杉醇是治疗癌症的一种有效天然化学成分,但紫杉醇在红豆杉体内含量很低,在万分之一以下。产紫杉醇的红豆杉植物资源十分有限,红豆杉已被列为国家保护植物,不能任意砍伐;栽培红豆杉生长也十分缓慢,还存在着紫杉醇含量更低的问题。目前已有人从红豆杉中分离得到产紫杉醇的内生真菌,不过产率很低,还达不到工业化生产水平。紫杉醇合成的一种前体物是巴卡亭III,由巴卡亭III可方便地人工合成紫杉醇,但巴卡亭III在红豆杉体内含量也很低,生产廉价的巴卡亭III是解决紫杉醇药源紧缺问题的一个有效技术措施。
发明内容:从太行山区的南方红豆杉植物体内分离得到一株产巴卡亭III(baccatinIII)的内生菌,经鉴定种名为Diaporthephaseolorum(Cooke&Ellis)Sacc。该菌种已于2012年12月6日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏单位地址为北京市朝阳区北辰西路1号院3号,保藏编号为CGMCCNo.6952。用液体PDA培养基在室温条件下按照每分钟80-100转的震荡培养方式培养这种菌8-10天后,包含培养液和菌丝的培养物中巴卡亭III含量达到0.8mg/L以上。该菌株液体培养产生的巴卡亭III能够用于人工合成紫杉醇。
具体实施方式:先制备出本专利菌株的种子液,再把种子液用液体PDA培养基震荡培养(80-100转/分钟)8-10天后,从培养物中提取巴卡亭III。

1.一株产巴卡亭III的红豆杉内生真菌,菌株保藏号为CGMCCNo.6952,菌种为Diaporthephaseolorum(Cooke&Ellis)Sacc,其特征在于能够产生化学物质巴卡亭III(baccatinIII)。
2.根据权利要求1所述的产巴卡亭III菌株,其特征在于用液体PDA培养基在室温条件下按照每分钟80-100转的震荡培养方式培养这种菌8-10天后,包含培养液和菌丝的培养物中巴卡亭III含量达到0.8mg/L以上。
3.根据权利要求1所述的产巴卡亭III菌株,其特征在于这种真菌为从红豆杉植物体内分离得到的内生菌,菌丝在PDA培养基上室温条件下能够正常生长,在PDA培养基上生长的菌丝白色,菌落呈放射状,有基生菌丝和气生菌丝,气生菌丝稍稀,菌落贴着培养基。
