本专利为一株高产紫杉醇的红豆杉内生真菌,种类是苜蓿茎点霉Phoma medicaginis。用液体PDA培养基在室温条件下按照每分钟80‑100转的震荡培养方式培养这种菌8‑10天后,包含培养液和菌丝的培养物中紫杉醇(paclitaxel)含量达到1.5mg/L以上。

Description

一株高产紫杉醇的红豆杉内生真菌
技术领域:本专利属于生物工程技术领域,具体地说属于一个真菌菌株,可用于液体发酵生产紫杉醇。
背景技术:紫杉醇是治疗癌症的一种有效天然化学成分,但紫杉醇在红豆杉体内含量很低,在万分之一以下。产紫杉醇的红豆杉植物资源十分有限,红豆杉已被列为国家保护植物,不能任意砍伐;栽培红豆杉生长也十分缓慢,还存在着紫杉醇含量更低的问题。在此之前已有人从红豆杉中分离得到产紫杉醇的内生真菌,不过产率很低,还达不到工业化生产水平。分离选育出高产紫杉醇的内生真菌并用于液体发酵生产是解决紫杉醇药源紧缺问题的一个有效技术措施。
发明内容:从太行山区的南方红豆杉植物体内分离得到一株产紫杉醇(paclitaxel)的内生菌,经鉴定种名为苜蓿茎点霉Phoma medicaginis。该菌种已于2015年12月17日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏单位地址为北京市朝阳区北辰西路1号院3号,保藏编号为CGMCC No.11812。用液体PDA培养基在室温条件下按照每分钟80-100转的震荡培养方式培养这种菌8-10天后,包含培养液和菌丝的培养物中紫杉醇含量达到1.5mg/L以上。
具体实施方式:先制备出本专利菌株的种子液,再把种子液用液体PDA培养基震荡培养(80-100转/分钟)8-10天后,从培养物中提取紫杉醇。

1.一株高产紫杉醇的红豆杉内生真菌,其特征在于,该真菌为苜蓿茎点霉(Phomamedicaginis),能够高效产生化学物质紫杉醇,保藏号为CGMCC No.11812。
2.根据权利要求1所述的高产紫杉醇的红豆杉内生真菌,其特征在于,用液体PDA培养基在室温条件下按照每分钟80-100转的震荡培养方式培养8-10天后,包含培养液和菌丝的培养物中紫杉醇含量达到1.5mg/L以上。
3.根据权利要求1所述的高产紫杉醇的红豆杉内生真菌,其特征在于,真菌为从红豆杉植物体内分离得到的内生菌,菌丝在PDA培养基上室温条件下能够正常生长,在PDA培养基上生长的菌丝白色,菌落呈圆形,有基生菌丝和气生菌丝,气生菌丝浓密,呈棉绒状。
