现有技术
脂肪组织是重要的内分泌器官,分泌多种代谢作用活跃的被称作脂肪细胞因子的重要蛋白。一些已知的脂肪细胞因子为瘦蛋白、肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6、降脂蛋白和脂连蛋白。尽管脂肪组织中其它类型的细胞能够分泌某些脂肪细胞因子,但是大多数脂肪细胞因子是由脂肪细胞本身分泌的。

脂连蛋白分子在脂肪细胞内经历翻译后修饰,聚集为多聚体的形式,包括三聚体、六聚体和具有高分子量的低聚体。在血流中不会经常检测到单体的脂连蛋白。
低血浆脂连蛋白水平与有害的代谢疾病状态相关,该疾病状态例如糖尿病、代谢综合征、血脂异常、脂肪代谢障碍和动脉硬化心血管疾病。已经发现,在这些病症中,血浆和脂肪组织中脂连蛋白mRNA表达也降低;这已经被认为可能是由于改变的脂肪细胞功能引起的。

术语“直链C1-C6烷基”是指由碳原子和氢原子组成的直链烃基,其不含有不饱和性,具有1至6个碳原子并且通过单键连接在分子的其余部分上。烷基的实例包括甲基、乙基、丙基、丁基、戊基和己基。
术语“芳基”是指芳香烃基,例如苯基、萘基或蒽基,优选为苯基。
术语“C1-C6烷氧基”是指通式-ORa的基团,其中Ra为如上所定义的烷基,例如甲氧基、乙氧基、丙氧基等。
除非另有说明,本发明中所用的化合物旨在包括仅在一种或多种同位素富集原子的存在上有差异的化合物。例如,除了用氘或氚取代氢,或者用13C-或14C-富集的碳或15N-富集的氮取代碳,具有本发明结构的化合物在本发明的范围内。

在具体的实施方案中,当在通式(I)化合物中X基团为吲唑时,根据如下方法合成。将溶解在THF(四氢呋喃)中的1.5当量的CDI(N,N’-羰基二咪唑)加入通式(III)的相应的3-吲唑酸的无水THF溶液中。将所得到的混合物搅拌4至5小时。这段时间过后,加入溶解在THF中的通式(V)的噻唑的相应衍生物并将反应在室温下搅拌10至12小时。一旦反应时间结束,用二氯甲烷稀释溶剂并用水洗涤数次。用无水Na2SO4干燥有机相,将溶剂蒸发,得到粗产物。必要时通过色谱柱、洗涤或重结晶的方法纯化该粗产物。

通式(I)化合物、其药物可接受的盐、前药和/或溶剂合物以及含有它们的药物组合物能够与可用于预防和/或治疗表现出改变的脂连蛋白水平或其中需要改变脂连蛋白水平的疾病的其它另外的药物一起使用。所述另外的药物能够形成相同药物组合物的一部分,或者能够以用于与包含通式(I)化合物、其药物可接受的盐、前药或溶剂合物的药物组合物同时或不同时给药的独立组合物的形式提供。
在本发明的范围内,“表现出改变的脂连蛋白水平或其中需要改变脂连蛋白水平的疾病”的表达涉及任何疾病、病症或疾病状态,其中能够在血浆中测定的脂连蛋白水平处于某个值之外,所述值被认为是正常的并且以在不具有这些疾病的人群中的血浆脂连蛋白浓度的平均值来获得,或者其中脂连蛋白水平的改变引起该疾病、病症或疾病状态的改善。所述疾病优选选自糖尿病、肥胖症、代谢综合征、动脉硬化心血管疾病、血脂异常和脂肪代谢障碍。
本发明的另一方面是用于治疗和/或预防表现出改变的脂连蛋白水平或其中需要改变脂连蛋白水平的疾病或病症的方法,该方法包括对需要这样治疗的患者给予治疗有效量的至少一种如上定义的通式(I)化合物或其药物组合物。
所述疾病或病症优选选自但不限于糖尿病、肥胖症、代谢综合征、动脉硬化心血管疾病、血脂异常和脂肪代谢障碍。
本说明书上下文中的术语“治疗(treatment)”或“治疗(treat)”是指本发明的化合物与制剂的给药,用于预防、缓解或消除疾病或与所述疾病有关的一种或多种症状。“治疗(treatment)”也包括预防、缓解或消除所述疾病的生理后遗症。
本发明上下文中的术语“缓解”应当理解为是指所治疗患者的情况的任何改善-无论是主观上(患者的感受或对患者的感觉)还是客观上(测定的参数)。
通过以下实施例进一步解释本发明。由于在权利要求中定义了本发明的范围,因此该解释绝不能被理解为对本发明范围的限制。
实施例
实施例1.1H-吲唑-3-羧酸的噻唑-2-基酰胺化合物(化合物1)的合成
将溶解在THF中的N,N’-羰基二咪唑(4.5mol,729mg)加入3-吲唑羧酸(3mmol,486.5mg)的无水THF溶液中。将所得到的混合物搅拌4至5小时。这段时间过后,加入溶解在THF中的2-氨基-噻唑(3mmol,300.5mg)并将反应在室温下搅拌10至12小时。一旦反应时间结束,用二氯甲烷稀释溶剂并用水洗涤数次。用无水Na2SO4干燥有机相,将溶剂蒸发,得到粗产物。采用1/2乙酸乙酯/己烷作为洗脱液,通过硅胶柱色谱纯化该粗产物。
得到期望产物,其为白色固体(纯度98%-100%)。重量=180mg。
收率(Y.):24%

