一、技术领域
本发明涉及饮料的制备,更具体地是以菊花秸秆为原料利用膜分离技术制备清凉饮料的方法。
二、背景技术
菊花是多年生菊科草本植物,其头状花序,是常用中药,也是卫生部规定的药食兼用植物,具有疏风、清热、明目、解毒之功效。主要治疗头痛、眩晕、目赤、心胸烦热、疔疮、肿毒等症。现代药理研究表明,菊花具有治疗冠心病、降低血压、预防高血脂、抗菌、抗病毒、抗炎、抗衰老等多种药理活性。国内外对其分类、鉴定、化学、临床、药理等方面均有报道。研究发现,菊花的化学成分比较复杂,其中黄酮类化合物、三萜类化合物和挥发油是其主要有效成分。
菊花秸秆中含有绿原酸,绿原酸(chlomgenic acid)是一种缩酚酸,属酚类化合物,是植物在有氧呼吸过程中,经莽草酸途径形成的一种苯丙素类化合物。绿原酸是一种重要的生理活性物质,具有抗菌、抗病毒、升高白细胞、保肝利胆、抗肿瘤、降血压、降血脂、清除自由基等作用。其广泛存在于许多植物中,如葵花籽(仁、粕)、水果、蔬菜、大豆、小麦、可可豆、咖啡豆中,尤其葵花籽、可可豆、咖啡豆、沙棘果及传统中草药,如金银花、杜仲中其含量较高。它是许多药材和中成药的主要有效成分之一,绿原酸含量多少又是某些成药的质量指标,也是许多果汁饮料保健成分之一。
每年菊花收获季节都有大量秸秆废弃田间,如能加以利用,可以变废为宝,减少秸秆焚烧对环境的污染。
三、发明内容
本发明的目的是提供一种用菊花秸秆制备清凉饮料及其制备方法,该饮料具有消暑,消炎等功效。
本发明技术方案如下:
一种用菊花秸秆制备清凉饮料的方法,该清凉饮料按重量份由如下物质组成:菊花秸秆提取液100份,蔗糖2份,蜂蜜0.3份,蛋白糖0.04份,薄荷0.00016份;其制备方法按如下步骤进行:
(1)提取将菊花秸秆进行粉碎,过20-80目筛;粉碎后的原料按料液比1:30(m:v)加入纯水,于常温下浸泡0.5小时,于90℃下搅拌提取0.75小时,重复提取三次,静置分离得到水提液,经真空抽滤机过滤得到提取液;
(2)调配按上述重量份比向菊花秸秆提取液中加入蔗糖、蜂蜜、蛋白糖以及薄荷;常温下搅拌摇匀后得调配液;
(3)微滤将上述调配液用孔径为50nm的陶瓷膜进行微滤得到微滤液,所述微滤操作温度为22-30℃、压力为0.5-1.2Mpa;
(4)灌装杀菌将微滤液灌装后100℃沸水高温灭菌;然后常温保存。
与已有技术相比,本发明有益效果体现在:
1、本发明以菊花秸秆为原料,对菊花秸秆充分加以利用,不仅能变废为宝,而且可以避免因菊花秸秆焚烧引发的环境污染。
2、由于菊花秸秆在水浸提过程中长时间受热,其组织结构已部分解体,所以饮料提取液中含有大量大分子物质及胶体物质和少量固体微粒,如多糖(果胶、纤维素等)、鞣质或其复合物、蛋白质等。本发明利用陶瓷膜对调配液进行过滤,除去菊花秸秆提取液中的颗粒杂质、蛋白质、果胶、纤维素等大分子物质,而使活性小分子物质通过,从而达到纯化提取液和除杂的目的,实验结果表明选用孔径为50nm的陶瓷膜,对提取饮料的除杂和纯化效果最好,且其中绿原酸的含量得到很大限度的保留,陶瓷膜过滤后,饮料不仅减轻了涩味,而且口感更清甜、柔和。
3、本发明饮料颜色淡黄,清透明亮,有一股菊花香味,口感纯正,酸甜适中,有清凉感,而且具有消暑,消炎等功效。

具体实施方式
