Abstract

本发明涉及一种格氏乳酸杆菌的液体深层发酵方法,包括(1)摇瓶培养格氏乳酸杆菌种子;(2)发酵罐中装入发酵培养基后实罐灭菌,当发酵罐罐温降至38~40℃时,以体积比8~12%的接种量接种,在以下条件发酵:发酵温度35-37℃;发酵全过程pH6.2~6.7,以浓氨水自动调整;转速80-120转/分钟;氮气通气量0.01~0.03vvm;罐压0~0.03MPa;补料液为葡萄糖溶液,葡萄糖含量为100~200g/l,恒速补料,补料总量为初始培养基体积的0.2~0.3倍;发酵总时间26~28小时。最终获得格氏乳酸杆菌的活菌体数量为5~8×1010cfu/ml。

一种格氏乳酸杆菌的液体深层发酵方法
技术领域
本发明涉及一种饲料和食品添加剂技术,具体是一种格氏乳酸杆菌的液体深层发酵方法。
技术背景
乳酸杆菌在自然界中分布广泛,格氏乳酸杆菌(Lactobacillus gasseri)作为乳酸杆菌的一种,是普遍存在于人和动物肠道中的厌氧型益生菌。已有研究表明格氏乳酸杆菌可定植于动物肠道内,正常生长代谢时产生乳酸和乙酸等,可造成肠道低pH环境,促进新陈代谢,抑制有害菌在动物肠道的生长和繁殖。因此,格氏乳酸杆菌在食品和饲料添加剂上具有潜在的应用前景。
现有生产采用格氏乳酸杆菌的技术主要是厌氧发酵和采用常规的培养基和培养条件进行摇瓶培养得到,厌氧发酵虽然工艺简单,能耗小,生产周期短,但是往往最终获得的菌体数量不高,一般其活菌体数量为5~8×1010cfu/ml,而导致生产效率降低。格氏乳酸杆菌的摇瓶培养工艺均采用常规的培养基和培养条件,其菌体数量也不高,一般其活菌体数量为5~8×1010cfu/ml,导致生产效率降低。
发明内容
本发明的目的在于针对现有技术存在的缺陷,提供一种格氏乳酸杆菌的液体深层发酵方法。
本发明的格氏乳酸杆菌的液体深层发酵方法包括如下步骤:
(1)摇瓶培养格氏乳酸杆菌种子,种子培养基和培养条件按常规;
(2)发酵罐中装入发酵培养基后实罐灭菌,当发酵罐罐温降至38~40℃时,以体积比8~12%的接种量接种,在以下条件发酵:发酵温度35-37℃;发酵全过程pH6.2~6.7,以浓氨水自动调整;转速80-120转/分钟;氮气通气量0.01~0.03vvm;罐压0~0.03MPa;补料液为葡萄糖溶液,葡萄糖含量为100~200g/l,恒速补料,补料总量为初始培养基体积的0.2~0.3倍;发酵总时间26~28小时。
上述液体深层发酵工艺最终获得的活菌体数量为5~8×1010cfu/ml。
作为优选,所述格氏乳酸杆菌来源于猪肠道。发酵培养基配方为:葡萄糖20~50g/l,酵母浸粉5~10g/l,大豆蛋白胨5~10g/l,硫酸铵10~15g/l,乙酸钠3~7g/l,柠檬酸钠3~7g/l,磷酸氢二钾2~8g/l,七水硫酸镁0.3~0.7g/l,一水硫酸锰0.05~0.2g/l。,补料时间控制在4~6小时。
本发明与现有技术相比,具有如下优点:
1、得到的格氏乳酸杆活菌体数量高,达到5~8×1010cfu/ml;
2、工艺简单,能耗小,生产周期短;
3、培养基由常用原料组成,培养基成本低廉;
具体实施方法
实施例1
乳酸杆菌的分离与鉴定
