Abstract

本发明提供了用于构建血浆DNA测序文库的方法和试剂盒。本发明的方法包括：抽提血浆DNA；将血浆DNA与测序接头连接，并对连接产物进行纯化；对纯化的连接产物进行PCR扩增，并对PCR扩增产物进行纯化，以获得所述血浆DNA测序文库，其中，该方法不包括对血浆DNA进行5’末端磷酸化的步骤。本发明的试剂盒包含：-将血浆DNA与测序接头连接的试剂，包括测序接头、连接酶、和连接缓冲液；以及对连接产物进行纯化的试剂和器械；-对纯化的连接产物进行PCR扩增的试剂，以及对PCR扩增产物进行纯化的试剂和器械；其中，所述试剂盒不包含用于对血浆DNA进行5’末端磷酸化的试剂。本发明简化了血浆DNA测序文库的构建流程，简化了实验步骤，使血浆样品文库构建更加低成本、高效、快速，便于大规模使用。