以下通过具体实施方式对本发明技术方案作进一步说明。
实施例中的蛋白糖、蔗糖、蜂蜜、薄荷均为市售常规产品,其中蛋白糖购自安徽天利泰食品科技发展公司,其型号TLT-60A;蜂蜜为荆花和洋槐的混合蜜,购自南京九蜂堂产品有限公司;菊花秸秆采自安徽省黄山市,名徽菊,又名贡菊;菊花秸秆经自然晒干,含水量在5-7%左右。真空抽滤机的生产厂家及型号;郑州长城科工贸有限公司,SHB-Ⅲ;陶瓷膜的生产厂家及型号;合肥世杰膜工程有限公司生产的规格为SJM-30-19陶瓷膜。绿原酸含量的测定采用waters公司高效液相色谱仪,waters2489紫外检测器;安捷伦C18柱:5um4.6*250mm。
1、纯水的制备
自来水用反渗透膜技术进行纯化,操作温度为25-45℃,压力为10-15MPa。
2、粉碎
菊花秸秆经两道工序粉碎。先用机械粉碎机粉碎三次,再用万能粉碎机粉碎两次;过20-80目筛。
3、提取
将粉碎后的菊花秸秆加30倍重量的纯水,于常温下浸泡0.5小时,于90℃下搅拌提取0.75小时,重复提取三次,分离得到水提取液用真空抽滤机过滤得到提取液,合并提取液。
4、调配
主料:菊花秸秆提取液100份。
辅料:蔗糖2份,蜂蜜0.3份,蛋白糖0.04份,薄荷0.00016份。常温下搅拌摇匀后得调配液;
5、微滤
将上述调配液用孔径为50nm的陶瓷膜进行微滤得到微滤液,实验结果表明果胶,纤维素,多糖,鞣质等大分子物质被去除,而绿原酸物质仅少量被截留。陶瓷膜是复合膜,规格为SJM-30-19;操作温度22-30℃、压力0.5-1.2Mpa;渗透测流量平均在700ml/min。过陶瓷膜前后,饮料中的固形物含量基本没有变化,为2.5%,饮料的pH为6~7,属于弱酸近中性饮料。
6、灌装、杀菌
灌装:饮料灌瓶,液体灌至离瓶口6-8mm处;
杀菌:高温杀菌法,轻轻加盖,高温灭菌锅中,100℃沸水中杀菌15min。
倒置再杀菌:旋紧瓶盖,倒置于100℃沸水中杀菌3min。
7、绿原酸含量的测定
采用高效液相色谱仪,流动相确定为甲醇:水:冰醋酸=24:75:1,流速:1mL/min;柱温:25℃;检测波长325nm,进样量为10μL。对所得水提液和微滤液中的绿原酸含量进行检测,并将所得谱图与绿原酸标准品得出的谱图相对照,测得饮料成品中含有绿原酸成分。且调配前菊花秸秆水提液中的绿原酸含量为1.7082mg/100ml,饮料成品中绿原酸的含量为1.5469mg/100ml。经过陶瓷膜除杂的调配液中,绿原酸的保留率达80.66%。
该饮料料颜色淡黄,清透明亮,有一股菊花香味,口感纯正,酸甜适中,有清凉感,而且具有消暑,消炎等功效。

(1)提取将菊花秸秆进行粉碎,过20-80目筛;粉碎后的原料按料液比1:30(m:v)加入纯水,于常温下浸泡0.5小时,于90℃下搅拌提取0.75小时,重复提取三次,静置分离得到水提取液经真空抽滤机过滤得到提取液;
(2)调配按上述重量份比向菊花秸秆提取液中加入蔗糖、蜂蜜、蛋白糖以及薄荷;常温下搅拌摇匀后得调配液;
(3)微滤将上述调配液用孔径为50nm的陶瓷膜进行微滤得到微滤液,所述微滤操作温度为22-30℃、压力为0.5-1.2Mpa;
(4)灌装杀菌将微滤液灌装后100℃沸水高温灭菌;然后常温保存。