实施例2:1H-吲唑-3-羧酸(5-甲基-噻唑-2-基)-酰胺(化合物5)的合成
将溶解在THF中的N,N’-羰基二咪唑(4.5mmol,729mg)加入3-吲唑羧酸(3mmol,486.5mg)的无水THF溶液中。将所得到的混合物搅拌4至5小时。这段时间过后,加入溶解在THF中的2-氨基-5-甲基-噻唑(3mmol,342.5mg)并将反应在室温下搅拌10至12小时。一旦反应时间结束,用二氯甲烷稀释溶剂并用水洗涤数次。用无水Na2SO4干燥有机相,蒸发溶剂,得到粗产物。当将粗产物重新溶解在CH2Cl2/MeOH的混合物中时,出现完全不溶的白色沉淀。将其过滤并用MeOH洗涤数次后,得到具有粉末质地的期望产物,其为白色固体(223.5mg,Y.29%)。

实施例3.吡啶-2-羧酸(5-甲基-噻唑-2-基)-酰胺(化合物4)的合成
将溶解在THF中的N,N ’-羰基二咪唑(4.5mmol,729mg)加入吡啶甲酸(3mmol,369mg)的无水THF溶液中。将所得到的混合物搅拌4至5小时。这段时间过后,加入溶解在THF中的2-氨基-5-甲基-噻唑(3mmol,342.5mg)并将反应在室温下搅拌10至12小时。一旦反应时间结束,用二氯甲烷稀释溶剂并用水洗涤数次。用无水Na2SO4干燥有机相,将溶剂蒸发,得到粗产物。采用1/1乙酸乙酯/己烷作为洗脱液,通过硅胶柱色谱纯化该粗产物。得到期望产物,其为白色固体(纯度99%)。重量=490mg。Y.:74%。

将溶解在THF中的N,N’-羰基二咪唑(4.5mmol,729mg)加入吡啶甲酸(3mmol,369mg)的无水THF溶液中。将所得到的混合物搅拌4至5小时。这段时间过后,加入溶解在THF中的(2-氨基-噻唑-4-基)-乙酸甲酯(3mmol,516.5mg)并将反应在室温下搅拌10至12小时。一旦反应时间结束,用二氯甲烷稀释溶剂并用水洗涤数次。用无水Na2SO4干燥有机相,将溶剂蒸发,得到粗产物。采用2/1乙酸乙酯/己烷作为洗脱液,通过硅胶柱色谱纯化该粗产物。得到期望产物,其为白色固体(纯度100%)。重量=354.5mg。Y.:43%

实施例6.生物测定
已经测试了如下化合物:

已经采用下述方案来分析化合物1-5对肥胖症、2型糖尿病和动脉硬化(ODA代谢疾病)的潜在治疗活性:
-制备细胞系,该细胞系已经稳定地整合了ODA疾病的发展中的主基因的人启动子;
-扩增源自商购的人基因组DNA的靶基因的人启动子序列;
-将启动子克隆入合适的载体中,该载体采用荧光素酶基因作为报道基因,使得能够获知启动子序列的转录活性;
-制备针对克隆入报道载体中的每个启动子的稳定的细胞系;
-采用本发明的化合物在稳定的细胞系上进行筛选试验;
-适当地验证采用某种化合物所得到的效果,该化合物能够改善启动子的转录活性。
所选用的细胞系是C2C12,对应于源自小鼠肌肉组织的成肌细胞。
因此,在第一天,在所述细胞的培养基中,将细胞接种在P6(6孔)中。在这种情况下,将每孔160,000个C2C12细胞接种在完全DMEM+10%胎牛血清(FBS)中。
在第二天,采用2.5μg的含有所关注的构建体(启动子+报道基因(荧光素酶))和0.5μg的含有潮霉素抗性基因的pH1c质粒共转染细胞。所用的转染为:
TRANSFAST(Promega):1∶3比率,μg DNA:μl转染试剂(Transfast)。
1.将DMEM培养基(每孔600μl)与上文确定的DNA混合。将反应在室温下孵育5分钟。
2.将TransFast试剂加入混合物中,并在室温下进行15分钟的孵育。
3.这段时间过后,将培养基从孔中除去并将转染容量逐滴加入细胞中。
4.在37℃下孵育混合物1小时;然后将转染混合物除去并加入完全培养基。
采用GFP(“绿色荧光蛋白”)报道基因和β-半乳糖苷酶,在与所关注的构建体相同的条件下,将双阳性对照一直用于转染过程。如下所述,这两个报道基因还使得能够估算转染过程的效率。
在第三天,观测转染效率:
a)采用荧光显微镜,通过GFP基因表达来观测转染效率:荧光细胞(在观察它们时采用相应的过滤器)已经正确地整合了报道基因,即它们已经被正确地转染。
b)采用一旦细胞被染色就能够在显微镜下观察到该细胞的特异性染色试剂盒,通过β-半乳糖苷酶基因表达来观测转染效率。已经整合了报道基因的细胞能够利用反应底物产生蓝色物质作为所述化学反应的产物。
两种方法都能够估算已经被成功转染的细胞的百分率(根据细胞是否分别已经整合了GFP或β-半乳糖苷酶,这些细胞为绿的或蓝的)。推而广之,假如采用相同的条件,这种成功转染百分率能够适用于我们所关注的构建体的情况。
考虑40-60%的高转染百分率,并且只有达到所述的百分率,才在细胞上进行1/100和1/150的传代(“稀释”),将它们接种到新的P6板上。每孔加入的潮霉素的量为700μg/ml(1.33mM)。
之前就已经采用接种在P6中的细胞制作了不同药物浓度下的潮霉素抗性曲线,从而选择待用于筛选的合适的药物量。
获得克隆.
在随后的几天中,一直采用潮霉素来维持细胞,并在显微镜下观察细胞来观看可能的克隆的演变。当在显微镜下观察时克隆占据了十倍的放大镜头,这时可以认为该克隆足够大了,足以从板上分离出来并建立源自它的稳定的细胞系。为了避免克隆在建立稳定的细胞系时可能的混合,选择在孔中彼此不十分接近的克隆是很重要的。用来分离每个克隆的方法如下所述:
1.除去培养基。
2.用PBS1×洗涤细胞两次。
3.将克隆环放置(采用凡士林)在每个克隆上,并将它们用夹具按压从而通过凡士林将它们粘住。
4.在每个环中加入20-30μl的胰岛素并且将反应在37℃下孵育5分钟。一旦已经证实所有的克隆细胞已经被适当地胰蛋白酶化时,加入100μl的培养基以使所述反应停止。
5.小心地上下移液以便不移动所述环并加入必要的体积使P24中的培养基的体积达到1ml;该加入体积会与潮霉素一起作为培养基来继续稳定的细胞筛选。
从P24至P6和从P6至T25连续逐步升级。当在T25中克隆的细胞达到汇合时,将小瓶冷冻并通过荧光素酶报道基因来测量它们已经稳定地整合的启动子的转录活性。在研究中呈阳性的克隆会继续保持在潮霉素中,定期测量荧光素酶以便控制克隆不失去所述活性。
根据如下方法测量荧光素酶活性:
a)按50000和1200000细胞/孔(并且一式两份)接种P24板。
b)大约24小时以后,将培养基从细胞中除去并用PBS 1×洗涤每个孔(由于DMEM干扰报道基因的活性测量)。
c)向每个孔中加入100μl的荧光素酶显影剂(luciferase developingreagent,Bright-GloTM荧光素酶检测系统-Promega)。该试剂携带报道基因底物并发光作为酶促反应的产物。当存在所述底物时,已经获得报道基因并由此获得靶启动子的细胞会发光。为了记录荧光素酶活性的测量结果,加入显影剂后立即将该板摇动30秒并且2分钟之后,收集体积并将所述体积转移到96孔板中(P96),其能够在照度计中读出。
d)选择合适的发光程序,使得每孔的读数持续5秒,对于每个得到的克隆,或者换句话说,对于每个稳定的细胞系,记录随机选择的发光单元。
反应底物是光敏的,因此从加入试剂的时刻起直到在每个实验中记录或读出荧光素酶活性,将板保持在黑暗中是非常重要的。
进行筛选试验:
在筛选中所用的克隆是:C2C12 Adipo(脂连蛋白启动子)、C2C12UCP3(UCP3启动子)、C2C12 AdR1(脂连蛋白受体1启动子)和C2C12Nmu(神经调节肽启动子)。
在任何筛选实验之前,首先必须提供适当数量的用于进行这样的实验的细胞。为此,经常接种两个T75(每个瓶中约1×106个细胞),这样四天后,细胞已经达到汇合,并容易进行下述筛选方法。
1.第一天:在白色P96板中接种,每孔30000个细胞;这些板可以放大发光信号并改进读数。将细胞培养于100μl不合潮霉素的培养基中。
2.第二天:接种24小时后,用化合物1-5刺激细胞。
通过下图所示的筛选方法将化合物1-5在1、5和10μg/ml的浓度下进行一式两份的测定:
在所标示的位置中,在靶启动子的已知商购的激活剂存在下,记录待研究的启动子的转录活性。这些孔具有双重功能:a)它们可用作转录活化作用和筛选方法的阳性对照;b)当筛选那些提高待研究的启动子的活性的阳性化合物时,它们可用作参照,因为它们提供如下计算的活化比:通过商购的激活剂存在下增加的平均发光信号除以媒介物存在下靶启动子的基线活性提供的平均发光信号。之所以最初选择罗格列酮(RGZ)作为商购的激活剂,是由于它在文献中被描述成脂连蛋白基因转录激活剂。制作剂量-反应曲线来确定达到最大反式激活时的浓度,所述浓度为20μM。然后用诸如染料木黄酮和黄苷元的其它商购的化合物代替RGZ。这两种化合物在文献中被描述为可能的PPARγ激活剂。制作剂量-反应曲线,该曲线与RGZ的剂量-反应曲线相同,达到靶启动子的最大反式激活时的浓度为20μM。
●在所标示的位置中,在每孔中存在1、5和10μg/ml的本发明的化合物1-5的情况下,一式两份地记录待研究的启动子的转录活性。
3.第四天:用化合物1-5刺激48小时后,按照之前所述的方法显影由靶启动子引导的荧光素酶活性,引入如下的改进:
-一旦已经从细胞中除去培养基并且已经用PBS 1×洗涤细胞,将40μl的荧光素酶显影剂加入到每一孔中。
-两分钟以后,在相同的P96板中进行读数。
结果分析
考虑到以上所示的用于筛选实验的示意图,获得一系列的发光值RLU(“相对荧光素酶单元”),其用来计算之前提到的由在每个实验中被测定的靶启动子负责的转录活化比。用在每个已经被测定的化合物1-4存在下的发光信号的增加对基线条件下(在媒介物存在下)获得的发光信号平均值作图来绘制所述比率。其中用商购的激活剂处理细胞时获得活化比≥2的任何测定,均被认为是有效的。对于所有测定的化合物,获得的转录活化比均大于1.2,因此能够认为它们是阳性化合物。
根据第一次测定所得到的结果,在0.5μg/ml至25μg/ml之间的三个新浓度下重复测定。
用4种所提到的化合物处理细胞时获得的活化比,以及商购的化合物的值详细列举如下:

Claims (15)
2.根据权利要求1所述的用途,其中表现出改变的脂连蛋白水平或其中需要改变脂连蛋白水平的所述疾病选自糖尿病、肥胖症、代谢综合征、动脉硬化心血管疾病、血脂异常和脂肪代谢障碍。
3.根据权利要求2所述的用途,其中所述疾病选自血脂异常和脂肪代谢障碍。
4.根据权利要求1至3所述的用途,其中R1和R2独立地选自H、直链C1-C6烷基和(CH2)nCO2R8,其中n为1、2或3,且R8选自H和直链C1-C6烷基。
5.根据权利要求1至4中任一权利要求所述的用途,其中基团R1和R2中的至少一个为H。
6.根据权利要求1至5中任一权利要求所述的用途,其中R1和R2为H。
7.根据权利要求1至4中任一权利要求所述的用途,其中n为1。
8.根据权利要求1至4中任一权利要求所述的用途,其中R1为CH2CO2R8,其中R8为H或甲基。
9.根据权利要求1至8中任一权利要求所述的用途其中R2为烷基,优选为甲基。
10.根据权利要求1至9中任一权利要求所述的用途,其中R3、R4、R5、R6和R7为氢。
13.药物组合物,其包含权利要求12中定义的通式(I)化合物或其药物可接受的盐、前药或溶剂合物,及至少一种药物可接受的载体、佐剂和/或媒介物。
14.化妆品组合物,其包含权利要求12中定义的通式(I)化合物或其化妆品可接受的盐、前药或溶剂合物,及至少一种化妆品可接受的载体、佐剂和/或媒介物。
15.如权利要求12中定义的通式(I)化合物,其用作医药产品。