将健康育肥猪宰杀后立即取其小肠中部粘膜,无菌条件下冲洗除去粘膜上的食糜,再刮取粘膜黏附物3次,3次的刮取物均混入20ml无菌生理盐水中,充分摇匀。取混合后的生理盐水0.2ml均匀涂布于MRS平板培养基上,置于二氧化塘培养箱中,37℃培养24-48小时,挑取形态不同的菌落在MRS培养基上进行纯培养,然后置于4℃保藏备用。
挑选纯培养物进行染色,镜检;选取无芽孢、革兰氏阳性杆菌进行接触酶试验,再取接触酶试验为阴性的培养物进行糖醇发酵试验,初步确定菌种种属后送中国工业微生物菌种保藏中心进行检验和鉴定,鉴定结果为格氏乳酸杆菌。
实施例2
摇瓶培养种子,种子培养基和培养条件按常规。
50L发酵罐中,装入30L发酵培养基后实罐灭菌。
当发酵罐罐温降至38℃时,以体积比12%的接种量接种,在以下条件发酵:发酵温度35℃;发酵全过程pH6.2~6.7,以浓氨水自动调整;转速120转/分钟;氮气通气量0.01vvm;罐压0.03MPa;补料液为葡萄糖溶液,葡萄糖含量为100g/l,恒速补料,补料总量为初始培养基体积的0.3倍;发酵总时间26小时,补料时间控制在6小时。
发酵培养基配方为:葡萄糖20g/l,酵母浸粉10g/l,大豆蛋白胨5g/l,硫酸铵15g/l,乙酸钠3g/l,柠檬酸钠7g/l,磷酸氢二钾2g/l,七水硫酸镁0.7g/l,一水硫酸锰0.05g/l。
发酵结束时取发酵液检测活菌数为8×1010cfu/ml。
实施例3
种子罐扩大培养种子,种子培养基和培养条件按常规。
15立方发酵罐中,装入8立方发酵培养基后实罐灭菌。
当发酵罐罐温降至40℃时,以体积比8%的接种量接种,在以下条件发酵:发酵温度37℃;发酵全过程pH6.2~6.7,以浓氨水自动调整;转速120转/分钟;氮气通气量0.03vvm;补料液为葡萄糖溶液,葡萄糖含量为200g/l,恒速补料,补料总量为初始培养基体积的0.2倍;发酵总时间28小时,补料时间控制在4小时。
发酵培养基配方为:葡萄糖50g/l,酵母浸粉5g/l,大豆蛋白胨10g/l,硫酸铵10g/l,乙酸钠7g/l,柠檬酸钠3g/l,磷酸氢二钾8g/l,七水硫酸镁0.3g/l,一水硫酸锰0.2g/l。
发酵结束时取发酵液检测活菌数为5×1010cfu/ml。
实施例4
发酵结束后,取发酵液进行离心浓缩或者膜过滤浓缩,得到活菌浓缩液;再经过吸附干燥或冷冻干燥等低温干燥工艺可制得格氏乳酸杆菌的活菌制剂。该活菌制剂通过拌料或饮水进入动物肠道后可发挥相应的益生作用,无毒副作用。

1.一种格氏乳酸杆菌的液体深层发酵方法,其特征在于包括如下步骤:
(1)摇瓶培养格氏乳酸杆菌种子;
(2)发酵罐中装入发酵培养基后实罐灭菌,当发酵罐罐温降至38~40℃时,以体积比8~12%的接种量接种步骤(1)获得的培养物,在以下条件发酵:发酵温度35-37℃;发酵全过程pH6.2~6.7,以浓氨水自动调整;转速80-120转/分钟;氮气通气量0.01~0.03vvm;罐压0~0.03MPa;补料液为葡萄糖溶液,葡萄糖含量为100~200g/l,恒速补料,补料总量为初始培养基体积的0.2~0.3倍;发酵总时间26~28小时,补料时间控制在4~6小时;
