Claims (14)
- 本发明要申明的权利是:1.小RNA美白产品中包括下述的所有或部分组成成份:·一个或多个抑制黑色素形成的寡聚核苷酸成份;·一个或多个洛氨酸酶的抑制成份;·一个药效上能接受的载体;·一种或2-3种还原剂;·一种或2-3种促进老批脱落的弱酸;·一种或2-3种抗氧化和防紫外线物质;·一种或2-3种有益于皮肤的中草药提取液;·其它的活性成份,辅助成份或添加剂。
- 2.在权利要求(1)中的抑制黑色素形成的寡聚核苷酸成份,它是一个19-25个核苷酸长的双链寡聚核苷酸(dsRNA,dsDNA,或RNA/DNA)或是一个10-30个核苷酸长的单链寡聚核苷酸,如RNA,DNA,或RNA/DNA的杂合体,并能有效地抑制人体内的与黑色素形成有关的基因的表达。
- 3.在权利要求(1或2)中,寡聚核苷酸成份是指任何一段与黑色素形成有关的基因的同源序列和/或互补序列,这些单链寡聚核苷酸或相互补的双链寡聚核苷酸的靶基因包括但不局限于:Gp75/Tyrp1,Tyrp2,Tyr,P-protein,AIM1,MC1R,PAR2,Myo5a,Rab27a,OA1 protein,MITF,gp100/pMel17/Silver,Kinesin,Agouti signal protein,Slc24a5和POMC。
- 4.在权利要求(1,2或3)中的寡聚核苷酸成份,它是一系列的能有效地灭活相应的mRNA分子的10-30个核苷酸长的寡聚核苷酸,包括但不局限于下列序号1到228的反义链或双链寡聚核苷酸分子。
- 5.在权利要求(1)的配方中,寡聚核苷酸成份是一种混合物,由一种或多种不同形式的单链和/或双链寡聚核苷酸分子,以等量或不等量的方式,或以任何其它的配伍形式所组成,特别适用于美白不同类型的肤色。
- 6.在权利要求(1,2或3)中的寡聚核苷酸分子,它是一种自然的单链和/或双链寡聚核苷酸分子,它也可是一种杂合式的含有修饰核苷酸成分的寡聚核苷酸分子。
- 7.在权利要求(1,2或3)中的杂合式寡聚核苷酸分子,它是指核苷酸与脱氧核苷酸形成的杂合式的单链或双链寡聚核苷酸分子,或是指单链或双链寡聚核苷酸分子中的单核苷酸或单脱氧核苷酸是自然的和/或经不同方法修饰过的,如肽核苷酸(PNA)、闭锁核苷酸(LNA)、或甲氧-或乙氧修饰的核苷酸单体或双体。
- 8.在权利要求(2,3,或4)中的双链寡聚核苷酸的伴随链的第17和18位常引入非互补的突变核苷酸或正常的互补核苷酸.
- 9.在权利要求(1,2,3,或4)中的双链寡聚核苷酸可被插入质粒载体或病毒载体,用于皮肤细胞表达而抑制靶基因的活性.
- 10.在权利要求(1)的配方中,寡聚核苷酸成份的组成可为最终基因皮肤美白产品总量的0.0000001%到99%。
- 11.一种皮肤美白产品的配方,它是指配置单纯的或复合的小RNA美白产品中各种成份的组成,所占的比列,和混合的程序,如配方1,2,3和4.
- 12.一种能有效地抑制人的皮肤黑色素细胞中的黑色素的产生的方法,它是指用小RNA美白产品中含有的脂质体,多聚体,多肽,或其它的透皮剂来转递一定量的抑制黑色素形成的寡聚核苷酸成份进入黑色素细胞从而抑制黑色素的形成。
- 13.在权利要求(10)的方法中,序号1到25的多肽的组成可为最终小RNA美白产品总量的0.00001%到10%。
- 14.在权利要求(10)的方法中,单纯的基因皮肤美白产品主要由一定量的抑制黑色素形成的寡聚核苷酸成份和一种药效上可接受的载体或一种稀释剂组成的复合物,而复合的基因皮肤美白产品可由多种成份组成,如●一个或多个抑制黑色素形成的寡聚核苷酸成份;●一个或多个洛氨酸酶的抑制成份;●一个或多个药效上能接受的载体;●一种或多种还原剂;●一种或多种促进老批脱落的弱酸;●一种或多种抗氧化和防紫外线物质;●一种或多种有益于皮肤的中草药提取液;●其它的活性成份,辅助成份或添加剂。
